The behavior of phalangeal bone ultrasound was studied, measured by amplitude-dependent speed of sound (Ad-SOS) in meters per second, in 324 normal women (mean age 48.9 +/- 13.7 years) classified by gonadal status (premenopausal, perimenopausal and postmenopausal) and body mass index (BMI, thin, normal, overweight and obese). Ad-SOS differed significantly with gonadal status and BMI (p<0.0001 for all). In the overall group of women, Ad-SOS correlated negatively with age (r=-0.84, p<0.0001), weight (r=-0.16, p<0.005), BMI (r=-0.27, p<0.0001), and tartrate-resistant acid phosphatase concentration (TRAP) (r=-0.35, p<0.0001). The negative correlation remained significant in the groups separated by gonadal status, but to a lesser extent. After adjusting for confounding variables such as age and weight, Ad-SOS was dependent on age (but not on weight or BMI) in the overall group of women and in the gonadal status groups. In conclusion, Ad-SOS values differed significantly with gonadal status and BMI, and correlated negatively with TRAP. The plot of Ad-SOS against age differed significantly with gonadal status as well as BMI.

Objective: This paper studied the influence of several gynecological factors (years since menopause (YSM), age at menarche and gynecological age or reproductive life) simultaneously with anthropometric factors as determinants of bone mass in 189 healthy postmenopausal women., Methods: Bone mass was determined by peripheral quantitative computed tomography., Results: An overall evaluation showed that YSM correlated negatively with trabecular and cortical bone density (BMDTrab and BMDCorti) (P<0.05 in both cases). Age at menarche correlated negatively with BMDCorti (P<0.05) and gynecological age correlated positively with BMDTrab (P<0.05). Classifying the women according to their body mass index (BMI), the YSM correlation persisted in those subjects whose BMI was >25 kg/m(2), and in age at menarche and gynecological age of women whose BMI was <25 kg/m(2) (P<0.05). After separating women according to their age at menarche, their gynecological age and BMI, the only significant difference that persisted was in BMDTrab which was lower in the group with gynecological age <33 years, with a BMI <25 kg/m(2) (P=0.020). Parity and smoking had no impact on our results. By multiple regression, with BMD as the dependent variable and the gynecological factors as independent variables, we only observed significance between YSM and BMDCorti (P<0.005). The same was observed after separating women according to their BMI in the >25 kg/m(2) group (P<0.05)., Conclusions: Our data stress the importance of YSM on BMDTrab and BMDCorti, of age at menarche on BMDCorti and of gynecological age on BMDTrab. However, YSM is the gynecological factor that mainly determines BMD. The differences observed between measurements taken with pQCT and other methods commonly used to estimate bone mass indicate that results obtained with one technique cannot be extrapolated to other methods.

Objective: The aim of this study was to examine the effect of manganese (Mn) alone and with the addition of copper (Cu) in the inhibition of osteopenia induced by ovariectomy (OVX) in rats. STUDY CONDITIONS: Four lots of 100-day-old female Wistar rats were divided into experimental groups of 15 each. One group received a diet supplemented with 40 mg/kg of Mn per kilogram of feed (OVX+Mn). The second group received the same diet as the first, but with an additional 15 mg/kg of copper (OVX+Mn+Cu). The third group of 15 OVX and the fourth group of 15 Sham-OVX received no supplements. At the conclusion of the 30-day experiment, the rats were slaughtered and their femurs and fifth lumbar vertebrae were dissected. Femoral and vertebral length were measured with caliper and bones were weighed on a precision balance. The bone mineral content (BMC) and bone density (BMD) of the femur (F-BMC, mg and F-BMD, mg/cm(2)) and the fifth lumbar vertebra (V-BMC, mg and V-BMD, mg/cm(2)) were measured separately with dual energy X-ray absorptiometry., Results: The F-BMD, mg/cm(2) was lower in the OVX than in the Sham-OVX group (P<0.0001) and in the other two groups receiving mineral supplements (P<0.005 in both). F-BMC, mg was significantly lower in the OVX group than in the other three (P<0.0001 in all cases). Calculations for V-BMC, mg and V-BMD, mg/cm(2) are similar to findings in the femur., Conclusions: These data show that a Mn supplement is an effective inhibitor of loss of bone mass after OVX, both on the axial and the peripheral levels, although this effect is not enhanced with the addition of Cu.

The effect of silicon (Si) supplement on preventing bone mass loss induced by ovariectomy (OVX) in rats was investigated. Three groups of 15, 100-day-old female Wistar rats each, with a mean initial weight of approximately 260 g per animal, were selected for the present study. One of the experimental group consisting of 15 OVX rats was fed a diet supplemented with 500 mg of Si per kg of feed (Si + OVX). The other two groups consisting of 15 OVX and 15 sham-OVX rats did not receive these supplements. Morphometric (weight and length) and densitometric studies with dual-energy X-ray absorptiometry were performed on the whole femur and 5th lumbar vertebra of each animal 30 days after the experiment. The Si + OVX rats did not show a loss of bone mass induced by OVX at axial level (5th lumbar vertebra) or periphery (femur). Nonetheless, a significant increase (ANOVA with Bonferroni/Dunn post hocs test) of longitudinal development of the femur (P < 0.0001) was patent. These results, obtained through the measurements of axial and peripheral bones, warrant closer scrutiny in connection with the Si inhibitory effect on bone mass loss as well as the stimulatory effect on bone formation. Both actions, namely, inhibition of resorption and stimulation of formation, infer that Si may have a potential therapeutic application in the treatment of involutive osteoporosis.

The effects of salmon calcitonin and clodronate were compared in ovariectomised rats. Sixty female Wistar rats ( 260 g in weight) were fed the same diet and had the same living conditions. The rats were divided into the following groups: 15 rats with sham ovariectomy and no drug treatment (Sham-OVX); 45 rats with bilateral ovariectomy subdivided into 15 rats not receiving drug treatment (OVX group), 15 rats treated with subcutaneous salmon calcitonin, 2 U/kg/day every 2 days (OVX + CT group) and 15 rats treated with subcutaneous clodronate, 5 mg/kg/day every 2 days (OVX + Cl group). Sixty days after surgery, the rats were sacrificed and their femurs and fifth lumbar vertebrae were dissected and cleaned of soft tissue. Femur length, vertebral height, and bone mineral content and bone mineral density of the femur and fifth lumbar vertebra by dual-energy X-ray absorptiometry were measured. Calcitonin had a significant and stronger effect in preventing ovariectomy-induced osteopenia in the femur (OVX + CT vs OVX groups, p < 0.0001); both calcitonin and clodronate had a significant effect on the fifth lumbar vertebra, which was greater in the calcitonin group (OVX + CT vs OVX + Cl groups, p<0.005). These findings indicate that calcitonin has a protective effect on both the axial (trabecular bone) and peripheral (cortical bone) skeletons, but clodronate only has a protective effect on the axial skeleton.

The effect of promethazine on bone is debated. We studied the effect of promethazine on bone and the mechanism of action involved by densitometric and histomorphometric measurements in female Wistar rats (100 days old, mean weight 25 +/- 20 g). A control group of 15 rats was not manipulated. An experimental group of 15 rats were ovariectomized (OVX) at 100 days of life and fed a diet supplemented with 4.8 mg/kg promethazine hydrochloride (OVX + Prom). The group that underwent OVX and a group of 15 rats that underwent sham ovariectomy (Sham-OVX) were not treated with promethazine. After 30 days, all the rats were killed. Their femur and 5th lumbar vertebra were dissected and cleaned of soft tissue. Femoral length and vertebral height were measured with a caliper and bones were weighed on a precision balance. The bone mineral content (BMC) and bone mineral density (BMD) of the whole right femurs and 5th lumbar vertebras were measured by dual-energy X-ray absorptiometry (DXA). Trabecular bone volume (Cn-BV-TV%), trabecular number (Tb-N mm(-1)), trabecular thickness (Tb-Th microm), and trabecular separation (Tb-Sp microm) were measured in the femurs by histomorphometric study of nondecalcified bone. Our results showed that promethazine significantly inhibited postovariectomy loss of bone mass (P < 0. 0001) by significantly reducing bone resorption, as shown by the smaller trabecular spaces observed in the treated OVX rats (P < 0. 0001).

We administered a potassium bicarbonate supplement to rats on strenuous treadmill training in order to determine the effect on bone mass and the metabolic acidosis seen with this type of training. A sample of 45 93-day-old female Wistar rats with a mean initial weight of 267 +/- 17 g were studied. The control group (15 rats) was not exercised or given potassium bicarbonate (Ex- PB-). The experimental group (30 rats) was randomly divided into two subgroups of 15 rats each, one that exercised and did not receive potassium bicarbonate (Ex+ PB-) and one that exercised and received potassium bicarbonate (Ex+ PB+), at a dose of 0.05 mg/kg/day administered by esophageal catheter on exercise days. Training consisted of treadmill running on 5 out of 7 days for a period of 11 weeks. Running time, treadmill speed, and the percent grade were gradually increased until week 7, then maintained until rats were sacrificed at the ened of 11 weeks. The bone mineral content (BMC) and bone mineral density (BMD) of the whole right femur and 5th lumbar vertebra were measured. Femoral and vertebral length were also measured. Femur length, weight, BMC, and BMD, and femur BMC/final weight ratio, and vertebral weight, BMD, and BMC, and vertebral BMC/final weight ratio were lower in the Ex+ PB- group than in either the controls or the Ex+ PB+ group (P < 0.01-P < 0.0001); the length of the 5th lumbar vertebra did not differ between groups.

Rationale and Objectives: The authors determined the effect of obesity on measurements of amplitude-dependent speed of bone ultrasound (Ad-SOS [m/sec]) and compared them to the total body bone mineral content (TBBMC/g)., Methods: A total of 25 women were studied (mean age 41.8 +/- 10.2 years). In all the subjects, body mass index (BMI) exceeded 30 kg/m2 (range, 31.12-47.47 kg/m2); mean body weight was 104 +/- 17 kg. Ad-SOS was measured at the proximal phalanges and TBBMC in whole body with dual-energy x-ray absorptiometry., Results: Correlation study (Fisher's r to z) showed that Ad-SOS correlated negatively with weight (r = -0.85, P < 0.0001) and with TBBMC (r = -0.71, P < 0.0001). The correlation between TBBMC and weight was r = 0.76, P < 0.0001. Body fat percentage correlated partially with TBBMC (r = 0.40, P < 0.05) and negatively with Ad-SOS (r = -0.75, P < 0.0001). When the correlation test was adjusted for weight (partial correlation), the correlation between Ad-SOS and TBBMC was not significant (r = -0.21, P = NS), and the correlation between Ad-SOS and weight continued to be inversely significant (r = -0.67, P < 0.0001)., Conclusions: The results showed a clearly negative effect of weight on Ad-SOS measurements and indicated the limitations of this technique when employed in overweight and obese patients. Broad-band ultrasound attenuation and speed of sound, two commonly measured variables in bone ultrasound studies, may be differently affected by soft tissue.

The ability of alprazolam to diminish cortisol response and favor ovarian function could make it useful in the prevention of osteopenia in athletes in selected cases. A sample of 45 female Wistar rats, all 93 days old and with a mean initial weight of 267 +/- 17 g, were studied. Rats were exposed to a high-performance level of exercise and were divided into two groups-one group received an alprazolam supplement and one did not-and compared with controls to determine the effect of alprazolam on bone mass as measured by dual-energy X-ray absorptiometry (DKA). Exercise consisted of treadmill running on 5 out of 7 days during a period of 11 weeks. A steep grade treadmill inclination was used to stimulate high-intensity muscle activity. Final inclination was 17.5 degrees and treadmill speed was 45 cm/second. Upon completion of the experiment, all the rats were killed and the femur and 5th lumbar vertebra were dissected and cleaned. Length, weight, bone mineral content (BMC), and density (BMD) of the whole right femur and 5th lumbar vertebra were measured. In the exercise only group (no alprazolam), the length, weight, BMC, BMD, and femur BMC/final rat weight ratio of the femur, and the vertebral weight, vertebral BMD and BMC, and vertebral BMC/final rat weight ratio were lower than in the control and the exercise-alprazolam groups (P < 0.0167 - < 0.0001). Alprazolam preserves bone mass in rats exposed to intense exercise.

Bone metabolism parameters were studied in 18 elite marathon runners (11 men and 7 women) who participated in the Marathon World Cup held at San Sebastian, Spain in 1993. Measurements were made before the race, immediately after the race, and 24 hours after the race. The most interesting finding was increased alkaline phosphatase (P < 0. 0001) and decreased tartrate-resistant acid phosphatase (P = 0.0035), which suggests that exercise produced uncoupling of the bone cell metabolism. Serum calcium corrected for proteins did not increase with exercise and at the end of the race there was a negative correlation between cortisol, which was significantly higher (P < 0. 0001), and corrected serum calcium (r = 0.53, P = 0.026) that was not present at baseline. Running time showed a significant negative correlation with baseline serum cortisol (r = -0.67, P = 0.0015) and a significant positive correlation with body mass index (r = 0.53, P = 0.0207). The increase in alkaline phosphatase persisted 24 hours after the race, which suggests that exercise produced an intense and sustained effect on osteogenic capacity.

Objective: To evaluate the effect of surgical uterine retroversion on bone mass in rats., Study Design: Forty-five female Wistar rats were assigned randomly to three groups: 15 unmanipulated rats, 15 rats that underwent uterine retroversion, and 15 rats that underwent sham uterine retroversion (exposure of the uterus to air followed by closure of the abdominal cavity). Sixty days later the rats were killed and their femurs were dissected. Femurs were weighed and measured, and femoral bone mineral content (BMC) and bone mineral density (BMD) were determined by dual energy X-ray absorptiometry., Results: In the group of rats that underwent uterine retroversion, BMC, BMD, and BMC corrected for final body weight were significantly lower (P<0.001) than in the unmanipulated control and sham uterine retroversion groups., Conclusion: Our findings indicate that uterine retroversion induced a loss of bone mass. We could not determine the mechanism of bone loss; in our opinion, these problem merits further investigations, which currently occupy our interest.

Intramucosal pH (pHi) in splanchnic organs is a reliable index of local tissular perfusion, and can be measured by tonometry. At the Surgical Intensive Care Unit we used tonometry to determine tissular perfusion in patients who underwent major digestive surgery. We report a prospective study of 20 patients with elective and emergency surgery. All of them underwent gastric tonometry and 10 of them, who had colonic disease, also underwent sigmoid tonometry. The values below pHi = 7.30 were associated with increased morbidity and mortality.

Objective: Longitudinal changes in bone mass were evaluated with use of ultrasonography and bone remodeling markers in 40 normal pregnant women in relation to their calcium intake., Study Design: The study took place at the University of Alcalá Hospital in Madrid. Biochemical markers of bone remodeling and ultrasonographic bone propagation velocity in the proximal phalanxes of fingers 2 to 5 were measured in all three trimesters of pregnancy. Wilcoxon, unpaired and paired t tests, and analysis of variance were used., Results: Ultrasonographic bone propagation velocity (meters per second) was lower in the second and third trimesters of pregnancy (p < 0.0001) compared with the respective preceding trimesters and in the third trimester in the overall group of pregnant women. Tartrate-resistant acid phosphatase and alkaline phosphatase levels increased significantly (p < 0.0001) in parallel with the ultrasonographic bone propagation velocity decrease., Conclusions: Gestation was accompanied by a reduction in ultrasonographic bone propagation velocity that was greater in women with low calcium intake.

Based on the hypothesis that the underlying osteoporotic mechanism of Colles' fracture in postmenopausal women is similar to that of other osteoporotic fractures, that is, cortical bone resorption as opposed to cancellous bone resorption, the rate of corticoendosteal bone loss was compared in 40 normal postmenopausal women [average age 68.4 +/- 7.1 years; 20 +/- 4 years since menopause (YSM)], in 35 postmenopausal women with Colles' fracture (age 69.4 +/- 7.5 years, 22 +/- 8 YSM), in 35 normal postmenopausal women with vertebral crush fracture (age 69.4 +/- 7.5 years, 22 +/- 8 YSM, and in 35 normal premenopausal women (age 36.1 +/- 7.9 years). Radiogrammetry by digital radiography of the second metacarpal was used to measure external (ED) and internal (ID) diameter, cortical thickness (CCT), cortical area (CA), and the ratio of cortical area to total area (CA/TA). The ID values of the groups of postmenopausal women were subtracted from the ID value of the premenopausal women and the result was divided by YSM to obtain the rate of corticoendosteal resorption/year (DeltaC), CA resorption year (DeltaCA) and CA/TA resorption/year (DeltaCA/TA). ID, DeltaC, DeltaCA, and DeltaCA/TA all were larger in the postmenopausal women with Colles' and vertebral crush fractures than in the normal postmenopausal women (ANOVA: all P < 0.0001). ID, CCT, DeltaC, CA, DeltaCA, and DeltaCA/TA did not differ between the two groups of postmenopausal women with fractures. DeltaC was 87% greater in postmenopausal women with vertebral crush fracture and 116% greater in women with Colles' fracture than in normal postmenopausal women. These results indicate that the loss of cortical bone is an important factor in Colles' fracture in postmenopausal women.

The Candida albicans MKC1 gene encodes a mitogen-activated protein (MAP) kinase, which has been cloned by complementation of the lytic phenotype associated with Saccharomyces cerevisiae slt2 (mpk1) mutants. In this work, the physiological role of this MAP kinase in the pathogenic fungus C. albicans was characterized and a role for MKC1 in the biogenesis of the cell wall suggested based on the following criteria. First, C. albicans mkc1 delta/mkc1 delta strains displayed alterations in their cell surfaces under specific conditions as evidenced by scanning electron microscopy. Second, an increase in specific cell wall epitopes (O-glycosylated mannoprotein) was shown by confocal microscopy in mkc1 delta/mkc1 delta mutants. Third, the sensitivity to antifungals which inhibit (1,3)-beta-glucan and chitin synthesis was increased in these mutants. In addition, evidence for a role for the MKC1 gene in morphological transitions in C. albicans is presented based on the impairment of pseudohyphal formation of mkc1 delta/mkc1 delta strains on Spider medium and on the effect of its overexpression on Sacch. cerevisiae colony morphology on SLADH medium. Using the two-hybrid system, it was also demonstrated that MKC1 is able to interact specifically with Sacch. cerevisiae Mkk1p and Mkk2p, the MAP-kinase kinases of the PKC1-mediated route of Sacch. cerevisiae, and to activate transcription in Sacch. cerevisiae when bound to a DNA-binding element. These results suggest a role for this MAP kinase in the construction of the cell wall of C. albicans and indicate its potential relevance for the development of novel antifungals.

Three techniques of bone mass measurement were evaluated in the diagnosis of postmenopausal osteoporosis; the overlap in the measurements and the capacity for discriminating was determined among 51 postmenopausal normal (mean age 66.6 +/- 8.4 years) and 42 postmenopausal osteoporotic women (mean age 68.5 +/- 7.5 years). All bone mass was evaluated by total body bone mineral content (BMCTB), density (BMDTB), ultrasound bone velocity (UBV) in proximal phalanxes 2-5 of the nondominant hand (UBV = mean value of all ultrasound measurements), and peripheral quantitative computed tomography of the nondominant forearm (pQCT). BMCTB was found to be significantly better (P < 0.0001) for diagnosing postmenopausal osteoporosis than the other methods; both cortical and trabecular pQCT measurements were more discriminating than the corresponding UBV measurements (P < 0.001). T-score values in normals, subjects versus osteoporotic ones were BMCTB -1.15 +/- 0.79 versus -3.17 +/- 0.74; BMDTB -1.01 +/- 0.97 versus -3.28 +/- 0.81; UBV -1.51 +/- 1.02 versus -2.34 +/- 1.21; trabecular-pQCT -0.40 +/- 0.72 versus -1.57 +/- 0.37; cortical-pQCT -1.00 +/- 0.87 versus -2.67 +/- 0.53; and total-pQCT -0.65 +/- 1.01 versus -2.34 +/- 0.27, respectively. The overlap in values between the postmenopausal normal and postmenopausal osteoporotic groups was 50% with UBV, 6% with BMCTB, 9% with BMDTB, 25% with cortical pQCT, and 42% with trabecular pQCT. BMCTB, BMDTB, UBV, and pQCT correlated well with each other as measurements of bone mass, but BMCTB was more discriminating than the other measurements in the diagnosis of osteoporosis.

Rationale and Objectives: The authors determine the relative effect of soft-tissue compartments, body fat (percent [%Bfat] and weight [Bfat kg]) and fat-free mass (FFM kg), on measurements of ultrasound bone velocity (UBV m/second)., Methods: The authors measured UBV in proximal phalanxes and body fat and fat-free mass by near infrared interactance in 40 healthy premenopausal women (mean age +/- standard deviation 28.2 +/- 3.8 years)., Results: Correlation study (Fisher's r to z) showed that UBV correlated negatively with %Bfat (r = -0.61, P < 0.0001), Bfat kg (r = -0.56, P = 0.0001) and marginally with body weight (r = -0.33, P = 0.0403), but did not correlate with FFM kg or H2O L (both r = -0.08, P not significant). When the correlation test was adjusted for weight and age (partial correlation), the negative correlation between UBV and %Bfat persisted (r = -0.54, P < 0.0005; and r = -0.63, P < 0.0001, respectively) and the correlation with FFM kg, adjusted for weight, became positive and significant (r = 0.55, P < 0.0005)., Conclusions: These results, to our knowledge, are the first to be obtained by in vivo evaluation of UBV in relation to body fat and fat-free mass. Body fat, but not fat-free mass, was the main factor affecting UBV. This confirms the deficiency of UBV measurements, considering that obesity is a protective factor for bone mass.

The influence of body mass index (BMI) on T scores for total body bone mineral content (TBBMC) and regional bone mineral content (RBMC) was studied in 186 healthy women: 100 postmenopausal, 35 perimenopausal, and 51 premenopausal. The three groups were divided by BMI >25 kg/m2 and BMI <25 kg/m2 and the postmenopausal women were further subdivided by years since menopause (YSM): <10, 10-20, and >20. Tartrate-resistant acid phosphatase (TRAP) concentration was higher in perimenopausal and postmenopausal women with BMI <25 kg/m2 (P < 0.001). T scores for TBBMC and for axial or peripheral RBMC differed (P < 0.05 in all) between women with BMI >25 kg/m2 and BMI <25 kg/m2. The rate of perimenopausal and postmenopausal age-related slope of BMC, as reflected in all measurements, differed with BMI. In the overall group of women, the T score for TBBMC correlated significantly with BMI (r = 0.46, P < 0.0001); this correlation increased when adjusted for age (r = 0.62, P < 0.0001). BMI correlated with TRAP only in postmenopausal women (r = 0.57, P < 0. 0001). Yearly TBBMC decline was twice as high in postmenopausal women with BMI <25 kg/m2 (P = 0.0004) than in those with BMI >25 kg/m2; the decline of trunk RBMC was more significant (P < 0.0001). These findings confirm the influence of BMI and gonadal status on bone mass.

Changes in total body bone mineral content (BMCTB) and density (BMDTB), and the T-score of BMCTB and (BMDTB) were evaluated in relation to the number of vertebral fractures in women with postmenopausal osteoporosis for the purpose of defining deviations in these parameters that could be predictive of the occurrence of vertebral fracture. The study group consisted of 62 women diagnosed with postmenopausal osteoporosis. All of them had two or more spinal fractures. Regression analysis of the number of fractures against each parameter studied indicated that the following were predictive of the risk of fracture: a reduction of -0.5 in the T-score of both BMCTB and BMDTB (P < 0.0001) and a loss of about 135 g of BMCTB or 0. 058 g/cm2 of BMDTB (P < 0.0001). The fact that such changes were found during the follow-up of women with osteoporosis highlights the importance of bone mass measurements during follow-up.

The effect of electromagnetic fields on bone is debated. In an experimental study of this effect, we compared two lots of growing female rates (both lots n = 15, age 3 weeks, average weight 23.2 +/- 3.3 g), one of which was exposed to a 3-mT, 100-Hz, Helmholtz-type electromagnetic field for 24 hours a day for 30 days, and the other of which served as the control. Bone development and bone mass were evaluated by morphometry, densitometry, and histomorphometry. The rats were killed at 30 days and weighed. The right femurs were dissected, measured, and weighed; bone densitometry was used to determine femoral bone mineral content (BMC) and density (BMD), and histomorphometry of the nondecalcified bone was used to determine trabecular bone volume (Cn-BV-TV%), number (Tb-N mm) and thickness (Tb-Th microm), intertrabecular space (Tb-Sp microm) and growth cartilage thickness (Gc-Th microm). In the rats exposed to the electromagnetic field, BMC and BMD (P = 0.019 and P = 0.002, respectively) and Cn-BV-TV, Tb-N, Tb-Th (P = 0.005, P = 0.036, and P = 0.027, respectively) all were decreased, whereas Tb-Sp was increased (P = 0.002). There were no significant differences in initial and final body weight, or in final femur weight, femur length, and GC-Th. These findings indicate that electromagnetic fields of the type used here reduced bone formation and increased bone resorption without affecting bone development in rats.

In 45 women with Colles' fracture, two types of complementary medical treatment (calcitonin with calcium [SCT+Ca] and calcium alone [Ca]) were compared with placebo. Consecutive patients were assigned randomly to one of the three study groups at the time of inclusion in the study: 15 women (68.6 +/- 5.7 years) were given 100 IU/day I.M. of SCT plus 1200 mg of elemental Ca for 10 successive days each month; 15 women (71.7 +/- 6.1 years) were given only 1200 mg of elemental Ca for 10 days each month; and 15 women (66.9 +/- 7. 9 years) were treated with placebo. Biochemical and radiogrammetric studies were made at baseline and after 1 year of treatment. In the SCT+Ca group tartrate-resistant acid phosphatase decreased (Wilcoxon test, P = 0.014) and the metacarpal index and the cortical and total area (CA/TA) ratio increased (both P = 0.001). In the group treated with Ca alone, no changes were observed. In the placebo group, the metacarpal index and CA/TA decreased (P = 0.015 and P = 0.007, respectively). Ca alone, at the dosage used here, inhibited bone loss after Colles' fracture. The addition of SCT to Ca administration not only impeded bone loss but significantly increased cortical bone mass.

Because of the importance of nutrition in the development of bone mass, we studied the nutritional state, and bone state by means of metacarpal radiogrammetric measurements and vertebral morphometry in a group of 40 premenopausal women born between 1960 and 1970, mean age 29 +/- 5 years, and in another group of 40 postmenopausal women born between 1934 and 1944, mean age 55 +/- 4 years. Both groups were considered normal, the main characteristic distinguishing them being that the women born between 1934 and 1944 grew up in a period of widespread malnutrition in Spain and the women born between 1960 and 1970 grew up in a period of normal nutrition. Protein, carbohydrate and fat intake in these two periods differed significantly (p < 0.0001 in the three cases by Fischer's exact test). The values of the metacarpal measurements, anterior height of the dorsal vertebrae from T-4 to T-12, and posterior height from L-1 to L-4 between the premenopausal and postmenopausal groups of women were significantly different (p < 0.001) (Anova test). These findings show the importance of nutrition in the development of bone mass during childhood.

A study was made of 110 women: 35 healthy premenopausal, 40 healthy postmenopausal, and 35 women diagnosed as having postmenopausal osteoporosis. The postmenopausal women had similar ages and years since menopause (YSM). In all of the women, total bone mass was evaluated by dual-energy X-ray absorptiometry and metacarpal morphometry was evaluated by radiogrammetry on the second metacarpal of the nondominant hand, performed by computed radiography. An external metacarpal diameter of >/=7.4 mm was required as proof of having developed an adequate peak bone mass. The endosteal diameter, which is indicative of bone resorption in both groups of postmenopausal women, obtained in the postmenopausal groups was subtracted from the endosteal diameter obtained in the premenopausal group and the resulting figure was divided by the years since menopause to calculate the rate of cortical bone resorption/year for each group. The endosteal diameters values differed in the three groups studied (P < 0.0001): 3.2 +/- 0.7 mm in the healthy premenopausal women; 3.9 +/- 0.6 mm in the healthy postmenopausal women; and 4.7 +/- 0.5 mm in the osteoporotic postmenopausal women. The rate of cortical bone resorption was 0.068 +/- 0.002 mm/YSM (years since menopause) in the osteoporotic postmenopausal women and 0.033 +/- 0.003 mm/YSM in the healthy postmenopausal women (P < 0. 0001). These figures reflect the importance of bone resorption, as opposed to deficient bone formation, as a cause of osteoporosis.

The effect of exposure to lead on the longitudinal development of bone and on bone mass was studied in rats. A group of 35, 50-day-old female Wistar rats was divided into a control group of 15 rats and an experimental group of 20 rats fed a diet supplemented with 17 mg of lead acetate per kg feed for 50 days. Total body bone densitometry (TBBMC) was performed the day before ending the 50-day experiment. On day 50, all rats were killed and their right femur and 5th lumbar vertebra were dissected. The bones were cleaned of soft tissue and femoral length and vertebral length were measured with a caliper and all bones were weighed on a precision scale. Final body weight (P < 0.05), TBBMC (P < 0.005), and femur weight (P < 0.005) were significantly lower in the control group. Femur length did not differ between groups, but the length of the 5th lumbar vertebra was greater in the control group (P < 0.05). Histomorphometry of the femur showed that Cn-BV/TV, Tb-N, Tb-Th were lower (P < 0.05 in all) and Tb-Sp was higher (P < 0.05) in the group given the lead-supplemented diet. These findings suggested lead-induced inhibition of axial bone development and a histomorphometric decrease in bone mass, produced mainly by enhanced resorption, and a densitometric increase in bone mass, produced by lead accumulation in bone.

Candida albicans cell wall components were analyzed by ethylenediamine (EDA) treatment. Based on their different solubility properties, the cell wall components produced three fractions (A, B, and C). Fractions B (EDA-soluble, water-insoluble) and C (EDA-insoluble) contained glucan, chitin, and protein in different proportions. After zymolyase (mainly a beta-glucanase complex) or chitinase treatment of fractions B and C, more polysaccharides and proteins were solubilized by a second EDA treatment, suggesting that the solubility of the polymers in EDA depends on the degree of polymer interactions. Western blot analysis using two monoclonal antibodies (1B12 and 4C12) revealed electrophoretic patterns that were similar in mycelial and yeast morphologies, except that in material obtained from mycelial walls, an additional band was detected with MAb 1B12. Fluorescence microscopy of cell wall fractions treated with FITC-labeled Con-A, Calcofluor white, and FITC-labeled agglutinin showed that glucan and mannoproteins are uniformly distributed in fractions B and C, while chitin is restricted to distinct patches. Transmission electron microscopy demonstrated that fraction C maintained the original shape of the cells, with an irregular thickness generally wider than the walls. When fraction C was treated with chitinase, the morphology was still present and was maintained by an external glucan layer, with an internal expanded fibrillar material covering the entire cellular lumen. Degradation of the glucan skeleton of fraction C with zymolyase resulted in the loss of the morphology.

Having observed that serum beta(2)-microglobulin concentration correlates with serum tartrate-resistant acid phosphatase (TRAP) concentration in postmenopausal osteoporosis, and that metacarpal endosteal diameter is dependent on bone resorption, we correlated the two biochemical parameters with the radiographic parameter to determine if beta(2)-microglobulin behaves like a biological marker of bone remodelling. In 105 women (mean age 68 +/- 4 years) consisting of 60 normal postmenopausal women and 55 osteoporotic postmenopausal women, there was a significant positive correlation between metacarpal endosteal diameter and these two biochemical values (r = 0.66 with beta(2)-microglobulin and r = 0.68 with TRAP in the osteoporotic postmenopausal women; r = 0.48 with beta(2)-microglobulin and r = 0.56 with TRAP in the normal postmenopausal women; P < 0.001 for all comparisons). All three measurements were significantly higher (P < 0.001) in the osteoporotic postmenopausal women than in the normal postmenopausal women. These findings show that serum beta(2)-microglobulin behaves like a biological marker of remodelling.

Recent reports suggest the presence of osteopenia in a high percentage of patients with Parkinson's disease (PD). These data contrast with previous reports of our group, perhaps due to the different methods used. We studied bone mass in 52 PD patients (28 males, 24 females) and in 80 age- and sex-matched controls (40 males, 40 females) who had no other disease that could affect bone mass. We measured the totally body bone mineral content (TBBMC) and the ultrasound bone velocity (UBV) of transmission in phalanx, and performed metacarpal radiogrammetry with computerized radiography (CCT). We also measured serum levels of total alkaline phosphatase and tartrate-resistant acid phosphatase, which were significantly increased in PD patients compared with controls (P < 0.0001). TBBMC was significantly lower in males (P < 0.05) and females (P < 0.05) with PD with respect to their controls. CCT did not differ significantly between the study groups. UBV was significantly lower in males with PD (P < 0.005), but similar in female PD and controls. These data suggest that the changes reported in bone mass in PD patients can depend on the sex and the study methods. We only found severe osteopenia in one male (3.6%) and five females (20.8%) with PD according to z-score. Only in women was no relationship found between TBBMC and severity of PD.

Rationale and Objectives: Bone mass as measured by dual energy x-ray absorptiometry varies with body weight. The authors studied the behavior of bone mass measurements made by ultrasound bone velocity and metacarpal radiogrammetry in relation to body weight., Methods: Eighty healthy women were studied: 40 post-menopausal and 40 premenopausal (mean age 60 +/- 6 and 38 +/- 8 years, respectively). The authors performed in every subject the following studies: a radiograph of the nondominant hand for metacarpal cortical thickness, a study of the 2nd to 5th proximal phalanges for ultrasound bone velocity, and total bone mineral content by dual energy x-ray absorptiometry., Results: The measurements obtained with the three methods correlated significantly with each other (P < 0.0001). The only parameter related significantly with weight were total bone mineral content (P < 0.0001). In the premenopausal women, age did not correlate with any measurement. In the postmenopausal women, age correlated significantly with the three measured parameters (P < 0.0058 to P < 0.0001)., Conclusions: Weight did not influence ultrasound bone velocity and metacarpal cortical thickness measurements.

Total body bone mineral content (TBBMC) and regional bone mineral content (BMC) was measured in the members of the Spanish National Dance Company (15 female, mean age 25.1 +/- 3.8 years, and 15 male, mean age 28.2 +/- 2.1 years) and in 30 controls (15 women, mean age 26.1 +/- 1.8 years, and 15 men, mean age 28.0 +/- 1.5 years). Ca, P, and Mg intake were greater in the group of ballet dancers than in controls analysis of variance (ANOVA, all P < 0.0001). BMC was similar in the group of ballet dancers and controls except in the trunk without pelvis (P < 0.001). Both male and female dancers weighed less than controls (P < 0.05). The BMC of the male dancers was less than that of male controls only in the trunk (P < 0.05) and in the trunk without pelvis (P < 0.005); BMC was lower in female dancers than in female controls only in the arms and in the trunk without pelvis (P < 0.05 and P < 0.005, respectively). TBBMC, adjusted for weight and age, was correlated partially with caloric intake (kcal/day) and with Ca, P, Mg, and Zn intake (g/day), and yielded significant differences between the dancers and controls only in P intake (P < 0.01), and between male dancers and male controls only in caloric intake and in Ca, P, and Zn intake (all P < 0.01, except for Ca, P < 0.05). The lower trunk bone mass observed in the female dancers is a risk factor for eventual osteoporosis.

Objective: This work studies the effects of Cyclosporine A (CsA) upon the activation and proliferation of mononuclear cells (MNC) from the peripheral blood (PB) of patients with chronic autoimmune arthritis and from healthy controls, and from the synovial fluid (SF) of patients., Methods: In vitro studies of activation, proliferation, mRNA expression and lymphokine production were carried out., Results: We found in the PB and SF MNCs from patients with autoimmune arthritis that CsA inhibits the proliferative response, activation antigen expression, IL-2 mRNA expression and IL-2 production induced by polyclonal mitogens in a dose dependent manner., Conclusion: CsA blocks lymphocyte activation in PB and SF MNCs from patients with autoimmune arthritis.

Rationale and Objectives: To investigate bone mass measurements by ultrasound bond velocity (UBV) in bone specimens obtained from experimental animals., Methods: The authors made UBV measurements in 40 femurs and tibias dissected from Sprague-Dawley rats (14 weeks-old, mean weight 290 g) and compared them with bone densitometric measurements made on the same material using dual-energy x-ray absorptiometry (DXA)., Results: The coefficient of variation for UBV measurements, based on values obtained in five femurs and five tibias at different times, was 0.2% and 0.3% respectively. Regression studies yielded a correlation between UBV and bone mineral density in femur of r = 0.87 (P < 0.0001) and with bone mineral content of r = 0.65 (P < 0.0001); in the tibia similar levels of significance were obtained. The correlation between femur weight and UBV was r = 0.51 (P < 0.0005) and with bone mineral content it was r = 0.79 (P < 0.0001). Partial correlation between UBV and femur bone mineral density, with respect to bone weight, was r = 0.68 (P < 0.001), and with femur bone mineral content was r = 0.71 (P < 0.0001). In the tibia measurements were similarly significant., Conclusions: Measurements of bone mass made with ultrasound transmission velocity are precise correlate well with DXA measurements.

Rationale and Objectives: We sought to determine the value of peripheral quantitative computed tomography (pQCT) in measuring bone mineral density., Methods: In 50 healthy, eugonodal premenopausal women, we correlated measurements of total bone mineral content (BMCTB), made with dual-energy X-ray absorptiometry (DXA), and bone mineral density, determined by pQCT., Results: The partial correlations, adjusted for weight and age, between BMCTB and cortical bone density, total bone density, and trabecular bone density were .71 (p < .0001), .63 (p < .0001), and .32 (p < .05), respectively., Conclusion: These results and the advantages of pQCT--providing precise bone density determinations for trabecular and compact bone separately, having a high spatial resolution that allows a "compartmental" analysis of bone structure, having a low coefficient of variation, and having a minimal radiation dose (< 5 mrem)--confirm the adequacy of using this method for bone mass studies.

Beta2-microglobulin has been observed to behave as a biological marker of bone remodeling. We measured beta2-microglobulin and tartrate-resistant acid phosphatase (TRAP), a specific biological marker of bone remodeling, in 225 women: healthy premenopausal controls, healthy postmenopausal control, and patients with diseases characterized by enhanced bone turnover (postmenopausal osteoporosis, primary hyperparathyroidism, primary hyperthyroidism, polyostotic Paget's bone disease), and in other Paget's group before and after calcitonin treatment. Beta2-microglobulin levels differed significantly between the healthy premenopausal women (n = 20) compared with all the other groups. However, beta2-microglobulin levels did not differ significantly between healthy postmenopausal women (n = 38) and patient's with Paget's bone disease (n = 40)(P = 0.5095), or between women with postmenopausal osteoporosis (n = 30) and women with hyperthyroidism (n = 20)(P = 0.7890). TRAP concentrations differed significantly in all the groups paired except for women with Paget's bone disease and women with either hyperparathyroidism or hyperthyroidism (P = 0.5179 and 0.6993, respectively); likewise, TRAP levels did not differ significantly between the women with hyperparathyroidism and those with hypothyroidism (P = 0.7804). After calcitonin treatment, there was a 22% increase in beta2-microglobulin, a 17% decrease in TRAP, and a 39% decrease in alkaline phosphatase, all of which were significant at P < 0.0001. Our findings indicate that serum beta2-microglobulin, like osteocalcin, behaves as a biological marker of remodeling in a number of diseases with enhanced bone remodeling but not in Paget's bone disease.

We compared bone mass in premenopausal, perimenopausal, and postmenopausal women using peripheral quantitative computed tomography to measure separately cortical, trabecular, and total bone density. Trabecular bone density was lower (p < 0.05) in the perimenopausal women than in the healthy premenopausal women. In perimenopausal women, trabecular bone density showed a significant linear regression with age (r = 0.554, p < 0.001), but cortical bone density did not (r = 0.130, p = 0.447). The group of postmenopausal women had significantly lower bone mass in all three bone compartments than the premenopausal and perimenopausal women. These findings confirm the existence of a significant loss of trabecular bone mass in the perimenopausal period.

Seventy-two postmenopausal osteoporotic women having more than one nontraumatic vertebral crush fracture were studied. Thirty-six of them, aged 68.8 +/- 1.2 years (18 +/- 4 YSM-years since menopause), were treated with 100 IU/day of salmon calcitonin i.m. plus 500 mg of elemental calcium for 10 days each month. The remaining 36 patients, aged 69.6 +/- 1.4 years (19 +/- 3 YSM), were given only 500 mg of elemental calcium for 10 days each month. All patients underwent clinical and analytical evaluation every 3 months. Radiological evaluation, assessment of vertebral deformities, and metacarpal radiogrammetry were done every 6 months. Densitometric measurements of total and regional bone mass were made every 12 months. At 24 months, the calcitonin group showed a 60% reduction in the number of new fractures and the group receiving only calcium had a 45% increase (P < 0.001). The incidence of vertebral fractures was 0.07 per patient-year in the group treated with calcitonin and 0.45 per patient-year in the group treated with calcium (P < 0.001). At 2 years, the calcitonin group showed a 12% increase in cortical bone mass on metacarpal radiogrammetry, a 16% increase in the axial skeleton on trunk densitometry, a 3.5% increase in total body bone mineral content, a 30.7% increase in pelvic bone mineral content, and a 6.2% increase in arm bone mineral content (all P < 0.001). In the group treated with calcium alone there was a loss of bone mass in every region. These findings suggest that salmon calcitonin is effective in the treatment of osteoporosis and show that it acts on cortical and trabecular bone.

We studied nutritional deficits, using as markers the levels of transferrin, retinol-binding protein, and prealbumin, in 20 women with osteoporotic hip fractures (type II), 40 women with vertebral fractures (type I), and two groups of age-matched control subjects. The concentrations of all three nutritional markers were lower in the two groups of patients than in their matched controls, and in type-I as compared with type-II osteoporosis. In the osteoporotic patients, simple linear regression showed a significant correlation between the variables which we studied (r2 ranged from 0.5 to 0.7; p < 0.001), the best correlation being between prealbumin and retinol-binding protein in type-II osteoporosis. Our results suggest that there is a more marked nutritional deficit in type-II than in type-I osteoporosis.

We evaluated the precision of metacarpal radiogrammetry by computed radiography as a technique for bone mass measurement. Measurements obtained using this method were compared with densitometric measurements of total body bone mass using dual-energy X-ray absorptiometry. Our results showed that the coefficients of variation for radiogrammetric measurements were similar in 60 women with osteoporosis and in 100 normal women: 0.7% in both groups for external diameter of the second metacarpal, 2.41% in the normal women and 2.03% in the women with osteoporosis for internal diameter, and 2.3% and 2.4%, respectively, for cortical thickness. Except for external metacarpal diameter, which did not differ between normal and osteoporotic women, all the measurements were smaller in the women with osteoporosis (p < 0.001). The correlation between metacarpal cortical thickness and total body bone mineral content was r2 = 0.462 in the women with osteoporosis, r2 = 0.476 in the controls, and r2 = 0.522 in the two groups combined (all p < 0.001). There was a significant correlation (p < 0.001) between cortical thickness and densitometric metacarpal measurements of both the diaphysis and epiphysis. These results confirm the value of radiogrammetry as a technique for quantitating bone mass under normal and pathological conditions. Additional advantages of the method are minimal radiation dose delivered, possibility for storing the image and data, and technical facility.

Seventy-two eugonadal women, mean age 39 +/- 4 years, with a z score for total body bone mineral content (BMC) of less than -1.5 (mean -1.80 +/- 0.12) were selected from a previous screening study of normal values of total body and regional BMC in women using dual-energy X-ray absorptiometry. The women were distributed into two groups: 36 women undergoing treatment with 1 g/day of elemental calcium in the form of calcium pidolate and 36 women not treated. One year later, total body BMC, regional BMC in arms, legs and trunk, 24-hour urinary calcium excretion (Ca 24 hour), fasting urinary calcium/creatinine ratio, and serum tartrate-resistant acid phosphatase concentration were measured in both groups. There was a significant increase in total body and regional BMC (P < 0.001) in the group taking calcium pidolate. The increase was greater in arms and legs (2.5%) than in trunk (1.7%), being 2.6% for total body BMC. The z score increased from -1.81 +/- 0.13 to -1.52 +/- 0.12 (16%, P < 0.001). The corporal index did not change, and total body BMC corrected for fat-free body mass increased by 6.7% (P < 0.001). These changes were accompanied by an increase in 24-hour urinary calcium excretion and a decrease in urinary calcium/creatinine ratio and serum tartrate-resistant acid phosphatase concentration (P < 0.05 for all). There was a correlation between cumulative calcium dose at the end of treatment and gain in total body BMC (r2 = 0.925, P < 0.001). Objective: We compared the diphosphonates etidronate and clodronate to find out which was more effective in inhibiting the loss of bone mass after ovariectomy in rats., Methods: Five lots of female Sprague Dawley rats, each containing 15 rats (14 wks old; mean weight 290 g), were kept under identical living conditions (12 hrs of light and 12 hrs of darkness), habitat, and diet following the current norms of the European Economic Community. One lot was kept as a control; one lot underwent sham ovariectomy (Sham-OVX); and the remaining 3 lots were anesthesized and underwent double ovariectomy (OVX). One lot of ovariectomized rats was given etidronate by esophageal tube, 5 mg/kg/day (OVX-E); one lot was treated with intraperitoneal clodronate, 5 mg/kg/3 days (OVX-Cl). The remaining lot was not manipulated (OVX). The animals' right femurs were then dissected for measurements of the bone mineral content., Results and Conclusion: Bone densitometry revealed that loss of bone mass 2 months after ovariectomy in the OVX-Cl group was 2.3%, not differing from that of the control group; bone loss in the OVX-E group was 11.7%, significantly different from the control group and from the OVX-Cl group (p < 0.001). These results show that clodronate is superior to etidronate in inhibiting osteopenia after ovariectomy in rats.

We studied the behavior of radiogrammetric and densitometric measurements in relation to season and body weight in a group of 30 healthy premenopausal women. Measurements were made at 6-month intervals, in summer/fall when bone density increases and in winter/spring when bone density declines. Total body bone mineral content (TBBMC) and regional bone mineral content (RBMC) were measured using dual-energy X-ray absorptiometry (DXA). Metacarpal radiogrammetry was carried out with computed radiography. Weight and body mass index increased significantly in winter (P < 0.05) and total body and RBMC decreased (P < 0.001). The opposite occurred in summer: weight and body mass index decreased significantly (P < 0.05) and total body and regional bone mineral content increased (P < 0.001). Differences in TBBMC persisted when the measurement was corrected for weight (TBBMC/W) (P < 0.001), but not for metacarpal cortical thickness corrected for weight. In the first measurement made there were significant relations between weight and both TBBMC (P < 0.001) and metacarpal cortical thickness (P < 0.005). The relation between weight and TBBMC remained significant in later measurements, but the relation between weight and metacarpal cortical thickness ceased to be significant in the second and fourth measurements. Our results show that there is an important seasonal variation in bone mass and that DXA is more sensitive than radiogrammetry in registering these changes.

Bone density measurements based on the rate of propagation of ultrasound waves provide information on bone elasticity, which is largely dependent on cortical bone structure. Both bone density and cortical bone present variations dependent on age and sex. As far as we know, bone elasticity in patients on hemodialysis has not been studied, so we used ultrasound bone velocity and metacarpal radiogrametry to study 23 adult patients with end-stage renal disease. The patients' results were compared with those of a group of 23 healthy subjects matched by age and sex. The measurements obtained by both ultrasound bone velocity and metacarpal radiogrametry were lower in hemodialyzed patients (p < 0.025) than in controls and there was a good correlation between the two measurements (r2 = 0.561; p < 0.0007). Neither of the measurements correlated with age or time on dialysis. These results show that bone elasticity in dialyzed patients is diminished, which could be an important factor in the bone lesions of these patients.

Changes in cortical and trabecular bone mass occurring in primary hyperparathyroidism are the subject of controversy. We measured the total and regional bone mass in a group of patients and controls and analyzed the observed changes with respect to sex and other characteristics. Parathormone, serum calcium, serum phosphorus, tubular resorption of phosphates, alkaline phosphatase, and tartrate-resistant acid phosphatase differed significantly (p < 0.001 for all) between patients and controls. All the patients had significantly less total and regional bone mass; the regional bone mass of the arms did not differ. When analyzed according to sex, we found that women had a significant reduction in bone mass in all the zones studied (p < 0.001), whereas men had a significant decrease in bone mass only in the head (p < 0.025). Our findings show that hyperparathyroidism is accompanied by a loss of bone mass except in the arms, and that predominantly both cortical bone (legs, pelvis and skull) and trabecular bone (trunk) are affected. This means that the skeleton suffers generalized bone loss and a significant reduction in total body calcium content. Although we did not study many men, their bone involvement was less intense, and there was a clear difference between the sexes in this respect.

Dual energy X-ray absorptiometry measurements of total body bone mineral content (TBBMC), fat body mass (FBM) and fat mass percentage (%FM), lean body mass (LBM) and body weight (BW) were performed on 168 normal postmenopausal females. They were matched regarding life style and habits and had body mass index under 30. Their TBBMCs were correlated with these measurements, with chronological age (CA) and with the number of years since menopause (YSM). There was no correlation between TBBMC and %FM and LBM, but there was with BW (p < 0.001). There was a significant and negative correlation (r = -0.453, p < 0.001) between TBBMC and CA and to a higher range (r = -0.697, p < 0.001) with YSM. Menopausal females over 60 (n = 87) presented less bone mass than younger females (n = 81) (p < 0.01). These data suggest that regarding TBBMC, menopausal onset is a more important factor in bone mass loss, which persists rather markedly even during periods of time far from menopause and that TBBMC depends more on BW than on LBM and FM in women.

Total body bone mineral content (TBBMC), total body bone mineral density (TBBMD), and regional bone mineral content (BMC) were determined with dual energy X-ray absorptiometry and correlated with different anthropometric variables in 324 normal male subjects aged 15 to 85 years, in order to obtain further knowledge of the controversial course of bone mass in males. These subjects showed increased TBBMC and TBBMD up to the age of 25-29 years. A positive and significant (p < 0.001) correlation was observed between TBBMC and TBBMD and with BMC with age (p < 0.001), as well as between age and body weight (p < 0.001) and between the latter and body fat (p < 0.001). Age did not correlate with weight or TBBMC, TBBMD, and regional BMC in subjects older than 29 years, while a significant and positive correlation between body weight and muscle mass was observed with bone mass (p < 0.001). By multiple regression analysis, TBBMC was significantly independent of fat-free mass (FFM) and lean body mass (LBM) (p < 0.01). These findings led us to point out that bone mass increases until the age of 29 years and that changes depend more on weight that on age in men. The rate of bone mass loss with age was 0.23%, that of soft tissue 0.35%, while fat mass increased 0.7% per year.
