253. An in vivoapproach to the reported effects of phenolic acids and flavonoids on the pancreatic α-amylase activity

- Abstract
Reduction in the activity of the α-amylases which hydrolyze starch and glycogen has been proposed for diminishing the incidence of obesity and diabetes. Many plant extracts contain inhibitors of α-amylases and α-glucosidases and several molecules with this activity have been identified. The present work attempts at investigating if several natural compounds (flavonoids and phenolic acids) that have been reported to be inhibitors of those hydrolytic enzymes can also inhibit starch absorption in vivo. The starch tolerance test in mice was used for assaying starch absorption after administration of 100 mg/kg of five flavonoids (epicatechin, catechin, rutin, quercetin, naringenin) and five phenolic acids (gallic, syringic, vanillic, sinapic, ferulic), which are known in vitroinhibitors of α-amylases. Among the flavonoids, only rutin inhibited starch absorption in mice, even though its effect was only 33% of that one found for the positive control acarbose. Among the five phenolic acids tested only syringic acid produced a clear diminution in starch absorption, which was 47% of that one found with acarbose. On the other hand, other three phenolic acids, namely gallic, sinapic and ferulic acid, caused delay in the absorption of starch, which was characterized by a shift in the time peak of maximal blood glucose concentration, with no modification in the excess of glucose that effectively entered the circulation. Attempts at detecting eventual synergisms by administrating mixtures of flavonoids and phenolic acids were unsuccessful. These results recommend careful translation of in vitrodata on the inhibition of α-amylases and α-glucosidases to the in vivoconditions.

The objective of this study was to evaluate the effect of feed restriction on some physical and blood parameters in horses kept outdoors under natural conditions. Twenty horses were deprived of food for 48 h. They were closely monitored and examined, and blood samples were taken at the beginning (0) of the experiment and 6, 12, 18, 24, 30, 36, 42 and 48 hours afterward. During the experimental period, the control group (12 animals) had free access to water and hay, while the restricted group had free access to water only. Data were submitted to two-way analysis of variance with repeated measures, and statistical significance was P ≤ 0.05. The horses tolerated feed restriction without complications. Feed restriction had no effect on body mass and body condition score, heart rate, respiratory rate, capillary filling time and body temperature. - Abstract
This is a qualitative study that aims at analyzing the Primary Health Care Strategic Planning in a continuing education process, as well as the professional’s formation to work as facilitators in it. Data was obtained through interviews with 11 nurses that had acted as the plan’s facilitators in a municipality within Belo Horizonte. The results indicate that the experience as facilitators allowed them to reflect on the work process and this practice contributed to the incorporation of new tools to the primary health care system. The participants reported the difficulties faced when conducting the experience and the gap in the professionals’ formation to act in the PHC and to put into practice the processes of continuing education on a day to day basis. In conclusion, the Planning represents an important continuing education strategy and it is significance to transform processes and practices in the primary health care service. IKr current, a major component of cardiac repolarization, is mediated by human Ether-à-go-go-Related Gene (hERG, Kv11.1) potassium channels. The blockage of these channels by pharmacological compounds is associated to drug-induced long QT syndrome (LQTS), which is a life-threatening disorder characterized by ventricular arrhythmias and defects in cardiac repolarization that can be illustrated using cardiomyocytes derived from human-induced pluripotent stem cells (hiPS-CMs). This study was meant to assess the modification in hiPS-CMs excitability and contractile properties by BeKm-1, a natural scorpion venom peptide that selectively interacts with the extracellular face of hERG, by opposition to reference compounds that act onto the intracellular face. Using an automated patch-clamp system, we compared the affinity of BeKm-1 for hERG channels with some reference compounds. We fully assessed its effects on the electrophysiological, calcium handling, and beating properties of hiPS-CMs. By delaying cardiomyocyte repolarization, the peptide induces early afterdepolarizations and reduces spontaneous action potentials, calcium transients, and contraction frequencies, therefore recapitulating several of the critical phenotype features associated with arrhythmic risk in drug-induced LQTS. BeKm-1 exemplifies an interesting reference compound in the integrated hiPS-CMs cell model for all drugs that may block the hERG channel from the outer face. Being a peptide that is easily modifiable, it will serve as an ideal molecular platform for the design of new hERG modulators displaying additional functionalities.

259. Índices plaquetários em indivíduos com doença hepática alcoólica crônica Platelet indices in chronic alcoholic liver disease patients with thrombocytopenia

OBJETIVO: Detectar alterações nos índices plaquetários em indivíduos com doença hepática alcoólica crónica e trombocitopenia e determinar o seu grau de correlação com outros parâmetros hematológicos. MÉTODOS: Foram estudados um total de 65 indivíduos, divididos em dois grupos: controlos (n = 35) e com doença hepática alcoólica crónica e trombocitopenia (n = 30). O grupo controlo foi emparelhado por idade e sexo com o grupo de doentes. A todos eles, foi efectuado hemograma completo, incluindo índices plaquetários. RESULTADOS: Os doentes com doença hepática alcoólica crónica apresentaram contagem de eritrócitos, leucócitos e plaquetas significativamente inferiores aos encontrados na população controlo. O mesmo se verificou com os valores de hemoglobina, hematócrito e valores absolutos de linfócitos e neutrófilos. O volume globular médio, a hemoglobina globular média e o RDW ("red cell distribution width") revelaram-se significativamente superiores no grupo de doentes. Relativamente aos índices plaquetários, foi encontrado aumento estatisticamente significativo do PDW ("platelet distribution width") e uma diminuição estatisticamente significativa no plaquetrócrito no grupo de doentes quando comparado com o grupo controlo. Em relação ao volume plaquetário médio não se encontraram diferenças estatisticamente significativas entre os dois grupos. Foi encontrada correlação estatisticamente significativa entre a contagem de plaquetas e os outros parâmetros hematológicos analisados. O plaquetrócito revelou-se o índice plaquetário que mais se relaciona com os restantes parâmetros hematológicos. CONCLUSÃO: Os doentes com doença hepática alcoólica crónica apresentam diminuição nas três linhas hematopoiéticas, possivelmente associada com o hiperesplenismo que estes doentes apresentam. Adicionalmente às alterações numéricas, encontraram-se alterações morfológicas a nível dos eritrócitos e plaquetas, demonstráveis pelos índices eritrocitários e plaquetários.AIM: To detect alterations in platelet indices in patients with chronic alcoholic liver disease and thrombocytopenia, and its correlation with other haematological parameters. METHODS: We studied 65 individuals separated in two groups: controls (n = 35) and chronic alcoholic liver disease patients with thrombocytopenia (n = 30). The control group was age and gender matched with patients group. In all, controls and patients, a haematological evaluation was done, including platelets indices. RESULTS: In the patients group we found a low number of erythrocytes, leucocytes and platelet when we compare with controls. The same is true when we compare haemoglobin, hematocrit and absolute count of lymphocyte and neutrophil. The mean globular volume, mean globular haemoglobin and red cell distribution width where significantly higher in patients group. Platelet indices showed a statistical significant increased in platelet distribution width and decreased in platelet crit in the patient group. No differences where found on mean platelet volume between the two groups. Correlation between platelet number and other haematological parameters was found. CONCLUSION: Chronic alcoholic liver disease patients showed a decrease in all haematopoietic cell lines, probably associated with hypersplenism found in those patients. Additionally to the numeric alterations the erythrocyte and platelets showed morphologic alteration revelled by respective indices.

260. Importance of multidisciplinary trauma prevention program for youth

OBJECTIVE: present the experience of the P.A.R.T.Y. program in Campinas, thereby changing the habits of young people.METHODS: The organizers visited the participating schools talking to the students, who are aged between 14-18 years. These students spent an afternoon at the Clinics Hospital of Unicamp, where, for four hours, they attended lectures of the organizers, partners and municipal sectors, and also visited the hospital, talking with trauma victims. Questionnaires were evaluated between2010-2012, being applied before and after the project.RESULTS:2,450 high school students attended the program. The mean age is 16 ± 0,99 years and 37.6% were male. 3.6% of males already drive while drunk versus 0.8% of women. Before the project 116 (11.3%) thought that drunk driving wasn't a risk, and only 37 (3.6%) knew the alcohol effects. After the project, 441 (43%) began to consider drunk driving a risk and 193 (18.8%) know the alcohol effects when driving. 956 (93.3%) considered that prevention projects have a huge impact on their formation.CONCLUSION: It's expected that the attendees will act as multipliers of information, conveying the message of prevention to their entire social circles resulting in reduction in the number of trauma events involving the young, in the long term.
