We here report a case of successful endovascular therapy for penile gangrene in a patient with calciphylaxis and a long history of diabetes mellitus and end-stage renal disease on dialysis. The internal iliac artery, with 75% stenosis, was treated with balloon-expandable bare metal stent implantation, whereas the inferior gluteal artery, 75% stenosis, was treated with balloon dilatation. After endovascular therapy, the intractable penile pain immediately resolved, and penile salvage with complete wound healing was achieved 6 months after the procedure.

An attempt was made to assign five genes, CPS1, OTC, ASS, CRYD2, and ARG2, to chicken chromosomes (GGA) by radiation-hybrid mapping. OTC was assigned to GGA1; ARG2 to GGA5; CPS1 to GGA7; and CRYD2 to GGA19. ASS was not, however, assigned to a specific chromosomal position.

The purpose of this study is to derive a rational design method and connection details for socket base column-to-foundation connections for earthquake-resistant buildings. Cyclic loading tests on one-half-scale model specimens were conducted to investigate the structural behavior of socket base connections. From experimental observations, modeling of the resultant force transfer mechanism of socket base connections was also carried out. Finally, a practical design method of connection details for socket base connections was proposed using the aforementioned model.

Chymotrypsinogen is widely present in various animal pancreases. To study evolutionary relationship of chymotrypsinogen gene in species, we used a cDNA probe of human prechymotrypsinogen to investigate the species distribution of chymotrypsinogen gene, and designed oligodeoxynucleotide primers to investigate the genomic organization in the three domains of active sites. The genomic analyses showed that chymotrypsinogen gene is evolutionary conserved in species. On the basis of the deduced amino acid residues, a three-dimensional model for human chymotrypsinogen was further built by computer graphics. The model showed high similarity to the X-ray crystal structure of bovine chymotrypsinogen A, thus, demonstrated that the three-dimensional structure is more conserved in evolution than protein sequences.

International audience; DNA fingerprints of Japanese quail male and female pure line breeders were obtained with probes 33.6, 33.15, and R18.1 and they yielded a total of 59 scoreable bands. Bandsharing ($0 < {\rm BS} < 1$) was calculated within and between six quail lines of two origins, and under reciprocal recurrent (AA and BB), within-line (DD and EE) or no (PP and FF) selection. Twenty one pair types were compared. BS was 0.30 higher within line than between lines. BS with the control line was smaller for reciprocal recurrent selection lines than for lines under individual selection. Bandsharing between the two reciprocal recurrent selection lines was 0.19 lower than between lines under individual selection. These results indicate that the two selection methods had different effects on the genetic constitution of the lines, in agreement with previous observations made from the analysis of biochemical polymorphisms with the same set of birds. Egg production and weight traits of pure and crossbred progeny from fingerprinted quail were obtained and compared, and a linear relationship with the measure of bandsharing was estimated. No significant regression coefficient of performance on BS was found over all progeny genetic types. Heterosis from individual matings could also be estimated under the two selection methods since the same birds were parents of both pure and crossbred performance-tested quail. The association of heterosis with the difference between BS of parents of the purebreds and BS of parents of their half-sib crossbreds was favourable and significant for early production traits in lines DD and EE, but no relationship was found in lines AA and BB. These results indicate that the high level of heterosis obtained through reciprocal recurrent selection, and the heterosis observed under within-line selection may have, partly at least, a different genetic determinism. Therefore, the relationship of heterosis with BS may also depend on the past history of selection in the lines.; Ressemblance des empreintes génétiques et liaisons avec les performances et l'hétérosis chez des cailles japonaises de deux origines et sous sélection directe ou réciproque récurrente pour la ponte précoce. Les sondes 33.6, 33.15 et R18.1 ont été utilisées pour obtenir les empreintes génétiques à partir de l'ADN de cailles japonaises reproductrices mâles et femelles de six lignées en sélection récurrente réciproque (AA et BB), en sélection individuelle (DD et EE) ou maintenues sans sélection (PP et FF), et provenant de deux origines. Les 21 types de paires possibles ont été comparés à l'aide du coefficient de partage de bande BS ($0 < {\rm BS} < 1$). BS était plus élevé de 0,30 intra-lignée qu'inter-lignée. La ressemblance génétique avec la lignée contrôle non sélectionnée, mesurée par BS, était moindre pour les lignées AA et BB que pour DD et EE. De plus, BS entre les deux lignées sous sélection réciproque récurrente était inférieur de 0,19 à la valeur obtenue entre celles sous sélection individuelle. Ces résultats indiquent que les deux méthodes de sélection ont bien eu chacune un effet différent sur la constitution génétique des lignées. Ils confirment ainsi des observations antérieures issues de l'analyse des polymorphismes biochimiques du même échantillon d'animaux. La descendance femelle pure et croisée des cailles aux empreintes génétiques connues a été mise en épreuve de ponte de façon à étudier la relation linéaire entre les performances de chaque caille et le BS de ses parents mais aucun lien significatif n'a été mis en évidence. L'hétérosis des croisements individuels a pu être estimé car les mêmes parents ont eu une descendance pure et croisée. L'association de l'hétérosis avec la différence de BS entre les paires de cailles parentes de la descendance pure et celles parentes de la descendance de leurs demi-sœurs croisées était significative et favorable pour les caractères mesurés en début d'épreuve de ponte chez les lignées DD et EE, mais aucune relation n'a été trouvée chez les lignées AA et BB. Ces résultats indiquent que le fort hétérosis obtenu par la sélection réciproque récurrente et l'hétérosis observé dans les lignées sous sélection individuelle pourraient avoir, au moins en partie, un déterminisme génétique différent. Plus généralement, il semble que la relation de l'hétérosis avec BS puisse dépendre aussi de la sélection pratiquée dans les lignées.

A genetic variation was found in pancreatic proteinase of Japanese quail. In eight bands of proteinase isozymes, the variation of band 5 (presence or absence) was detected among quails. Band 5 was identified as a chymotrypsin. The presence and absence of band 5 are controlled by a pair of allelic genes (Prt-5A and Prt-5a) on an autosomal locus, and gene Prt-5A, causing expression of band 5, is dominant to gene Prt-5a, a null allele for band 5. Zymograms of proteinases and their zymogens were also compared; no variation of chymotrypsinogen; corresponding to chymotrypsin Prt 5 were detected. It is suggested that the Prt-5 variant of chymotrypsin may be formed during activation of chymotrypsinogen.

ニワトリ末梢リンパ球を用いて変異原性試験を行なう際の基礎情報を得る目的で,その細胞周期について分析を行なった.供試鶏には白色レグホン種を用いた.培養は,尺側皮静脈より採取した血液を分離した後,BrdUを含む培地で行なった.培養開始後24～72時間までの期間,経時的にコルセミドを加え細胞分裂の状態を調べた.培養開始後24時間では,中期細胞はほとんど観察されなかったが,分裂指数は徐々に増加の傾向を示し42時間後に11.25%に達した.その後44時間まで幾分減少したが,再び増加を始め,66時間で13.95%と培養期間中最も高い値を示した.66～72時間では分裂指数の低下が認められた.BrdU-Giemsa染色法により増殖動態を分析すると,24～36時間までは1回目の分裂中期細胞が大部分(100～80%)を占めていたが,42時間以降極端に減少し72時間ではほとんど観察されなくなった.2回目の分裂を示す細胞は30時間より出現しはじめ42時間でピーク(57.25%)に達した後,減少していった.3回目以上の分裂に移行した細胞は36時間以後に観察され,時間の経過とともに増加を続け,72時間では分裂中期細胞の86%を占めるに至った.

4種の野鶏すなわち赤色野鶏,灰色野鶏,セイロン野鶏および緑襟野鶏について,末梢リンパ球培養法により染色体数および核型の同定を試みた.血液は,採血後400-500rpmで分離を行ない,基礎培地としてRPMI-1640,分裂促進剤としてPHAを用い,39°C 5% CO2+95% Airの条件下で培養した.分析は重なりが少なく分散のない分裂中期細胞を各野鶏とも100個選び各々の細胞を写真にとり染色体数および核型を調べた.4種の野鶏はすべて2n=78の染色体数を示し,さらに核型においても4種間に著しい違いは認められなかった.78本の染色体は11対の大型染色体群と28対の微小染色体群に分類できた.大型染色体群はメタセントリック,サブメタセントリsックおよびアクロセントリックより構成されていた.一方,微小染色体群は非常に小さく個々の同定は困難であったが,おそらくアクロセントリックであろうと思われた.これらの結果より,野鶏の染色体はこれまで報告されている家鶏のものとほぼ同じであることが示唆された.

育成初期における日本ウズラの筋肉蛋白質代謝回転速度について遺伝的パラメータの分析を行なった。代謝回転速度はNτ-methylhistidine (Nτ-MH)の排せつ量により測定し,遺伝的パラメータは両親の分散共分散成分により推定した。ウズラの育成期におけるNτ-MH排せつ量,筋肉蛋白質の分解速度(Kd)および合成速度(Ks)は2週齢においては父内母家系間で,4週齢においては父家系間で有意差が見られ,両親の分散成分によるそれぞれの形質の遣伝率は2週齢で.49±.36, .55±.25および.60.26, 4週齢で.46±.25, .65±.16および.25±.21と推定された。各形質間の遺伝相関係数を計算した結果,Ksと平均一日増体量,成長速度および飼料効率との間は2週齢および4週齢共に正の高い相関を示し,KsがKdに比べて,成長や飼料効率と深い関連を有していることが示唆された。これらの結果から,筋肉蛋白質代謝回転速度は遺伝的支配を受けていることが明らかとなり,さらに,それらの代謝速度を選抜の指標として利用し得ることが示唆された。

Two experiments were conducted to elucidate the genetic variation in the liver lipid content of coturnix quail. In Experiment 1, the change of the liver lipid content with advancing age was studied. In Experiment 2, the genetic variation of the liver lipid content at 4 weeks of age was analyzed. Liver lipid was extracted with ethanol-ether solution and weighed. Genetic parameters of liver lipid content, body weight, and liver weight were estimated from sib analysis. The means of liver lipid content at 2 and 4 weeks of age were approximately 25% on a dry weight basis in both sexes. After 6 weeks of age, the liver lipid content of the female was higher than that of the male. The liver lipid content of the egg-laying female was slightly higher than that of the nonlaying female. Heritability (h2sire) of liver lipid content from 4-week-old coturnix quail was estimated as h2sire: .62 +/- .73.

Fractional rates (%/day) of degradation of muscle protein were determined by measuring the output of NT-methylhistidine (NT-MH) in the excreta at 2 and 10 weeks of age in three lines of quail, a random-bred line and two lines selected for body size, one for increased and the other for decreased size. In all lines, fractional rates of degradation of muscle protein at 2 weeks of age were higher than those at 10 weeks of age. The fractional rate of degradation at 2 weeks of age was highest for the RR line, 9.1-9.2%/day. However, at 10 weeks of age, the rank order changed, and the RR line showed the lowest rate, 1.8-1.9%/day. The SS line (5.8-6.2%/day at 2 weeks and 5.8-5.9%/day at 10 weeks of age) was significantly higher than the LL line (4.1-4.2%/day at 2 weeks and 2.1-2.2%/day at 10 weeks of age). Acid protease activities in supernatants of homogenized muscle of the three lines of quail at 2 and 10 weeks of age were measured. In all lines, the acid protease activities in supernatant of homogenized muscle decreased from 2 to 10 weeks of age. At 2 weeks, the protease activity of the RR line was significantly higher than that of the LL and SS lines, which did not differ significantly. However, at 10 weeks of age, the SS line had higher activity in both sexes than the LL and RR lines. The results suggest that selection for body size brings about significant changes in both fractional degradation rate and acid protease activity in the muscle.

Background Recurrence after catheter ablation of persistent atrial fibrillation (AF) remains an unsolved issue. This study aimed to explore the association between the left atrial appendage peak flow velocity (LAAV) and AF recurrence after ablation in persistent AF patients. Methods Fifty-three consecutive patients who underwent an initial catheter ablation of persistent AF were enrolled [age, 65 ± 10 years; male, 42 (79%)]. The LAAV was obtained by transesophageal echocardiography before ablation. All the patients underwent pulmonary vein isolation and were followed up for 12 months. The LAAV and other clinical factors (AF duration, CHA 2 DS 2 VASc score, left atrial diameter, left atrial volume, and left ventricular ejection fraction) were tested using a Cox proportional hazards regression analysis as predictors of AF recurrence during the 1-year follow-up. Results AF recurrence occurred in 16 (30%) patients. The patients with AF recurrences had lower LAAVs (23.3 ± 7.2 cm/s vs. 33.3 ± 15.1 cm/s, p = 0.002) than those without AF recurrence. In the multivariable analysis, a low LAAV independently predicted AF recurrence (hazard ratio, 3.04; 95% confidence interval, 1.05–8.79; p = 0.040). A Kaplan–Meier analysis also demonstrated a lower survival rate free from AF recurrence in the low LAAV group than in the high LAAV group ( p = 0.030). Conclusion A low LAAV was associated with AF recurrence after the initial catheter ablation of persistent AF.
