Predictions of drought onset and termination at subseasonal (from 2 weeks to 1 month) lead times could provide a foundation for more effective and proactive drought management. We used reforecasts archived in NOAA’s Subseasonal Experiment (SubX) to force the Noah Multiparameterization (Noah-MP), which produced forecasts of soil moisture from which we identified drought levels D0–D4. We evaluated forecast skill of major and more modest droughts, with leads from 1 to 4 weeks, and with particular attention to drought termination and onset. We find usable drought termination and onset forecast skill at leads 1 and 2 weeks for major D0–D2 droughts and limited skill at week 3 for major D0–D1 droughts, with essentially no skill at week 4 regardless of drought severity. Furthermore, for both major and more modest droughts, we find limited skill or no skill for D3–D4 droughts. We find that skill is generally higher for drought termination than for onset for all drought events. We also find that drought prediction skill generally decreases from north to south for all drought events.

With the development of vehicle network technology, transportation companies have generated GPS-based driver behavior data for operating vehicles, providing potential for research on driver behavior and road safety risk perception. Research on road safety risk perception at the present stage generally involves fuzziness and subjectivity, the degree of risk is difficult to quantify, and big data-based road safety risk perception has not yet been used practically. To address this problem, this paper proposes a method of quantifying the degree of road safety risk based on the hierarchical analysis method and fuzzy comprehensive evaluation method under the drive of abnormal driving behavior data, constructs a scientific and reasonable urban road safety risk identification comprehensive evaluation model, and explores important factors affecting road safety. The research results have a certain reference effect for traffic management departments to accurately locate dangerous sections in the urban road network.

Our previous studies have shown that unexplained dizziness may be related to patent foramen ovale (PFO). This study aims to explore whether the degree of shunt is correlated with the degree of unexplained dizziness and looking for the possible clinical intervention for patients with unexplained dizziness.Methods: This study was a large single-center, prospective, controlled study. From March 2019 to March 2022, patients with unexplained dizziness and explained dizziness and healthy controls were recruited. Contrast-enhanced transcranial Doppler sonography (c-TCD) was performed to detect the existence of right-to-left shunt (RLS) and shunt grading. The dizziness handicap inventory (DHI) was completed to evaluate the dizziness. Unexplained dizziness patients with large amount of PFO were volunteered to receive medication treatment and transcatheter PFO closure and followed up six months.Results: A total of 387 patients (132 unexplained, 123 explained and 132 controls) were enrolled. There was a statistical difference in the RLS grading with three groups (p < 0.001). The Spearman correlation coefficient of RLS grading and DHI scores in unexplained dizziness patients (r=0.122, p=0.163) and explained dizziness patients (r=0.067, p=0.460). In the unexplained group, there were 49 cases with massive grading RLS. Of which 25 patients received percutaneous PFO closure treatment and 24 cases received medication treatment. Followed up six months after treatment, the amount of DHI scores change in patients who received the percutaneous PFO closure was significantly higher than that in the medication treatment group (p < 0.001).Conclusion: RLS may play an important role in unexplained dizziness. For unexplained dizziness patients, PFO closure may contribute to better outcomes. Osteosarcoma (OS) is a type of tumor with high malignant behaviors. Increasing investigates have confirmed that long non-coding RNA HLA complex group 18 (lncRNA HCG18) acted as a tumor-promoting factor in multiple tumors. Nevertheless, the underlying mechanism of HCG18 on OS remains largely unclear. HCG18, miR-34a, and runt-related transcription factor 2 (RUNX2) expressions were detected by quantitative real-time PCR (RT-qPCR) or western blotting assays, respectively. The underlying tumorigenic phenotypes were detected by MTT, wound healing, transwell invasion, western blotting assays. Molecular interactions were verified by dual-luciferase report assay. HCG18 and RUNX2 were notably enhanced, whereas miR-34a was decreased in OS tumor tissues and cell lines. Functional experiments uncovered that HCG18 silencing significantly inhibited the capabilities of proliferation, migration, and invasion, while overexpression of HCG18 play the opposite roles. Furthermore, HCG18 directly bound to miR-34a, and miR-34a was confirm to be a negative regulator of RUNX2. Interestingly, the anti-tumor effects of HCG18 silencing were attenuated by miR-34a inhibitor and RUNX2 overexpression. Taken together, the present study suggested that HCG18 promoted the malignant biological behaviors of OS through regulating the miR-34a/RUNX2 pathway, implying HCG18 might serve as a new target for OS treatment.

We analyzed the sealing effectiveness of cap rocks bearing different types of stylolites using a combination of petrographic, petrophysical, pore structure, and sealing capacity characterization techniques. This study was based on examples of carbonate cap rocks that seal ultradeep hydrocarbon reservoirs of the Tarim Basin (China). Samples from both drill cores and their outcrop analogues were investigated to quantify how morphology influences the sealing capacity of different types of stylolite-bearing rocks. The study cap rocks consisted of mudstone, wackestone to packstone, grainstone, and dolomitic limestone. Four types of stylolites were identified: rectangular layer, seismogram pinning, suture and sharp peak, and simple wave-like types. The difference in the sealing capacity of carbonate cap rocks is attributed to their pore structure connections and the types of stylolites they develop. Samples bearing simple wave-like stylolites showed the best sealing capacity, followed by those with rectangular layer and suture and sharp peak types, whereas carbonates hosting seismogram pinning types had the lowest sealing capacity. The impact of stylolite segments on the rock sealing properties, however, differed from one segment to another. Rectangular layer–type stylolites could be divided into three distinct segments (with good, moderate, and poor sealing, respectively). Both the seismogram pinning and suture and sharp peak stylolite types were divided in two parts, with the former one having moderate and poor sealing and the latter exhibiting good and moderate sealing. The simple wavelike type had a good sealing capacity all along the pressure-solution seam. The most effective sealing barriers for vertical fluid flow form when (1) calcite and siliceous cements are pervasively distributed in the vicinity of stylolites, forming highly cemented zones with lower porosity and permeability than their surrounding host rocks; (2) stylolites are enriched in insoluble residues; and (3) rare microfractures and dissolution vugs are found along the stylolites. This work provides useful examples for the prediction of the sealing potential of stylolite-bearing carbonate rocks according to stylolite morphology in other geologic settings.

Present studies have found that PFO is a risk factor that can lead to many diseases. However, few studies have examined the relationship between PFO and dizziness.Methods: This study was a large single-center, prospective, controlled study. From March 2019 to March 2021, we recruited patients with dizziness were divided into two groups: “explained” and “unexplained”. All patients were submitted to contrast-enhanced transcranial Doppler ultrasound and screened for PFO. Transesophageal heart ultrasound or right heart catheterization was used to confirm PFO. Additionally, multiple factors were collected and statistical analysis was performed between the two groups.Results: Among the 244 patients included, 123 were in the “explained” group and 121 were in the “unexplained” group. The prevalence of PFO in the “explained” group was 34 (27.4%) compared to 79 (64.7%) in the “unexplained” group. In the “explained” group, 7 were RLS level IV, 6 were level III, 7 were level II, and 14 were level I. For the “unexplained” group, the numbers of patients with levels IV, III, II and I were 27, 26, 12 and 14, respectively. Univariate analysis revealed that PFO (χ2= 34.77, P < 0.001) and age (t = − 3.49, P < 0.001) seemed to be potential risk factor candidates for “unexplained” dizziness. In multiple regression analysis, age (OR = 0.97; 95% CI 0.95– 0.99) and the prevalence of PFO (OR = 4.37; 95% CI 2.50– 7.63) were statistically significant. Massively shunted PFO showed more pronounced risk factors (OR = 8.76; 95% CI 4.04– 19.03).Conclusion: There was a high prevalence of PFO and a greater RLS level in unexplained dizziness. PFO and age were independent risk factors for unexplained dizziness. Marine–continental transitional shale is one of the most promising targets for shale gas exploration in the Lower Yangtze region. To investigate the sedimentary environments and the regularity of the enrichment of the Longtan shale, multiple techniques including core and thin-section observations, geochemical and elemental analyses, X-ray diffraction, scanning electron microscopy (SEM), and low-pressure nitrogen adsorption (LPNA) were used to analyze the sedimentology, mineralogy, and pore structure of the Longtan shale. The core descriptions and thin-section observations showed that the Longtan shale was deposited in marine–delta transitional environments including delta-front, shore swamp, mixed tidal flat and shallow shelf environments. The Sr/Cu, V/Cr, CIA, EF (Mo), EF (U), and other major and trace element results indicated warm and moist climates and water-reducing conditions in the Longtan period. Both the climate and water conditions were favorable for organic matter production and preservation. The geochemical results showed that the Longtan shale was in the overmature stage (Ro values ranging from 2.4% to 3.57%) and that the average total organic carbon (TOC) content was 5.76%. The pore system of the Longtan shale consisted of inorganic pores with a small number of organic pores and microfractures. The porosity and specific surface area were mainly affected by the TOC and clay mineral contents. An effective combination of brittle mineral particles, organic matter, and clay minerals provided the necessary conditions for pore preservation. The organic pores, intergranular pores in clay minerals, and brittle mineral pores formed the main network system for the Longtan shale. In summary, the lithological combinations, organic geochemistry, and pore structure system were all affected by the sedimentary environments.

The prognosis for colon carcinoma is poor, it is a highly aggressive tumor, and its early symptoms are subtle. Hence, the need for more colon cancer prognostic indicators is urgent. Many long non-coding RNAs (lncRNAs) are known to actively contribute to the detection, prognosis, and treatment of tumors. On the other hand, immunogenic cell death (ICD) is vital for the prevention of tumors. This study seeks to research the mechanism of action of ICD-related lncRNAs in colon cancer and integrate ICD-related genes with lncRNAs. In order to investigate ICD-related lncRNAs, this research suggests a multi-constraint canonical correlation analysis. The prognostic model of the most significant lncRNAs is also constructed and verified in this paper using univariate and Lasso-Cox regression. In this study, colon cancer samples are split into high-risk and low-risk groups based on the model's risk score, and the impact of the model is assessed using the TCGA queue and the GEO queue, respectively. Also, we thoroughly examined the parallels and discrepancies in immune infiltration, functional enrichment, and medication sensitivity across the two risk groups. In conclusion, the ICD-related lncRNAs risk model developed in this research can serve as a guide for predicting the prognosis and therapeutic sensitivity of colon cancer patients.

Toroidal vortices, also known as vortex rings, are whirling, closed-loop disturbances that form a characteristic ring shape in liquids and gases and propagate in a direction that is perpendicular to the plane of the ring. They are well-studied structures and commonly found in various fluid and gas flow scenarios in nature, for example in the human heart, underwater air bubbles and volcanic eruptions1–3. Here we report the experimental observation of a photonic toroidal vortex as a new solution to Maxwell’s equations, generated by the use of conformal mapping4–7. The resulting light field has a helical phase that twists around a closed loop, leading to an azimuthal local orbital angular momentum density. The preparation of such an intriguing state of light may offer insights for exploring the behaviour of toroidal vortices in other disciplines and find important applications in light–matter interactions, optical manipulation, photonic symmetry and topology, and quantum information8–17.

The intestinal tract is a complex ecosystem where numerous cell types of epithelial, immune, neuronal, and endothelial origin coexist in an intertwined, highly organized manner. The functional equilibrium of the intestine relies heavily on the proper crosstalk and cooperation among each cell population. Furthermore, macrophages are versatile, innate immune cells that participate widely in the modulation of inflammation and tissue remodeling. Emerging evidence suggest that macrophages are central in orchestrating tissue homeostasis. Herein, we describe how macrophages interact with epithelial cells, neurons, and other types of mesenchymal cells under the context of intestinal inflammation, followed by the therapeutic implications of cellular crosstalk pertaining to the treatment of inflammatory bowel disease.

Inflammasome contributes to the pathogenesis of various inflammatory diseases, but the epigenetic mechanism controlling its activation remains elusive. Here, we found that the histone methyltransferase Ezh2 mediates the activation of multiple types of inflammasomes in macrophages/microglia independent of its methyltransferase activity and thus promotes inflammasome-related pathologies. Mechanistically, Ezh2 functions through its SANT2 domain to maintain the enrichment of H3K27 acetylation in the promoter region of the long noncoding RNA (lncRNA) Neat1, thereby promoting chromatin accessibility and facilitating p65-mediated transcription of Neat1, which is a critical mediator of inflammasome assembly and activation. In addition, the tumour suppressor protein p53 competes with Ezh2 for the same binding region in the Neat1 promoter and thus antagonises Ezh2-induced Neat1 transcription and inflammasome activation. Therefore, loss of Ezh2 strongly promotes the binding of p53, which recruits the deacetylase SIRT1 for H3K27 deacetylation of the Neat1 promoter and thus suppresses Neat1 transcription and inflammasome activation. Overall, our study demonstrates an epigenetic mechanism involved in modulating inflammasome activation through an Ezh2/p53 competition model and highlights a novel function of Ezh2 in maintaining H3K27 acetylation to support lncRNA Neat1 transcription.
