Understanding the metabolic interactions between bacteria in natural habitat at the single-cell level and the contribution of individual cell to their functions is essential for exploring the dark matter of uncultured bacteria. The combination of Raman-activated cell sorting (RACS) and single-cell Raman spectra (SCRS) with unique fingerprint characteristics makes it possible for research in the field of microbiology to enter the single cell era. This review presents an overview of current knowledge about the research progress of recognition and assessment of single bacterium cell based on RACS and further research perspectives. We first systematically summarize the label-free and non-destructive RACS strategies based on microfluidics, microdroplets, optical tweezers, and specially made substrates. The importance of RACS platforms in linking target cell genotype and phenotype is highlighted and the approaches mentioned in this paper for distinguishing single-cell phenotype include surface-enhanced Raman scattering (SERS), biomarkers, stable isotope probing (SIP), and machine learning. Finally, the prospects and challenges of RACS in exploring the world of unknown microorganisms are discussed. Spinal injury is among the most severe and feared injuries an athlete may face. We present an up-to-date review of the recent literature, stratifying recommendations based on injury location (cervical, thoracic, and lumbar spine) and type, as well as, the level of competitive play (high school, collegiate, professional). A literature search was completed to identify all publications reporting return to play guidelines for athletic injuries or injury-related surgery irrespective of the study design. Publication dates were not restricted by year. Search terms used included "return to play" and "spinal injury" on National Library of Medicine (PubMed) and Google Scholar. Selection criteria for literature included axial spine injury guidelines for athletic participation post-injury or post-surgery. Literature found from the search criteria was sorted based on level of competition and location of axial spine injury involved. It was found that professional athletes are more likely to suffer severe spinal injuries, require surgery, and necessitate a longer return to play (RTP), with high school and college athletes usually returning to play within days or weeks. Injuries occur mainly within contact sports and concordance exists between initial and subsequent spinal injuries. Adequate rest, rehabilitation, and protective equipment alongside the education of athletes and coaches are recommended. In conclusion, a multidisciplinary approach to patient management is required with consideration for the emotional, social, and perhaps financial impact that spinal injury may have upon the athlete. Consensus from the literature states that in order for an athlete to safely return to play, that athlete should not be actively suffering from pain, should have a full range of motion, and complete return of their strength in the absence of neurological deficit. Kin recognition plays an important role in social behavior and evolution, but the proximate mechanisms by which individuals recognize kin remain poorly understood. In many species, individuals form a "kin template" that they compare with conspecifics' phenotypes to assess phenotypic similarity—and, by association, relatedness. Individuals may form a kin template through self-inspection (i.e., self-referencing) and/or by observing their rearing associates (i.e., family referencing). However, despite much interest, few empirical studies have successfully disentangled self-referencing and family referencing. Here, we employ a novel set of breeding crosses using the Trinidadian guppy (Poecilia reticulata) to disentangle referencing systems by manipulating exposure to kin from conception onward. We show that guppies discriminate among their full and maternal half siblings, which can be explained only by self-referencing. Additional behavioral experiments revealed no evidence that guppies incorporate the phenotypes of their broodmates or mother into the kin template. Finally, by manipulating the format of our behavioral tests, we show that olfactory communication is both necessary and sufficient for kin discrimination. These results provide robust evidence that individuals recognize kin by comparing the olfactory phenotypes of conspecifics with their own. Craniofacial morphogenesis depends on proper migration of neural crest cells and their interactions with placodes and other cell types. Hox genes provide positional information and are important in patterning the neural crest and pharyngeal arches (PAs) for coordinated formation of craniofacial structures. Hox genes are expressed in the surface ectoderm and epibranchial placodes, their roles in the pharyngeal epithelium and their downstream targets in regulating PA morphogenesis have not been established. We altered the Hox code in the pharyngeal region of the Hoxb3 Tg /+ mutant, in which Hoxb3 is driven to ectopically expressed in Hoxb2 domain in the second pharyngeal arch (PA2). In the transgenic mutant, ectopic Hoxb3 expression was restricted to the surface ectoderm, including the proximal epibranchial placodal region and the distal pharyngeal epithelium. The Hoxb3 Tg /+ mutants displayed hypoplasia of PA2, multiple neural crest-derived facial skeletal and nerve defects. Interestingly, we found that in the Hoxb3 Tg /+ mutant, expression of the Notch ligand Jag1 was specifically up-regulated in the ectodermal pharyngeal epithelial cells of PA2. By molecular experiments, we demonstrated that Hoxb3 could bind to an upstream genomic site S2 and directly regulate Jag1 expression. In the Hoxb3 Tg/+ mutant, elevated expression of Jag1 in the pharyngeal epithelium led to abnormal cellular interaction and deficiency of neural crest cells migrating into PA2. In summary, we showed that Hoxb3 regulates Jag1 expression and proposed a model of pharyngeal epithelium and neural crest interaction during pharyngeal arch development. [ABSTRACT FROM AUTHOR]

Neural crest (NC) cells give rise to a wide variety of cell types and tissues, such as neurons and glial cells in the peripheral nervous system. Sox2, which encodes an HMG‐box transcription factor, is known to mediate pluripotency of primordial germ cells and embryonic stem (ES)/induced pluripotent stem (iPS) cells, and to regulate central nervous system development. Previous studies have revealed that Sox2 is also an important regulator of NC development. This review summarizes the well‐established inhibitory roles of Sox2 in NC formation and subsequent neuronal differentiation of NC‐derived cells. This review also covers recent studies suggesting additional roles for Sox2 in early NC development, neurogenesis, and glial differentiation of NC‐derived cells. [ABSTRACT FROM AUTHOR]

In this first Editorial for 2021, and our 50th overall (!), we highlight four Original Articles describing (1) the intracellular localization of the guanine nucleotide exchange factor EGFP-fused DENND1B in several different cell lines by live-cell confocal microscopy); (2) the phenotypic and functional characterization of cardiac macrophages during heart development; (3) the increased level of the redox protein thioredoxin 1 (Trx1) in the bone and bone marrow following ischemi-reperfusion injury; and (4) description of a new protocol for double whole-mount ISH combined with immunoperoxidase staining for analyzing cellular gene expression patterns during forelimb development in avian embryos. Cell migration plays a crucial role in early embryonic development. The chemokine receptor CXCR4 has been reported to guide migration of neural crest cells (NCCs) to form the dorsal root ganglia (DRG) and sympathetic ganglia (SG). CXCR4 also plays an important part during the formation of limb and cloacal muscles. NCCs migration and muscle formation during embryonic development are usually considered separately, although both cell lineages migrate in close neighbourhood and have markers in common. In this study, we present a new method for the simultaneous detection of CXCR4, mesodermal markers and NCCs markers during chicken embryo developmental stages HH18–HH25 by combining double whole-mount in situ hybridization (ISH) and immunostaining on floating vibratome sections. The simultaneous detection of CXCR4 and markers for the mesodermal and neural crest cells in multiple labelling allowed us to compare complex gene expression patterns and it could be easily used for a wide range of gene expression pattern analyses of other chicken embryonic tissues. All steps of the procedure, including the preparation of probes and embryos, prehybridization, hybridization, visualization of the double labelled transcripts and immunostaining, are described in detail. Mathematical and computational models can assist in gaining an understanding of cell behavior at many levels of organization. Here, we review models in the literature that focus on eukaryotic cell motility at 3 size scales: intracellular signaling that regulates cell shape and movement, single cell motility, and collective cell behavior from a few cells to tissues. We survey recent literature to summarize distinct computational methods (phase-field, polygonal, Cellular Potts, and spherical cells). We discuss models that bridge between levels of organization, and describe levels of detail, both biochemical and geometric, included in the models. We also highlight links between models and experiments. We find that models that span the 3 levels are still in the minority. Author summary: In this review paper, we summarize the literature on computational models for cell motility, from the biochemical networks that regulate it, to the behavior of 1 and many cells. We discuss the distinct approaches used at each level, and how models can build bridges between the different size scales. We find models at many different levels of biological detail, and discuss their relative contributions to our understanding of single and collective cell behavior. The mammalian middle ear comprises a chain of ossicles, the malleus, incus, and stapes that act as an impedance matching device during the transmission of sound from the tympanic membrane to the inner ear. These ossicles are derived from cranial neural crest cells that undergo endochondral ossification and subsequently differentiate into their final functional forms. Defects that occur during middle ear development can result in conductive hearing loss. In this review, we summarize studies describing the crucial roles played by signaling molecules such as sonic hedgehog, bone morphogenetic proteins, fibroblast growth factors, notch ligands, and chemokines during the differentiation of neural crest into the middle ear ossicles. In addition to these cell‐extrinsic signals, we also discuss studies on the function of transcription factor genes such as Foxi3, Tbx1, Bapx1, Pou3f4, and Gsc in regulating the development and morphology of the middle ear ossicles. New York City's Rikers jail complex is gripped by a crisis of legitimacy. Following a series of investigations, it has been denounced as a major symbol of criminal justice dysfunction, with calls for its closure and replacement with new smaller "state of the art" jails. Yet, when it opened, Rikers was hailed as a "model" facility, at the cutting edge of prison design and prisoner rehabilitation. Biologically-informed neural networks (BINNs), an extension of physics-informed neural networks [1], are introduced and used to discover the underlying dynamics of biological systems from sparse experimental data. In the present work, BINNs are trained in a supervised learning framework to approximate in vitro cell biology assay experiments while respecting a generalized form of the governing reaction-diffusion partial differential equation (PDE). By allowing the diffusion and reaction terms to be multilayer perceptrons (MLPs), the nonlinear forms of these terms can be learned while simultaneously converging to the solution of the governing PDE. Further, the trained MLPs are used to guide the selection of biologically interpretable mechanistic forms of the PDE terms which provides new insights into the biological and physical mechanisms that govern the dynamics of the observed system. The method is evaluated on sparse real-world data from wound healing assays with varying initial cell densities [2]. Author summary: In this work we extend equation learning methods to be feasible for biological applications with nonlinear dynamics and where data are often sparse and noisy. Physics-informed neural networks have recently been shown to approximate solutions of PDEs from simulated noisy data while simultaneously optimizing the PDE parameters. However, the success of this method requires the correct specification of the governing PDE, which may not be known in practice. Here, we present an extension of the algorithm that allows neural networks to learn the nonlinear terms of the governing system without the need to specify the mechanistic form of the PDE. Our method is demonstrated on real-world biological data from scratch assay experiments and used to discover a previously unconsidered biological mechanism that describes delayed population response to the scratch. This article re-examines the ILO's normative outlook on prison labour, arguing that it is out of touch with the realities on the ground, where public/private hybrid forms of prison labour are proliferating. The authors bring to light the controversy surrounding the position taken by the ILO, as member States repeatedly demand that it relax, and increasingly defy, its dichotomous stance. They illustrate the heavy price to be paid if the ILO stays on its current course, but also if it adopts the position favoured by some of these member States. The article explores EDTA-induced platelet agglutination is much more commonly observed with an estimated incidence being as high as 0.1%.1 Leukoagglutination is a rare EDTA-dependent event, which usually involves neutrophils, resulting in pseudoneutropenia. There seem to be at least three different kinds of mechanisms that can lead to spurious thrombocytopenia and neutropenia.

The neural crest (NC) cells and cranial placodes are two ectoderm-derived innovations in vertebrates that led to the acquisition of a complex head structure required for a predatory lifestyle. They both originate from the neural border (NB), a portion of the ectoderm located between the neural plate (NP), and the lateral non-neural ectoderm. The NC gives rise to a vast array of tissues and cell types such as peripheral neurons and glial cells, melanocytes, secretory cells, and cranial skeletal and connective cells. Together with cells derived from the cranial placodes, which contribute to sensory organs in the head, the NC also forms the cranial sensory ganglia. Multiple in vivo studies in different model systems have uncovered the signaling pathways and genetic factors that govern the positioning, development, and differentiation of these tissues. In this literature review, we give an overview of NC and placode development, focusing on the early gene regulatory network that controls the formation of the NB during early embryonic stages, and later dictates the choice between the NC and placode progenitor fates. Directed cell migration is essential all along an individual's life, from embryogenesis to tissue repair and cancer metastasis. Thus, due to its biomedical relevance, directed cell migration is currently under intense research. Directed cell migration has been shown to be driven by an assortment of external biasing cues, ranging from gradients of soluble (chemotaxis) to bound (haptotaxis) molecules. In addition to molecular gradients, gradients of mechanical properties (duro/mechanotaxis), electric fields (electro/galvanotaxis) as well as iterative biases in the environment topology (ratchetaxis) have been shown to be able to direct cell migration. Since cells migrating in vivo are exposed to a challenging environment composed of a convolution of biochemical, biophysical, and topological cues, it is highly unlikely that cell migration would be guided by an individual type of "taxis." This is especially true since numerous molecular players involved in the cellular response to these biasing cues are often recycled, serving as sensor or transducer of both biochemical and biophysical signals. In this review, we confront literature on Xenopus cephalic neural crest cells with that of other cell types to discuss the relevance of the current categorization of cell guidance strategies. Furthermore, we emphasize that while studying individual biasing signals is informative, the hard truth is that cells migrate by performing a sort of "mixotaxis," where they integrate and coordinate multiple inputs through shared molecular effectors to ensure robustness of directed cell motion. Cardiac neural crest cells (cNCCs) are a migratory cell population that stemfromthe cranial portion of the neural tube. They undergo epithelialto- mesenchymal transition and migrate through the developing embryo to give rise to portions of the outflow tract, the valves and the arteries of the heart. Recent lineage-tracing experiments in chick and zebrafish embryos have shown that cNCCs can also give rise to mature cardiomyocytes. These cNCC-derived cardiomyocytes appear to be required for the successful repair and regeneration of injured zebrafish hearts. In addition, recent work examining the response to cardiac injury in the mammalian heart has suggested that cNCC-derived cardiomyocytes are involved in the repair/regeneration mechanism. However, the molecular signature of the adult cardiomyocytes involved in this repair is unclear. In this Review, we examine the origin,migration and fates of cNCCs. Investigations of the cellular and molecular mechanisms that mediate the development of the autonomic nervous system have identified critical genes and signaling pathways that, when disrupted, cause disorders of the autonomic nervous system. This review summarizes our current understanding of how the autonomic nervous system emerges from the organized spatial and temporal patterning of precursor cell migration, proliferation, communication, and differentiation, and discusses potential clinical implications for developmental disorders of the autonomic nervous system, including familial dysautonomia, Hirschsprung disease, Rett syndrome, and congenital central hypoventilation syndrome. Background: Vitamin A regulates patterning of the pharyngeal arches, cranial nerves, and hindbrain that are essential for feeding and swallowing. In the LgDel mouse model of 22q11.2 deletion syndrome (22q11DS), morphogenesis of multiple structures involved in feeding and swallowing are dysmorphic. We asked whether changes in maternal dietary Vitamin A intake can modify cranial nerve, hindbrain and pharyngeal arch artery development in the embryo as well as lung pathology that can be a sign of aspiration dysphagia in LgDel pups. Methods: Three defined amounts of vitamin A (4, 10, and 16 IU/g) were provided in the maternal diet. Cranial nerve, hindbrain and pharyngeal arch artery development was evaluated in embryos and inflammation in the lungs of pups to determine the impact of altering maternal diet on these phenotypes. Results: Reduced maternal vitamin A intake improved whereas increased intake exacerbated lung inflammation in LgDel pups. These changes were accompanied by increased incidence and/or severity of pharyngeal arch artery and cranial nerve V (CN V) abnormalities in LgDel embryos as well as altered expression of Cyp26b1 in the hindbrain. Conclusions: Our studies demonstrate that variations in maternal vitamin A intake can influence the incidence and severity of phenotypes in a mouse model 22q11.2 deletion syndrome. Collective migration, themovement of groups inwhich individuals affect the behaviour of one another, occurs at practically every scale, from bacteria up to whole species' populations. Universal principles of collective movement can be applied at all levels. In this review, we will describe the rules governing collectivemotility, with a specific focus on the neural crest, an embryonic stem cell population that undergoes extensive collective migration during development. A great deal has been written about the changing nature and direction of criminology over the past two decades, including claims that we are moving into a "new penology." Many of these claims are suggestive rather than authoritative. In contrast to most commentaries on the subject, this article provides longer historical overview and attempts to sketch out how the central structures or "pillars" of the criminal justice system have become weakened and eroded over the last 200 years and how the emergence of body of "new crimes" and their regulation is challenging what might be called the "old criminology." The emergence of new relations between victims and offenders, criminal justice and social justice, as well as the development of innovative modes of regulation are, it is argued, changing the social and criminological landscape. The generation of a feature matrix is the first step in conducting machine learning analyses on complex data sets such as those containing DNA, RNA or protein sequences. These matrices contain information for each object which have to be identified using complex algorithms to interrogate the data. They are normally generated by combining the results of running such algorithms across various datasets from different and distributed data sources. Thus for non-computing experts the generation of such matrices prove a barrier to employing machine learning techniques. Further since datasets are becoming larger this barrier is augmented by the limitations of the single personal computer most often used by investigators to carry out such analyses. Here we propose a user friendly system to generate feature matrices in a way that is flexible, scalable and extendable. Additionally by making use of The Berkeley Open Infrastructure for Network Computing (BOINC) software, the process can be speeded up using distributed volunteer computing possible in most institutions. The system makes use of a combination of the Grid and Cloud User Support Environment (gUSE), combined with the Web Services Parallel Grid Runtime and Developer Environment Portal (WS-PGRADE) to create workflow-based science gateways that allow users to submit work to the distributed computing. This report demonstrates the use of our proposed WS-PGRADE/gUSE BOINC system to identify features to populate matrices from very large DNA sequence data repositories, however we propose that this system could be used to analyse a wide variety of feature sets including image, numerical and text data. Objective: To investigate the assessment of feasibility and safety of laparoscopic decortication of simple renal cysts in elderly patients. Methods: The data of 114 patients who underwent laparoscopic decortication for simple renal cyst between October 2011 and May 2019 were retrospectively evaluated. Patients' age, gender and ASA scores; operation time; cyst size and location; complications; hospital stays and success rates of operations were recorded. Results: The mean age of the patients were 73.8 ± 8.2 years. Forty-eight (42.1%) of the patients were female and 66 (57.9%) were male. Cysts were localized on the right side in 61 (53.5%) patients and on the left side in 53 (46.5%) patients; and mean cyst size was 72.83 ± 31.13 mm. Also, they were localized on the anterior in 88 (77.2%) patients and on the posterior of the kidney in 26 (22.8%) patients. Preoperative ASA score distributions were ASA-I in 12 patients (10.5%), ASA-II in 31 patients (27.2%), ASA-III in 68 patients (59.7%), and ASA-IV in 3 patients (2.6%). Mean operative time was 41.4 ± 5.6 min. Clavien grade 1 complications were observed in 5 (10.4%) patients. Median hospitalization time was 1 day. No recurrence was occurred in any patient in the postoperative period. Conclusions: Although laparoscopic surgery has some risks and complications for elderly patients, laparoscopic decortication can be applied safely and effectively in treatment of simple renal cyst in that patient population. Laser‐induced breakdown spectroscopy (LIBS) and Raman spectroscopy are powerful key techniques for the geoanalytical exploration of extraterrestrial bodies, especially when combined. Their data are complementary, which motivates the question of how it can be best combined to maximize the scientific output. For this study, LIBS and Raman data from pure sulfates and their mixtures as well as from other Mars‐relevant salts such as carbonates, chlorides, perchlorates, and sulfates in a basaltic matrix were measured and investigated. All measurements were performed with miniaturized setups, and LIBS experiments were done in simulated Martian atmospheric conditions. Multivariate data analysis (MVA) techniques such as principal component analysis (PCA) and partial least squares discriminant analysis (PLS‐DA) were employed to evaluate the potential for identifying the sulfates or the salts in the basalt with LIBS and Raman data alone and with their low‐level fused data. We found that low‐level data fusion, that is, combination of LIBS and Raman spectra at the data level, can improve the identification of sulfates and salts. Although the approach of low‐level data fusion aims to use all relevant information from both techniques, we observed that not all benefits from the single models are completely represented by the fused model. The computation and performance of appropriate MVA models are affected by the weighting of the single spectra in the combined one, by the dimensionality of the MVA models, and in case of PLS‐DA, by the given input data. From this study, we conclude that generally, data fusion of LIBS and Raman is an advantage for the identification of unknown samples but that more levels, especially, high‐level data fusion (decision level), should be further investigated. This article examines the troubled career of Black inner-city principal Dr. Roy Alexander West from Richmond, Virginia. In conjunction with recently organized school board minutes, personal interviews, newspapers accounts, court cases, and archival collections, the article argues that Black public school administrators struggled among themselves over the use of proactive, tough-on-crime discipline during the 1980s; a time when urban schools shifted their focus from integration to educational reform. This article challenges the scholarly consensus by Hinton (2016), Sudler (2017), Simmons (2017), and Agyapong (2018) that the school-to-prison pipeline evolved into a fixed network where underfunded public schools worked hand-in-hand with police, courts, and legislators to criminalize and incarcerate Black children. The urban Black educational crisis in the 1980s should be studied by urban, African American, and education scholars because it reflects how punishing Black children became a vital part of America's response to the poverty, crime, and de facto segregation caused by the failures of the modern Civil Rights Movement. Cleft lip and palate are the most common congenital abnormalities that occur early in pregnancy. The majority of cranial mesenchyme is derived from cranial neural crest cells that differentiate into odontoblasts, cartilage, craniofacial bone, and connective tissue. A subset of these cells differentiates into cranial ganglia. We have previously reported an induction protocol of cranial neural crest cell-like cells from human pluripotent stem cells. This study tested detection of the cytotoxic sensitivities of dental materials, including titanium ions, palladium ions, and hydroxyethyl methacrylate, on the cell viability of induced cranial neural crest cell-like cells (iNC-LCs) derived from Tic human induced pluripotent stem cell (hiPSC) line. Further, the sensitivity was compared with those of human fetal lung fibroblastic cell line MRC-5, which is origin of Tic hiPSC, and osteoblastic cell line MC3T3-E1 which was derived from mouse calvaria. The results suggested that this cell-based assay system using iNC-LCs is a potential method for in vitro screening as an alternative to animal testing to predict toxic effects of dental materials on early craniofacial development. The application of laser-induced breakdown spectroscopy (LIBS) for classification of rocks with the total alkali-silica (TAS) diagram is considered in this work. The attractive feature of LIBS compared to other spectrochemical techniques used for TAS classification in geological studies and especially for Mars exploration, such as X-ray fluorescence (XRF) and alpha particle X-ray spectrometry (APXS), consists in its ability for in situ stand-off analysis of samples in their natural state. The analysis accuracy of LIBS for geological samples, rocks, crusts, and soils, is however, seriously affected by the matrix effects due to the various chemical compositions and the different surface physical states of the samples. In this work with an originally designed experiment, a collection of 20 rocks were analyzed in the three different states: natural rock, with a polished surface and pressed pellets. This allowed the simulation of a real application scenario where a prediction model built with calibration standards prepared in a laboratory in the form of pressed pellets, is used to predict chemical compositions with the LIBS spectra acquired from geological samples in their natural state. In the framework of the TAS classification, the concentration of the major compounds in the rocks, SiO2, Na2O, and K2O, was thus determined first with univariate calibration models which were clearly affected by both the chemical and physical matrix effects. Multivariate calibration models were then developed based on machine learning to predict the concentrations of the above 3 compounds and perform the TAS classification. An efficient reduction of the chemical matrix effect has been demonstrated, and the influence of the physical matrix effect has been investigated in the cases of both univariate and multivariate calibration models. Recent reviews on sexual dichromatism in frogs included Mannophryne trinitatis as the only example they could find of dynamic dichromatism (males turn black when calling) within the family Aromobatidae and found no example of ontogenetic dichromatism in this group. We demonstrate ontogenetic dichromatism in M. trinitatis by rearing post-metamorphic froglets to near maturity: the throats of all individuals started as grey coloured; at around seven weeks, the throat became pale yellow in some, and more strongly yellow as development proceeded; the throats of adults are grey in males and variably bright yellow in females, backed by a dark collar. We demonstrated the degree of throat colour variability by analysing a large sample of females. The red: green (R:G) ratio ranged from ~1.1 to 1.4, reflecting variation from yellow to yellow/orange, and there was also variation in the tone and width of the dark collar, and in the extent to which the yellow colouration occurred posterior to the collar. Female M. trinitatis are known to be territorial in behaviour. We show a positive relationship between throat colour (R:G ratio) and escape performance, as a proxy for quality. Our field observations on Tobago's M. olmonae showed variability in female throat colour and confirmed that males in this species also turn black when calling. Our literature review of the 20 Mannophryne species so far named showed that all females have yellow throats with dark collars, and that male colour change to black when calling has been reported in eight species; in the remaining 12 species, descriptions of males calling are usually lacking so far. We predict that both dynamic and ontogenetic sexual dichromatism are universal in this genus and provide discussion of the ecological role of dichromatism in this genus of predominantly diurnal, non-toxic frogs, with strong paternal care of offspring. Display of a peptide or protein of interest on the filamentous phage (also known as bacteriophage), a biological nanofiber, has opened a new route for disease diagnosis and therapy as well as proteomics. Earlier phage display was widely used in protein–protein or antigen–antibody studies. In recent years, its application in nanomedicine is becoming increasingly popular and encouraging. We aim to review the current status in this research direction. For better understanding, we start with a brief introduction of basic biology and structure of the filamentous phage. We present the principle of phage display and library construction method on the basis of the filamentous phage. We summarize the use of the phage displayed peptide library for selecting peptides with high affinity against cells or tissues. We then review the recent applications of the selected cell or tissue targeting peptides in developing new targeting probes and therapeutics to advance the early diagnosis and targeted therapy of different diseases in nanomedicine. We also discuss the integration of antibody phage display and modern proteomics in discovering new biomarkers or target proteins for disease diagnosis and therapy. Finally, we propose an outlook for further advancing the potential impact of phage display on future nanomedicine. This article examines how we might best examine the most extreme and inaccessible corners of imprisonment. Drawing on a study of life imprisonment without parole (LWOP) in California, this article argues that prisoners' letters can shed exceptional light on the hidden and unspoken experiences of imprisonment. It provides an in-depth methodological and reflective outline of how to use prisoners' letters, a familiar tactic in prison researchers' and advocates' toolkits, but a research implement that is rarely discussed explicitly. By better understanding how to do prison research with letters and the emotions these might provoke in the process, this article demonstrates the very specific and distinct value of the method, in particular how letters can contribute to knowledge about extreme penal severity. Directional migration during embryogenesis and tumor progression faces the challenge that numerous external signals need to converge to precisely control cell movement. The Rho guanine exchange factor (GEF) Trio is especially well suited to relay signals, as it features distinct catalytic domains to activate Rho GTPases. Here, we show that Trio is required for Xenopus cranial neural crest (NC) cell migration and cartilage formation. Trio cell-autonomously controls protrusion formation of NC cells and Trio morphant NC cells show a blebbing phenotype. Interestingly, the Trio GEF2 domain is sufficient to rescue protrusion formation and migration of Trio morphant NC cells. We show that this domain interacts with the DEP/C-terminus of Dishevelled (DVL). DVL - but not a deletion construct lacking the DEP domain - is able to rescue protrusion formation and migration of Trio morphant NC cells. This is likely mediated by activation of Rac1, as we find that DVL rescues Rac1 activity in Trio morphant embryos. Thus, our data provide evidence for a novel signaling pathway, whereby Trio controls protrusion formation of cranial NC cells by interacting with DVL to activate Rac1. [ABSTRACT FROM AUTHOR]

Tumours evolve to cope with environmental stresses or challenges such as nutrient starvation, depletion of survival factors, and unbalanced mechanical forces. The uncontrolled growth and aberrant deregulation of core cell homeostatic pathways induced by genetic mutations create an environment of stress. Here, we explore how the adaptations of tumours to the changing environment can drive changes in the motility machinery of cells, affecting migration, invasion, and metastasis. Tumour cells can invade individually or collectively, or they can be extruded out of the surrounding epithelium. These mechanisms are thought to be modifications of normal processes occurring during development or tissue repair. Cation substitution is one of the effective ways to improve Cu2ZnSn(S,Se)4 (CZTSSe) photovoltaic performance. However, the commonly reported substitutes, Ag and Cd, are not ideal as they detract from the earth‐abundant and nontoxic motivation of CZTSSe. Herein, the role of Mn substitution in sulfide and sulfoselenide films are compared in terms of optoelectronic properties and device performance. CZT(S,Se) + CMZT(S,Se) double‐layered structures are fabricated by a sol–gel spin‐coating method with variations in the CMZT(S,Se) layer thickness. It is found that a smaller amount of Mn is required to achieve the highest photovoltaic performance in sulfoselenide films in comparison with sulfide‐based films. All device parameters (particularly Voc and fill factor) of the sulfoselenide films are improved as compared with the sulfide system. Using a combination of capacitance–voltage, drive‐level capacitance profiling, and photoluminescence (PL), it is found that the sulfoselenide film has a smaller interface defect density and higher hole mobility and PL intensity, which suggest much more effective charge separation and transport. The article develops a new understanding of neoliberal security provision on the basis of available accounts of three different "states"; the penal state, the regulatory state, and the activating welfare state. I argue that these forms of state intervention provide individuals with security in the sense that anxiety is temporarily alleviated, while stabilizing the conflictual dynamic of global power structures. Marketization and organizational control account for the specifically neoliberal character. Such an understanding matters because it directs attention to the dynamic between state practices and individual experience, and the multitude of mechanisms, which not only promise but also provide security, however temporary and partial. Matrix metalloproteinases have a broad spectrum of substrates ranging from extracellular matrix components and adhesion molecules to chemokines and growth factors. Despite being mostly secreted,MMPs have been detected in the cytosol, the mitochondria or the nucleus. Although most of the attention is focused on their role in matrix remodeling, the diversity of their substrates and their complex trafficking open the possibility for non-canonical functions. Yet in vivo examples and experimental demonstration of the physiological relevance of such activities are rare. Here, we have used chick neural crest (NC) cells, a highly migratory stem cell population likened to invasive cancer cells, as a model for physiological epithelialmesenchymal transition (EMT). We demonstrate that MMP14 is required for NC delamination. Interestingly, this role is independent of its cytoplasmic tail and of its catalytic activity. The quality of stem cells obtained through serial subcultivation is the pivotal factor determining the therapeutic effectiveness of regenerative medicine. However, an effective quality monitoring system for cell culture is yet to be established. Detailed parameter studies of the migratory behavior of stem cells at different passages may provide insight into the deterioration of stemness. Thus, this study aimed to evaluate the feasibility of quantitative bioimage analysis for monitoring stem cell quality during in vitro culture and to explore the passaging effects on stem cell migration. An image‐based analytical tool using cell tracking, cytometric analyses, and gating with time‐lapse microscopy was developed to characterize the migratory behavior of human mesenchymal stem cells (hMSCs) isolated from human adipose tissue (hADAS) and placenta (hPDMC) cultured on chitosan membranes. Quantitative analysis was performed for the single cells and assembled spheroids selected from 15 videos of Passages 3, 5, and 11 for hADAS and those from 12 videos of Passages 7, 11, and 16 for hPDMC. These passages were selected to represent the young, matured, and degenerated stem cells, respectively. Migratory behavior varied with cell passages. The mobility of single hMSCs decreased at degenerated passages. In addition, enhancement of mobility, due to transformation from single cells to spheroids, occurred at each passage. The young hMSCs seemed more likely to move as single cells rather than as aggregates. Once matured, they tended to aggregate with strong 3D spheroid formability and increased mobility. However, the spheroid formability and mobility decreased at late passage. The increase in aggregation rate with passaging may be a compensatory mechanism to enhance the declining mobility of hMSCs through cell coordination. Cranial neural crest cells are multipotent cells that migrate into the pharyngeal arches of the vertebrate embryo and differentiate into various craniofacial organ derivatives. Therefore, migrating cranial neural crest cells are considered one of the most attractive candidate cell sources in regenerative medicine. We generated cranial neural crest like cell (cNCCs) using mouse-induced pluripotent stem cells cultured in neural crest-inducing medium for 14 days. Subsequently, we conducted RNA sequencing experiments to analyze gene expression profiles of cNCCs at different time points after induction. cNCCs expressed several neural crest specifier genes; however, some previously reported specifier genes such as paired box 3 and Forkhead box D3, which are essential for embryonic neural crest development, were not expressed. Moreover, ETS proto-oncogene 1, transcription factor and sex-determining region Y-box 10 were only expressed after 14 days of induction. Finally, cNCCs expressed multiple protocadherins and a disintegrin and metalloproteinase with thrombospondin motifs enzymes, which may be crucial for their migration. [ABSTRACT FROM AUTHOR]

Rocks are known to fail in a time‐dependent manner similar to many brittle and quasi‐brittle materials. However, observed time dependence of hydraulic fracture initiation has ubiquitously been attributed to fluid flow‐related mechanisms without consideration of the intrinsic time dependence of rock failure. Laboratory delayed hydraulic fracturing breakdown experiments on three rocks (granite, sandstone, and limestone) show sensitivity to rock properties associated with subcritical crack growth as well as fluid viscosity and ambient confining pressure applied to the specimens. A new hydraulic fracture initiation model accounting for subcritical crack growth and fluid flow in a poroelastic medium, as well as the additional energy dissipation required for fluid flow in the rough fracture tip, is then used to estimate the value of the subcritical index n based on characterization experiments. Given this characterization data, the model is shown to be capable of predicting time‐dependent hydraulic fracture initiation with variation of fluid viscosity and confining stress. Background: Cytochrome P450 1b1 (Cyp1b1) deletion and dietary retinol deficiency during pregnancy (GVAD) affect perinatal liver functions regulated by Srebp. Cyp1b1 is not expressed in perinatal liver but appears in the E9.5 embryo, close to sites of retinoic acid (RA) signaling. Hypothesis: Parallel effects of Cyp1b1 and retinol on postnatal Srebp derive from effects in the developing liver or systemic signaling. Approach: Cluster postnatal increases in hepatic genes in relation to effects of GVAD or Cyp1b1 deletion. Sort expression changes in relation to genes regulated by Srebp1 and Srebp2.Test these treatments on embryos at E9.5, examining changes at the site of liver initiation. Use in situ hybridization to resolve effects on mRNA distributions of Aldh1a2 and Cyp26a1 (RA homeostasis); Hoxb1 and Pax6 (RA targets). Assess mice lacking Lrat and Rbp4 (DKO mice) that severely limits retinol supply to embryos. Results: At birth, GVAD and Cyp1b1 deletion stimulate gene markers of hepatic stellate cell (HSC) activation but also suppress Hamp. These treatments then selectively prevent the postnatal onset of genes that synthesize cholesterol (Hmgcr, Sqle) and fatty acids (Fasn, Scd1), but also direct cholesterol transport (Ldlr, Pcsk9, Stard4) and retinoid synthesis (Aldh1a1, Rdh11). Extensive support by Cyp1b1 is implicated, but with distinct GVAD interventions for Srebp1 and Srebp2. At E9.5, Cyp1b1 is expressed in the septum transversum mesenchyme (STM) with β-carotene oxygenase (Bco1) that generates retinaldehyde. STM provides progenitors for the HSC and supports liver expansion. GVAD and Cyp1b1-/- do not affect RA-dependent Hoxb1 and Pax6. In DKO embryos, RA-dependent Cyp26a1 is lost but Hoxb1 is sustained with Cyp1b1 at multiple sites. Conclusion: Cyp1b1-/- suppresses genes supported by Srebp. GVAD effects distinguish Srebp1 and Srebp2 mediation. Srebp regulation overlaps appreciably in cholesterol and retinoid homeostasis. Metazoan animals are characterized by restricted phenotypic heterogeneity (i.e. morphological disparity) of organisms within various species, a feature that contrasts sharply with intra-species morphological diversity observed in the plant kingdom. Robust emergence of morphogenic blueprint in metazoan animals reflects restricted autonomy of individual cells in adoption of fate outcomes such as differentiation. Fates of individual cells are linked to and influenced by fates of neighboring cells at the population level. Such coupling is a common property of all self-organising systems and propels emergence of order from simple interactions between individual cells without supervision by external directing forces. As a consequence of coupling, expected functional relationship between the constituent cells of an organ system is robustly established concurrent with multiple rounds of cell division during morphogenesis. Notably, the molecular regulation of multicellular coupling during morphogenic self-organisation remains largely unexplored. Here, we review the existing literature on multicellular self-organisation with particular emphasis on recent discovery that β-catenin is the key coupling factor that programs emergence of multi-cellular self-organisation by regulating synchronised cycling of individual cells. Our understanding of the neural crest, a key vertebrate innovation, is built upon studies of multiple model organisms. Early research on neural crest cells (NCCs) was dominated by analyses of accessible amphibian and avian embryos, with mouse genetics providing complementary insights in more recent years. The zebrafish model is a relative newcomer to the field, yet it offers unparalleled advantages for the study of NCCs. Specifically, zebrafish provide powerful genetic and transgenic tools, coupled with rapidly developing transparent embryos that are ideal for high‐resolution real‐time imaging of the dynamic process of neural crest development. While the broad principles of neural crest development are largely conserved across vertebrate species, there are critical differences in anatomy, morphogenesis, and genetics that must be considered before information from one model is extrapolated to another. Here, our goal is to provide the reader with a helpful primer specific to neural crest development in the zebrafish model. We focus largely on the earliest events—specification, delamination, and migration—discussing what is known about zebrafish NCC development and how it differs from NCC development in non‐teleost species, as well as highlighting current gaps in knowledge.
