l‐galactono‐1,4‐lactone dehydrogenase (GALDH) catalyzes the terminal step of vitamin C biosynthesis in plant mitochondria. Here we investigated the communication between Arabidopsis thaliana GALDH and its natural electron acceptor cytochrome c (Cc). Using laser‐generated radicals we observed the formation and stabilization of the GALDH semiquinone anionic species (GALDHSQ). GALDHSQ oxidation by Cc exhibited a nonlinear dependence on Cc concentration consistent with a kinetic mechanism involving protein–partner association to form a transient bimolecular complex prior to the electron transfer step. Oxidation of GALDHSQ by Cc was significantly impaired at high ionic strength, revealing the existence of attractive charge–charge interactions between the two reactants. Isothermal titration calorimetry showed that GALDH weakly interacts with both oxidized and reduced Cc. Chemical shift perturbations for 1H and 15N nuclei of Cc, arising from the interactions with unlabeled GALDH, were used to map the interacting surface of Cc. For Arabidopsis Cc and yeast Cc, similar residues are involved in the interaction with GALDH. These residues are confined to a single surface surrounding the heme edge. The range of chemical shift perturbations for the physiological Arabidopsis Cc–GALDH complex is larger than that of the non‐physiological yeast Cc–GALDH complex, indicating that the former complex is more specific. In summary, the results point to a relatively low affinity GALDH–Cc interaction, similar for all partner redox states, involving protein–protein dynamic motions. Evidence is also provided that Cc utilizes a conserved surface surrounding the heme edge for the interaction with GALDH and other redox partners.

NADPH-dependent thioredoxin reductases (NTRs) contain a flavin cofactor and a disulfide as redox-active groups. The catalytic mechanism of standard NTR involves a large conformational change between two configurations. Oxygenic photosynthetic organisms possess a plastid-localized NTR, called NTRC, with a thioredoxin module fused at the C terminus. NTRC is an efficient reductant of 2-Cys peroxiredoxins (2-Cys Prxs) and thus is involved in the protection against oxidative stress, among other functions. Although the mechanism of electron transfer of canonical NTRs is well established, it is not yet known in NTRC. By employing stopped-flow spectroscopy, we have carried out a comparative kinetic study of the electron transfer reactions involving NTRC, the truncated NTR module of NTRC, and NTRB, a canonical plant NTR. Whereas the three NTRs maintain the conformational change associated with the reductive cycle of catalysis, NTRC intramolecular electron transfer to the thioredoxin module presents two kinetic components (kET of ∼2 and 0.1 s−1), indicating the occurrence of additional dynamic motions. Moreover, the dynamic features associated with the electron transfer to the thioredoxin module are altered in the presence of 2-Cys Prx. NTRC shows structural constraints that may locate the thioredoxin module in positions with different efficiencies for electron transfer, the presence of 2-Cys Prx shifting the conformational equilibrium of the thioredoxin module to a specific position, which is not the most efficient

Under nitroxidative stress, a minor fraction of cytochrome c can be modified by tyrosine nitration. Here we analyze the specific effect of nitration of tyrosines 46 and 48 on the dual role of cytochrome c in cell survival and cell death. Our findings reveal that nitration of these two solvent-exposed residues has a negligible effect on the rate of electron transfer from cytochrome c to cytochrome c oxidase, but impairs the ability of the heme protein to activate caspase-9 by assembling a non-functional apoptosome. It seems that cytochrome c nitration under cellular stress counteracts apoptosis in light of the small amount of modified protein. We conclude that other changes such as increased peroxidase activity prevail and allow the execution of apoptosis.

Plastocyanin and cytochrome c 6, the alternate donor proteins to photosystem I, can be acidic, neutral or basic; the role of electrostatics in their interaction with photosystem I vary accordingly for cyanobacteria, algae and plants. The effect of different crowding agents on the kinetics of the reaction between plastocyanin or cytochrome c 6 and photosystem I from three different cyanobacteria, Synechocystis PCC 6803, Nostoc PCC 7119 and Arthrospira maxima, and a green alga, Monoraphidium braunii, has been investigated by laser flash photolysis, in order to elucidate how molecular crowding affects the interaction between the two donor proteins and photosystem I. The negative effect of viscosity on the interaction of the two donors with photosystem I for the three cyanobacterial systems is very similar, as studied by increasing sucrose concentration. Bovine serum albumin seems to alter the different systems in a specific way, probably by means of electrostatic interactions with the donor proteins. Ficoll and dextran behave in a parallel manner, favouring the interaction by an average factor of 2, although this effect is somewhat less pronounced in Nostoc. With regards to the eukaryotic system, a strong negative effect of viscosity is able to overcome the favourable effect of any crowding agent, maybe due to stronger donor/photosystem I electrostatic interactions or the structural nature of the eukaryotic photosystem I-enriched membrane particles.

Construcción génica que codifica para el citocromo c y procedimiento de obtención. Se describe una construcción génica que comprende (i) una primera secuencia de ácido nucleico que comprende una secuencia de nucleótidos que codifica para un cito cromo c, y (ii) una segunda secuencia de ácido nucleico que comprende una secuencia de nucleótidos que codi fica para un péptido señal; en donde el extremo 5’ de di cha primera secuencia de ácido nucleico está unido al ex tremo 3’ de dicha segunda secuencia de ácido nucleico. Dicha construcción génica puede ser utilizada para cons truir vectores útiles para transformar, junto con un vec tor que comprende los genes necesarios para la madura ción del citocromo c, una célula hospedadora y producir citocromo c.

Con el presente trabajo de Tesis se pretende establecer las diferencias funcionales entre sistemas de transferencia de electrones homólogos pertenecientes a diferentes organismos, así como profundizar en el conocimiento de las interacciones moleculares responsables de la formación del complejo transitorio y la posterior transferencia de electrones. En definitiva, el fin último de este trabajo es a portar nuevos datos que ayuden a comprender los factores que controlan la relación estructura-función, así como el modo de interacción y formación de complejos interproteicos, con especial énfasis en los rasgos que les confieren a las proteínas su capacidad para interaccionar especifica y transitoriamente con otras.Como sistema modelo para abordar el estudio se eligió, dados los antecedentes, la interacción del par Cf-Pc en la cianobacteria Nostoc sp. PCC 7119. Los estudios funcionales y estructurales que se habían realizados en trabajos previos sobre la transferencia electrónica desde la Pc hasta el PSI en dicho organismo, los realizados sobre el complejo Cf-Pc en otras cianobacterias y en plantas, así como los recientes estudios estructurales mediante RMN del propio sistema silvestre de Nostoc, permitían asegurar que el análisis funcional de dicho sistema aportaría datos novedosos sobre el mismo y permitiría concluir los trabajos previos realizados en dicho organismo. Con dichos antecedentes y dirigidos por los planteamientos mencionados, en este trabajo de tesis se decidió llevar a cabo la caracterización, desde el punto de vista funcional, de la transferencia de electrones desde el Cf a uno de sus aceptores fisiológicos, la Pc, en la cianobacteria Nostoc sp. PCC 7119, llevando a cabo un análisis comparado de los resultados obtenidos con complejos homólogos de otros organismos. In cyanobacteria, cytochrome c6 and plastocyanin are able to replace each other as redox carriers in the photosynthetic and respiratory electron transport chains with the synthesis of one or another protein being regulated by the copper concentration in the culture medium. However, the presence of a third unidentified electron carrier has been suggested. To address this point, we have constructed two deletion mutants of the cyanobacterium Synechocystis sp. PCC 6803, each variant lacking either the petE or petJ gene, which respectively codes for the copper or heme protein. The photoautotrophic and heterotrophic growth rate of the two mutants in copper-free and copper-supplemented medium as well as their photosystem I reduction kinetics in vivo were compared with those of wild-type cells. The two mutant strains grow at equivalent rates and show similar in vivo photosystem I reduction kinetics as wild-type cells when cultured in media that allow the expression of just one of the two electron donor proteins, but their ability to grow and reduce photosystem I is much lower when neither cytochrome c6 nor plastocyanin is expressed. These findings indicate that the normal functioning of the cyanobacterial photosynthetic and respiratory chains obligatorily depends on the presence of either cytochrome c6 or plastocyanin.

Photosystem I reduction by the soluble metalloproteins cytochrome c6 and plastocyanin, which are alternatively synthesized by some photosynthetic organisms depending on the relative availability of copper and iron, has been investigated in cyanobacteria, green algae and plants. The reaction mechanism is classified in three different types on the basis of the affinity of the membrane complex towards its electron donor protein. The role of electrostatic interactions in forming an intermediate transient complex, as well as the structural and functional similarities of cytochrome c6 and plastocyanin are analysed from an evolutionary point of view. The proposal made is that the heme protein was first “discovered” by nature, when iron was much more abundant on the Earth's surface, and replaced by plastocyanin when copper became available because of the oxidizing conditions of the new atmosphere.

The influence of the amino acid residues sandwiching the flavin ring in flavodoxin (Fld) from the cyanobacteriumAnabaena sp. PCC 7119 in complex formation and electron transfer (ET) with its natural partners, photosystem I (PSI) and ferredoxin-NADP+ reductase (FNR), was examined in mutants of the key residues Trp57 and Tyr94. The mutants' ability to form complexes with either FNR or PSI is similar to that of wild-type Fld. However, some of the mutants exhibit altered kinetic properties in their ET processes that can be explained in terms of altered flavin accessibility and/or thermodynamic parameters. The most noticeable alteration is produced upon replacement of Tyr94 by alanine. In this mutant, the processes that involve the transfer of one electron from either PSI or FNR are clearly accelerated, which might be attributable to a larger accessibility of the flavin to the reductant. However, when the opposite ET flow is analyzed with FNR, the reduced Y94A mutant transfers electrons to FNR slightly more slowly than wild type. This can be explained thermodynamically from a decrease in driving force due to the significant shift of 137 mV in the reduction potential value for the semiquinone/hydroquinone couple (E 1) of Y94A, relative to wild type (Lostao, A., Gómez-Moreno, C., Mayhew, S. G., and Sancho, J. (1997) Biochemistry 36, 14334–14344). The behavior of the rest of the mutants can be explained in the same way. Overall, our data indicate that Trp57 and Tyr94 do not play any active role in flavodoxin redox reactions providing a path for the electrons but are rather involved in setting an appropriate structural and electronic environment that modulates in vivo ET from PSI to FNR while providing a tight FMN binding.

Una de las líneas de investigación mas intereses de la bioquímica contemporánea es el estudio de la relación entre la estructura y la función de las macromoléculas biológicas, o reconocimiento molecular. Como sistema objeto de este estudio, se ha escogido el citocromo c6(Cit) y la plastocianina (Pc), dos proteínas con estructuras diferentes pero con idéntica función, cual es transportar electrones entre los complejos de membrana citocromo b6-f y fotosistema I(PSI). La conexión entre ambos complejos la realiza una metaloproteina soluble, que puede ser la Pc en plantas o el Cit en cianobacterias, aunque existen algas verdes (Eucarióticas) y otras cianobacterias capaces de sintetizar tanto Pc como Cit. La siguiente pregunta se plantea de inmediato: ¿Por qué el Cit se ha visto desplazado por la Pc durante el curso evolutivo? Cit y Pc consituyen un excelente ejemplo para el estudio de la evolución biológica a nivel molecular. Ambas proteínas poseen propiedades bioquímicas similares, y exhiben un mecanismo de reacción con el PSI distinto de un organismo a otro pero similar dentro de un mismo organismo. En este proyecto se han clonado, mutado y expresado en la enterobacteria Excherichia coli los genes que codifican la Pc(p e tE) y el Cit(petJ) de la cianobacteria Anabaena sp. PC 7119. Posteriormente, se ha realizado un estudio estructural y funcional comparado de todas las proteínas recombinantes. Los resultados obtenidos son los siguientes: 1. Se han clonado, secuenciado y expresado correctamente en Escherichia coli los genes petJ y petE de la cianobacteria Anabaena sp PCC 7119. 2. Se ha diseñado un método de superproducción del citocromo c6, cuyo rendimiento ha permitido realizar estudios funcionales y estructurales, así como marcar homogeneamente con 15 N la hemoproteína. 3. Positively charged plastocyanin from Anabaena sp. PCC 7119 was investigated by site-directed mutagenesis. The reactivity of its mutants toward photosystem I was analyzed by laser flash spectroscopy. Replacement of arginine at position 88, which is adjacent to the copper ligand His-87, by glutamine and, in particular, by glutamate makes plastocyanin reduce its availability for transferring electrons to photosystem I. Such a residue in the copper protein thus appears to be isofunctional with Arg-64 (which is close to the heme group) in cytochrome c6 from Anabaena(Molina-Heredia, F. P., Dı́az-Quintana, A., Hervás, M., Navarro, J. A., and De la Rosa, M. A. (1999)J. Biol. Chem. 274, 33565–33570) and Synechocystis (De la Cerda, B., Dı́az-Quintana, A., Navarro, J. A., Hervás, M., and De la Rosa, M. A. (1999) J. Biol. Chem. 274, 13292–13297). Other mutations concern specific residues of plastocyanin either at its positively charged east face (D49K, H57A, H57E, K58A, K58E, Y83A, and Y83F) or at its north hydrophobic pole (L12A, K33A, and K33E). Mutations altering the surface electrostatic potential distribution allow the copper protein to modulate its kinetic efficiency: the more positively charged the interaction site, the higher the rate constant. Whereas replacement of Tyr-83 by either alanine or phenylalanine has no effect on the kinetics of photosystem I reduction, Leu-12 and Lys-33 are essential for the reactivity of plastocyanin.

Photosystem I (PSI) interacts with plastocyanin or cytochrome c 6 on the luminal side. To identify sites of interaction between plastocyanin/cytochromec 6 and the PSI core, site-directed mutations were generated in the luminal J loop of the PsaB protein fromSynechocystis sp. PCC 6803. The eight mutant strains differed in their photoautotrophic growth. Western blotting with subunit-specific antibodies indicated that the mutations affected the PSI level in the thylakoid membranes. PSI proteins could not be detected in the S600R/G601C/N602I, N609K/S610C/T611I, and M614I/G615C/W616A mutant membranes. The other mutant strains contained different levels of PSI proteins. Among the mutant strains that contained PSI proteins, the H595C/L596I, Q627H/L628C/I629S, and N638C/N639S mutants showed similar levels of PSI-mediated electron transfer activity when either cytochrome c 6 or an artificial electron donor was used. In contrast, cytochromec 6 could not function as an electron donor to the W622C/A623R mutant, even though the PSI activity mediated by an artificial electron donor was detected in this mutant. Thus, the W622C/A623R mutation affected the interaction of the PSI complex with cytochrome c 6. Biotin-maleimide modification of the mutant PSI complexes indicated that His-595, Trp-622, Leu-628, Tyr-632, and Asn-638 in wild-type PsaB may be exposed on the surface of the PSI complex. The results presented here demonstrate the role of an extramembrane loop of a PSI core protein in the interaction with soluble electron donor proteins.

A number of surface residues of cytochromec 6 from the cyanobacterium Anabaenasp. PCC 7119 have been modified by site-directed mutagenesis. Changes were made in six amino acids, two near the heme group (Val-25 and Lys-29) and four in the positively charged patch (Lys-62, Arg-64, Lys-66, and Asp-72). The reactivity of mutants toward the membrane-anchored complex photosystem I was analyzed by laser flash absorption spectroscopy. The experimental results indicate that cytochrome c 6 possesses two areas involved in the redox interaction with photosystem I: 1) a positively charged patch that may drive its electrostatic attractive movement toward photosystem I to form a transient complex and 2) a hydrophobic region at the edge of the heme pocket that may provide the contact surface for the transfer of electrons to P700. The isofunctionality of these two areas with those found in plastocyanin (which acts as an alternative electron carrier playing the same role as cytochromec 6) are evident.

This paper reports the first site-directed mutagenesis analysis of any cytochrome c 6, a heme protein that performs the same function as the copper-protein plastocyanin in the electron transport chain of photosynthetic organisms. Photosystem I reduction by the mutants of cytochromec 6 from the cyanobacteriumSynechocystis sp. PCC 6803 has been studied by laser flash absorption spectroscopy. Their kinetic efficiency and thermodynamic properties have been compared with those of plastocyanin mutants from the same organism. Such a comparative study reveals that aspartates at positions 70 and 72 in cytochrome c 6 are located in an acidic patch that may be isofunctional with the well known “south-east” patch of plastocyanin. Calculations of surface electrostatic potential distribution in the mutants of cytochromec 6 and plastocyanin indicate that the changes in protein reactivity depend on the surface electrostatic potential pattern rather than on the net charge modification induced by mutagenesis. Phe-64, which is close to the heme group and may be the counterpart of Tyr-83 in plastocyanin, does not appear to be involved in the electron transfer to photosystem I. In contrast, Arg-67, which is at the edge of the cytochrome c 6 acidic area, seems to be crucial for the interaction with the reaction center.

El presente trabajo forma parte del realizado por el grupo al que el doctorando se encuentra adscrito, en el que se busca una mejor compresión de la relación estructura-función en proteínas redox. En este sentido, se están realizando análisis comparados del citocromo c6 y la plastocianina como donadores de electrones del fotosistema I (Hervás et al., 1992a; 1992c; 1994; Medina et al., 1993; Díaz et al., 1994), así como de la ferredoxina y la flavodoxina como aceptores de electrones de dicho fotosistema (Hervás et al., 1992b; 1992c; Medina at al., 1992). El estudio comparado de dichas proteínas tiene interés desde el punto de vista evolutivo, no sólo por la selección de la plastocianina y la ferredocina como donador y aceptor de electrones -respectivamente- del PSI en plantas superiores, en lugar de sus respectivos análogos, sino por el esclarecimiento de una posible evolución en los mecanismos de reacción de dichas proteínas (Hervás et al., 1994). Por otro lado, se pretende determinar qué residuos intervienen en la interacción de las metaloproteínas con el PSI mediante el estudio de proteínas modificadas por mutagénesis dirigida, para lo que se ha clonado el gen petE de Synechocystis sp. PCC 6803, que codifica la plastociannina (Hervás et al., 1993). En el presente trabajo se muestra un estudio sobre la naturaleza de la interacción del PSI de Monoraphidium con sus donadores de electrones y se compara con resultados obtenidos con PSI y metaloproteínas obtenidas de Anabaena, donde las fuerzas electrostáticas de largo alcance son atractivas, en lugar de repulsivas. Además, se ha clonado en E. coli el gen petJ de Synechocystis sp. PCC 6803, que codifica el citocromo c6, y se ha demostrado que la proteína se sintetiza y madura correctamente en la enterobacteria. En este trabajo se ha abordado el estudio de la localización y función del citocromo b-559 en la cadena fotosintética de transporte de electrones. En primer lugar, se ha llevado a cabo una caracterización, sobre todo desde el punto de vista redox y acido-base, de este citocromo en partículas del fotosistema II. Los resultados obtenidos en este sentido, así como los extraídos de la purificación y el análisis de algunas propiedades físico-químicas de esta proteína, nos ha permitido establecer que este citocromo es una única especie molecular que puede existir como dos pares interconvertibles con diferente potencial de oxido-reducción: uno de alto potencial lábil y protonado y otro de bajo potencial estable y desprotonado. Asimismo, hemos caracterizado la estabilidad diferencial de las formas oxidada y reducida del par de alto potencial. Nuestros experimentos, en fin, nos han permitido cerrar el ciclo redox y acido-base de este citocromo, consiguiendo todas las transformaciones posibles entre las cuatro formas de los dos pares. Por otra parte, el problema de la localización ha sido abordado, en cloroplastos, mediante el estudio comparativo del efecto de agentes inhibidores muy específicos y desacoplantes sobre los cambios redox inducidos por luces selectivas de los fotosistemas I y II en los citocromos F y b-559. Los resultados obtenidos permiten concluir que el citocromo b-559 es reducido por el fotosistema II a través de QA, QB y plastoquínona; y que se oxida por el fotosistema I, interviniendo en esta oxidación la plastocianína y, muy probablemente, el citocromo F. Además, los resultados obtenidos con desacoplantes sugieren que este citocromo se encuentra implicado en el sitio de acoplamiento de la cadena lineal de transporte de electrones. En definitiva, la conclusión final de este trabajo es que el citocromo b-559 se encuentra localizado en la cadena principal de transporte de electrones entre la plastoquinona y la plastocianina. En este trabajo se ha tratado de profundizar en el posible papel en la transducción de la energía del citocromo b-564 de la cadena respiratoria, así como del citocromo b-559 de la cadena fotosintética. Para ello, se ha realizado un estudio potenciométrico y espectrofotométrico de ambos citrocromos, tratando de confirmar la existencia de estos citrocromos en dos formas de potencial, así como el hecho de que son realmente una única proteína. Se ha estudiado el efecto de distintos tratamientos que afectan al metabolismo energético, y en particular a la síntesis de ATP, sobre el estado redox y la proporción en las dos formas de potencial de estos citocromos. Así mismo, se ha realizado un estudio del comportamiento de estos citocromos respecto al pH. En el caso del citocromo b-564 de mitocondrias de levadura, se ha aislado el complejo III del que forma parte, estudiándose este citocromo en el complejo aislado, así como cuando éste ha sido incorporado en vesículas artificiales de lípidos. En el caso del citocromo b-559 se han realizado estudios tratando de relacionar la interconversión entre las dos formas de potencial con la capacidad de translocación de protones por vesículas tilacoidales. La localización de estos dos citocromos de tipo “b” en sitios de fosforilación, así como su peculiar comportamiento hacen de ellos candidatos idóneos para llevar a cabo el acoplamiento entre la energía redox y ácido-base en estos sitios de acoplamiento.
