Abstract

Studies on the glomerular epithelial cells (GEC) suggest that immune complexes formed on the cell surface are endocytosed and are subjected to processes undergone by a ligand-receptor complex. After internalization in vesicles which acidify the complexes, the release of the dissociated antibody to the medium occurs. The inability of GEC to degrade the immune complexes may cause the chronic inflammation observed subsequent to IgG trapping.

Abstract

Abstract Background Single nucleotide polymorphism (SNP) validation and large-scale genotyping are required to maximize the use of DNA sequence variation and determine the functional relevance of candidate genes for complex stress tolerance traits through genetic association in rice. We used the bead array platform-based Illumina GoldenGate assay to validate and genotype SNPs in a select set of stress-responsive genes to understand their functional relevance and study the population structure in rice. Results Of the 384 putative SNPs assayed, we successfully validated and genotyped 362 (94.3%). Of these 325 (84.6%) showed polymorphism among the 91 rice genotypes examined. Physical distribution, degree of allele sharing, admixtures and introgression, and amino acid replacement of SNPs in 263 abiotic and 62 biotic stress-responsive genes provided clues for identification and targeted mapping of trait-associated genomic regions. We assessed the functional and adaptive significance of validated SNPs in a set of contrasting drought tolerant upland and sensitive lowland rice genotypes by correlating their allelic variation with amino acid sequence alterations in catalytic domains and three-dimensional secondary protein structure encoded by stress-responsive genes. We found a strong genetic association among SNPs in the nine stress-responsive genes with upland and lowland ecological adaptation. Higher nucleotide diversity was observed in indica accessions compared with other rice sub-populations based on different population genetic parameters. The inferred ancestry of 16% among rice genotypes was derived from admixed populations with the maximum between upland aus and wild Oryza species. Conclusions SNPs validated in biotic and abiotic stress-responsive rice genes can be used in association analyses to identify candidate genes and develop functional markers for stress tolerance in rice.

Thus, we aimed to prepare novel 50:50 poly(lactic-co-glycolic acid) (PLGA)-loaded B nanoparticles (BNP) and to compare its anti-hepatocellular carcinoma (HCC) activity with parent B. Methods: BNP were synthesized and characterized using different methods such as scanning electron microscopy (SEM), fourier-transform infrared (FTIR) spectrometry and particle size analyses. Particle size of BNP was optimized through the application of the stabilizer, polyvinyl alcohol (PVA). The anti-HCC response was evaluated through in vitro cell line study using Hep-G2 cells, confocal microscopy, in vivo oral pharmacokinetics and animal studies. Further, quantitative reverse transcription polymerase chain reaction (qRT-PCR) analysis was conducted to observe the changes in the expression of specific genes. Results: Particle size of BNP was optimized through the application of the stabilizer, polyvinyl alcohol. Physicochemical characterization exhibited particle size of 257.1 nm with zeta potential -0.170 mV (optimized batch B, BNP). SEM and FTIR analyses of BNP showed that cylindrical particles of B converted to spherical particles in BNP and there were no interaction between B and used polymers. The release study of optimized BNP was highest (≥80%) than any other formulation. Later, in vitro cell culture analysis using Hep-G2 cells and confocal microscopy studies revealed that BNP had the highest inhibition and penetration properties than parent B. Oral pharmacokinetics studies using albino Wistar rats at single 100 mg dose again exhibited BNP had the higher 50% of plasma concentration (t1/2), a higher maximum plasma concentration (Cmax) and took longer to reach the maximum plasma concentration (Tmax) than parent B. Next, our in vivo study using nitrosodiethyl amine (NDEA)-induced HCC model documented BNP decreased in number of nodules, restored body weight, oxidative stress parameters, liver marker enzymes and histological architecture than parent B. Lastly, qRT-PCR studies further demonstrated that anti-HCC properties of BNP may be due to over expression of antiapoptotic caspases i.e., caspase 3 and 8. Conclusion: The prepared BNP showed a better therapeutic response against HCC and could be attributed as future candidate molecule for HCC treatment. Abstract

In an imbalanced dataset with binary response, the percentages of successes and failures are not approximately equal. In many real world situations, majority of the observations are “normal” (i.e., success) with a much smaller fraction of failures. The overall probability of correct classification for extremely imbalanced data sets can be very high but the probability of correctly predicting the minority class can be very low. Consider a fictitious example of a dataset with 1,000,000 observations out of which 999,000 are successes and 1,000 failures. A rule that classifies all observations as successes will have very high accuracy of prediction (99.9%) but the probability of correctly predicting a failure will be 0. In many situations, the cost associated with incorrect prediction of a failure is high, and it is therefore important to improve the prediction accuracy of failures as well. Literature suggests that over-sampling of the minority class with replacement does not necessarily predict the minority class with higher accuracy. In this article, we propose a simple over-sampling method which bootstraps a subset of the minority class, and illustrate the bootstrap over-sampling method with several examples. In each of these examples, an improvement in prediction accuracy is seen.

Abstract

The number of countries and cities implementing smoking bans has increased significantly over the last couple decades with smoking bans becoming more inclusive with less exemptions being given. Casinos though still have an exemption in most areas since there is uncertainty of the effect to gaming volume but there is a new call to action due to COVID-19. This study evaluates the long-term effects of a smoking ban in Kangwon Land Casino in South Korea. A time series ARIMAX model for April 2003 to May 2013 data was conducted to determine if the new smoking ban in public gaming areas significantly affected table games drop or slot coin-in, which are the two most common indicators of gaming volume for casinos. Results show that the smoking ban did not significantly impact table games drop or slot coin-in. The results of this study indicate that in the long-term a smoking ban in public areas does not significantly decrease gaming volume. These results add to the minimal studies on the long-term effects of a smoking ban while also being the first study to evaluate an Asian casino. [ABSTRACT FROM AUTHOR]

Abstract

This article aims to explore the impressive impact of emerging parameters on transient fully evolved free convective flow inside a vertical cylinder containing a porous material. The mathematical formulation of the model related to the considered physical circumstance is presented under compatible boundary conditions. Closed‐form solutions are received for the velocity field, the temperature distribution, mass flux, skin friction, and the Nusselt number in terms of Bessel functions and modified Bessel functions of the first kind. Impressive effects of parameters such as the Darcy number Da, Prandtl number Pr, viscosity ratio M, and also time t on both the velocity and temperature distribution have been explored employing graphs and tables. It is irradiated by analysis that flow erection, heat transfer rate, skin friction, and mass flux are admirably impacted by the Prandtl number, the Darcy number, viscosity ratio parameter, and time. It is found that both the velocity and temperature field profiles rise with the rising value of time and ultimately attain their steady state. Moreover, the Prandtl number and the viscosity ratio parameter reduce the velocity profiles, while the reverse phenomenon occurs with the Darcy number. [ABSTRACT FROM AUTHOR]
