Photoexcited semiconductor nanoparticles undergo charge equilibration when they are in contact with metal nanoparticles. Such a charge distribution has direct influence in dictating the energetics of the composite by shifting the Fermi level to more negative potentials. The transfer of electrons to Au nanoparticles has now been probed by exciting TiO(2) nanoparticles under steady-state and laser pulse excitation. Equilibration with the C(60)/C(60)(-) redox couple provides a means to determine the apparent Fermi level of the TiO(2)-Au composite system. The size-dependent shift in the apparent Fermi level of the TiO(2)-Au composite (20 mV for 8-nm diameter and 40 mV for 5-nm and 60 mV for 3-nm gold nanoparticles) shows the ability of Au nanoparticles to influence the energetics by improving the photoinduced charge separation. Isolation of individual charge-transfer steps from UV-excited TiO(2) --> Au --> C(60) has provided mechanistic and kinetic information on the role of metal in semiconductor-assisted photocatalysis and size-dependent catalytic activity of metal-semiconductor nanocomposites.

An electroanalytical method has been developed to investigate the uptake of redox-active species by the humic acid substances. The Suwannee River humic acid (SHA) films were first cast on a glassy carbon electrode using an electrophoretic approach. The binding of a series of redox-active species to these SHA films was then probed using cyclic voltammetry at a rotating disk electrode. Neutral molecules such as hydroquinone and cationic species such as methyl viologen are able to bind with the humic membrane and exhibit high redox activity within the film. On the other hand, anionic species such as ferrocyanide are unable to attach themselves to the SHA films and thus exhibit negligible electrochemical activity. Cyclic voltammetric study of SHA films has also facilitated the determination of the partitioning constants and identification of the effect of coadsorbed ions (Ca2+) on the binding of redox species. The electroanalytical method described in this study opens up new avenues to examine the interactions and transport of charged species in a humic acid environment.

Carbon monoxide dehydrogenase/acetyl-CoA synthase (CODH/ACS) utilizes a unique Ni-M bimetallic site in the biosynthesis of acetyl-CoA, where a square-planar Ni ion is coordinated to two thiolates and two deprotonated amides in a Cys-Gly-Cys motif. The identity of M is currently a matter of debate, although both Cu and Ni have been proposed. In an effort to model ACS's unusual active site and to provide insight into the mechanism of acetyl-CoA formation and the role of each of the metals ions, we have prepared and structurally characterized a number of Ni(II)-peptide mimic complexes. The mononuclear complexes Ni(II) N, N'-bis(2-mercaptoethyl)oxamide (1), Ni(II) N, N'-ethylenebis(2-mercaptoacetamide) (2), and Ni(II) N, N'-ethylenebis(2-mercaptopropionamide) (3) model the Ni(Cys-Gly-Cys) site and can be used as synthons for additional multinuclear complexes. Reaction of 2 with MeI resulted in the alkylation of the sulfur atoms and the formation of Ni(II) N, N'-ethylenebis(2-methylmercaptoacetamide) (4), demonstrating the nucleophilicity of the terminal alkyl thiolates. Addition of Ni(OAc)(2).4H(2)O to3 resulted in the formation of a trinuclear species (5), while 2 crystallizes as an unusual paddlewheel complex (6) in the presence of nickel acetate. The difference in reactivity between the similar complexes 2 and 3 highlights the importance of ligand design when synthesizing models of ACS. Significantly,5 maintains the key features observed in the active site of ACS, namely a square-planar Ni coordinated to two deprotonated amides and two thiolates, where the thiolates bridge to a second metal, suggesting that 5 is a reasonable structural model for this unique enzyme.

Delhi has the highest cluster of small-scale industries (SSI) in India. There are generally less stringent rules for the treatment of waste in SSI due to less waste generation within each individual industry. This results in SSI disposing of their wastewater untreated into drains and subsequently into the river Yamuna, which is a major source of potable water in Delhi, thus posing a potential health and environmental risk to the people living in Delhi and downstream. To study the quantity, quality and distribution of heavy metals in liquid waste from industrial areas, wastewater, suspended materials and bed sediments were collected from industrial areas and from the river Yamuna in Delhi. This study has also focused on the efficiency of production processes in small-scale industries in India. Heavy metals such as Fe, Mn, Cu, Zn, Ni, Cr, Cd, Co and Pb were detected using a GBC 902 atomic absorption spectrometer. The concentration of heavy metals observed was as follows: Fe 2-212, Mn 0.3-39, Cu 0.2-20, Zn 0.2-5, Ni 0.6-6, Cr 0.2-53, Cd 0.08-0.2, Co 0.013-0.55, Pb 0.3-0.7 mg L(-1) in wastewater; Fe 5842-78 000, Mn 585-10 889, Cu 206-7201, Zn 406-9000, Ni 22-3621, Cr 178-10 533, Co 17-114, Cd 13-141, Pb 67-50 171 mg kg(-1) in suspended material; and Fe 3000-84000, Mn 479-1230, Cu 378-8127, Zn 647-4010, Ni 164-1582, Cr 139-3281, Co 20-54, Cd 37-65, Pb 228-293 mg kg(-1) in bed residues. This indicates that SSI could be one of the point sources of metals pollution in the river system.

A perimenopausal patient presented with a history of irregular vaginal bleeding. Clinical examination revealed lower abdominal mass, and the pre operative diagnosis was an ovarian cyst. At laparotomy the ovaries and tubes were normal, and the cyst was anatomically attached to the uterus with a short pedicle. There was no obvious sign of malignancy at laparotomy. The histopathology of the cyst was a benign Mullerian type cyst. Bilateral tubal ligation performed at the same time revealed normal fallopian tubes. The patient was followed up 6 weeks and 6 months later, and she remains symptom free. The unusual anatomical location of the cyst is discussed.

Polydisperse humic acid thin films on optically transparent electrodes (OTEs) have been prepared by electrophoretic deposition from a solution of Suwanee River humic acid (SHA) in ethanol/acetonitrile. The thickness of the film and the rate of deposition of SHA are dependent on the applied voltage and the concentration of the solution. Tapping-mode atomic force microscopy (TM-AFM) confirms the assembly of SHA aggregates on the electrode surface. The ability of these thin films to incorporate redox-active species such as ferrocene from solution is demonstrated by cyclic voltammetry experiments. A linear dependence of the peak current for the oxidation of ferrocene as a function of scan rate indicates that the ferrocene is incorporated into the humic membrane.

Protein synthesis in eukaryotic cytoplasm and in archaebacteria is initiated with methionine, whereas, that in eubacteria and in eukaryotic organelles, such as mitochondria and chloroplasts, is initiated with formylmethionine. In view of this clear distinction, we have investigated whether protein synthesis in the eukaryotic cytoplasm can be initiated with formylmethionine, and, if so, what the consequences are to the cell. For this purpose, we have expressed in an inducible manner the Escherichia coli methionyl-tRNA formyltransferase (MTF) in the cytoplasm of the yeast Saccharomyces cerevisiae. Expression of active MTF, but not of an inactive mutant, leads to formylation of methionine attached to the yeast cytoplasmic initiator tRNA to the extent of about 70%. As a consequence, the yeast strain grows slowly. Coexpression of the E. coli polypeptide deformylase (DEF), which removes the formyl group from the N-terminal formylmethionine in a polypeptide, rescues the slow-growth phenotype, whereas, coexpression of an inactive mutant of DEF does not. These results suggest that the cytoplasmic protein-synthesizing system of yeast, like that of eubacteria, can at least to some extent utilize formylated initiator Met-tRNA to initiate protein synthesis and that initiation of proteins with formylmethionine leads to the slow-growth phenotype. Removal of the formyl group in these proteins by DEF would explain the rescue of the slow-growth phenotype.

Tetanus toxin and the seven serologically distinct botulinal neurotoxins (BoNT/A to BoNT/G) abrogate synaptic transmission at nerve endings through the action of their light chains (L chains), which proteolytically cleave VAMP (vesicle-associated membrane protein)/synaptobrevin, SNAP-25 (synaptosome-associated protein of 25 kDa), or syntaxin. BoNT/C was reported to proteolyze both syntaxin and SNAP-25. Here, we demonstrate that cleavage of SNAP-25 occurs between Arg198 and Ala199, depends on the presence of regions Asn93 to Glu145 and Ile156 to Met202, and requires about 1,000-fold higher L chain concentrations in comparison with BoNT/A and BoNT/E. Analyses of the BoNT/A and BoNT/E cleavage sites revealed that changes in the carboxyl-terminal residues, in contrast with changes in the amino-terminal residues, drastically impair proteolysis. A proteolytically inactive BoNT/A L chain mutant failed to bind to VAMP/synaptobrevin and syntaxin, but formed a stable complex (KD = 1.9 x 10(-7) M) with SNAP-25. The minimal essential domain of SNAP-25 required for cleavage by BoNT/A involves the segment Met146-Gln197, and binding was optimal only with full-length SNAP-25. Proteolysis by BoNT/E required the presence of the domain Ile156-Asp186. Murine SNAP-23 was cleaved by BoNT/E and, to a reduced extent, by BoNT/A, whereas human SNAP-23 was resistant to all clostridial L chains. Lys185Asp or Pro182Arg mutations of human SNAP-23 induced susceptibility toward BoNT/E or toward both BoNT/A and BoNT/E, respectively.

The importance of soluble N-ethyl maleimide (NEM)-sensitive fusion protein (NSF) attachment protein (SNAP) receptors (SNAREs) in synaptic vesicle exocytosis is well established because it has been demonstrated that clostridial neurotoxins (NTs) proteolyze the vesicle SNAREs (v-SNAREs) vesicle-associated membrane protein (VAMP)/brevins and their partners, the target SNAREs (t-SNAREs) syntaxin 1 and SNAP25. Yet, several exocytotic events, including apical exocytosis in epithelial cells, are insensitive to numerous clostridial NTs, suggesting the presence of SNARE-independent mechanisms of exocytosis. In this study we found that syntaxin 3, SNAP23, and a newly identified VAMP/brevin, tetanus neurotoxin (TeNT)-insensitive VAMP (TI-VAMP), are insensitive to clostridial NTs. In epithelial cells, TI-VAMP-containing vesicles were concentrated in the apical domain, and the protein was detected at the apical plasma membrane by immunogold labeling on ultrathin cryosections. Syntaxin 3 and SNAP23 were codistributed at the apical plasma membrane where they formed NEM-dependent SNARE complexes with TI-VAMP and cellubrevin. We suggest that TI-VAMP, SNAP23, and syntaxin 3 can participate in exocytotic processes at the apical plasma membrane of epithelial cells and, more generally, domain-specific exocytosis in clostridial NT-resistant pathways.

We have investigated the effect of botulinum neurotoxin (BoNT) C1 light chain (LC) on insulin exocytosis from the clonal beta-cell line HIT-T15. In streptolysin-O permeabilized cells, the beta-cell impermeant BoNT C1 cleaved mainly syntaxin 1 and inhibited Ca2+ as well as GTPgammaS induced exocytosis. To study the effect of BoNTs in intact cells, we transiently coexpressed the BoNT LC together with a reporter gene for insulin release. BoNT C1 inhibited K+ induced insulin secretion by 95% but reduced insulin release stimulated by glucose only by 25%. Thus a component of glucose stimulated insulin release is insensitive to BoNT C1.

The effect of arachidonic acid (AA) on the activity of diacylglycerol (DG) kinase in neural membranes was investigated. When rat brain cortical membranes were incubated with 0.5 mM dipalmitin and [gamma-32P]ATP, formation of phosphatidic acid (PA) was observed. It was linear up to 5 min, and the initial rate was approximately 1.0 nmol/min/mg of protein. The DG kinase activity was stimulated twofold by 0.25 mM AA. The stimulation was apparent at the earliest time point measured (1 min) and with the lowest concentration of AA tested (62.5 microM). The stimulation was proportional to the concentration of AA up to 250 microM. AA was the most potent stimulator of DG kinase, and linolenic acid showed approximately 40% stimulation. Oleic acid showed no effect, whereas linoleic and the saturated fatty acids tested were inhibitory. AA stimulation of DG kinase was observed only with membranes of cerebrum, cerebellum, and myelin and not with brain cytosol or liver membranes. AA also stimulated the formation of PA in the absence of added dipalmitin (endogenous activity) with membranes prepared from whole brain. DG kinase of neural membranes was extracted with 2 M NaCl, which on dialysis yielded a precipitate. Both the precipitate and the supernatant showed DG kinase activity, but only the enzyme in the precipitate was stimulated by AA at concentrations as low as 25 microM. It is suggested that AA, through its effect on DG kinase, regulates the level of DG in neural membranes, which in turn regulates protein kinase C activity.

Incubation of glyceraldehyde-3-phosphate dehydrogenase (GAPD) with sodium nitroprusside (SNP) decreased its activity in concentration- and time-dependent fashion in the presence of a thiol compound, with DTT being more effective than GSH. Both forward and backward reactions were effected. Coinciding with this, HgCl2-sensitive labelling of the protein by [32P]NAD+ also increased, indicating the stimulation of ADP-ribosylation. Treatment with SNP of GAPD samples from rabbit muscle, sheep brain and yeast inactivated the dehydrogenase activity of the three, but only the mammalian proteins showed ADP-ribosylation activity. The SNP-modified protein of rabbit muscle GAPD, freed from the reagent by Sephadex filtration showed a concentration-dependent restoration of the dehydrogenase activity on preincubation with DTT and GSH. Such thiol-treated preparations also gave increased ADP-ribosylation activity with DTT, and to a lesser extent with GSH. The SNP-modified protein was unable to catalyze this activity with the native yeast enzyme and native and heat-inactivated muscle enzyme. It was possible to generate the ADP-ribosylation activity in muscle GAPD, by an NO-independent mechanism, on dialysis in Tris buffer under aerobic conditions, and on incubating with NADPH, but not NADH, in muscle and brain, but not yeast, enzymes. The results suggest that the inverse relationship of the dehydrogenase and ADP-ribosylation activities is coincidental but not correlated.
