Bioassay-guided isolation of spiroaspertrione A from cultures of Aspergillus sp. TJ23 in 2017 demonstrated potent resensitization of oxacillin against methicillin-resistant Staphylococcus aureus by lowering the oxacillin minimal inhibitory concentration up to 32-fold. Abstract. Background:. Innovative coronavirus disease 2019 (COVID-19) vaccines, with elevated global manufacturing capacity, enhanced safety and efficacy, simplified dosing regimens, and distribution that is less cold chain-dependent, are still global imperatives for tackling the ongoing pandemic. A previous phase I trial indicated that the recombinant COVID-19 vaccine (V-01), which contains a fusion protein (IFN-PADRE-RBD-Fc dimer) as its antigen, is safe and well tolerated, capable of inducing rapid and robust immune responses, and warranted further testing in additional clinical trials. Herein, we aimed to assess the immunogenicity and safety of V-01, providing rationales of appropriate dose regimen for further efficacy study. Methods:. A randomized, double-blind, placebo-controlled phase II clinical trial was initiated at the Gaozhou Municipal Centre for Disease Control and Prevention (Guangdong, China) in March 2021. Both younger (n = 440; 18–59 years of age) and older (n = 440; ≥60 years of age) adult participants in this trial were sequentially recruited into two distinct groups: two-dose regimen group in which participants were randomized either to follow a 10 or 25 μg of V-01 or placebo given intramuscularly 21 days apart (allocation ratio, 3:3:1, n = 120, 120, 40 for each regimen, respectively), or one-dose regimen groups in which participants were randomized either to receive a single injection of 50 μg of V-01 or placebo (allocation ratio, 3:1, n = 120, 40, respectively). The primary immunogenicity endpoints were the geometric mean titers of neutralizing antibodies against live severe acute respiratory syndrome coronavirus 2, and specific binding antibodies to the receptor binding domain (RBD). The primary safety endpoint evaluation was the frequencies and percentages of overall adverse events (AEs) within 30 days after full immunization. Results:. V-01 provoked substantial immune responses in the two-dose group, achieving encouragingly high titers of neutralizing antibody and anti-RBD immunoglobulin, which peaked at day 35 (161.9 [95% confidence interval [CI]: 133.3–196.7] and 149.3 [95%CI: 123.9–179.9] in 10 and 25 μg V-01 group of younger adults, respectively; 111.6 [95%CI: 89.6–139.1] and 111.1 [95%CI: 89.2–138.4] in 10 and 25 μg V-01 group of older adults, respectively), and remained high at day 49 after a day-21 second dose; these levels significantly exceed those in convalescent serum from symptomatic COVID-19 patients (53.6, 95%CI: 31.3–91.7). Our preliminary data show that V-01 is safe and well tolerated, with reactogenicity predominantly being absent or mild in severity and only one vaccine-related grade 3 or worse AE being observed within 30 days. The older adult participants demonstrated a more favorable safety profile compared with those in the younger adult group: with AEs percentages of 19.2%, 25.8%, 17.5% in older adults vs. 34.2%, 23.3%, 26.7% in younger adults at the 10, 25 μg V-01 two-dose group, and 50 μg V-01 one-dose group, respectively. Conclusions:. The vaccine candidate V-01 appears to be safe and immunogenic. The preliminary findings support the advancement of the two-dose, 10 μg V-01 regimen to a phase III trial for a large-scale population-based evaluation of safety and efficacy. Abstract Addressing microstructure-property relations of polymer nanocomposites is vital for designing advanced dielectrics for electrostatic energy storage. Here, we develop an integrated phase-field model to simulate the dielectric response, charge transport, and breakdown process of polymer nanocomposites. Subsequently, based on 6615 high-throughput calculation results, a machine learning strategy is schemed to evaluate the capability of energy storage. We find that parallel perovskite nanosheets prefer to block and then drive charges to migrate along with the interfaces in x-y plane, which could significantly improve the breakdown strength of polymer nanocomposites. To verify our predictions, we fabricate a polymer nanocomposite P(VDF-HFP)/Ca2Nb3O10, whose highest discharged energy density almost doubles to 35.9 J cm−3 compared with the pristine polymer, mainly benefit from the improved breakdown strength of 853 MV m−1. This work opens a horizon to exploit the great potential of 2D perovskite nanosheets for a wide range of applications of flexible dielectrics with the requirement of high voltage endurance.

Abstract As a cathode for sodium‐ion batteries (SIBs), Na3V2(PO4)2F3 (NVPF) with 3D open framework is a promising candidate due to its high working voltage and large theoretical capacity. However, the severe capacity degradation and poor rate capability hinder its practical applications. The present study demonstrated the optimization of Na‐storage performance of NVPF via delicate lattice modulation. Aliovalent substitution of V3+ at Na+ in NVPF induces the generation of electronic defects and expansion of Na+‐migration channels, resulting in the enhancement in electronic conductivity and acceleration of Na+‐migration kinetics. It is disclosed that the formed stronger NaO bonds with high ionicity than VO bonds lead to the significant increase in structural stability and ionicity in the Na+‐substituted NVPF (NVPF‐Nax). The aforementioned effects of Na+ substitution achieve the unprecedented electrochemical performance in the optimized Na3.14V1.93Na0.07(PO4)2F3 (NVPF‐Na0.07). As a result, NVPF‐Na0.07 delivers a high‐rate capability (77.5 mAh g−1 at 20 C) and ultralong cycle life (only 0.027% capacity decay per cycle over 1000 cycles at 10 C). Sodium‐ion full cells are designed using NVPF‐Na0.07 as cathode and Se@reduced graphene oxide as anode. The full cells exhibit excellent wide‐temperature electrochemical performance from −25 to 25°C with an outstanding rate capability (96.3 mAh g−1 at 20 C). Furthermore, it delivered an excellent cycling performance over 300 cycles with a capacity retention exceeding 90% at 0.5 C under different temperatures. This study demonstrates a feasible strategy for the development of advanced cathode materials with excellent electrochemical properties to achieve high‐efficiency energy storage.

Abstract Background The molecular pathogenesis of endometrial cancer is not completely understood. CypB upregulated in many cancers, however, its role in endometrial carcinoma has not been studied. Here, we determine the effect of CypB on the growth of endometrial cancer. Methods In this study, we examined the expression of CypB in endometrial cancer tissues using immunohistochemistry. CypB silenced in HEC-1-B cell line by shRNA. CCK-8, colony formation assays, wound healing assays, and transwell analysis were performed to assess its effect on tumor cell proliferation and metastasis. Furthermore, microarray analysis was carried out to compare the global mRNA expression profile between the HEC-1-B and CypB-silenced HEC-1-B cells. Gene ontology and KEGG pathway enrichment analysis were performed to determine the potential function of differentially expressed genes related to CypB. Results We found that CypB was upregulated in endometrial cancer, inhibit CypB expression could significantly suppress cell proliferation, metastasis, and migration. Regulation of gene expression by DNA-binding transcription factors is essential for proper control of growth and development in all organisms. In this study, we annotate and characterize growth and developmental phenotypes for transcription factor genes in the model filamentous fungus Neurospora crassa We identified 312 transcription factor genes, corresponding to 3.2% of the protein coding genes in the genome. The largest class was the fungal-specific Zn2Cys6 (C6) binuclear cluster, with 135 members, followed by the highly conserved C2H2 zinc finger group, with 61 genes. Viable knockout mutants were produced for 273 genes, and complete growth and developmental phenotypic data are available for 242 strains, with 64% possessing at least one defect. The most prominent defect observed was in growth of basal hyphae (43% of mutants analyzed), followed by asexual sporulation (38%), and the various stages of sexual development (19%). Two growth or developmental defects were observed for 21% of the mutants, while 8% were defective in all three major phenotypes tested. Analysis of available mRNA expression data for a time course of sexual development revealed mutants with sexual phenotypes that correlate with transcription factor transcript abundance in wild type. Inspection of this data also implicated cryptic roles in sexual development for several cotranscribed transcription factor genes that do not produce a phenotype when mutated.

Enabling motor control by epidural electrical stimulation of the spinal cord is a promising therapeutic technique for the recovery of motor function after a spinal cord injury (SCI). Although epidural electrical stimulation has resulted in improvement in hindlimb motor function, it is unknown whether it has any therapeutic benefit for improving forelimb fine motor function after a cervical SCI. We tested whether trains of pulses delivered at spinal cord segments C6 and C8 would facilitate the recovery of forelimb fine motor control after a cervical SCI in rats. Rats were trained to reach and grasp sugar pellets. Immediately after a dorsal funiculus crush at C4, the rats showed significant deficits in forelimb fine motor control. The rats were tested to reach and grasp with and without cervical epidural stimulation for 10weeks post-injury. To determine the best stimulation parameters to activate the cervical spinal networks involved in forelimb motor function, monopolar and bipolar currents were delivered at varying frequencies (20, 40, and 60Hz) concomitant with the reaching and grasping task. We found that cervical epidural stimulation increased reaching and grasping success rates compared to the no stimulation condition. Bipolar stimulation (C6- C8+ and C6+ C8-) produced the largest spinal motor-evoked potentials (sMEPs) and resulted in higher reaching and grasping success rates compared with monopolar stimulation (C6- Ref+ and C8- Ref+). Forelimb performance was similar when tested at stimulation frequencies of 20, 40, and 60Hz. We also found that the EMG activity in most forelimb muscles as well as the co-activation between flexor and extensor muscles increased post-injury. With epidural stimulation, however, this trend was reversed indicating that cervical epidural spinal cord stimulation has therapeutic potential for rehabilitation after a cervical SCI.

Background: Stanniocalcin-2 (STC2) is a secreted glycoprotein which plays an important role in regulating the homeostasis of calcium, glucose homeostasis, and phosphorus metastasis. Accumulating evidence suggests that STC2 is implicated in cancer mechanisms. However, the effects of STC2 on cancer development and progression across pan-cancer are not yet completely known.Methods: Data were downloaded from The Cancer Genome Atlas database to obtain differentially expressed genes significantly associated with prognosis (key genes). A gene was selected for subsequent correlation studies by integrating the significance of prognosis and the time-dependent ROC curve. Gene expression of different tumor types was analyzed based on the UCSC XENA website. Furthermore, our study investigated the correlation of STC2 expression between prognosis, immune cell infiltration, immune checkpoint genes (ICGs), mismatch repair genes (MMRs), tumor mutation burden (TMB), microsatellite instability (MSI), and drug sensitivity in various malignant tumors. Gene set enrichment analysis (GSEA) was conducted for correlated genes of STC2 to explore potential mechanisms.Results: A total of 3,429 differentially expressed genes and 397 prognosis-related genes were identified from the TCGA database. Twenty-six key genes were found by crossing the former and the latter, and the highest risk gene, STC2, was selected for subsequent correlation studies. STC2 had good diagnostic performance for HNSCC, and was closely related to the survival status and clinicopathological stage of HNSCC patients. In pan-cancer analysis, STC2 was upregulated in 20 cancers and downregulated in seven cancers. STC2 overexpression was overall negatively correlated with overall survival, disease-free survival, disease-specific survival, and progress-free survival. STC2 was profoundly correlated with the tumor immune microenvironment, including immune cell infiltration, ICGs, MMRs, TMB, and MSI. Moreover, STC2 was significantly negatively correlated with the sensitivity or resistance of multiple drugs.Conclusion: STC2 was a potential prognostic biomarker for pan-cancer and a new immunotherapy target.

The current classification and evaluation methods of volcanic rock reservoirs have low accuracy and cannot effectively provide guidance for the selection of volcanic rock gas reservoirs, which have efficient properties for gas production. In this research, we have analyzed the lithology, lithofacies, reservoir space type, pore combination mode, and reservoir microscopic characteristics of volcanic reservoirs using the energy storage coefficient as a constraint. Then, the method of reservoir classification was proposed. The results showed the following: 1) The energy storage coefficient can better characterize the single-layer productivity of gas wells. The volcanic rock reservoirs in the Wangfu gas field can be subdivided into three categories by considering the energy storage coefficient. 2) Type I reservoirs mainly develop structural fractures–matrix dissolution pores, structural fractures–intercrystalline micropores, and matrix dissolution pores. Type II reservoirs mainly develop matrix dissolution pores and residual intergranular pores, and Type III reservoirs are dominated by structural fractures. From Type I to Type III reservoirs, the skewness of the mercury intrusion curve and the sorting coefficient deteriorated, and the physical properties of the reservoir and the maximum mercury saturation and other parameters also decreased, whereas the displacement pressure and the median saturation pressure increased. 3) The characteristics of conventional and special logging curves of typical reservoirs were comprehensively analyzed, and the combination of sensitivity parameters reflecting gas-bearing properties and logging curves was optimized. Furthermore, a reservoir classification chart was established, and the results enabled to confirm the choice of the reservoir and demonstrated that the standard classification has high accuracy. 4) The diagenesis processes such as weathering, leaching, and dissolution improved the physical properties of the reservoir. The research results can effectively provide guidance for the evaluation of the “sweet spot” of volcanic rock reservoirs.

Good’s syndrome (GS) is an immunodeficiency disease, causing thymoma, low or absent B-cells, hypogammaglobulinemia, and defects in cell-mediated immunity. The most common clinical presentation is recurrent infection, followed by refractory diarrhea, due to the immunodeficiency. However, there are only few reports on intestinal endoscopy and pathology.

The coordination complex bis(1H-benzimidazole-κN3)cadmium(II) dibenzoate has been synthesized and characterized by single crystal diffraction analysis. Cadmium center is six coordinated and formed a distorted octahedron coordinated geometry. The Hirshfeld analysis shows that in the dnorm-surface of the compound, there are dark red spots near the hydrogen-bonds acceptor and donor atoms, while intermolecular interactions result in faint-red spots. The AIM analysis was performed, there exist a BCP in each N(C)–H∙∙∙O hydrogen bond, the bond paths also can be seen, the |V(b)|/G(b) < 1 and the H(b) > 0, the interaction is indicative of being a closed shell. Polymer blends are regarded as a straightforward and effective method to enhance the energy storage performance of dielectric film capacitors. However, how the components and structures within the blend systems affect the energy density and efficiency remains insufficiently explored in-depth. In this discourse, employing a polymer blend of ferroelectric and linear dielectric phases as a paradigm, we perform phase-field simulations to elucidate the effects of ferroelectric phase volume fractions, geometrical dimensions, and the dielectric constant of the linear phase on the energy storage capabilities. Concurrently, we have devised six divergent blending microstructures to probe the ramifications of structural variances on the overarching performance metrics. We also analyze the domain configurations and switching dynamics under varying electric fields to understand the performance variations and delineate the determinants conducive to superior energy density and efficiency. This paper theoretically establishes the component–content–structure–performance relationships of different polymer blend systems, which is expected to better guide the innovative design of new polymer blend dielectrics.
