252. Self-propelling and rolling of a sessile-motile aggregate of the bacterium Caulobacter crescentus.

Abstract

Active dispersal of microorganisms is often attributed to the cells' motile organelles. However, much less is known about whether sessile cells can access such motility through aggregation with motile counterparts. Here, we show that the rosette aggregates of the bacterium Caulobacter crescentus, although predominantly sessile, can actively disperse through the flagellar motors of motile members. Comparisons in kinematics between the motile rosettes and solitary swimming cells indicate that the rosettes can be powered by as few as a single motor. We further reconstructed the 3D movements of the rosettes to reveal that their proximity to a solid-liquid interface promotes a wheel-like rolling, as powered by the flagellar torque. This rolling movement also features a sequence of sharp turns, a reorientation mechanism distinct from that of swimming cells. Overall, our study elucidates an unexplored regime of aggregation-based motility that can be widely applied to sessile-motile composites.

253. A Tale of Winglets: Evolution of Flight Morphology in Stick Insects

Abstract

The evolutionary transition between winglessness and a full-winged morphology requires selective advantage for intermediate forms. Conversely, repeated secondary wing reductions among the pterygotes indicates relaxation of such selection. However, evolutionary trajectories of such transitions are not well-characterized. The stick insects (Phasmatodea) exhibit diverse wing sizes at both interspecific and intersexual levels, and thus provide a system for examining how selection on flight capability, along with other selective forces, drives the evolution of flight-related morphology. Here, we examine variation in relevant morphology for stick insects using data from 1,100+ individuals representing 765 species. Although wing size varies along a continuous spectrum, taxa with either long or miniaturized wings are the most common, whereas those with intermediate-sized wings are relatively rare. In a morphological space defined by wing and body size, the aerodynamically relevant parameter termed wing loading (the average pressure exerted on the air by the wings) varies according to sex-specific scaling laws; volant but also flightless forms are the most common outcomes in both sexes. Using phylogenetically-informed analyses, we show that relative wing size and body size are inversely correlated in long-winged insects regardless of sexual differences in morphology and ecology. These results demonstrate the diversity of flight-related morphology in stick insects, and also provide a general framework for addressing evolutionary coupling between wing and body dimensions. We also find indirect evidence for a “fitness valley” associated with intermediate-sized wings, suggesting relatively rapid evolutionary transitions between wingless and volant forms.

Abstract

Abstract Genome editing is an advanced genetic modification tool that facilitates the removal, insertion, or substitution of nucleotides in the genome of an organism for mutation induction. CRISPR/Cas9 (clustered regularly interspaced short palindromic repeats/CRISPR‐associated protein 9) is the most widely used genome editing tool that can precisely modify the specific sequence of a genome. Significant progress has been made in the use of CRISPR/Cas9 for the genetic improvement of food crops. However, in forage crops, the use of CRISPR/Cas9 is still in the infant stage with limited understanding and application. This article reviews current advances in the establishment and application of the CRISPR/Cas9 system in alfalfa, summarizes the application of CRISPR/Cas9 in several other forage legume and grass species, and discusses the prospects of CRISPR/Cas9 in the genetic improvement of forage crops.
