262. Near-infrared light absorption and scattering based on a mono-layer of gold nanoparticles

Abstract

We report fabrication and characterization of large-area ultrathin near-infrared light absorbers and scatterers based on a mono-layer of gold nanoparticles laying on top of a dielectric spacer and an aluminum reflector. The nanoparticles are formed through thermal annealing of an evaporated continuous gold film. Through optimization of initial gold-film thickness, spacer thickness, as well as annealing temperature we obtained samples that exhibit very low (similar to 2%) broadband specular reflectance at near-infrared (NIR) wavelength range. By considering also diffuse reflection, we identify that the low specular reflectance can be due to either relatively high light absorption (similar to 70%) or high light scattering (over 60%), with the latter achieved for samples having relatively sparse gold nanoparticles. Both strong absorption and scattering of NIR light are not inherent properties of the bulk materials used for fabricating the samples. Abstract

Protein Numb, first identified as a cell-fate determinant in Drosophila, has been shown to promote the development of neurites in mammals and to be cotransported with endocytic receptors in clathrin-coated vesicles in vitro. Nevertheless, its function in mature neurons has not yet been elucidated. Here we show that cerebellar Purkinje cells (PCs) express high levels of Numb during adulthood and that conditional deletion of Numb in PCs is sufficient to impair motor coordination despite maintenance of a normal cerebellar cytoarchitecture. Numb proved to be critical for internalization and recycling of metabotropic glutamate 1 receptor (mGlu1) in PCs. A significant decrease of mGlu1 and an inhibition of long-term depression at the parallel fiber-PC synapse were observed in conditional Numb knockout mice. Indeed, the trafficking of mGlu1 induced by agonists was inhibited significantly in these mutants, but the expression of ionotropic glutamate receptor subunits and of mGlu1-associated proteins was not affected by the loss of Numb. Moreover, transient and persistent forms of mGlu1 plasticity were robustly induced in mutant PCs, suggesting that they do not require mGlu1 trafficking. Together, our data demonstrate that Numb is a regulator for constitutive expression and dynamic transport of mGlu1.

266. FOSL1 enhances growth and metastasis of human prostate cancer cells through epithelial mesenchymal transition pathway.

Abstract

OBJECTIVE: To investigate the effect of highly-expressed FOSL1 on the tumorigenesis and metastasis of prostate cancer. PATIENTS AND METHODS: Researches were carried out in human prostate cancer tissues and cell lines. In prostate cancer tissues, the expression of FOSL was detected by immunohistochemistry. In vitro cell line experiments, we constructed a prostate cancer cell model with FOSL1 stable knockdown and tested cell proliferation and metastasis before and after knockdown of FOSL1. Finally, the epithelial-mesenchymal transition (EMT) markers before and after interference of FOSL1 were also analyzed. RESULTS: FOSL1 was confirmed to have a high expression in prostate cancer. Transwell experiments demonstrated that FOSL1 could enhance prostate cancer metastasis, while in vivo experiments revealed an accelerated progression of prostate cancer caused by FOSL1. In addition, Western blot analysis revealed an elevated level of N-cadherin and Snail1 and a reduced level of E-cadherin that was induced by FOSL1. 267. Cost-effectiveness analysis of oral probiotics for the prevention of Clostridium difficile-associated diarrhoea in children and adolescents.

Abstract

Background: The incidence of Clostridium difficile-associated diarrhoea (CDAD) in hospitalized children and adolescents has been increasing year-on-year. Paediatric CDAD places a significant economic burden on healthcare systems. Probiotics are live organisms thought to improve the microbial balance of the host, counteract disturbances in intestinal flora, and reduce the risk of colonization by pathogenic bacteria.Aim: A cost-effectiveness analysis was conducted to assess the economy of probiotics for the prevention of CDAD in children and adolescents receiving antibiotics.Methods: A decision tree model combining clinical effectiveness, utility and cost data was used. Sensitivity analyses were conducted to determine the robustness of the model outcomes.Finding: The 'oral probiotics' strategy and 'no probiotics' strategy offered patients 0.05876 and 0.056 quality-adjusted life years (QALYs) at a cost of $16,668.70 and $20,355.28, respectively. The oral probiotics strategy exhibited higher QALY and lower cost, and represents the cost-saving strategy. The results were robust for sensitivity analyses.Conclusion: From the perspective of the medical system, oral probiotics as a preventive strategy for CDAD in hospitalized children and adolescents receiving a therapeutic course of antibiotics reduced the risk of CDAD and represents a cost-saving strategy. Abstract

Oral submucous fibrosis (OSMF), a potentially malignant oral cavity disorder that causes speech and mastication problems, lacks an established treatment regimen; moreover, no treatment can effectively reverse the course of OSMF. A meta‐analysis was conducted to investigate the efficacy of the peripheral vasodilator, pentoxifylline, in the treatment of OSMF. We searched five different databases for studies meeting our eligibility criteria (up to June 30, 2017). Statistical analyses were performed using RevMan 5.3 software. Three randomized controlled trials (247 OSMF patients) were selected. Pentoxifylline increased the objective sign maximal mouth opening (MMO; WMD: −4.59, 95% CI: −8.65, −0.53; p < 0.05) following short‐term (under 1 month; WMD: −1.94, 95% CI: −3.12, −0.77; p < 0.05) and long‐term (over 1 month; WMD: −5.44, 95% CI: −6.81, −4.07; p < 0.05) application. Pentoxifylline improved the subjective symptom burning sensation (WMD: −0.11, 95% CI: −0.17, −0.05; p < 0.05) only following long‐term application (WMD: 0.42, 95% CI: 0.18, 0.96; p < 0.05). The efficacy on MMO and burning sensation increased with time. Although OSMF lacks definitive treatment modalities, our meta‐analysis shows that pentoxifylline effectively improves the objective signs and subjective symptoms of OSMF and its therapeutic efficacy increases with time. 269. Effects of paclitaxel combined with miR-448 on growth and proliferation of bladder cancer EJ cells.

Abstract

OBJECTIVE: Bladder cancer is one of the most common malignant tumors of the urinary system characterized by a high recurrence rate after treatment. Paclitaxel is a new type of anti-neoplastic agent used for treatment by inhibiting the proliferation of bladder cancer cells. This study aimed to explore the mechanism of paclitaxel on the inhibition of bladder cancer cell proliferation by applying paclitaxel combined with miR-448 on bladder cancer cells. MATERIALS AND METHODS: Bladder cancer EJ cells were divided into 5 groups, including control group, paclitaxel group, negative control (NC) group, microRNA-448 (miR-448) mimic group, and paclitaxel combined with miR-448 mimic group. Cell apoptosis was tested by flow cytometry. Cell proliferation was determined by cell counting kit 8 (CCK8) assay. The regulation of miR-448 on B cell lymphoma 2 (Bcl-2) gene was assessed by dual luciferase reporter assay. Bcl-2 protein expression was detected by Western blot. RESULTS: The apoptotic rate of cells was significantly increased, while the cell proliferation ability was significantly reduced in paclitaxel group and miR-448 mimic group compared with control (p<0.05). Cell apoptotic rate in paclitaxel combined with miR-448 mimic group was markedly higher than that of paclitaxel group (p<0.05). MiR-448 can bind to the 3'UTR region of Bcl-2 mRNA and regulate the expression of Bcl-2. The expression of Bcl-2 protein in paclitaxel group and miR-448 mimic group was significantly lower than that in control group and higher than paclitaxel combined with miR-448 mimic group (p<0.05). CONCLUSIONS: Paclitaxel combined with miR-448 promoted EJ cell apoptosis and inhibited cell proliferation by suppressing Bcl-2 gene expression. [ABSTRACT FROM AUTHOR]

270. Analysis and Prediction for Time Series on Torque Friction of Rolling Bearings.

Abstract

Based on the Cao, mutual information, and small-data methods, the embedding dimension, delay time, and maximum Lyapunov exponent are calculated, respectively, to analyze the chaos characteristics of rolling bearings. The curve attractor of x(t)-x(t+(m-1)t)x(t) is constructed to parse the dynamics features on friction torque in the phase space of time series. According to the five prediction methods utilized--one-rank local-region, adding-weight one-rank local-region, improved adding-weight one-rank local-region, dial basis function (RBF) neural network, and Volterra series--the time series of bearings A, B, and C are forecasted with the first 400 experiment data as training values and the latter 57 data as test values to verify the prediction models' feasibility. Finally, the bootstrap-maximum-entropy method is proposed to effectively fuse the results of these five prediction methods, and obtain the estimation interval and true value of friction torque. Experimental investigation shows that the friction torque phase trajectory has a linear increasing trend. These five forecasting models are effective for friction torque time series prediction with small error and high precision. The range of the fused estimation interval is relatively small, and the maximum error between the estimated true value and the experiment value is only 5.183%, so the fluctuation information and the change trends of friction torque are accurately described. Moreover, the proposed models do not consider the probability distribution and trend information of the research system, breaking from the features of traditional statistical models. Abstract

The external beam particle-induced X-ray emission (PIXE) setup has been upgraded by introducing a new silicon drift detector with the aim of obtaining better minimum detection limits (MDLs) at the 2 × 1.7 MV Tandetron accelerator of the Beijing Normal University in China. The upgraded external beam PIXE setup is equipped with two silicon drift detectors for PIXE analysis of low and high Z elements. A surface barrier detector for Rutherford backscattering spectrometry monitors beam and helium flow. Two kinds of aerosol filter samples (quartz fiber filters and Teflon filters) were studied. A 200-μm thick Mylar absorber in front of the medium-high energy X-rays detector was adopted, and it got the best MDLs for atmospheric aerosol samples analysis. Multielemental analysis of quartz fiber filter aerosol samples was achieved. For more accurate and better MDLs of low Z elements in PIXE analysis, it is necessary to keep sufficient helium flow behind the thin samples. [ABSTRACT FROM AUTHOR]

Abstract

Multisite longitudinal neuroimaging designs are used to identify differential brain structural change associated with onset or progression of disease. The reliability of neuroanatomical measurements over time and across sites is a crucial aspect of power in such studies. Prior work has found that while within-site reliabilities of neuroanatomical measurements are excellent, between-site reliability is generally more modest. Factors that may increase between-site reliability include standardization of scanner platform and sequence parameters and correction for between-scanner variations in gradient nonlinearities. Factors that may improve both between- and within-site reliability include use of registration algorithms that account for individual differences in cortical patterning and shape. In this study 8 healthy volunteers were scanned twice on successive days at 8 sites participating in the North American Prodrome Longitudinal Study (NAPLS). All sites employed 3 Tesla scanners and standardized acquisition parameters. Site accounted for 2 to 30% of the total variance in neuroanatomical measurements. Abstract

BACKGROUND This study was undertaken to determine whether use of the direct renin inhibitor aliskiren would reduce cardiovascular and renal events in patients with type 2 diabetes and chronic kidney disease, cardiovascular disease, or both. METHODS In a double-blind fashion, we randomly assigned 8561 patients to aliskiren (300 mg daily) or placebo as an adjunct to an angiotensin-converting-enzyme inhibitor or an angiotensin-receptor blocker. The primary end point was a composite of the time to cardiovascular death or a first occurrence of cardiac arrest with resuscitation; nonfatal myocardial infarction; nonfatal stroke; unplanned hospitalization for heart failure; end-stage renal disease, death attributable to kidney failure, or the need for renal-replacement therapy with no dialysis or transplantation available or initiated; or doubling of the baseline serum creatinine level. RESULTS The trial was stopped prematurely after the second interim efficacy analysis. After a median follow-up of 32.9 months, the primary end point had occurred in 783 patients (18.3%) assigned to aliskiren as compared with 732 (17.1%) assigned to placebo (hazard ratio, 1.08; 95% confidence interval [CI], 0.98 to 1.20; P = 0.12). Effects on secondary renal end points were similar. Systolic and diastolic blood pressures were lower with aliskiren (between-group differences, 1.3 and 0.6 mm Hg, respectively) and the mean reduction in the urinary albumin-to-creatinine ratio was greater (between-group difference, 14 percentage points; 95% CI, 11 to 17). The proportion of patients with hyperkalemia (serum potassium level, = 6 mmol per liter) was significantly higher in the aliskiren group than in the placebo group (11.2% vs. 7.2%), as was the proportion with reported hypotension (12.1% vs. 8.3%) (P CONCLUSIONS The addition of aliskiren to standard therapy with renin-angiotensin system blockade in patients with type 2 diabetes who are at high risk for cardiovascular and renal events is not supported by these data and may even be harmful. Abstract

To evaluate and compare the effects and toxicity of the domestic product of recombinant mutant human tumor necrosis factor (rmhTNF) combined with chemotherapy and chemotherapy alone in the treatment of patients with non-small cell lung cancer (NSCLC).Two hundred patients with NSCLC in multicenter were randomly devided into trial group (150 cases) and control group (50 cases). Chemotherapy with CAP regimen was given to the patients. Meanwhile, rmhTNF injection of 4×10⁶U/m² was also given from the 1st to 7th days, the 11th to 17th days on the chemotherapy cycle in the trial group. The control patients received chemotherapy alone. Twenty-one days were as a cycle, 2 cycles were given to each patient. The chemotherapeutic effects and toxicity were observed and compared between the two groups after the therapy.of the 200 patients, 5 cases in the trial group and 3 cases in the control group were out of the trial because of economy. The other 192 cases (145 cases in the trial group and 47 cases in the control group) could be analyzed and evaluated the clinical effects and toxicity. The response rate of chemotherapy was 46.90% (68/145) in the trial group and 17.02% (8/47) in the control group respectively ( P =0.001). The KPS scores was 86.02±9.74 in the trial group, and 80.14±9.10 in the control group ( P =0.025). No significant difference of degree III+IV toxicity was observed between the two groups ( P0.05). The side effects related to rmhTNF included slight fever, cold-like symptoms, pain and red and swelling in the injection site. All of them were mild and didn't need any treatment and disappeared after the therapy. There were no severe abnormality of liver and kidney function and ECG in both groups.The results demonstrate that the effects of domestic rmhTNF combined with chemotherapy are remarkably higher than that of chemotherapy alone in the treatment of NSCLC. rmhTNF can increase the sensitivity to chemotherapy and improve the quality of life of the patients with slight toxicity. Hence rmhTNF is worth expanding clinical use.

Abstract

The temperature dynamics of the radiative heat propagation in a multilayer structure is theoretically treated with a formalism combining the scattering matrix and Green's-functions methods. The time evolution of the temperature of parallel plates of silicon carbide in vacuum is simulated for different interplate distances and thicknesses of plates. The characteristic radiative heat exchange time and temperature of the plates at stationary state are determined from the time evolutions. The threshold interplate distance which separates heating and cooling regimes for the sink plate is found. We show that the variation of the interplate distance allows us to control the relaxation processes in the system of absorber and emitter., QC 20140818
