A primary goal of molecular physiology is to understand how conformational changes of proteins affect the function of cells, tissues, and organisms. Here, we describe an imaging method for measuring the conformational changes of the voltage sensors of endogenous ion channel proteins within live tissue, without genetic modification. We synthesized GxTX-594, a variant of the peptidyl tarantula toxin guangxitoxin-1E, conjugated to a fluorophore optimal for two-photon excitation imaging through light-scattering tissue. We term this tool EVAP (Endogenous Voltage-sensor Activity Probe). GxTX-594 targets the voltage sensors of Kv2 proteins, which form potassium channels and plasma membrane-endoplasmic reticulum junctions. GxTX-594 dynamically labels Kv2 proteins on cell surfaces in response to voltage stimulation. To interpret dynamic changes in fluorescence intensity, we developed a statistical thermodynamic model that relates the conformational changes of Kv2 voltage sensors to degree of labeling. We used two-photon excitation imaging of rat brain slices to image Kv2 proteins in neurons. We found puncta of GxTX-594 on hippocampal CA1 neurons that responded to voltage stimulation and retain a voltage response roughly similar to heterologously expressed Kv2.1 protein. Our findings show that EVAP imaging methods enable the identification of conformational changes of endogenous Kv2 voltage sensors in tissue.

Abstract Background Diabetic foot is a significant cause of morbidity in diabetic patients, with a rate that is approximately twice that of patients without foot ulcers. “Metabolic memory” represents the epigenetic changes induced by chronic hyperglycaemia, despite the correction of the glucose levels themselves. These epigenetic modifications appear to perpetuate the damage caused by persistently elevated glucose levels even in their absence, acting at various levels, mostly affecting the molecular processes of diabetic ulcer healing. Methods The aim of our cross-sectional study was to analyse a cohort of patients with diabetes with and without lower limb ulcers. We examined the effects of epigenetic changes on miRNA 126, 305, and 217 expression and the frequency of the SNPs of genes encoding inflammatory molecules (e.g., IL-6 and TNF-alpha) and their correlations with serum levels of proangiogenic molecules (e.g., ENOS, VEGF and HIF-1alpha) and several adipokines as well as with endothelial dysfunction, assessed noninvasively by reactive hyperaemia peripheral artery tonometry. Between March 2021 and June 2022, 110 patients were enrolled into the study: 50 diabetic patients with diabetic foot injuries, 40 diabetic patients without ulcerative complications and 20 nondiabetic patients as the control group. Results Diabetic subjects with lower limb ulcerative lesions exhibited higher levels of inflammatory cytokines, such as VEGF (191.40 ± 200 pg/mL vs. 98.27 ± 56.92 pg/mL vs. 71.01 ± 52.96 pg/mL; p = 0.22), HIF-1alpha (40.18 ± 10.80 ng/mL vs. 33.50 ± 6.16 ng/mL vs. 33.85 ± 6.84 ng/mL; p = 0.10), and Gremlin-1 (1.72 ± 0.512 ng/mL vs. 1.31 ± 0.21 ng/mL vs. 1.11 ± 0.19 ng/mL; p

Abstract Background We developed a 13-mer locked nucleic acid (LNA) inhibitor of miR-221 (LNA-i-miR-221) with a full phosphorothioate (PS)-modified backbone. This agent downregulated miR-221, demonstrated anti-tumor activity against human xenografts in mice, and favorable toxicokinetics in rats and monkeys. Allometric interspecies scaling allowed us to define the first-in-class LNA-i-miR-221 safe starting dose for the clinical translation. Methods In this first-in-human, open-label, dose-escalation phase 1 trial, we enrolled progressive cancer patients (aged ≥ 18 years) with ECOG 0–2 into 5 cohorts. The treatment cycle was based on a 30-min IV infusion of LNA-i-miR-221 on 4 consecutive days. Three patients within the first cohort were treated with 2 cycles (8 infusions), while 14 patients were treated with a single course (4 infusions); all patients were evaluated for phase 1 primary endpoint. The study was approved by the Ethics Committee and Regulatory Authorities (EudraCT 2017-002615-33). Results Seventeen patients received the investigational treatment, and 16 were evaluable for response. LNA-i-miR-221 was well tolerated, with no grade 3–4 toxicity, and the MTD was not reached. We recorded stable disease (SD) in 8 (50.0%) patients and partial response (PR) in 1 (6.3%) colorectal cancer case (total SD + PR: 56.3%). Pharmacokinetics indicated non-linear drug concentration increase across the dose range. Pharmacodynamics demonstrated concentration-dependent downregulation of miR-221 and upregulation of its CDKN1B/p27 and PTEN canonical targets. Five mg/kg was defined as the recommended phase II dose. Conclusions The excellent safety profile, the promising bio-modulator, and the anti-tumor activity offer the rationale for further clinical investigation of LNA-i-miR-221 (ClinTrials.Gov: NCT04811898).

Pregnancy is an immunologically regulated, complex process. A tightly controlled complement system plays a crucial role in the successful establishment of pregnancy and parturition. Complement inhibitors at the feto-maternal interface are likely to prevent inappropriate complement activation to protect the fetus. In the present study, we aimed to understand the role of Factor H (FH), a negative regulator of complement activation, in normal pregnancy and in a model of pathological pregnancy, i.e. preeclampsia (PE). The distribution and expression of FH was investigated in placental tissues, various placental cells, and in the sera of healthy (CTRL) or PE pregnant women via immunohistochemistry, RT-qPCR, ELISA, and Western blot. Our results showed a differential expression of FH among the placental cell types, decidual stromal cells (DSCs), decidual endothelial cells (DECs), and extravillous trophoblasts (EVTs). Interestingly, FH was found to be considerably less expressed in the placental tissues of PE patients compared to normal placental tissue both at mRNA and protein levels. Similar results were obtained by measuring circulating FH levels in the sera of third trimester CTRL and PE mothers. Syncytiotrophoblast microvesicles, isolated from the placental tissues of PE and CTRL women, downregulated FH expression by DECs. The present study appears to suggest that FH is ubiquitously present in the normal placenta and plays a homeostatic role during pregnancy.

IntroductionDuring the pandemic, the Cognitive Disorders Unit of San Raffaele Hospital (Milan, Italy) offered patients the opportunity to undergo neuropsychological evaluations and cognitive training through telemedicine.MethodWe conducted an investigation to assess how patients responded to this option and to determine if telemedicine could ensure continuity of care.ResultsBetween October 2019 and May 2022, a total of 5,768 telemedicine appointments and 8,190 in-person outpatient appointments were conducted, resulting in an increase in the rate of telemedicine activity from 16.81% in January 2020 to 23.21% in May 2022. Peaks in telemedicine activity reached 85.64% in May 2020 and 83.65% in February 2021, both representing a significant portion of the total activity. Interestingly, there was a notable positive correlation between telemedicine activity and the worsening of the Italian pandemic (r = 0.433, p = 0.027).DiscussionDuring the peaks of contagion, the total number of visits remained stable, highlighting that telemedicine effectively served as a valuable and efficient tool to ensure continuity of care for vulnerable patients. This was evident from the integration of remote visits with in-person appointments.

We aim at modelling the appearance of distinct tags in a sequence of labelled objects. Common examples of this type of data include words in a corpus or distinct species in a sample. These sequential discoveries are often summarised via accumulation curves, which count the number of distinct entities observed in an increasingly large set of objects. We propose a novel Bayesian nonparametric method for species sampling modelling by directly specifying the probability of a new discovery, therefore allowing for flexible specifications. The asymptotic behavior and finite sample properties of such an approach are extensively studied. Interestingly, our enlarged class of sequential processes includes highly tractable special cases. We present a subclass of models characterized by appealing theoretical and computational properties. Moreover, due to strong connections with logistic regression models, the latter subclass can naturally account for covariates. We finally test our proposal on both synthetic and real data, with special emphasis on a large fungal biodiversity study in Finland.

The $3 \times 1 \times 1$ m$^3$ demonstrator is a dual phase liquid argon time projection chamber that has recorded cosmic rays events in 2017 at CERN. The light signal in these detectors is crucial to provide precise timing capabilities. The performances of the photon detection system, composed of five PMTs, are discussed. The collected scintillation and electroluminescence light created by passing particles has been studied in various detector conditions. In particular, the scintillation light production and propagation processes have been analyzed and compared to simulations, improving the understanding of some liquid argon properties., Comment: 30 pages, 26 figures

Hyper-Kamiokande is the next generation underground water Cherenkov detector that builds on the highly successful Super-Kamiokande experiment. The detector which has an 8.4~times larger effective volume than its predecessor will be located along the T2K neutrino beamline and utilize an upgraded J-PARC beam with 2.6~times beam power. Hyper-K's low energy threshold combined with the very large fiducial volume make the detector unique, that is expected to acquire an unprecedented exposure of 3.8~Mton$\cdot$year over a period of 20~years of operation. Hyper-Kamiokande combines an extremely diverse science program including nucleon decays, long-baseline neutrino oscillations, atmospheric neutrinos, and neutrinos from astrophysical origins. The scientific scope of this program is highly complementary to liquid-argon detectors for example in sensitivity to nucleon decay channels or supernova detection modes. Hyper-Kamiokande construction has started in early 2020 and the experiment is expected to start operations in 2027. The Hyper-Kamiokande collaboration is presently being formed amongst groups from 19 countries including the United States, whose community has a long history of making significant contributions to the neutrino physics program in Japan. The Deep Underground Neutrino Experiment (DUNE) will be a powerful tool for a variety of physics topics. The high-intensity proton beams provide a large neutrino flux, sampled by a near detector system consisting of a combination of capable precision detectors, and by the massive far detector system located deep underground. This configuration sets up DUNE as a machine for discovery, as it enables opportunities not only to perform precision neutrino measurements that may uncover deviations from the present three-flavor mixing paradigm, but also to discover new particles and unveil new interactions and symmetries beyond those predicted in the Standard Model (SM). Of the many potential beyond the Standard Model (BSM) topics DUNE will probe, this paper presents a selection of studies quantifying DUNE's sensitivities to sterile neutrino mixing, heavy neutral leptons, non-standard interactions, CPT symmetry violation, Lorentz invariance violation, neutrino trident production, dark matter from both beam induced and cosmogenic sources, baryon number violation, and other new physics topics that complement those at high-energy colliders and significantly extend the present reach., Comment: 54 pages, 40 figures, paper based on the DUNE Technical Design Report (arXiv:2002.03005)

We report measurements by the T2K experiment of the parameters $\theta_{23}$ and $\Delta m^2_{32}$ which govern the disappearance of muon neutrinos and antineutrinos in the three-flavor PMNS neutrino oscillation model at T2K's neutrino energy and propagation distance. Utilizing the ability of the experiment to run with either a mainly neutrino or a mainly antineutrino beam, muon-like events from each beam mode are used to measure these parameters separately for neutrino and antineutrino oscillations. Data taken from $1.49 \times 10^{21}$ protons on target (POT) in neutrino mode and $1.64 \times 10^{21}$ POT in antineutrino mode are used. The best-fit values obtained by T2K were $\sin^2\left(\theta_{23}\right)=0.51^{+0.06}_{-0.07} \left(0.43^{+0.21}_{-0.05}\right)$ and $\Delta m^2_{32}=2.47^{+0.08}_{-0.09} \left(2.50^{+0.18}_{-0.13}\right)$\evmass for neutrinos (antineutrinos). No significant differences between the values of the parameters describing the disappearance of muon neutrinos and antineutrinos were observed. An analysis using an effective two-flavor neutrino oscillation model where the sine of the mixing angle is allowed to take non-physical values larger than 1 is also performed to check the consistency of our data with the three-flavor model. Our data were found to be consistent with a physical value for the mixing angle.

The Deep Underground Neutrino Experiment (DUNE), a 40-kton underground liquid argon time projection chamber experiment, will be sensitive to the electron-neutrino flavor component of the burst of neutrinos expected from the next Galactic core-collapse supernova. Such an observation will bring unique insight into the astrophysics of core collapse as well as into the properties of neutrinos. The general capabilities of DUNE for neutrino detection in the relevant few- to few-tens-of-MeV neutrino energy range will be described. KM3NeT is a research infrastructure being installed in the deep Mediterranean Sea. It will house a neutrino telescope comprising hundreds of networked moorings - detection units or strings equipped with optical instrumentation to detect the Cherenkov radiation generated by charged particles from neutrino-induced collisions in its vicinity. In comparison to moorings typically used for oceanography, several key features of the KM3NeT string are different: the instrumentation is contained in transparent and thus unprotected glass spheres; two thin Dyneema ropes are used as strength members; and a thin delicate backbone tube with fibre-optics and copper wires for data and power transmission, respectively, runs along the full length of the mooring. Also, compared to other neutrino telescopes such as ANTARES in the Mediterranean Sea and GVD in Lake Baikal, the KM3NeT strings are more slender to minimise the amount of material used for support of the optical sensors. Moreover, the rate of deploying a large number of strings in a period of a few years is unprecedented. For all these reasons, for the installation of the KM3NeT strings, a custom-made, fast deployment method was designed. Despite the length of several hundreds of metres, the slim design of the string allows it to be compacted into a small, re-usable spherical launching vehicle instead of deploying the mooring weight down from a surface vessel. After being lowered to the seafloor, the string unfurls to its full length with the buoyant launching vehicle rolling along the two ropes.The design of the vehicle, the loading with a string, and its underwater self-unrolling are detailed in this paper.

288. Learning Transferable Push Manipulation Skills in Novel Contexts

This paper is concerned with learning transferable forward models for push manipulation that can be applying to novel contexts and how to improve the quality of prediction when critical information is available. We propose to learn a parametric internal model for push interactions that, similar for humans, enables a robot to predict the outcome of a physical interaction even in novel contexts. Given a desired push action, humans are capable to identify where to place their finger on a new object so to produce a predictable motion of the object. We achieve the same behaviour by factorising the learning into two parts. First, we learn a set of local contact models to represent the geometrical relations between the robot pusher, the object, and the environment. Then we learn a set of parametric local motion models to predict how these contacts change throughout a push. The set of contact and motion models represent our internal model. By adjusting the shapes of the distributions over the physical parameters, we modify the internal model's response. Uniform distributions yield to coarse estimates when no information is available about the novel context (i.e. unbiased predictor). A more accurate predictor can be learned for a specific environment/object pair (e.g. low friction/high mass), i.e. biased predictor. The effectiveness of our approach is shown in a simulated environment in which a Pioneer 3-DX robot needs to predict a push outcome for a novel object, and we provide a proof of concept on a real robot. We train on 2 objects (a cube and a cylinder) for a total of 24,000 pushes in various conditions, and test on 6 objects encompassing a variety of shapes, sizes, and physical parameters for a total of 14,400 predicted push outcomes. Our results show that both biased and unbiased predictors can reliably produce predictions in line with the outcomes of a carefully tuned physics simulator., Comment: This work has been submitted to IEEE Transactions on Robotics journal in July 2020

The ProtoDUNE-SP detector is a single-phase liquid argon time projection chamber with an active volume of $7.2\times 6.0\times 6.9$ m$^3$. It is installed at the CERN Neutrino Platform in a specially-constructed beam that delivers charged pions, kaons, protons, muons and electrons with momenta in the range 0.3 GeV$/c$ to 7 GeV/$c$. Beam line instrumentation provides accurate momentum measurements and particle identification. The ProtoDUNE-SP detector is a prototype for the first far detector module of the Deep Underground Neutrino Experiment, and it incorporates full-size components as designed for that module. This paper describes the beam line, the time projection chamber, the photon detectors, the cosmic-ray tagger, the signal processing and particle reconstruction. It presents the first results on ProtoDUNE-SP's performance, including noise and gain measurements, $dE/dx$ calibration for muons, protons, pions and electrons, drift electron lifetime measurements, and photon detector noise, signal sensitivity and time resolution measurements. The measured values meet or exceed the specifications for the DUNE far detector, in several cases by large margins. ProtoDUNE-SP's successful operation starting in 2018 and its production of large samples of high-quality data demonstrate the effectiveness of the single-phase far detector design., Comment: 93 pages, 70 figures

We apply the notion of a full convex subcategory to a wide range of algebras including tilted, quasi-tilted, shod, weakly shod, left and right glued, laura, simply connected, strongly simply connected, left supported, and cluster-tilted. In particular, given an algebra $\Lambda$ from one of the aforementioned classes, we investigate certain factor algebras $\Lambda/I$ where $I$ is an ideal generated by a suitable idempotent.

The sensitivity of the Deep Underground Neutrino Experiment (DUNE) to neutrino oscillation is determined, based on a full simulation, reconstruction, and event selection of the far detector and a full simulation and parameterized analysis of the near detector. Detailed uncertainties due to the flux prediction, neutrino interaction model, and detector effects are included. DUNE will resolve the neutrino mass ordering to a precision of 5$\sigma$, for all $\delta_{\mathrm{CP}}$ values, after 2 years of running with the nominal detector design and beam configuration. It has the potential to observe charge-parity violation in the neutrino sector to a precision of 3$\sigma$ (5$\sigma$) after an exposure of 5 (10) years, for 50\% of all $\delta_{\mathrm{CP}}$ values. It will also make precise measurements of other parameters governing long-baseline neutrino oscillation, and after an exposure of 15 years will achieve a similar sensitivity to $\sin^{2} 2\theta_{13}$ to current reactor experiments., Comment: arXiv admin note: substantial text overlap with arXiv:2002.03005; Updated after referee comments

The Deep Underground Neutrino Experiment is a next-generation neutrino oscillation experiment that aims to measure $CP$-violation in the neutrino sector as part of a wider physics program. A deep learning approach based on a convolutional neural network has been developed to provide highly efficient and pure selections of electron neutrino and muon neutrino charged-current interactions. The electron neutrino (antineutrino) selection efficiency peaks at 90% (94%) and exceeds 85% (90%) for reconstructed neutrino energies between 2-5 GeV. The muon neutrino (antineutrino) event selection is found to have a maximum efficiency of 96% (97%) and exceeds 90% (95%) efficiency for reconstructed neutrino energies above 2 GeV. When considering all electron neutrino and antineutrino interactions as signal, a selection purity of 90% is achieved. These event selections are critical to maximize the sensitivity of the experiment to $CP$-violating effects., Comment: 39 pages, 11 figures

We report measurements of the flux-integrated $\bar{\nu}_\mu$ and $\bar{\nu}_\mu+\nu_\mu$ charged-current cross-sections on water and hydrocarbon targets using the T2K anti-neutrino beam, with a mean neutrino energy of 0.86 GeV. The signal is defined as the (anti-)neutrino charged-current interaction with one induced $\mu^\pm$ and no detected charged pion nor proton. These measurements are performed using a new WAGASCI module recently added to the T2K setup in combination with the INGRID Proton module. The phase space of muons is restricted to the high-detection efficiency region, $p_{\mu}>400~{\rm MeV}/c$ and $\theta_{\mu}<30^{\circ}$, in the laboratory frame. Absence of pions and protons in the detectable phase space of "$p_{\pi}>200~{\rm MeV}/c$ and $\theta_{\pi}<70^{\circ}$", and "$p_{\rm p}>600~{\rm MeV}/c$ and $\theta_{\rm p}<70^{\circ}$" is required. In this paper, both of the $\bar{\nu}_\mu$ cross-sections and $\bar{\nu}_\mu+\nu_\mu$ cross-sections on water and hydrocarbon targets, and their ratios are provided by using D'Agostini unfolding method. The results of the integrated $\bar{\nu}_\mu$ cross-section measurements over this phase space are $\sigma_{\rm H_{2}O}\,=\,(1.082\pm0.068(\rm stat.)^{+0.145}_{-0.128}(\rm syst.)) \times 10^{-39}~{\rm cm^{2}/nucleon}$, $\sigma_{\rm CH}\,=\,(1.096\pm0.054(\rm stat.)^{+0.132}_{-0.117}(\rm syst.)) \times 10^{-39}~{\rm cm^{2}/nucleon}$, and $\sigma_{\rm H_{2}O}/\sigma_{\rm CH} = 0.987\pm0.078(\rm stat.)^{+0.093}_{-0.090}(\rm syst.)$. The $\bar{\nu}_\mu+\nu_\mu$ cross-section is $\sigma_{\rm H_{2}O} = (1.155\pm0.064(\rm stat.)^{+0.148}_{-0.129}(\rm syst.)) \times 10^{-39}~{\rm cm^{2}/nucleon}$, $\sigma_{\rm CH}\,=\,(1.159\pm0.049(\rm stat.)^{+0.129}_{-0.115}(\rm syst.)) \times 10^{-39}~{\rm cm^{2}/nucleon}$, and $\sigma_{\rm H_{2}O}/\sigma_{\rm CH}\,=\,0.996\pm0.069(\rm stat.)^{+0.083}_{-0.078}(\rm syst.)$.

The KM3NeT research infrastructure is currently under construction at two locations in the Mediterranean Sea. The KM3NeT/ORCA water-Cherenkov neutrino detector off the French coast will instrument several megatons of seawater with photosensors. Its main objective is the determination of the neutrino mass ordering. This work aims at demonstrating the general applicability of deep convolutional neural networks to neutrino telescopes, using simulated datasets for the KM3NeT/ORCA detector as an example. To this end, the networks are employed to achieve reconstruction and classification tasks that constitute an alternative to the analysis pipeline presented for KM3NeT/ORCA in the KM3NeT Letter of Intent. They are used to infer event reconstruction estimates for the energy, the direction, and the interaction point of incident neutrinos. The spatial distribution of Cherenkov light generated by charged particles induced in neutrino interactions is classified as shower- or track-like, and the main background processes associated with the detection of atmospheric neutrinos are recognized. Performance comparisons to machine-learning classification and maximum-likelihood reconstruction algorithms previously developed for KM3NeT/ORCA are provided. It is shown that this application of deep convolutional neural networks to simulated datasets for a large-volume neutrino telescope yields competitive reconstruction results and performance improvements with respect to classical approaches.

This paper reports the first simultaneous measurement of the double differential muon neutrino charged-current cross section on oxygen and carbon without pions in the final state as a function of the outgoing muon kinematics, made at the ND280 off-axis near detector of the T2K experiment. The ratio of the oxygen and carbon cross sections is also provided to help validate various models' ability to extrapolate between carbon and oxygen nuclear targets, as is required in T2K oscillation analyses. The data are taken using a neutrino beam with an energy spectrum peaked at 0.6 GeV. The extracted measurement is compared with the prediction from different Monte Carlo neutrino-nucleus interaction event generators, showing particular model separation for very forward-going muons. Overall, of the models tested, the result is best described using Local Fermi Gas descriptions of the nuclear ground state with RPA suppression.

In this study, we present an innovative technique for speaker adaptation in order to improve the accuracy of segmentation with application to unit-selection Text-To-Speech (TTS) systems. Unlike conventional techniques for speaker adaptation, which attempt to improve the accuracy of the segmentation using acoustic models that are more robust in the face of the speaker's characteristics, we aim to use only context dependent characteristics extrapolated with linguistic analysis techniques. In simple terms, we use the intuitive idea that context dependent information is tightly correlated with the related acoustic waveform. We propose a statistical model, which predicts correcting values to reduce the systematic error produced by a state-of-the-art Hidden Markov Model (HMM) based speech segmentation. Our approach consists of two phases: (1) identifying context-dependent phonetic unit classes (for instance, the class which identifies vowels as being the nucleus of monosyllabic words); and (2) building a regression model that associates the mean error value made by the ASR during the segmentation of a single speaker corpus to each class. The success of the approach is evaluated by comparing the corrected boundaries of units and the state-of-the-art HHM segmentation against a reference alignment, which is supposed to be the optimal solution. In conclusion, our work supplies a first analysis of a model sensitive to speaker-dependent characteristics, robust to defective and noisy information, and a very simple implementation which could be utilized as an alternative to either more expensive speaker-adaptation systems or of numerous manual correction sessions., Comment: isbn:978-88-7870-619-4

The electron (anti-)neutrino component of the T2K neutrino beam constitutes the largest background in the measurement of electron (anti-)neutrino appearance at the far detector. The electron neutrino scattering is measured directly with the T2K off-axis near detector, ND280. The selection of the electron (anti-)neutrino events in the plastic scintillator target from both neutrino and anti-neutrino mode beams is discussed in this paper. The flux integrated single differential charged-current inclusive electron (anti-)neutrino cross-sections, $d\sigma/dp$ and $d\sigma/d\cos(\theta)$, and the total cross-sections in a limited phase-space in momentum and scattering angle ($p > 300$ MeV/c and $\theta \leq 45^{\circ}$) are measured using a binned maximum likelihood fit and compared to the neutrino Monte Carlo generator predictions, resulting in good agreement.
