Background Affinity peptides, as a core part of affinity chromatography, play an important role in the purification of target molecules. Methods Here we describe the use of molecular docking technology for virtual screening of affinity peptides that specifically recognize the PCV2 Cap protein for the first time. Thirteen candidate peptides with high scores were obtained and then further characterized. Experimentally, the affinity and sensitivity of the peptides studied were identified by ELISA and LSPR, respectively. In order to investigate the purification effect of a selected peptide (L11) for the recombinant PCV2 Cap protein, it was coupled to NHS agarose magnetic beads as an affinity adsorbent (NaMB-L11); and the ligand density of the affinity adsorbent and pH value in the purification of the recombinant PCV2 Cap protein were optimized. Results Our data showed that the peptide L11- DYWWQSWE has the smallest KD = 103 nM with higher specificity for PCV2 Cap protein recognition. The NaMB-L11 affinity adsorbent yielded a purified Cap sample with 98% purity at 90% recovery in a single step. Conclusion Based on the structure, we obtained a high affinity peptide L11 binding to the PCV2 Cap protein by molecular docking technology. It not only provides a theoretical basis for the design of PCV2 Cap affinity peptide, but a new method for the purification of the PCV2 Cap protein.

Abstract Canine parvovirus (CPV) has been considered to be an important pathogen, which can cause acute infectious disease in canids. Although current vaccines are effective in preventing CPV infection, safety problems still remain unsolved. In this study, a subunit vaccine against CPV based on virus-like particles (VLPs) with good safety and immunogenicity is reported. Soluble CPV VP2 protein was produced by co-expression of chaperone trigger factor (Tf16) in Escherichia coli (E.coli), and assembled into CPV VLPs which could be affected by NaCl and pH. At 250 mM NaCl pH 8.0, the VLPs co-expressed with Tf16 had similar size (25 nm) and shape with the authentic virus capsid under the transmission electron microscopy (TEM), which is also in accordance with the dynamic light scattering (DLS) data. Immunization with these particles could induce high-titer hemagglutination inhibition (1:12288) and neutralizing antibodies (1:6144) in guinea pigs. Splenic cells of them could secrete IFN-γ and IL-4 after stimulation by CPV. Thus, the VLPs produced by the new approach with high yield and immunogenicity could be a potential candidate for CPV vaccine.

Cracking of electrodes caused by large volume change and the associated lithium diffusion-induced stress during electrochemical cycling is one of the main reasons for the short cycle life of lithium-ion batteries using high capacity anode materials, such as Si and Sn. In this work, we study the fracture behavior and cracking patterns in amorphous Si thin film electrodes as a result of electrochemical cycling. A modified spring-block model is shown to capture the essential features of cracking patterns of electrode materials, including self-similarity. It is shown that cracks are straight in thick films, but show more wiggles in thin films. As the thickness of film decreases, the average size of islands separated by cracks decreases. A critical thickness bellow which material would not crack is found for amorphous Si films. The experimental and simulation results of this work provide guidelines for designing crack free thin-film lithium ion battery electrodes during cycling by patterning the electrode and reducing the film thickness. A novel ligand tuning strategy for the synthesis and assembly of ZnTe nanocrystals is proposed in this paper: a specific ligand is selected to work with the reaction system to regulate (passivate or activate) the reactivity of zinc precursors, as well as the growth and the assembly of resulting nanocrystals in a coordinate way. By utilization of this strategy, high-quality ZnTe nanodots, branched-nanorods (including nanotetrapods), nanowires and microspheres are obtained. Furthermore, by using ZnTe microspheres as building blocks, ordered two-dimensional (2D) and three-dimensional (3D) arrays and well-defined hollow microspheres are fabricated. The size, morphology, and crystal structure of as-prepared ZnTe nanocrystals are well characterized. The underlying mechanisms for ligand-tuned synthesis and assembly of ZnTe nanocrystals are also intensively discussed. Finally, the shape-dependent optical, structural, and electrochemical properties of those ZnTe nanocrystals are systemically investigated; their band edge positions are studied by cyclic voltammetry. In this paper, we report a novel and cost-effective fabrication technique to produce electrode arrays that can be used for monitoring and electrical manipulaton of the molecular orientation of DNA self-assembled monolayers (SAMs) on gold. The electrode arrays were prepared from gold coated glass sides or compact discs (CD-Rs) by using standard office inkjet printers without any hardware or software modifications. In this method, electrode arrays of varied shape and size (from submillimeter to centimeter) can be rapidly fabricated and are suitable for standard electrochemical measurements. We were able to use a dual-channel potentiostat to control the electrodes individually and a fluorescence (FL) scanner to image the electrode array simultaneously. With such an integrated modulation setup, the structural switching behavior (from "lying" to "standing" position) and the enhanced hybridization reactivity of thiolate DNA SAMs on gold under potential control have been successfully demonstrated. A mild and efficient surface activation protocol to convert polycarbonate (PC) substrates, e.g., plastic bases of compact disks, to biochip platforms for DNA probe immobilization and target detection is described. The preparation procedure (activation, patterning, and coupling) is simple and effective; the on-chip hybridization is sensitive and selective. Particularly, UV/ozone treatment of PC sheets produces a hydrophilic surface with a high density of reactive carboxylic acid groups [(4.8 ± 0.2) × 10-10 mol/cm²] in less than 10 mm at ambient conditions, and no significant aging or physical damage to the substrate is observed. Covalent immobilization of DNA probes via both passive (reagent-less photopatterning and coupling in bulk solution phase) and flow-through (creation of microarrays with microfluidic channel plates) procedures has been demonstrated. Subsequent hybridization shows uniform and strong fluorescent signals for complementary target DNA and allows clear discrimination between fully complementary targets and strands with a single base-pair mismatch. The surface chemistry described herein will facilitate the development of disposable plastic biochips (not limited to DNA microarrays) and the fabrication of biomedical devices that are readable with conventional optical drives. A series of ternary tetrapodal nanocrystals of CdSexTe1?xwith x= 0?(CdTe), 0.23, 0.53, 0.78, 1 (CdSe) were synthesized and used to fabricate hybrid nanocrystal/polymer solar cells. Herein, the nanocrystals acted as electron acceptors, and poly(2-methoxy-5-(2'-ethyl-hexyloxy)-1,4-phenylene vinylene) (MEH-PPV) was used as an electron donor. It was found that the open circuit voltage (Voc), short-circuit current (Jsc) and power conversion efficiency (?) of the devices all increased with increasing Se content in the CdSexTe1?xnanocrystals under identical experimental conditions. The solar cell based on the blend of tetrapodal CdSe nanocrystals and MEH-PPV (9:1?w/w) showed the highest power conversion efficiency of 1.13% under AM 1.5, 80?mW?cm?2, and the maximum incident photon to converted current efficiency (IPCE) of the device reached 47% at 510?nm. The influence of nanocrystal composition on the photovoltaic properties of the hybrid solar cells was explained by the difference of the band level positions between MEH-PPV and the nanocrystals. Classical swine fever (CSF), caused by CSF virus (CSFV), is one of the most devastating viral epizootic diseases of swine in many countries. To control the disease, highly efficacious and safe live attenuated vaccines have been used for decades. However, the main drawback of these conventional vaccines is the lack of differentiability of infected from vaccinated animals (DIVA concept). Advances in biotechnology and our detailed knowledge of multiple basic science disciplines have facilitated the development of effective and safer DIVA vaccines to control CSF. To date, two types of DIVA vaccines have been developed commercially, including the subunit vaccines based on CSFV envelope glycoprotein E2 and chimeric pestivirus vaccines based on infectious cDNA clones of CSFV or bovine viral diarrhea virus (BVDV). Although inoculation of these vaccines successfully induces solid immunity against CSFV, none of them could ideally meet all demands regarding to safety, efficacy, DIVA potential, and marketability. Due to the limitations of the available choices, researchers are still striving towards the development of more advanced DIVA vaccines against CSF. This review summarizes the present status of candidate CSFV vaccines that have been developed. The strategies and approaches revealed here may also be helpful for the development of new-generation vaccines against other diseases.

Quantum resource theories have been widely studied to systematically characterize the nonclassicality of quantum systems. Most resource theories focus on quantum states and study their interconversions. Although quantum channels are generally used as a tool for state manipulation, such a manipulation capability can be naturally regarded as a generalized quantum resource, leading to an open research direction in the resource theories of quantum channels. Various resource-theoretic properties of the channels have been investigated, however, without treating the channels themselves as operational resources that can also be manipulated and converted. In this Rapid Communication, we address this problem by first proposing a general resource framework for quantum channels and introducing resource monotones based on general distance quantifiers of the channels. We study the interplay between the channel and state resource theories by relating the resource monotones of a quantum channel to its manipulation power of the state resource. Regarding channels as operational resources, we introduce asymptotic channel distillation and dilution, the most important tasks in an operational resource theory, and show how to bound the conversion rates with the channel resource monotones. Finally, we apply our results to quantum coherence as an example and introduce the coherence of channels, which characterizes the coherence generation ability of channels. We consider asymptotic channel distillation and dilution with maximally incoherent operations and find the theory asymptotically irreversible, in contrast to the asymptotic reversibility of the coherence of states.

Foot-and-mouth disease (FMD) is a highly contagious disease of cloven-hoofed animals that has caused tremendous economic losses worldwide. In this study, we designed a chimeric nanoparticles (CNPs) vaccine that displays the predominant epitope of the serotype O foot-and-mouth disease virus (FMDV) VP1 131-160 on the surface of MS2 phage. The recombinant protein was expressed in Escherichia Coli and can self-assemble into CNPs with diameter at 25–30 nm in vitro. A tandem repeat peptide epitopes (TRE) was prepared as control. Mice were immunized with CNPs, TRE and commercialized synthetic peptide vaccines (PepVac), respectively. The ELISA results showed that CNPs stimulated a little higher specific antibody levels to PepVac, but was significantly higher than the TRE groups. Moreover, the results from specific IFN-γ responses and lymphocyte proliferation test indicated that CNP immunized mice exhibited significantly enhanced cellular immune response compared to TRE. These results suggested that the CNPs constructed in current study could be a potential alternative vaccine in future FMDV control.

Pharmacogenomics is the study of the impact of genetic variations or genotypes of individuals on their drug response or drug metabolism. Compared to traditional genomics research, pharmacogenomic research is more closely related to clinical practice. Pharmacogenomic discoveries may effectively assist clinicians and healthcare providers in determining the right drugs and proper dose for each patient, which can help avoid side effects or adverse reactions, and improve the drug therapy. Currently, pharmacogenomic approaches have proven their utility when it comes to the use of cardiovascular drugs, antineoplastic drugs, aromatase inhibitors, and agents used for infectious diseases. The rapid innovation in sequencing technology and genome-wide association studies has led to the development of numerous data resources and dramatically changed the landscape of pharmacogenomic research. Porcine reproductive and respiratory syndrome (PRRS) is identified as one of the most important etiological agents in multifactorial respiratory disease of swine and can predispose pigs to secondary infections by other pathogens, usually bacteria. To understand the mechanism for an increased susceptibility to secondary bacterial infections, we investigated the antibody-dependent phagocytosis behaviour and killing ability of PMNs after infection by PRRSV strains BJ-4 or HN07-1. PMN's antibody-dependent phagocytosis and their ability to kill E.coli were both noticeably decreased following PRRSV infection, in particular with the highly pathogenic strain HN07-1. As the change in this function of the PMNs may reflect a variation in the expression of FcγRs, the expression profiles of the activating and the inhibitory FcγRs were examined. We found that RNA expression of the inhibitory receptor FcγRIIB was up-regulated post-infection, and this was greater after infection with the more virulent PRRSV strain HN07-1. The activating receptor FcγRIIIA RNA expression was on the other hand inhibited to the same extent by both PRRSV strains. Neutralizing antibody titers post-infection by PRRSV strains BJ-4 or HN07-1 were also detected. All of the pigs in infection groups showed viraemia by the end of the study (56 DPI). These observations may help to understand the mechanism of increased susceptibility to secondary bacterial infections following PRRSV infection.

A facile route for synthesizing CuInSe2nanocrystals was developed using an alkanethiol as ligand and noncoordinating octadecene as solvent. The CuInSe2nanoparticles and nanoplates were obtained, depending on the reaction temperature and time. When the reaction temperature was set at 180?°C and reaction for an hour, CuInSe2nanoparticles with an average size of 6.2 nm were produced. As the reaction time was prolonged to 3 h, nanoplates appeared inside the nanoparticles. When the reaction temperature was elevated to 210?°C and reaction for 3 h, triangular or hexagonal CuInSe2nanoplates with sharp edges were synthesized. It was proposed that the Ostwald ripening induced the morphology evolution of the CuInSe2nanoparticles into nanoplates at the higher temperature. The nanoparticles and nanoplates were characterized by transmission electron microscopy (TEM), selected area electron diffraction (SAED), x-ray diffraction (XRD) and x-ray photoelectron spectroscopy (XPS). The UV-vis absorption spectra of the CuInSe2nanoparticles were significantly blue-shifted in comparison with the bulk material due to the quantum confinement. [ABSTRACT FROM AUTHOR]
