Deep basins create uniquely favorable conditions for causing air pollution, and the Sichuan Basin (SCB) in Southwest China is such a basin featuring frequent heavy pollution. A wintertime heavy haze pollution event in the SCB was studied with conventional and intensive observation data and the WRF-Chem model to explore the 3D distribution of PM2.5 to understand the impact of regional pollutant emissions, basin circulations associated with plateaus, and downwind transport to the adjacent areas. It was found that the vertical structure of PM2.5 over the SCB was characterized by a remarkable hollow sandwiched by high PM2.5 layers at heights of 1.5–3 km and a highly polluted near-surface layer. The southwesterlies over the Tibetan Plateau (TP) and Yunnan-Guizhou Plateau (YGP) resulted in a lee vortex over the SCB, which helped form and maintain heavy PM2.5 pollution. The basin PM2.5 was lifted into the free troposphere and transported outside of the SCB. At the bottom of the SCB, high PM2.5 concentrations were mostly located in the northwestern and southern regions. Due to the blocking effect of the plateau terrain on the northeasterly winds, PM2.5 gradually increased from northeast to southwest in the basin. In the lower free troposphere, the high PM2.5 centers were distributed over the northwestern and southwestern SCB areas, as well as the central SCB region. For this event, the regional emissions from the SCB contributed 75.4 %–94.6 % to the surface PM2.5 concentrations in the SCB. The SCB emissions were the major source of PM2.5 over the eastern regions of the TP and the northern regions of the YGP, with contribution rates of 72.7 % and 70.5 %, respectively, during the dissipation stage of heavy air pollution over the SCB, which was regarded as the major pollutant source affecting atmospheric environment changes in Southwest China.

With the development of infrared technology, large-array infrared focal plane detector is getting more and more popular. In order to solve the problem of reduced reliability when the cooling head is packaged with large-array infrared detector, a new design with reinforcement structures on cold platform and cold shield is introduced in this paper. The finite element simulation is used to analyze the optimized structure, and the results show that this design can fully meet the mechanical requirements of the project. By using the reinforcement structures, a Dewar was designed and processed to package a 2.7K×2.7K-15μm large-array infrared detector. After all the packaging process, the total mass of the Dewar is only 800g, and the cooling head is more than 90g. Under the circumstances of a heavy cooling head, the Dewar successfully passed the environmental adaptability assessment, and there is no problem with all the indexes of the infrared focal plane detector, which verified the reliability of the reinforcement structure.

Brassica rapa is an important Chinese vegetable crop that is beneficial to human health. The primary factor affecting B. rapa yield is low temperature, which promotes bolting and flowering, thereby lowering its commercial value. However, quickened bolting and flowering can be used for rapid breeding. Therefore, studying the underlying molecular mechanism of vernalization in B. rapa is crucial for solving production-related problems. Here, the transcriptome of two B. rapa accessions were comprehensively analyzed during different vernalization periods. During vernalization, a total of 974,584,022 clean reads and 291.28 Gb of clean data were obtained. Compared to the reference genome of B. rapa, 44,799 known genes and 2280 new genes were identified. A self-organizing feature map analysis of 21,035 differentially expressed genes was screened in two B. rapa accessions, &lsquo, Jin Wawa&rsquo, and &lsquo, Xiao Baojian&rsquo, The analysis indicated that transcripts related to the plant hormone signal transduction, starch and sucrose metabolism, photoperiod and circadian clock, and vernalization pathways changed notably at different vernalization periods. Moreover, different expression patterns of TPS, UGP, CDF, VIN1, and seven hormone pathway genes were observed during vernalization between the two accessions. The transcriptome results of this study provide a new perspective on the changes that occur during B. rapavernalization, as well as serve as an excellent reference for B. rapa breeding.

Background Proline-rich extension-like receptor protein kinases (PERKs) are an important class of receptor kinases located in the plasma membrane, most of which play a vital role in pollen development. Results Our study identified 25 putative PERK genes from the whole Brassica rapa genome (AA). Phylogenetic analysis of PERK protein sequences from 16 Brassicaceae species divided them into four subfamilies. The biophysical properties of the BrPERKs were investigated. Gene duplication and synteny analyses and the calculation of Ka/Ks values suggested that all 80 orthologous/paralogous gene pairs between B. rapa and A. thaliana, B. nigra and B. oleracea have experienced strong purifying selection. RNA-Seq data and qRT-PCR analyses showed that several BrPERK genes were expressed in different tissues, while some BrPERKs exhibited high expression levels only in buds. Furthermore, comparative transcriptome analyses from six male-sterile lines of B. rapa indicated that 7 BrPERK genes were downregulated in all six male-sterile lines. Meanwhile, the interaction networks of the BrPERK genes were constructed and 13 PERK coexpressed genes were identified, most of which were downregulated in the male sterile buds. Conclusion Combined with interaction networks, coexpression and qRT-PCR analyses, these results demonstrated that two BrPERK genes, Bra001723.1 and Bra037558.1 (the orthologs of AtPERK6 (AT3G18810)), were downregulated beginning in the meiosis II period of male sterile lines and involved in anther development. Overall, this comprehensive analysis of some BrPERK genes elucidated their roles in male sterility.

Cucumber is a major vegetable crop grown in facilities, and its seedlings are sensitive to salinity. Thus, it is important for cucumber cultivation to research the effects of Ca(NO3)2. In this study, we investigated salt-sensitive Chunqiu (CQ) and salt-tolerant BoMei 49 (BM) seedlings, the changes to the enzymes involved in the mitochondria antioxidant system in the seedlings, and the changes in the MPTP (Mitochondrial Permeability Transition Pore) opening, mitochondrial membrane fluidity, mitochondrial membrane potential depolarization, mitochondrial electron transfer chain, and NO metabolism in response to Ca(NO3)2. Ca(NO3)2 stresses inhibited the growth of the cucumber seedlings, which showed a reduced root length, a decreased surface area and a decreased number of root tips, but a significantly increased root diameter. The mitochondrial malondialdehyde (MDA) content, H2O2 content, and O2&minus, producing rate of the two cucumber roots were gradually increased in the Ca(NO3)2 treatment. The activity of superoxide dismutase (SOD) and peroxidation enzyme (POD) gradually increased, and catalase (CAT) activity gradually decreased. The electron transport chain activity of &ldquo, BM&rdquo, was higher than &ldquo, CQ&rdquo, under Ca(NO3)2 stress. Ca(NO3)2 stress destroyed the membrane structure of the mitochondria, so that the mitochondrial body tended to bend, causing double-membrane digestion and a hollow interior. Under the stress of Ca(NO3)2, the NO3- content of the seedlings increased significantly. The contents of NH4+ and NO, as well as the activities of nitrate reductase (NR), nitrite reductase (NIR), and nitric oxide synthase (NOS) decreased significantly. These findings provide physiological insights into root damage in response to salt stress for salt tolerance in cucumber.
