304. The pennate benthic diatoms of the sand beach of Kruglaya Bay (the Black Sea)

Abstract

For the first time in the Kruglaya Bay of the Black Sea, 25 taxa of Bacillariophyta from 17 genera were found, three of them were new for the Crimea and the Black Sea Halamphora tenerrima, Amphora tenuissima, and Navicula antonii. Marine (40%), coastalbrackish (32%) and boreal-tropical (32%), arctic-boreal-tropical (24%), boreal and cosmopolitan (20% and 16%, respectively) prevailed. The morphological characteristics of the species in the SEM (scanning electronic microscope) and LM (light microscope), as well as the occurrence, ecology, phytogeography, and general distribution in various regions of the World Ocean are given.

Abstract

Abstract A solution for a coupled thermomechanical boundary value problem of the theory of large deformations that simultaneously takes into account the intense deformation of an elastoviscoplastic material and its heating due to such deformation and near-wall friction is constructed. The conditions for the emergence and development of a viscoplastic flow in the material of a cylindrical layer enclosed between rigid surfaces, one of which has the ability to rotate under the action of a changing given load relative to the axis of the viscometer, are calculated. At all stages of the process from its start to stop, the stress-strain state and temperature distributions in the areas of flow and reversible deformation are calculated, including flow stop, unloading and cooling, with the calculation of residual stresses and deformations. The yield strength is assumed to be temperature dependent.

325. MUC13 Cell Surface Mucin Limits Salmonella Typhimurium Infection by Protecting the Mucosal Epithelial Barrier

Abstract

BACKGROUND & AIMS: MUC13 cell surface mucin is highly expressed on the mucosal surface throughout the intestine, yet its role against bacterial infection is unknown. We investigated how MUC13 impacts Salmonella typhimurium (S Tm) infection and elucidated its mechanisms of action. METHODS: Muc13-/- and wild-type littermate mice were gavaged with 2 isogenic strains of S Tm after pre-conditioning with streptomycin. We assessed clinical parameters, cecal histology, local and systemic bacterial load, and proinflammatory cytokines after infection. Cecal enteroids and epithelial cell lines were used to evaluate the mechanism of MUC13 activity after infection. The interaction between bacterial SiiE and MUC13 was assessed by using siiE-deficient Salmonella. RESULTS: S Tm-infected Muc13-/- mice had increased disease activity, histologic damage, and higher local and systemic bacterial loads. Mechanistically, we found that S Tm binds to MUC13 through its giant SiiE adhesin and that MUC13 acts as a pathogen-binding decoy shed from the epithelial cell surface after pathogen engagement, limiting bacterial invasion. In addition, MUC13 reduces epithelial cell death and intestinal barrier breakdown by enhancing nuclear factor kappa B signaling during infection, independent of its decoy function. CONCLUSIONS: We show for the first time that MUC13 plays a critical role in antimicrobial defense against pathogenic S Tm at the intestinal mucosal surface by both acting as a releasable decoy limiting bacterial invasion and reducing pathogen-induced cell death. This further implicates the cell surface mucin family in mucosal defense from bacterial infection.

Abstract

BACKGROUND AND AIMS: Ulcerative colitis [UC] is a major form of inflammatory bowel disease globally. Phenotypic heterogeneity is defined by several variables including age of onset and disease extent. The genetics of disease severity remains poorly understood. To further investigate this, we performed a genome wide association [GWA] study using an extremes of phenotype strategy. METHODS: We conducted GWA analyses in 311 patients with medically refractory UC [MRUC], 287 with non-medically refractory UC [non-MRUC] and 583 controls. Odds ratios [ORs] were calculated for known risk variants comparing MRUC and non-MRUC, and controls. RESULTS: MRUC-control analysis had the greatest yield of genome-wide significant single nucleotide polymorphisms [SNPs] [2018], including lead SNP = rs111838972 [OR = 1.82, p = 6.28 × 10-9] near MMEL1 and a locus in the human leukocyte antigen [HLA] region [lead SNP = rs144717024, OR = 12.23, p = 1.7 × 10-19]. ORs for the lead SNPs were significantly higher in MRUC compared to non-MRUC [p < 9.0 × 10-6]. No SNPs reached significance in the non-MRUC-control analysis (top SNP, rs7680780 [OR 2.70, p = 5.56 × 10-8). We replicate findings for rs4151651 in the Complement Factor B [CFB] gene and demonstrate significant changes in CFB gene expression in active UC. Detailed HLA analyses support the strong associations with MHC II genes, particularly HLA-DQA1, HLA-DQB1 and HLA-DRB1 in MRUC. CONCLUSIONS: Our MRUC subgroup replicates multiple known UC risk variants in contrast to non-MRUC and demonstrates significant differences in effect sizes compared to those published. Non-MRUC cases demonstrate lower ORs similar to those published. Additional risk and prognostic loci may be identified by targeted recruitment of individuals with severe disease.

Abstract

Most eukaryotic genes have been vertically transmitted to the present from distant ancestors. However, variable gene number across species indicates that gene gain and loss also occurs. While new genes typically originate as products of duplications and rearrangements of preexisting genes, putative de novo genes-genes born out of ancestrally nongenic sequence-have been identified. Previous studies of de novo genes in Drosophila have provided evidence that expression in male reproductive tissues is common. However, no studies have focused on female reproductive tissues. Here we begin addressing this gap in the literature by analyzing the transcriptomes of 3 female reproductive tract organs (spermatheca, seminal receptacle, and parovaria) in 3 species-our focal species, Drosophila melanogaster-and 2 closely related species, Drosophila simulans and Drosophila yakuba, with the goal of identifying putative D. melanogaster-specific de novo genes expressed in these tissues. We discovered several candidate genes, located in sequence annotated as intergenic. Consistent with the literature, these genes tend to be short, single exon, and lowly expressed. We also find evidence that some of these genes are expressed in other D. melanogaster tissues and both sexes. The relatively small number of intergenic candidate genes discovered here is similar to that observed in the accessory gland, but substantially fewer than that observed in the testis.

Abstract

To enrich spatio-temporal information on the distribution of alien, cryptogenic, and neonative species in the Mediterranean and the Black Sea, a collective effort by 173 marine scientists was made to provide unpublished records and make them open access to the scientific community. Through this effort, we collected and harmonized a dataset of 12,649 records. It includes 247 taxa, of which 217 are Animalia, 25 Plantae and 5 Chromista, from 23 countries surrounding the Mediterranean and the Black Sea. Chordata was the most abundant taxonomic group, followed by Arthropoda, Mollusca, and Annelida. In terms of species records, Siganus luridus, Siganus rivulatus, Saurida lessepsianus, Pterois miles, Upeneus moluccensis, Charybdis (Archias) longicollis, and Caulerpa cylindracea were the most numerous. The temporal distribution of the records ranges from 1973 to 2022, with 44% of the records in 2020–2021. Lethrinus borbonicus is reported for the first time in the Mediterranean Sea, while Pomatoschistus quagga, Caulerpa cylindracea, Grateloupia turuturu, and Misophria pallida are first records for the Black Sea; Kapraunia schneideri is recorded for the second time in the Mediterranean and for the first time in Israel; Prionospio depauperata and Pseudonereis anomala are reported for the first time from the Sea of Marmara. Many first country records are also included, namely: Amathia verticillata (Montenegro), Ampithoe valida (Italy), Antithamnion amphigeneum (Greece), Clavelina oblonga (Tunisia and Slovenia), Dendostrea cf. folium (Syria), Epinephelus fasciatus (Tunisia), Ganonema farinosum (Montenegro), Macrorhynchia philippina (Tunisia), Marenzelleria neglecta (Romania), Paratapes textilis (Tunisia), and Botrylloides diegensis (Tunisia).

330. Male-derived transcripts isolated from the mated female reproductive tract in Drosophila melanogaster.

Abstract

In species with internal fertilization, sperm, and seminal fluid are transferred from male to female during mating. While both sperm and seminal fluid contain various types of molecules, including RNA, the role of most of these molecules in the coordination of fertilization or in other possible functions is poorly understood. In Drosophila, exosomes from the accessory gland, which produces seminal fluid, are transferred to females, but their potential cargoes have not been described. Moreover, while the RNA composition of sperm has been described in several mammalian species, little work on this problem has occurred in Drosophila. Here we use single nucleotide polymorphism differences between males and females from a set of highly inbred lines of D. melanogaster, and transcriptome data from the female reproductive tract, sperm, testis, and accessory gland, to investigate the potential origin, male vs female, RNA molecules isolated from 3 female reproductive tract organs, the seminal receptacle and spermatheca, which store sperm, and the parovaria, which does not. We find that mated females carry male-derived transcripts from many genes, including those that are markers of the accessory gland and known seminal fluid proteins. Our observations also support the idea that intact sperm transcripts can be isolated from the female sperm storage organs.

331. Evolution of secondary cell number and position in the Drosophila accessory gland.

Abstract

In animals with internal fertilization, males transfer gametes and seminal fluid during copulation, both of which are required for successful reproduction. In Drosophila and other insects, seminal fluid is produced in the paired accessory gland (AG), the ejaculatory duct, and the ejaculatory bulb. The D. melanogaster AG has emerged as an important model system for this component of male reproductive biology. Seminal fluid proteins produced in the Drosophila AG are required for proper storage and use of sperm by the females, and are also critical for establishing and maintaining a suite of short- and long-term postcopulatory female physiological responses that promote reproductive success. The Drosophila AG is composed of two main cell types. The majority of AG cells, which are referred to as main cells, are responsible for production of many seminal fluid proteins. A minority of cells, about 4%, are referred to as secondary cells. These cells, which are restricted to the distal tip of the D. melanogaster AG, may play an especially important role in the maintenance of the long-term female post-mating response. Many studies of Drosophila AG evolution have suggested that the proteins produced in the gland evolve quickly, as does the transcriptome. Here, we investigate the evolution of secondary cell number and position in the AG in a collection of eight species spanning the entire history of the Drosophila genus. We document a heretofore underappreciated rapid evolutionary rate for both number and position of these specialized AG cells, raising several questions about the developmental, functional, and evolutionary significance of this variation.

Abstract

Fossil apes from the eastern Mediterranean are central to the debate on African ape and human (hominine) origins. Current research places them either as hominines, as hominins (humans and our fossil relatives) or as stem hominids, no more closely related to hominines than to pongines (orangutans and their fossil relatives). Here we show, based on our analysis of a newly identified genus, Anadoluvius, from the 8.7 Ma site of Çorakyerler in central Anatolia, that Mediterranean fossil apes are diverse, and are part of the first known radiation of early members of the hominines. The members of this radiation are currently only identified in Europe and Anatolia; generally accepted hominins are only found in Africa from the late Miocene until the Pleistocene. Hominines may have originated in Eurasia during the late Miocene, or they may have dispersed into Eurasia from an unknown African ancestor. The diversity of hominines in Eurasia suggests an in situ origin but does not exclude a dispersal hypothesis.

333. Diuretic treatment before and after transcatheter aortic valve implantation:A Danish nationwide study

Abstract

Objectives We examined loop diuretic treatment before and 1-year after transcatheter aortic valve implantation (TAVI), as a proxy for changes in symptom severity and secondly assessed how changes in loop diuretics related to mortality risk. Background Randomized clinical trials suggest that approximately one third of patients undergoing TAVI do not achieve symptom relief, but "all-comer"data are lacking. Methods Using Danish nationwide registries, we identified all citizens, who underwent TAVI from 2008 to 2019 and were alive at 1-year post-discharge. Loop diuretic treatment pre-TAVI and at 1-year post-TAVI were assessed and grouped as receiving 1) no-loop diuretics; 2) low: 1-40 mg of furosemide (or equivalent bumetanide) daily; 3) intermediate: 41-120 mg of furosemide daily; or 4) high: >120 mg furosemide daily. Results Among the 4431 patients undergoing TAVI, 2173 (49%) patients were not treated with loop diuretics at the time of TAVI, 918 (21%) had low-loop diuretics, 881 (20%) had intermediateloop diuretics, and 459 (10%) had high-loop diuretics. At 1-year post-TAVI, 893 (20%) patients had increased, 1010 (23%) had reduced, and 2528 (57%) had unchanged loop diuretic treatment. The cumulative 5-year risk of death in patients surviving one year, was 61% (95% CI: 56.4% to 65.3%) in patients with increased and 47% (95% CI: 44.9% to 49.9%) in patients with reduced/unchanged loop diuretic treatment, respectively. In multivariable Cox proportional hazard analysis, increased loop diuretic treatment was associated with a higher risk of death compared with reduced/unchanged loop diuretic treatment (Hazard ratio: 1.4; 95% CI: 1.22 to 1.52). Conclusions Among patients undergoing TAVI, surviving one year, one fifth of patients had increased loop diuretic treatment, and a little over one fifth had reduced loop diuretic treatment 1-year post-procedure. Abstract

Most eukaryotic genes have been vertically transmitted to the present from distant ancestors. However, variable gene number across species indicates that gene gain and loss also occurs. While new genes typically originate as products of duplications and rearrangements of pre-existing genes, putativede novogenes - genes born out of previously non-genic sequence - have been identified. Previous studies ofde novogenes inDrosophilahave provided evidence that expression in male reproductive tissues is common. However, no studies have focused on female reproductive tissues. Here we begin addressing this gap in the literature by analyzing the transcriptomes of three female reproductive tract organs (spermatheca, seminal receptacle, and parovaria) in three species - our focal species,D. melanogaster- and two closely related species,D. simulansandD. yakuba, with the goal of identifying putativeD. melanogaster-specificde novogenes expressed in these tissues. We discovered several candidate genes, which, consistent with the literature, tend to be short, simple, and lowly expressed. We also find evidence that some of these genes are expressed in otherD. melanogastertissues and both sexes. The relatively small number of candidate genes discovered here is similar to that observed in the accessory gland, but substantially fewer than that observed in the testis.

Abstract

Studies of allele-specific expression in interspecific hybrids have provided important insights into gene-regulatory divergence and hybrid incompatibilities. Many such investigations in Drosophila have used transcriptome data from whole animals or gonads, however, regulatory divergence may vary widely among species, sex, and tissues. Thus, we lack sufficiently broad sampling of tissues to be confident about the general principles of regulatory divergence. Here we seek to fill some of these gaps in the literature by characterizing regulatory evolution and hybrid misexpression in a somatic male sex organ, the accessory gland, in F1 hybrids betweenDrosophila melanogasterandD. simulans.The accessory gland produces seminal fluid proteins, which play an important role in male and female fertility and may be subject to adaptive divergence due to male-male or male-female interactions. We find thattransdifferences are relatively more abundant thancis, in contrast to most of the interspecific hybrid literature, though large effect-sizetransdifferences are rare. Seminal fluid protein genes have significantly elevated levels of expression divergence and tend to be regulated through bothcisandtransdivergence. We find limited misexpression in this organ compared to other Drosophila studies. As in previous studies, male-biased genes are overrepresented among misexpressed genes and are much more likely to be underexpressed. ATAC-Seq data show that chromatin accessibility is correlated with expression differences among species and hybrid allele-specific expression. This work identifies unique regulatory evolution and hybrid misexpression properties of the accessory gland and suggests the importance of tissue-specific allele-specific expression studies.

344. Selection and geography shape male reproductive tract transcriptomes in Drosophila melanogaster

Abstract

Transcriptome analysis of several animal clades suggests that male reproductive tract gene expression evolves quickly. However, the factors influencing the abundance and distribution of within-species variation, the ultimate source of interspecific divergence, are poorly known. Drosophila melanogaster, an ancestrally African species that has recently spread throughout the world and colonized the Americas in the last roughly 100 years, exhibits phenotypic and genetic latitudinal clines on multiple continents, consistent with a role for spatially varying selection in shaping its biology. Nevertheless, geographic expression variation in the Americas is poorly described, as is its relationship to African expression variation. Here, we investigate these issues through the analysis of two male reproductive tissue transcriptomes [testis and accessory gland (AG)] in samples from Maine (USA), Panama, and Zambia. We find dramatic differences between these tissues in differential expression between Maine and Panama, with the accessory glands exhibiting abundant expression differentiation and the testis exhibiting very little. Latitudinal expression differentiation appears to be influenced by the selection of Panama expression phenotypes. While the testis shows little latitudinal expression differentiation, it exhibits much greater differentiation than the accessory gland in Zambia vs American population comparisons. Expression differentiation for both tissues is non-randomly distributed across the genome on a chromosome arm scale. Interspecific expression divergence between D. melanogaster and D. simulans is discordant with rates of differentiation between D. melanogaster populations. Strongly heterogeneous expression differentiation across tissues and timescales suggests a complex evolutionary process involving major temporal changes in the way selection influences expression evolution in these organs.

347. Adverse Effects Associated with Diazoxide Use in Neonates

Abstract

Introduction: Diazoxide is the first-line treatment used to manage infants and children with hypoglycemia secondary to hyperinsulinemic hypoglycemia (HH), after initial stabilization of blood glucose. Recent changes in Pediatric Endocrine Society guidelines regarding definition of, and screening for hypoglycemia in the neonate led to an increase in the number of infants treated with diazoxide. Previous studies have noted various side effects of diazoxide. With the increased use of this drug in neonates, our aim is to further evaluate these side effects. Methods: We performed a retrospective case control analysis of all neonates admitted to a tertiary care center in New Jersey who were treated with diazoxide for hypoglycemia after a confirmed diagnosis of HH between 1/1/2015 and 9/1/2019. All subjects were younger than 6 months of age at treatment initiation with diazoxide. We collected data regarding general patient characteristics, diuretic dosing, findings on echocardiogram, blood counts, episodes of emesis, and quantity of feeds at time points before and after diazoxide initiation. Results: A total of 25 infants (64% males) met inclusion criteria and were included in the analysis. Based on echocardiogram results, one baby (4%) had pulmonary hypertension (PHTN) prior to initiation of diazoxide, and 20% after, with 5 cases (20%) of reopening of the ductus arteriosus (DA). Nine patients (36%) required a diuretic dose increase after diazoxide initiation. There was increased feeding intolerance with increased number of emesis events after diazoxide initiation (p=0.006). A week after initiation of diazoxide, there was no improvement in the percent of total oral intake as would typically be expected. Conclusion: New onset PHTN and re-opening of the DA were associated with initiation of diazoxide treatment. Significant increase in emesis after diazoxide initiation suggests that infants may be experiencing feeding difficulties associated with this medication. Larger studies are needed to further evaluate the extent and frequencies of these side effects.

348. Potential Years of Life Lost in the Population of the Orenburg Region in 2019–2020

Abstract

Introduction: Along with the mortality rate, the indicator of Potential Years of Life Lost (PYLL) is used to assess health losses in the population. Objective: To assess the structure and rates of mortality from diseases of the circulatory system and to estimate the number of potential years of life lost due to this category of disorders in the population of the Orenburg Region. Methods: We conducted a retrospective analysis of deaths registered among the population of the Orenburg Region in 2010– 2020 using a depersonalized database of medical death certificates within the mortality monitoring system of the Medical Information and Analytical Center of the Orenburg Region. The results were evaluated using parametric and nonparametric methods. Potential years of life lost due to the diseases of the circulatory system were estimated by 5-year age groups for the population aged 18–75 in the years 2019 and 2020. Results: We established that the greatest demographic and socio-economic damage was caused by the diseases of the circulatory system, external causes of death, and neoplasms. We also estimated that in 2019, the mortality rate from diseases of the circulatory system was 633.7 per 100,000 population, i.e. 0.7 % lower than that in 2018 (638.2 per 100,000). In 2010–2020, the circulatory disease mortality rate was the highest in 2010 (842.0 per 100,000 population) and the lowest in 2016 (608.2 per 100,000). The average age of the deceased was 72.9 ± 13.8 years, and we observed its increase from 72.0 ± 10.9 years in 2019 to 73.6 ± 9.4 years in the year 2020 (p = 0.014). In 2019, the indicator of premature mortality was 30,895 and 10,515 years for men and women, including 24,323 and 5,404 potential years of life lost in working-age men and women (< 59 and < 54 years of age), respectively. In 2020, the number of PYLL rose to 39,425 and 16,868 years in men and women, respectively. We found significant age and sex differences in the level of losses from premature mortality. Conclusion: Our findings show that the regional mortality rates from diseases of the circulatory system in 2010–2020 exceeded the national averages. The structure of causes of death from diseases of the circulatory system did not change. In 2020, the number of potential years of life lost increased compared to 2019 and was more than twice as high for men than for women.
