Neurogenesis occurs throughout life in the dentate gyrus of hippocampus and subventricular zone, but this phenomenon has rarely been observed in other brain regions of adult mammals. The aim of the current study was to investigate the cell proliferation process in the ischemically challenged region-at-risk after focal cerebral ischemia in the adult rat brain. A reversible photothrombotic ring stroke model was used, which features sustained hypoperfusion followed by late spontaneous reperfusion and a remarkable morphologic tissue recovery in the anatomically well defined somatosensory cortical region-at-risk. Twelve-week-old male Wistar rats received repeated intraperitoneal injections of the cell proliferation specific marker 5-bromodeoxyuridine (BrdU) after stroke induction. Immunocytochemistry of coronal brain sections revealed that the majority of BrdU-positive cells were of glial, macrophage, and endothelial origin, whereas 3% to 6% of the BrdU-positive cells were double-labeled by BrdU and the neuron-specific marker Map-2 at 7 and 100 days after stroke onset in the region-at-risk. They were distributed randomly in cortical layers II-VI. Three-dimensional confocal analyses of BrdU and the neuronal-specific marker Neu N by double immunofluorescence confirmed their colocalization within the same cells at 72 hours and 30 days after stroke induction. This study suggests that, as a potential pathway for brain repair, new neurons can be generated in the cerebral cortex of adult rats after sublethal focal cerebral ischemia.

Background Endovascular thrombectomy (EVT) is standard of care for anterior circulation acute ischemic stroke (AIS) caused by large vessel occlusion (LVO), but data on nationwide performance in routine healthcare are sparse. The study aims were to describe EVT patients with LVO AIS, analyze mortality and functional outcome, and compare results with randomized controlled trials (RCTs).Methods Data from the Riksstroke and the Swedish Endovascular Treatment of Acute Stroke Registry (RSEVAS) on pre-stroke independent patients, with LVO AIS in 2017–2019, defined as occlusion of the intracranial internal carotid artery, or the M1 or M2 segments of the middle cerebral artery, and groin puncture <6 h of onset, were compared to aggregated HERMES collaboration RCT data. We assessed 90-day survival and function, defined by the modified Rankin Scale. Specific analyzes were stratified by occlusion location.Results In all, 1011/2560 of RSEVAS patients matched RCT inclusion criteria. Compared with RCT data, patients were older (73 vs. 68), fewer received intravenous thrombolysis (63.1% vs. 83%), and M2 occlusions were more common (24.5% vs. 8%). 90-day survival in RSEVAS was 85.3%, 42.8% achieved good outcome and 5% had symptomatic intracerebral hemorrhage (sICH). Corresponding outcomes in RCT data were 84.7% survival, 46% good outcome, and 4.4% sICH. Functional outcome was most favorable following M2 occlusions.Conclusions EVT patients from our large real-world national dataset differed from RCT patients in several baseline factors including distribution of vascular occlusion site. However, the overall outcome of EVT in our Swedish cohort appeared to well match the pivotal trial findings.

•VIPVIZA is a pragmatic randomized controlled trial performed within the regular primary health care and targeting both individuals and physicians.•Beneficial effects on risk for cardiovascular disease regardless of sex and education level 3 years after providing pictorial information of subclinical atherosclerosis in addition to regular preventive information.•The results indicate that the intervention effect is a combined effect of pharmacological treatment and lifestyle modification.•The intervention effect was observed in the intermediate risk group, the group where most CVD events occur and in which sufficient prevention is often overlooked.

Because of the significance of potassium in the practice of medicine, it is important to consider factors that influence the integrity of intracellular and extracellular K. Clinicians today are skilled in managing hypokalemia as well as hyperkalemia. However, physicians may be less aware of the role of magnesium in K homeostasis.Experimental observations to date support the view that Mg and K metabolism are closely linked. Dietary Mg depletion in the rat is accompanied by muscle K depletion, which occurs despite provision of K.1,2 Concurrent K and Mg depletion results in a greater loss of muscle K than with K depletion alone.3 In K- and Mg-deficient rats, muscle-cell K repletion is not optimized unless both K and Mg deficits are simultaneously corrected.4 Thus, Mg appears to be important not only in maintenance of cell K but also in conservation and repletion of cell K.5 These experimental

The anteroposterior distribution of monoamine and monoamine metabolite concentrations was determined in subcortical brain nuclei of two cases of Sjögren-Larsson syndrome (SLS) and was compared to two control cases. The brains were divided into halves and sectioned coronally. For biochemical analyses, caudate nucleus, putamen, globus pallidus, substantia nigra, nucleus accumbens, amygdala, and hippocampus were dissected macroscopically. Monoamine and its metabolites were determined by reverse-phase liquid chromatography with electrochemical detection. The other hemisphere was studied neuropathologically.

The effects of bilateral carotid artery occlusion/recirculation on cortical CBF (cCBF) were studied in rats following the intravenous administration of either the nitric oxide synthase inhibitor NG-nitro-l-arginine methyl ester hydrochloride (l-NAME; 30 mg/kg) or an equivalent volume of saline (500 μl). Induction of bilateral carotid occlusion (BCO) in l-NAME-treated animals resulted in a reduction of cCBF to 30% of baseline. During recirculation subsequent to 20 min of BCO, cCBF in l-NAME-infused animals remained at 30% of baseline. In contrast, cCBF in saline-treated control animals returned to the original baseline level following a similar reduction to 30–40% of baseline during BCO. These results indicate that inhibition of nitric oxide generation limits normalization of regional cortical perfusion following occlusion of proximal large cerebral vessels.

Central hemodynamics and gas exchange were studied in six patients with chronic obstructive pulmonary disease (mean age, 60 years). Patients were selected for the study if the volume of blood was 1 L larger than the predicted normal value, if there was no history of infection, and if no drug assumed to influence pulmonary circulation had been given during the last four weeks. Measurements were first performed in the hypervolemic state. This was followed by a venesection of 0.5 L, and the measurements were repeated 15 minutes later (immediate effects of dehydration). After two weeks to two months of intensive diuretic therapy, new measurements were performed (long-term effects of dehydration). With hypervolemia, there was pulmonary hypertension, increased pulmonary vascular resistance, and normal cardiac output. The arterial oxygen tension (PaO2) was markedly reduced. The arterial carbon dioxide tension (PaCO2) was increased in two patients, and dead-space ventilation (VD/VT) increased in all. The sole immediate effect of venesection was a small increase in heart rate and a reduction in PaCO2. The long-term effects were a maintained low blood volume (on the average, 0.7 L less than before bloodletting) and lowered hematocrit reading (8 percent less than before bloodletting), a reduced and in some patients normalized pulmonary arterial pressure, a reduced pulmonary vascular resistance, and an unchanged cardiac output. The PaO2improved, while PaCO2did not change; neither did VD/VT. The changes correlated partly with the diminution in blood volume and partly with the reduction in hematocrit reading.

A comparison of the range of urinary excretion of 20 elements (Ag, As, Au, Br, Ce, Cd, Cs, Cu, Fe, Hg, La, Mg, Mo, Rb, Sb, Sc, Se, Sm, W, Zn) in healthy subjects as well as in patients with primary hyperparathyroidism before and after operation was performed by means of a recently developed ion-exchange technique following neutron activation analysis. The urinary levels of the 20 trace elements showed no distinct differences when the controls were compared with the untreated cases of hyperparathyroidism. Small differences in the urinary excretion of certain trace elements in patients with hyperparathyroidism before and after operation were, however, revealed. The biological significance of these findings is discussed.
