Abstract

Background: Antimicrobial Resistance (AMR) and Tuberculosis (TB) are global concern. According to the WHO fact sheet on tuberculosis, in 2017, 10 million people fell ill with TB, and 1.6 million including 230,000 children died from the disease. There is a critical need of design and development of novel chemotherapeutic agents to combat the emergence and increasing prevalence of resistant pathogens. In the present study, a new series of 1,3,4-oxadiazoles incorporating benzimidazole and pyridine scaffolds in a single molecular framework has been reported. Methods: The structures of the synthesized derivatives (4a to 4e) were assigned by IR, NMR and mass spectral techniques. The hybrid compounds were evaluated for their antimicrobial, antitubercular and antioxidant activities. In addition, docking simulations were performed to study ligand-protein interactions and to determine the probable binding conformations. Results: Molecule 4a has shown anti-tubular activities with MIC 1.6 μg/ml. As compared to ascorbic acid activities (IC50 = 62.91 μg/ml), molecule 4e exhibited better antioxidant activities (IC50 = 24.85 μg/ml). Also, molecule 4e has shown significant antimicrobial activities. Conclusion: The synthesized derivatives from 4a to 4e have exhibited various medicinal activities and could be emerged as lead compounds and further explored as potential therapeutic agents.

267. Open Tubular Capillary Electrochromatography-Mass Spectrometry and Liquid Chromatography-Mass Spectrometry for Targeted Metabolomics

Abstract

Tryptophan (TRP) is one of the nine essential amino acids metabolized through various metabolic pathways in the human body. The kynurenine pathway (KP) of TRP metabolism is known to be regulated differently in different disease conditions. The accurate measurement of the TRP metabolites and study of their regulation in biofluids under different disease conditions is of greaat importance. The work in this dissertation presents the development of repeatable and sensitive capillary electrochromatography (CEC) and liquid chromatography (LC) with tandem mass spectrometry (MS/MS) detction methods for quantitative analysis of tryptophan and its kynurenine pathway metabolites in human cerebrospinal fluid and plasma. The 4-VPBA was evaluated as a novel stationary phase and its applications in CEC-MS/MS separation and quantitative analysis of TRP and KYN in human plasma samples was deminstrated. The development of repeatable, selective, and highly sensitive LC-MS/MS method using reversed phase (C-8) for the simultaneous separation and quantitation of TRP and its eight metabolites in human cerebrospinal fluid (CSF) and plasma samples is reported. Using the developed reversed-phase separation method, the baseline separation of all nine target metabolites was achieved for the first time to our knowledge. The dynamic multiple reaction monitoring (MRM) feature of triple quadrupole MS was utilized to improve the selectivity and sensitivity of the developed multi-analyte method. Finally, this dissertation presents the correlations of TRP metabolites in two biofluids (CSF and plasma) and their regulations in neuroinflammation conditions.

Abstract

A series of new heterocyclic disperse dyes has been prepared by subsequent diazotization of 2-methyl-3-[3’-aminophthalimido]-4(3H)-quinazolinone and coupling with various mono- and di-N-substituted derivatives of aniline. All the disperse dyes were characterized by their percentage yield, melting point, UV-visible spectrum, elemental analysis, infrared spectrum and dyeing performance on nylon 66 and polyester fibres. The percentage dye bath exhaustion on different fibres was found to be reasonably good and acceptable. The dyed fibres showed fair to fairly good to good fastness to light and very good to excellent fastness to washing, rubbing, perspiration and sublimation.

Abstract

A new fused heterocyclic compound, 11-amino-13H-acenaphtho[1,2-e]pyridazino [3,2-b]quinazolin-13-one was synthesized and used to prepare a novel series of heterocyclic mono azo dyes by coupling with various naphthols. All the mono azo dyes were characterized by their melting point, elemental analysis, UV-visible spectrum, infrared spectrum and dyeing performance on nylon and polyester fibres. The percentage dye bath exhaustion on different fibres was found to be reasonably good and acceptable. The dyed fibres show fair to good fastness to light and very good to excellent fastness to washing, rubbing, perspiration and sublimation.

299. Investigation on electrical discharge coating of brass and copper powder.

Abstract

In this study, electrical discharge coating (EDC) has been performed using green compact electrode of brass and copper powder to fabricate thick coating onto the mild steel substrate. The influence of EDC process parameters on surface morphology, electrode wear, and tribological behavior of coating has been investigated. Experimental result shows fewer pores and micro-cracks on the coated sample with powder mixture ratio of brass:Cu/60:40, peak current of 7A and 70% duty cycle. It is found that the thickness of the coating layer increases with the peak current and the range of the obtained coating layer thickness is 136.19–216.68 µm. The X-ray diffraction profile of coating confirms the presence of elements Cu, CuZn and Fe in the deposited layer. Further, a significant reduction in corrosion resistance and friction coefficient has been observed in the coated sample compared to mild steel substrate. [ABSTRACT FROM AUTHOR]
