ObjectiveCXCL14 (C-X-C motif chemokine ligand-14) is a chemokine released by active brown fat, showing protective effects against insulin resistance in experimental models. Polycystic ovary syndrome (PCOS) in adolescent girls is usually related to hepato-visceral fat excess and insulin resistance, and associates with comorbidities such as type 2 diabetes. Treatment with a low-dose combination of one antiandrogen and antimineralocorticoid drug (spironolactone) and two insulin sensitizers (pioglitazone/metformin) (SPIOMET) is particularly effective in improving these metabolic derangements. Adipose tissue may be involved in the metabolic alterations of PCOS, and it is a likely target of therapeutic action. We investigated the alterations in CXCL14 levels and the effects of drugs composing SPIOMET treatment on CXCL14 in human adipocytes.Research design and methodsWe studied 51 adolescent patients with PCOS and 21 age-matched healthy controls. Thirty-one adolescent patients with PCOS under SPIOMET or oral contraception-based treatment were also studied. For studies in vitro, Simpson Golabi Behmel Syndrome (SGBS) adipose cells were used. Gene expression for CXCL14 and other genes was quantified using quantitative real-time PCR. The levels of CXCL14 and adipokines in serum and cell culture media were determined by ELISA.ResultsSerum CXCL14 levels are reduced in patients with PCOS. One-year SPIOMET treatment normalized CXCL14 concentrations and improved the metabolic status of patients with PCOS. Pioglitazone induced CXCL14 expression in differentiating human SGBS adipocytes, in parallel with the induction of marker genes of brown adipogenesis. Spironolactone induced CXCL14 expression and release in differentiated human adipocytes.ConclusionInsulin sensitization with SPIOMET normalizes the abnormally low levels of CXCL14 in girls with PCOS. This is consistent with the effects of pioglitazone and spironolactone inducing CXCL14 expression and promoting a brown-like phenotype in adipocytes. CXCL14 may be a novel biomarker for PCOS as well as a potential mediator of the beneficial effects of the SPIOMET combination and may hold promise as a therapeutic modulator of the disorder.Trial registration numbersISRCTN29234515 and ISCRCTN11062950.

T cells are activated by antigen-bearing dendritic cells (DC), but how DCs receive reciprocal activating signals from T cells is still unclear. Here the authors show that mitochondrial DNA-containing extracellular vesicles from activated T cells can prime DCs for anti-viral immunity via the cGAS/STING DNA sensing pathways.

This paper examines some aspects of the legislation on volunteering, concretely, this concept that is included in the Volunteer Law (Title I "Of volunteering").To do this, it begins with the definition and delimitation of this term trying to know the fundamental aspects that make this figure, then, it sees its historical background and its legislative evolution in Spain, the national Legislation is studied trying to know the causes of the changes that have taken place. Subsequently, it takes place a comparison between the different autonomic regulations, in order to try to know the legislative differences between the different communities.; En este trabajo se analizan algunos aspectos de la legislación sobre el voluntariado, en concreto su concepto, que aparece recogido en el Título I “Del voluntariado”.Para ello se comienza con la definición y delimitación del término, tratando de conocer los aspectos fundamentales que delimitan esta figura, observándose su evolución legislativa en España y comenzando con el análisis de la normativa estatal, tratando de conocer las causas de las modificaciones que han tenido lugar para posteriormente estudiar las distintas normativas autonómicas, también referidas a ese Título I, tratando de conocer las diferencias legislativas entre las distintas comunidades.; Lan honetan boluntariotzari buruzko legeriaren zenbait alderdi aztertzen dira, zehazki, haren kontzeptua, I. tituluan ("boluntariotza") jasota dagoena.Horretarako, terminoaren definizioarekin eta mugaketarekin hasten da, figura hori mugatzen duten funtsezko alderdiak ezagutzen saiatuz, Espainian izan duen legegintza-bilakaera behatuz eta Estatuko araudia aztertzen hasiz, ondoren egin diren aldaketen arrazoiak ezagutzen saiatuz, ondoren araudi autonomiko desberdinak aztertzeko, horiek ere I. Background: Ketogenic dietary therapies (KDT) are used as a treatment in childhood epilepsy. However, their mechanism has not yet been established. The main objective of this study was to determine the changes in the transcriptomic profile induced by KDT in children with epilepsy in order to shed light on its possible mechanisms. Methods: Eight children with refractory epilepsy were enrolled in the study. Peripheral blood mononuclear cells were obtained before and after the children were treated with KDT for a minimum of 6 months. RNA was extracted and mRNA and miRNA profiling were performed and analyzed. Results: Our intervention with KDT significantly reduced the seizure number in seven of the eight paediatric patients treated and caused important changes in their gene expression profile. Our study reveals modifications in the transcription of 4630 genes and 230 miRNAs. We found that the genes involved in the protection against epileptic crises were among those mainly changed. These genes collectively encode for ion channels, neurotransmitter receptors, and synapse structural proteins. Conclusions: Together our results explain the possible mechanisms of KDT and reinforce its clinical importance in the treatment of epilepsy.

Introduction: Polycystic ovary syndrome (PCOS) is often associated with metabolic-associated fatty liver disease (MAFLD). MAFLD has been associated with altered hepatic function, systemic dysmetabolism, and abnormal circulating levels of signaling molecules called organokines. Here, we assessed the effects of two randomized treatments on a set of organokines in adolescent girls with PCOS and without obesity, and report the associations with circulating biomarkers of liver damage, which were assessed longitudinally in the aforementioned studies as safety markers. Materials and methods: Liver enzymes [aspartate aminotransferase (AST), alanine aminotransferase (ALT), and gamma-glutamyl transferase (GGT)] were assessed as safety markers in previous randomized pilot studies comparing the effects of an oral contraceptive (OC) with those of a low-dose combination of spironolactone-pioglitazone-metformin (spiomet) for 1 year. As a post hoc endpoint, the organokines fibroblast growth factor-21 (FGF21), diazepambinding protein-1 (DBI), and meteorin-like protein (METRNL) were assessed by ELISA after 6 months of OC (N = 26) or spiomet (N = 28). Auxological, endocrinemetabolic, body composition (using DXA), and abdominal fat partitioning (using MRI) were also evaluated. Healthy, age-matched adolescent girls (N = 17) served as controls. Results: Circulating ALT and GGT levels increased during OC treatment and returned to baseline concentrations in the post-treatment phase; in contrast, spiomet treatment elicited no detectable changes in ALT and GGT concentrations. In relation to organokines after 6 months of treatment, (1) FGF21 levels were significantly higher in PCOS adolescents than in control girls; (2) DBI levels were lower in OC-treated girls than in controls and spiomet-treated girls; and (3) no differences were observed in METRNL concentrations between PCOS girls and controls. Serum ALT and GGT levels were directly correlated with circulating METRNL levels only in OC-treated girls (R = 0.449, P = 0.036 and R = 0.552, P = 0.004, respectively). Conclusion: The on-treatment increase in ALT and GGT levels occurring only in OC-treated girls is associated with circulating METRNL levels, suggesting enhanced METRNL synthesis as a reaction to the hepatic changes elicited by OC treatment. Background: The secretory properties of brown adipose tissue are thought to contribute to the association between active brown fat and a healthy metabolic status. Although a few brown adipokines have been identified, a comprehensive knowledge of the brown adipose tissue secretome is lacking.Methods: Here, to examine the effects of thermogenic activation of brown adipocytes on protein secretion, we used isobaric tags for relative and absolute quantification (iTRAQ) analysis to determine how the secreted proteome of brown adipocytes (that detected in cell culture medium) differed in response to cAMP.Results: Our results indicated that 56 secreted proteins were up-regulated in response to cAMP. Of them, nearly half (29) corresponded to extracellular matrix components and regulators. Several previously known adipokines, were also detected. Unexpectedly, we also found five components of the complement system. Only 15 secreted proteins were down-regulated by cAMP; of them three were ECM-related and none was related to the complement system. We observed a partial concordance between the cAMP-regulated release of proteins (both from proteomics and from antibody-based quantification of specific proteins) and the cAMP-mediated regulation of their encoding transcript for the up-regulated secreted proteins. However, a stronger concordance was seen for the down-regulated secreted proteins.Conclusions: The present results highlight the need to investigate previously unrecognized processes such as the role of extracellular matrix in thermogenic activation-triggered brown fat remodeling, as well as the intriguing question of how brown adipocyte-secreted complement factors contribute to the signaling properties of active brown adipose tissue.

The precise function of CERKL, a Retinitis Pigmentosa (RP) causative gene, is not yet fully understood. There is evidence that CERKL is involved in the regulation of autophagy, stress granules, and mitochondrial metabolism, and it is considered a gene that is resilient against oxidative stress in the retina. Mutations in most RP genes affect photoreceptors, but retinal pigment epithelium (RPE) cells may be also altered. Here, we aimed to analyze the effect of CERKL overexpression and depletion in vivo and in vitro, focusing on the state of the mitochondrial network under oxidative stress conditions. Our work indicates that the depletion of CERKL increases the vulnerability of RPE mitochondria, which show a shorter size and altered shape, particularly upon sodium arsenite treatment. CERKL-depleted cells have dysfunctional mitochondrial respiration particularly upon oxidative stress conditions. The overexpression of two human CERKL isoforms (558 aa and 419 aa), which display different protein domains, shows that a pool of CERKL localizes at mitochondria in RPE cells and that CERKL protects the mitochondrial network—both in size and shape—against oxidative stress. Our results support CERKL being a resilient gene that regulates the mitochondrial network in RPE as in retinal neurons and suggest that RPE cell alteration contributes to particular phenotypic traits in patients carrying CERKL mutations.

The aim of this article is to analyze the unstable identity of the main character of “Contra el amor en compañía” (1991), by Carme Riera, who as well as other stories from the same collection, shows a peculiar version about double and alter ego. The contradictory impulses of the protagonist's identity reflect the struggle between the creative, imaginative, passionate woman, in contrast to the real, domestic, mother-wife one, in a perennial struggle with her own flesh, wrapped in a body that, like her repressed sexual desire, overflows everywhere. The writer and the real woman live in a strange symbiosis of contradictory dependence: the literary Coral writes erotic poetry, while the other Coral is inexperienced and frigid in her marriage relationship, until the end, when she manages to harmonize body, sexuality and writing in homoeroticism. Coral Flora rejects heteronormative sexuality to opt for the practice of onanism through the image of the mirror, which produces a metamorphosis of herself. Our theoretical perspective is based on the assumptions of the ginocritic and, more specifically, on the concepts of identity and difference, present in the feminism of difference, which allow us to analyze the live-going development of the main character, from the internal contradiction which involves writing and living for others, to the reconciliation with herself in her own writing-sexuality.

Abstract Mitochondrial diseases due to mutations in the mitochondrial (mt) DNA are heterogeneous in clinical manifestations but usually include OXPHOS dysfunction. Mechanisms by which OXPHOS dysfunction contributes to the disease phenotype invoke, apart from cell energy deficit, maladaptive responses to mitochondria-to-nucleus retrograde signaling. Here we used five different cybrid models of mtDNA diseases to demonstrate that the expression of the nuclear-encoded mt-tRNA modification enzymes TRMU, GTPBP3 and MTO1 varies in response to specific pathological mtDNA mutations, thus altering the modification status of mt-tRNAs. Importantly, we demonstrated that the expression of TRMU, GTPBP3 and MTO1 is regulated by different miRNAs, which are induced by retrograde signals like ROS and Ca2+ via different pathways. Our data suggest that the up- or down-regulation of the mt-tRNA modification enzymes is part of a cellular response to cope with a stoichiometric imbalance between mtDNA- and nuclear-encoded OXPHOS subunits. However, this miRNA-mediated response fails to provide full protection from the OXPHOS dysfunction; rather, it appears to aggravate the phenotype since transfection of the mutant cybrids with miRNA antagonists improves the energetic state of the cells, which opens up options for new therapeutic approaches.

La resolución de los conflictos, a través de los años, se ha fundamentado en una serie de mecanismos específicos que permiten establecer soluciones a las disputas y enfrentamientos entre los actores involucrados, derivados de la cultura de los pueblos, de sus intereses, de los pactos y convenios realizados. No obstante, esa resolución de conflictos en la comunidad indígena wayúu tiene sus variantes precisas enfocadas en una praxis propia de su ámbito territorial, de conformidad con sus propias normas y procedimientos, pero cuya eficacia y legalidad depende de su condición de apego a las normas impuestas por el estado en esa territorialidad, la cual, no siempre es tomada en consideración al momento de la ocurrencia de una situación hostil. Bajo tales consideraciones, se fundamenta este estudio, con el objetivo de analizar la resolución de los conflictos al interior de la comunidad indígena wayúu. Con tales fines, se produce un estudio comparativo que versa sobre la praxis consuetudinaria y la cultura occidental. En tal sentido, se realiza una investigación documental, enfocada bajo el paradigma de investigación cualitativo, propio de las ciencias sociales, y se aplican métodos de la ciencia relativos a la hermenéutica, con la finalidad de hacer la interpretación de doctrinas y teorías recopiladas, la interpretación de las leyes que regulan la materia y el análisis de algunas entrevistas suscitadas. Los resultados encontrados, develan debilidades en cuanto a la justa implementación de las leyes nacionales, lo que obliga al investigador a formular sugerencias y recomendaciones diversas que intentan aportar ideas para racionalizar los conflictos suscitados entre las partes intervinientes en ellos.

El daño ambiental y la injusticia social son dos rasgos característicos del mundo contemporáneo. Ambos evidencian una falta de respeto hacia los valores naturales y humanos que la realidad, per se, posee. El descubrimiento de estos valores puede constituir una oportunidad para promover cambios de conducta que contribuyan a lograr una cultura más sostenible y justa. Tras examinar algunas posibles causas culturales de la insostenibilidad socio-ambiental, los autores plantean que la belleza que encierra la realidad puede desvelar a cada individuo valores naturales y humanos que antes no se percibían como tales. Este descubrimiento del valor interpela a la conducta personal e invita a la adopción libre de un compromiso de respeto hacia el mismo. Intertwined environmental and social impacts manifest a disrespect for environmental and social values intrinsic to reality. Discovering these values provides an opportunity to foster behavior changes towards a fair and sustainable culture and society. The authors analyze somecultural causes of present-day unsustainability and advance that the beauty found in nature and human beings may reveal actual natural and human values formerly unseen. Finding out these values may challenge personal behavior, inviting to commit oneself to a more respectful lifestyle.

En els últims anys, molts països i governs locals han prestat especial atenció a les circumstàncies i necessitats de les dones com a creadores i productores d'expressions artístiques diverses. La inclusió de la igualtat de gènere a l'agenda política ha emergit, en moltes ciutats, de la mà de moviments feministes i de l'activisme de professionals del sector cultural. Aquest article té com a objectiu explorar el paper de Barcelona com a ciutat laboratori de bones pràctiques en el camp de la cultura i el gènere. A partir d'un diagnòstic previ sobre les desigualtats de gènere en l'àmbit de les arts i la cultura a la ciutat de Barcelona i una recerca original enfocada des d'una metodologia qualitativa, basada en una sèrie d'entrevistes semi-estructurades amb professionals del sector referents dins l'àmbit de la cultura i el gènere, s'analitzen diferents casos d'experiències innovadores en el camp cultural localitzades en diversos barris de la ciutat. L'article conclou que, si bé els moviments feministes i el sector cultural de Barcelona han treballat per situar la igualtat de gènere a l'agenda política i institucional, encara queda un llarg camí per recórrer en la implementació d'accions programàtiques transformadores de la societat.

294. Obesity-Related Single-Nucleotide Polymorphisms and Weight Gain Following First-Line Antiretroviral Therapy

Background We studied the association of obesity-related single-nucleotide polymorphisms (OR-SNPs) with weight gain after antiretroviral therapy (ART) in people with human immunodeficiency virus (HIV; PWH). Methods Participants were ART-naive PWH from the Spanish HIV Research Cohort who started ART from 2014 onward and had blood/DNA deposited in the cohort Biobank. The primary outcome was change in weight at 96 weeks after starting ART. We genotyped 14 OR-SNPs from a meta-analysis of genome-wide association studies of body mass index (BMI) loci. Changes over time in weight and BMI were studied using adjusted linear mixed models. Results A total of 1021 PWH were included. The mean weight gain over 96 weeks was 2.90 (95% confidence interval, 2.54–3.26) kg. Factors associated with higher weight gain were female sex, birth in sub-Saharan Africa, prior AIDS, CD4+ 100 000 copies/mL, negative hepatitis C virus serology, and use of tenofovir alafenamide. A significant association was found between ZC3H4 rs3810291 GG genotype and BCDIN3D/FAIM2 rs7138803 GG genotype polymorphisms and weight and BMI increase. The estimated adjusted mean (standard error [SE]) of weight gain was 4.26 (0.56) kg in ZC3H4 rs3810291 GG carriers and 2.66 (0.19) kg in AA/AG carriers (P = .007). Likewise the estimated weight gain at 96 weeks was 3.35 (0.29) kg in BCDIN3D/FAIM2 rs7138803 GG carriers and 2.51 (0.24) kg in AG/AA carriers (P = .020). Conclusions Genetic factors may play a role in weight gain after ART initiation. Further work is needed to replicate our findings and understand how the identified SNPs lead to higher weight gain in this context.

Las competencias específicas son adquiridas por los estudiantes y certificadas mediante la superación de las diferentes materias de los planes de estudio. Sin embargo, las competencias transversales no se trabajan sistemáticamente y no se evalúan, lo que implica que no pueda garantizarse su adquisición. Sin embargo, dichas competencias son base fundamental en la formación superior y en la futura inserción laboral de los graduados universitarios. El actual paradigma metodológico de la Educación Superior muestra la importancia de la evaluación auténtica y alternativa de los aprendizajes, a partir de actividades o tareas de ejecución relevantes. El trabajo de AprenRED durante los últimos años se ha centrado en el campo de las competencias transversales. En este caso, el objetivo de este trabajo es optimizar y mejorar las estrategias e instrumentos de evaluación de las competencias transversales, en base a los resultados obtenidos en los últimos años, desde nuestra visión multidisciplinar.
