To explore the benefit of energy storage for countering high-level wind power fluctuations, a two-stage distributionally robust optimization model is proposed for wind farms and storage units (SUs) jointly operated power systems. First, the 1-norm and $\infty $ -norm confidence sets are presented to model the fluctuations of wind power output, then a two-stage distributionally optimization model is formed to minimize system total cost with secure operation constraints, where the ON-OFF status of generators and SUs are determined in the first stage by day-ahead dispatching, while the power output of generators, wind power curtailment, load shedding and SUs charging/discharging power are optimized in the second stage. Afterward, the column-and-constraint generation (CCG) algorithm is presented to solve the proposed two-stage model. Finally the influence of confidence level of confidence sets and SU capacities on system total cost is analyzed, and the effectiveness of the proposed model is also validated by the case studies.

The ban on bisphenol A (BPA) has led to a rapid increase in the use of BPA analogs, and they are increasingly being detected in the natural environment and biological organisms. Studies have pointed out that BPA analogs can lead to adverse health outcomes. However, their interference with ovarian tissue has not been fully elucidated. In this study, 7- to 8-week-old CD-1 mice were exposed to corn oil containing 300 μg/kg/day bisphenol B (BPB) or bisphenol AF (BPAF) through oral gavage, and ovarian tissues were collected at 14 and 28 days of exposure. Ovarian toxicity was evaluated by the ovarian index, ovarian area, and follicle number. mRNA-seq was used to identify differentially expressed genes (DEGs) and infer the association of DEGs with ovarian diseases. BPB or BPAF exposure induced morphological changes in ovarian tissue in CD-1 mice. In addition, Gene Ontology (GO) analysis revealed disturbances in biological processes associated with steroid biosynthetic process (GO:0006694) and cellular calcium ion homeostasis (GO:0006874). Subsequently, regulatory networks of contaminants-DEGs-ovarian diseases were constructed. Importantly, the expression levels of DEGs and transcription factors (TFs) associated with ovarian disease were altered. BPB or BPAF exposure causes damage to ovarian morphology through the synergistic effects of multiple biological processes and may be associated with altered mRNA expression profiles as a risk factor for ovarian diseases.

With the rapid development of hydropower facility construction, the total dissolved gas (TDG) generated by dam discharge is seriously threatening the survival of fish and has become an ecological environmental issue of global concern. However, how TDG affects fish physiology and the underlying molecular mechanism remain poorly known. In this study, Acipenser dabryanus, an ancient living fossil that is a flagship species of the Yangtze River, was exposed to water supersaturated with TDG at a level of 116% for 48 h. A comprehensive analysis was performed to study the effect of TDG supersaturation stress on A. dabryanus, including histopathological, biochemical, transcriptomic and metabolomic analyses. The histopathological results showed that mucosal-associated lymphoid tissues were seriously damaged after TDG supersaturation stress. Plasma catalase levels increased significantly under TDG supersaturation stress, while superoxide dismutase levels decreased significantly. Transcriptomic analysis revealed 289 upregulated genes and 162 downregulated genes in gill tissue and 535 upregulated and 104 downregulated genes in liver tissue. Metabolomic analysis revealed 63 and 164 differentially abundant metabolites between the control group and TDG group in gill and liver, respectively. The majority of heat shock proteins and genes related to ubiquitin and various immune-related pathways were significantly upregulated by TDG supersaturation stress. Integrated transcriptomic and metabolomic analyses revealed the upregulation of amino acid metabolism and glycometabolism pathways under TDG supersaturation stress. Glycerophospholipid metabolism was increased which might be associated with maintaining cell membrane integrity. This is the first study revealing the underlying molecular mechanisms of effects of TDG supersaturation on fish. Our results suggested that acute TDG supersaturation stress could enhance immune and antioxidative functions and activate energy metabolic pathways as an adaptive mechanism in A. dabryanus.

The aim of this study was to explore the effects and mechanisms of different starvation treatments on the compensatory growth of Acipenser dabryanus. A total of 120 fish (60·532 (sem 0·284) g) were randomly assigned to four groups (fasting 0, 3, 7 or 14 d and then refed for 14 d). During fasting, middle body weight decreased significantly with prolonged starvation. The whole-body and muscle composition, serum biochemical indexes, visceral indexes and digestive enzyme activities had been effected with varying degrees of changes. The growth hormone (GH) level in serum was significantly increased in 14D; however, insulin-like growth factor-1 (IGF-1) showed the opposite trend. The neuropeptide Y (npy) mRNA level in brain was significantly improved in 7D; peptide YY (pyy) mRNA level in intestine was significantly decreased during fasting. After refeeding, the final body weight, percentage weight gain, specific growth rate, feed intake, feed efficiency and protein efficiency ratio showed no difference between 0D and 3D. The changes of whole-body and muscle composition, serum biochemical indexes, visceral indexes and digestive enzyme activities had taken place in varying degrees. GH levels in 3D and 7D were significantly higher than those in the 0D; the IGF-1 content decreased significantly during refeeding. There was no significant difference in npy and pyy mRNA levels. These results indicated that short-term fasting followed by refeeding resulted in full compensation and the physiological and biochemical effects on A. dabryanus were the lowest after 3 d of starvation and 14 d of refeeding. Additionally, compensation in A. dabryanus may be mediated by appetite genes and GH, and the degree of compensation is also affected by the duration of starvation.

Atherosclerosis is characterized by the pathological accumulation of cholesterol-laden macrophages in the arterial wall. Atherosclerosis is also the main underlying cause of CVDs, and its development is largely driven by elevated plasma cholesterol. Strong epidemiological data find an inverse association between plasma β-carotene with atherosclerosis, and we recently showed that β-carotene oxygenase 1 (BCO1) activity, responsible for β-carotene cleavage to vitamin A, is associated with reduced plasma cholesterol in humans and mice. In this study, we explore whether intact β-carotene or vitamin A affects atherosclerosis progression in the atheroprone LDLR-deficient mice. Compared with control-fed Ldlr(−/−) mice, β-carotene-supplemented mice showed reduced atherosclerotic lesion size at the level of the aortic root and reduced plasma cholesterol levels. These changes were absent in Ldlr(−/−)/Bco1(−/−) mice despite accumulating β-carotene in plasma and atherosclerotic lesions. We discarded the implication of myeloid BCO1 in the development of atherosclerosis by performing bone marrow transplant experiments. Lipid production assays found that retinoic acid, the active form of vitamin A, reduced the secretion of newly synthetized triglyceride and cholesteryl ester in cell culture and mice. Overall, our findings provide insights into the role of BCO1 activity and vitamin A in atherosclerosis progression through the regulation of hepatic lipid metabolism.

Most variants in most genes across most organisms have an unknown impact on the function of the corresponding gene. This gap in knowledge is especially acute in cancer, where clinical sequencing of tumors now routinely reveals patient-specific variants whose functional impact on the corresponding gene is unknown, impeding clinical utility. Transcriptional profiling was able to systematically distinguish these variants of unknown significance (VUS) as impactful vs. neutral in an approach called expression-based variant-impact phenotyping (eVIP). We profiled a set of lung adenocarcinoma-associated somatic variants using Cell Painting, a morphological profiling assay that captures features of cells based on microscopy using six stains of cell and organelle components. Using deep-learning-extracted features from each cell’s image, we found that cell morphological profiling (cmVIP) can predict variants’ functional impact and, particularly at the single-cell level, reveals biological insights into variants which can be explored in our public online portal. Given its low cost, convenient implementation, and single-cell resolution, cmVIP profiling therefore seems promising as an avenue for using non-gene-specific assays to systematically assess the impact of variants, including disease-associated alleles, on gene function.

Copy Number Variation (CNV) is the major manner for the variation of genome structure, which is associated with numerous important traits. The heat shock factor 1 (HSF1) gene is a stress response transcriptional regulator. It participates in the heat shock response, simultaneously participated in the development of tissue. The objective of this research was to explore the influence of CNV of the HSF1 gene on the growth traits of the Ashidan yak. In this study, the growth traits (withers height, body weight, chest girth, and body length) of 274 Ashidan yaks were divided into four stages (6, 12, 18, and 30 months old). Moreover, quantitative polymerase chain reaction (qPCR) was exploited for determining the HSF1 gene relative expression level, and SPSS software was utilized for the statistical analysis. The outcomes indicated that HSF1-CNV was significantly associated with body length (p 0.05) and was extremely significant associated with withers height (p 0.01) of 18-month-old Ashidan yaks. Besides, the HSF1 relative expression in heart and muscle was higher than that existed in other tissues (p 0.01). The outcomes suggested that the CNV of HSF1 gene would affect the growth and development of the Ashidan yak, which is conducive to the early breeding of yak.

The large number of tailings ponds in my country, coupled with various man-made and natural factors in recent years, leads to the frequent occurrence of tailings pond accidents, causing serious harm. Mastering the number and distribution of tailings ponds is of great significance to prevent tailings pond accidents and carry out emergency management of tailings ponds. For the identification and monitoring of tailings ponds, the traditional survey methods are mainly based on ground surveys, and it is difficult to achieve large-scale and high-frequency detection. With the development of artificial intelligence technology, based on the support vector machine (SVM) method, the automatic extraction of remote sensing image (RSI) information has been realized, and remarkable achievements have been made in the field of remote sensing. This paper takes the tailings pond as the research goal, and based on the analysis of the characteristics of the tailings pond in the RSI, the method of SVM target detection is used to realize the rapid and high-precision identification of the tailings pond. The main contents of this paper are as follows: (1) The accident risk of tailings ponds at home and abroad and the status quo of tailings pond monitoring technology are introduced. (2) The relevant theory of SVM is learned, and the kernel function and corresponding parameter selection method of SVM are discussed in a multiclassification problem. (3) On the basis of determining the kernel function, parameters, and features, the trained model is compared with the original model. The results show that the SVM detection model proposed in this paper has excellent performance in tailings pond image recognition.

Objective. There are four widely-used formulae to calculate the perioperative glomerular filtration rate (GFR) of patients undergoing cardiac surgery. We assessed the predictive values of these formulae in the occurrence of postoperative acute kidney injury (AKI). Methods. Patients who underwent open-heart valvular surgery with cardiopulmonary bypass from January 2015 to October 2017 were enrolled in this retrospective study. Demographic data and perioperative serum creatinine levels were collected. The estimated GFR (eGFR) was calculated using four formulae: Cockcroft Gault (CG), body surface area (BSA) corrected CG, simplified modification of diet in renal disease (MDRD), and chronic kidney disease-epidemiology collaboration (CKD-EPI) formula. The incidence of post-operative AKI was calculated and the predictive capability of these formulae was evaluated. Results. A total of 290 patients were included. 136 patients (46.90%) developed AKI after surgery. The eGFR in the AKI group was significantly lower than those in the non-AKI group at all investigated time points. In addition, the eGFR in the non-AKI group increased temporarily on the day of operation, then decreased on the following days, and returned to preoperative level about one week after surgery. However, in the AKI group, the eGFR decreased from the day of operation, which still did not recover to the preoperative level by the end of the first week after surgery. The eGFR calculated by the BSA-standardized CG formula had the highest AUC ROC curves of 0.699 and 0.774 before operation and on the day of operation, respectively. While eGFR calculated by CKD-EPI formula had the highest AUC ROC of 0.874 and 0.887 at the first and second postoperative day. Conclusions. The eGFR formula is a powerful tool for perioperative renal function assessment. The BSA-corrected CG and CKD-EPI formula have better performance in predicting postoperative AKI after cardiopulmonary bypass than serum creatinine level and other formulae.

Cultivated chrysanthemum (Chrysanthemum × morifolium Ramat.) is a beloved ornamental crop due to the diverse capitula types among varieties, but the molecular mechanism of capitulum development remains unclear. Here, we report a 2.60 Gb chromosome-scale reference genome of C. lavandulifolium, a wild Chrysanthemum species found in China, Korea and Japan. The evolutionary analysis of the genome revealed that only recent tandem duplications occurred in the C. lavandulifolium genome after the shared whole genome triplication (WGT) in Asteraceae. Based on the transcriptomic profiling of six important developmental stages of the radiate capitulum in C. lavandulifolium, we found genes in the MADS-box, TCP, NAC and LOB gene families that were involved in disc and ray floret primordia differentiation. Notably, NAM and LOB30 homologs were specifically expressed in the radiate capitulum, suggesting their pivotal roles in the genetic network of disc and ray floret primordia differentiation in chrysanthemum. The present study not only provides a high-quality reference genome of chrysanthemum but also provides insight into the molecular mechanism underlying the diverse capitulum types in chrysanthemum.

PurposeTo measure the practice and management of proactive policing in local American police agencies and assess them in comparison to recommendations of the National Academies of Sciences (NAS) Committee on Proactive Policing.Design/methodology/approachA survey was conducted with a national sample of American police agencies having 100 or more sworn officers to obtain detailed information about the types of proactive work that officers engage in, to quantify their proactive work and to understand how the agencies measure and manage those activities. Responding agencies (n = 180) were geographically diverse and served populations of approximately half a million persons on average.FindingsProactivity as practiced is much more limited in scope than what the NAS envisions. Most agencies track only a few forms of proactivity and cannot readily estimate how much uncommitted time officers have available for proactive work. Measured proactivity is mostly limited to traffic stops, business and property checks and some form of directed or general preventive patrol. Many agencies have no formal policy in place to define or guide proactive activities, nor do they evaluate officer performance on proactivity with a detailed and deliberate rubric.Originality/valueThis is the first national survey that attempts to quantify proactive policing as practiced broadly in the United States. It provides context to the NAS recommendations and provides knowledge about the gap between practice and those recommendations.

The use of animal serum in culture medium brings safety concerns and batch-to-batch variability, and thus may restrict the clinical use of ex vivo expanded mesenchymal stromal cells (MSCs). Clinically compliant MSCs should be developed in adherence to serum-free, xeno-free and chemically defined medium (S&XFM-CD). In this study, we develop a S&XFM-CD by replacing all serum components with synthetic alternatives for the derivation of clinical-grade umbilical cord-derived MSCs (UCMSCs). The critical aspects including characterization, safety concerns, potency and exogenous factors contamination risk of UCMSCs in S&XFM-CD are compared with serum-containing medium (SCM). UCMSCs in S&XFM-CD retain fibroblastic-like morphology and immunophenotype of MSCs, and exhibit superior clone efficiency, proliferation capacity, and osteogenic and chondrogenic differentiation potential compared with SCM. Moreover, UCMSCs in S&XFM-CD retain similar immunosuppressive potential, and exhibit superior secretion levels of bFGF, PDGF-BB and IGF-1 compared with SCM. In addition, UCMSCs in S&XFM-CD do not undergo transformation, preserve the normal karyotypes and genomic stability, and are less prone to senescence process after long-term in vitro culture, which conforms to the current guidance of international and national evaluation standard. The S&XFM-CD developed here may serve as a GMP-grade production platform of UCMSCs for future clinical application.

This study aims to assess the evidential value of the knowledgebase in criminology after accounting for the presence of potential Type I errors. The present study examines the distribution of 1248 p-values (that inform 84 statistically significant outcomes across 26 systematic reviews) in meta-analyses on the topic of crime and justice published by the Campbell Collaboration (CC) using p-curve analysis. The distribution of all CC p-values have a significant cluster of p-values immediately below 0.05, which is indicative of p-hacking. Evidential value (right skewed p-curves) is detected in most meta-analytic topic areas but not motivational interviewing (substance use outcome), sex offender treatment (sexual/general recidivism), police legitimacy (procedural justice), street-level drug law enforcement (total crime), and treatment effectiveness in secure corrections (juvenile recidivism). More studies, especially carefully designed and implemented randomized experiments with sufficiently large sample sizes, are needed before we are able to affirm the presence of evidential value and replicability of studies in all CC topic areas with confidence.

Jiaozi Steamed Bread (JSB) has a unique aroma as a traditional staple food in China. The volatile compounds in JSB were extracted by simultaneous distillation and extraction (SDE) and headspace solid-phase microextraction (HS-SPME). These volatile substances were analyzed by gas chromatography-mass spectrometry (GC-MS) and gas chromatography-olfactometry-mass spectrometry (GC-O-MS). The results demonstrated that 61 volatile compounds were identified totally in samples, of which 15 were confirmed as potent aroma compounds with odor active values (OAVs) >1. The 15 potent aroma compounds were ethanol, 1-butanol, 1-pentanol, 1-hexanol, heptanol, 1-octen-3-ol, 3-methyl-1-butanol, hexanal, heptanal, nonanal, (E)-2-heptenal, benzaldehyde, (E, E)-2,4-decadienal, 2-pentylfuran and naphthalene. The SDE method had better linearity with coefficients of determination (R2) equal to or higher than 0.9991. Furthermore, the SDE method also achieved lower sensitivity and better repeatability and recovery than HS-SPME. This work provides reference method and parameters for future research on the flavor of JSB for commercial products. Anti-cancer uses of non-oncology drugs have occasionally been found, but such discoveries have been serendipitous. We sought to create a public resource containing the growth inhibitory activity of 4,518 drugs tested across 578 human cancer cell lines. We used PRISM, a molecular barcoding method, to screen drugs against cell lines in pools. An unexpectedly large number of non-oncology drugs selectively inhibited subsets of cancer cell lines in a manner predictable from the cell lines' molecular features. Our findings include compounds that killed by inducing PDE3A-SLFN12 complex formation; vanadium-containing compounds whose killing depended on the sulfate transporter SLC26A2; the alcohol dependence drug disulfiram, which killed cells with low expression of metallothioneins; and the anti-inflammatory drug tepoxalin, which killed via the multi-drug resistance protein ABCB1. The PRISM drug repurposing resource (https://depmap.org/repurposing) is a starting point to develop new oncology therapeutics, and more rarely, for potential direct clinical translation.

The 2017 National Academies of Sciences (NAS) Committee and Report on Proactive Policing highlighted what we know about the effects of proactive policing practices on crime prevention and police–community relations. However, the evaluation evidence reviewed by the NAS, which largely comes from case studies of carefully managed proactive initiatives, does not provide a basis for estimating how extensively these practices are used or whether they are used in the most effective ways. Accordingly, it is unclear whether police proactivity as practiced on an everyday basis reflects optimal strategies and implementation methods as recommended by the NAS. This study addresses this knowledge gap by analyzing almost 2 million computer-aided dispatch records from four agencies and systematically observing 84 officers for more than 180 hours to better understand the empirical realities of police proactivity. The findings indicate a major difference between the types of proactive interventions supported by research and the practice of everyday police proactivity. Specifically, proactive policing practices are limited in scope and are often implemented in less than optimal ways. A large proportion of proactive activities are also not recorded, rewarded, or supervised, indicating that patrol commanders may have little control over, or awareness of, proactive deployment. From an evidence-based policing perspective, much more effort is needed to record and track proactivity to measure its impacts (both positive and negative) and align it with what we now know about effective proactive activity from research.

Since China initiated new health-care reforms in early 2009, a variety of measures have been implemented to slow the growth of medical expenses. This study was conducted to investigate the effect of controlling medical expenses. Based on inpatients' medical expenses at the largest tertiary hospital in Shenzhen, China, from 2009 to 2017, this study analyzed the changes in medical expenses and expense structures according to payment sources (insured or self-financed), stratifying the medical expenses according to the ICD-10 classification chapters of the principal diagnoses of the inpatients in two years (2009 and 2017) in order to control for confounding diseases. The results showed that mean inpatient expenses continued to rise from 2009 to 2017, and the expenses of the self-financed group began to exceed those of the insured group after 2011. Drug and consumable expenses were still the main factors that affected inpatient expenses, and consumable expenses remarkably increased, becoming the highest proportion of expenses. New health-care reforms were effective in controlling growing medical expenses for insured patients but did not make a significant difference in the expenses of self-financed patients. The excessive use of consumables has become a new driver of growing medical expenses.

Purpose Empirical research suggests that traffic enforcement is the most common type of proactive activity police officers engage in on a daily basis. Further, agencies often use traffic enforcement to achieve both traffic safety and crime control. Given these goals, the purpose of this paper is to investigate whether (and to what extent) officers are accurately targeting their proactive traffic enforcement with crime and vehicle crashes in two agencies. Design/methodology/approach The study examines traffic enforcement patterns in two agencies to see whether proactive traffic enforcement aligns spatially with crime and vehicle crashes. This study employs negative binomial regression models with clustered standard errors to investigate this alignment at the micro-spatial level. Key variables of interest are measured with police calls for service data, traffic citation data and vehicle crash data from two law enforcement jurisdictions. Findings High levels of spatial association are observed between traffic accidents and crime in both agencies, lending empirical support to the underlying theories of traffic enforcement programs that also try to reduce crime (i.e. “DDACTS”). In both agencies, traffic accidents also appear to be the most prominent predictor of police proactive traffic enforcement activities, even across different times of day. However, when vehicle crashes are accounted for, the association between crime and traffic stops is weaker, even during times of day when agencies believe they are using proactive traffic enforcement as a crime deterrent. Originality/value No prior study to authors knowledge has examined the empirical association between police proactive traffic activities and crime and traffic accidents in practice. The current study seeks to fill that void by investigating the realities of traffic stops as practiced daily by police officers, and their alignment with crime and vehicle crashes. Such empirical inquiry is especially important given the prevalent use of traffic enforcement as a common proactive policing tool by police agencies to control both traffic and crime problems.

A new geminivirus was identified in common bean (Phaseolus vulgaris) showing severe stunt and leaf curling symptoms in Heilongjiang province of China, via sequencing and assembly of small RNAs. The genome of this geminivirus comprises 2,959 nucleotides (nt) and shares 21.77-54.97% nt sequence identity with other geminiviruses. The coat protein (CP) shares the highest amino acid (aa) sequence identity (23.5%) with that of sesame curly top virus (SeYMV; genus Turncurtovirus), whereas the C1 (Rep) shares the highest aa sequence identity (66.5%) with that of beet severe curly top virus (BSCTV; genus Curtovirus). This geminivirus neighbors the turncurtoviruses in phylogenetic trees based on the full genome sequence or the amino acid sequence of the Rep protein, but it forms a distinct clade in the phylogenetic tree based on the coat protein. Recombination analysis showed that parts of the C1 coding region of this geminivirus were recombined from a curtovirus or turncurtovirus. Based on these results, the name "common bean curly stunt virus" (CBCSV) is proposed for this virus.

The structure–activity relationship (SAR) of DNMDP analogs was evaluated using a phenotypic viability assay, resulting in several compounds with suitable pharmacokinetic properties for in vivo analysis. One of these compounds, BRD9500, was active in an SK-MEL-3 xenograft model of cancer.

Myocardial hypertrophy is a common heart disease that is closely associated with heart failure. The expression of F-box and WD repeat-containing protein 7 (FBW7) is significantly downregulated in angiotensin (Ang) II-induced cardiac fibroblasts, suggesting that it may possess an important function in cardiac development. The present study attempted to explore the role of FBW7 in Ang II-induced myocardial hypertrophic injury and its associated mechanism of action. Reverse transcription-quantitative PCR and western blotting were used to determine the expression levels of FBW7 in Ang II-induced H9C2 cells. The expression levels of autophagy-related and mTOR signaling pathway-related proteins were detected using western blotting. Cell viability was assessed using the Cell Counting Kit-8 assay. The apoptosis rate of H9C2 cells was detected using TUNEL assay and western blotting. Cellular hypertrophy and fibrosis were assessed using phalloidin staining and western blotting. Levels of inflammatory factors were examined using ELISA and western blotting, whereas levels of oxidative stress-related markers were detected by corresponding kits. The results indicated that FBW7 expression was downregulated in Ang II-induced H9C2 cells. FBW7 upregulation enhanced the expression levels of autophagy-related proteins and activated mTOR-mediated cellular autophagy. FBW7 overexpression promoted the cell viability, inhibited Ang II-induced apoptosis, cellular hypertrophy and fibrosis in H9C2 cells via the autophagic pathway, as well as inflammation and oxidative stress. Overall, the data indicated that FBW7 overexpression ameliorated Ang II-induced hypertrophic myocardial injury via the mTOR-mediated autophagic pathway.

Acipenser dabryanus is an endemic species restricted to the upper reaches of the Yangtze River and is listed as a national first-class protected animal in China. In the present study, two lipid metabolism-associated genes, namely, lipoprotein lipase (LPL) and apolipoprotein E (apoE), were identified from the full-length transcriptome analysis of A. dabryanus. The full-length genes encoded polypeptides with typical conserved motifs and functional sites shared with their homologues in other fish. Gene expression profile analysis showed that the LPL and apoE genes were constitutively expressed in A. dabryanus. The highest expression level of LPL was found in the muscle, while apoE was predominantly expressed in the liver. In the fasting and refeeding experiment, one hundred twenty fish were randomly assigned to four groups (fasting 0, 3, 7 or 14 d and then refed for 14 d). Fasting and refeeding significantly regulated the expression level of LPL with various expression patterns in different tissues, whereas starvation showed little effect on apoE expression. In addition, the expression levels of LPL and apoE were significantly affected by feeding frequency. The fish were bred for eight weeks at six different feeding frequencies groups (once a day, two times a day, three times a day, four times a day, five times a day and six times a day). Both LPL and apoE expression levels were significantly upregulated in the liver in the highest feeding frequency group. Our study suggests potential roles for LPL and apoE in lipid metabolism in A. dabryanus and will be useful for further study of nutrition regulation in fish.

Tianzhu white yak is a rare local yak breed with a pure white coat in China. In recent years, breeders have discovered long-haired individuals characterized by long hair on the forehead in the Tianzhu white yak, and the length and density of the hair on these two parts of the body are higher than that of the normal Tianzhu white yak. To elucidate the genetic mechanism of hair length in Tianzhu white yak, we re-sequence the whole genome of long-haired Tianzhu White yak (LTWY) (n = 10) and normal Tianzhu White yak (NTWY) (n = 10). Then, fixation index (FST), θπ ratio, cross-population composite likelihood ratio (XP-CLR), integrated haplotype score (iHS), cross-population extended haplotype homozygosity (XP-EHH), and one composite method, the de-correlated composite of multiple signals (DCMS) were performed to discover the loci and genes related to long-haired traits. Based on five single methods, we found two hotspots of 0.2 and 1.1 MB in length on chromosome 6, annotating two (FGF5, CFAP299) and four genes (ATP8A1, SLC30A9, SHISA3, TMEM33), respectively. Function enrichment analysis of genes in two hotspots revealed Ras signaling pathway, MAPK signaling pathway, PI3K-Akt signaling pathway, and Rap1 signaling pathway were involved in the process of hair length differences. Besides, the DCMS method further found that four genes (ACOXL, PDPK1, MAGEL2, CDH1) were associated with hair follicle development. Henceforth, our work provides novel genetic insights into the mechanisms of hair growth in the LTWY.

Increased drought intensity with rising atmospheric demand for water (hereafter VPD) increases the risk of tree mortality worldwide. Ecosystem-scale water-use strategy (WUSe), quantified here by canopy stomatal sensitivity to VPD (Sc), is increasingly recognized as a factor in drought-related ecosystem dysfunction. However, the links between Sc and ecosystem adaptation to and stability following droughts are poorly established. We examined how Sc regulates carbon sequestration, identifying ecosystems potentially susceptible to drought-induced mortality based on data from the global flux network, remote-sensing products, and plant functional-traits archive. We found that Sc is higher where ecosystem water availability is low in arid regions, reflecting conservative WUSe (i.e., hypersensitivity), but ecosystems of all regions converge on permissive WUSe (i.e., hyposensitivity) under ample water supply. During extreme droughts, hyposensitive and hypersensitive ecosystems achieved similar net ecosystem productivity employing considerably different structural-functional strategies. However, hyposensitive ecosystems, risking their hydraulic system with permissive WUSe, did not recover from extreme droughts as quickly. Models predicting current performance and future distributions of vegetation types should account for the greater vulnerability of hyposensitive ecosystems to intensifying atmospheric and soil drought.

Background Fractional flow reserve derived from computed tomography (FFRCT) has been demonstrated to improve identification of lesion-specific ischemia significantly compared with coronary computed tomography angiography (CCTA). It remains unclear whether the distribution of FFRCT values in obstructive stenosis between patients who received percutaneous coronary intervention (PCI) or not in routine clinical practice, as well as its association with clinical outcome. This study aims to reveal the distribution of FFRCT value in patients with single obstructive coronary artery stenosis and explored the independent factors for predicting major adverse cardiac events (MACE). Methods This was a retrospective study of adults with non-ST-segment elevation acute coronary syndrome undergoing FFRCT assessment by using CCTA data from January 1, 2016 to December 31, 2020. Propensity score matching (PSM) method was used to account for patient selection bias. The risk factors for predicting MACE were evaluated by a Cox proportional hazards regression analysis. Results Overall, 655 patients with single obstructive (≥ 50%) stenosis shown on CCTA were enrolled and divided into PCI group (279 cases) and conservative group (376 cases) according to treatment strategy. The PSM cohort analysis demonstrated that the difference in history of unstable angina, Canadian Cardiovascular Society Class (CCSC) and FFRCT between PCI group (188 cases) and conservative group (315 cases) was statistically significant, with all P values P = 0.912). The incidence of MACE in PCI group and conservative group were 14.9% (28/188) and 23.5% (74/315) respectively, P = 0.020. Multivariate analysis of Cox proportional hazards regression revealed that history of unstable angina (adjusted odds ratio (adjOR), 3.165; 95% confidence interval (CI), 2.087–4.800; P P = 0.016), and PCI therapy (OR 0.481; 95% CI 0.305–0.758) were the independent factors for MACE. Conclusions History of unstable angina and FFRCT value of ≤ 0.8 were the independent risk factors for MACE, while PCI therapy was the independent protective factor for MACE.

Mammalian skeletal muscle is composed of various muscle fibers that exhibit different physiological and metabolic features. Muscle fiber type composition has significant influences on the meat quality of livestock. In this study, we comprehensively analyzed the whole transcriptome profiles of the oxidative muscle biceps femoris (BF) and the glycolytic muscle obliquus externus abdominis (OEA) of yak. A total of 1436 mRNAs, 1172 lncRNAs, and 218 circRNAs were differentially expressed in the oxidative muscles compared with the glycolytic muscles. KEGG annotation showed that differentially expressed mRNAs regulated by lncRNA and circRNA were mainly involved in PPAR signaling pathway, citrate cycle (TCA cycle), and PI3K-Akt signaling pathway, which reflect the different metabolic properties between oxidative and glycolytic muscles. In addition, regulatory networks associated with muscle fiber type conversion and mitochondria energy metabolism in muscles were constructed. Our study provides new evidence for a better understanding of the molecular mechanisms underlying skeletal muscle fiber determination and meat quality traits of yak.
