252. Transcriptomic profiling and identification of candidate genes in two Phoebe bournei ecotypes with contrasting cold stress responses.

Abstract

Key message: We identified a set of ecotype-specific cold-responsive genes, providing candidate genes for enhancing cold resistance in Phoebe bournei (Hemsl.) Yang and other woody plants.Abstract: Phoebe bournei (Hemsl.) Yang is an indigenous, endangered, precious timber and ornamental tree in China. Cold stress greatly affects the survival of Phoebe species. To elucidate the molecular mechanisms of cold response and identify candidate genes for improving cold resistance, we used RNA-Seq and qRT-PCR to compare the transcriptomic profiles of two ecotypes of P. bournei, cold-resistant Wuyuan (WY) and cold-sensitive Wuping (WP), under cold stress. A total of 3970 and 4433 genes were differentially expressed in WY and WP, respectively, with 2030 differentially expressed genes (DEGs) were specific to WY. Specifically, the categories “defense response”, “secondary metabolites”, and “ABC transporters” were enriched among WY-specific DEGs. Several transcription factor genes might involve in cold stress response, including RAP2-3, NAC2, NAC43, WRKY47, and WRKY51, were induced only in WY, and CBF3, ZAT10, MYB308, and WER exhibited greater cold stress-induced expression changes in WY compared with WP, which may partially explain the distinct transcriptomic changes induced by cold stress between the two ecotypes. Greater fold changes in the expression of starch metabolism-related genes such as BAM1/3 and lower fold changes in the expression of DPE2 were detected in WY, which was consistent with its greater abundance of total soluble sugars. The stronger transcriptional differentiation of related genes under cold stress in WY may explain its greater cold resistance compared with the WP ecotype, providing important genetic resources for improving cold-resistant characteristics in P. bournei. In addition, two interaction networks were constructed, resulting in identification of a series of hub genes, which facilitates functional dissection of the molecular mechanisms involved in the response to cold stress of P. bournei and extends our ability to improve cold resistance in this valued species. Abstract

The sulfidation-flotation method is commonly used for the treatment of zinc oxide minerals. However, few studies have been conducted on the promoting sulfidation-flotation behavior and mechanism of smithsonite. In this paper, we investigate the promoting sulfidation of smithsonite by zinc sulfide species increase with addition of ammonium chloride (NH 4 Cl) and its effect on flotation performance. The micro-flotation experiment results concluded that the addition of the ammonium chloride improved smithsonite flotation recovery by ∼17%. Zinc ion adsorption experiments and Visual MINTEQ model analysis showed that more zinc sulfide species were produced in the presence of NH 4 Cl. Zeta potential analysis indicated that additional sulfide ion species and isoamyl xanthate ions (IX − ) from the pulp solution adhered to the smithsonite surface as NH 4 Cl was added, and the SEM-EDS study and the contact angle results further confirmed the positive influence of NH 4 Cl on the smithsonite sulfidation process. The TOF-SIMS results provided strong evidence for an increase in zinc sulfide species on the smithsonite surface and an increase in the sodium isoamyl xanthate (NaIX) molecular adsorption on the surface of sulfidized smithsonite with the addition of NH 4 Cl. The thickness of zinc sulfide species formed after promoting sulfidation was ∼29.20 nm, and the thickness of xanthogenate species formed was a few nanometers. The promotion in sulfidation may be due to the combined effect of the improvement of sphalerite precipitation adsorption and the ion-exchange reaction between the sulfide ions and the carbonate ions on the surface of the smithsonite. Abstract

In plant-microbe interactions, plant sugars produced by photosynthesis are not only a carbon source for pathogens, but may also act as signals that modulate plant defense responses. Here, we report that decreasing sorbitol synthesis in apple (Malus domestica) leaves by antisense suppression of ALDOSE-6-PHOSPHATE REDUCTASE (A6PR) leads to downregulation of 56 NUCLEOTIDE BINDING/LEUCINE-RICH REPEAT (NLR) genes and converts the phenotypic response to Alternaria alternata from resistant to susceptible. We identified a resistance protein encoded by the apple MdNLR16 gene and a small protein encoded by the fungal HRIP1 gene that interact in both a yeast two-hybrid assay and a bimolecular fluorescence complementation assay. Deletion of HRIP1 in A. alternata enables gain of virulence on the wild-type control plant. Overexpression of MdNLR16 in two antisense A6PR lines increases resistance, whereas RNAi suppression of MdNLR16 in the wild-type control decreases resistance against A. alternata. MdWRKY79 transcriptionally regulates MdNLR16 by binding to the promoter of MdNLR16 in response to sorbitol, and exogenous sorbitol feeding partially restores resistance of the antisense A6PR lines to A. alternata. These findings indicate that sorbitol modulates resistance to A. alternata via the MdNLR16 protein that interacts with the fungal effector in a classic gene-for-gene manner in apple. 257. Design and test result of a superconducting double-spoke cavity

Abstract

Superconducting multi-spoke cavities are outstanding alternative choice for acceleration of heavy ions in medium velocity regimes. Based on the scheme of China ADS, several researches on the superconducting double-spoke cavities were done and two prototype cavities have been developed. In this paper, the RF design, the mechanical design and fabrication considerations of the bare cavity will be described in detail. After Buffered Chemical Polishing and High Pressure Rinsing, one of the prototype cavities was installed into the Vertical Test Stand for high gradient RF testing at 4.2 K. The measurement results of the quality factor as a function of the accelerating field and the maximum surface field will be presented. An accelerating gradient of more than 15 MV/m is achieved during the test, with maximum surface electric field of 58 MV/m, and maximum surface magnetic field of 117 mT.

Abstract

This work presents a comprehensive analysis of 3D cylindrical junction-less charge trapping memory device performance regarding continuous scaling of the structure dimensions. The key device performance, such as program/erase speed, vertical charge loss, and lateral charge migration under high temperature are intensively studied using the Sentaurus 3D device simulator. Although scaling of channel radius is beneficial for operation speed improvement, it leads to a retention challenge due to vertical leakage, especially enhanced charge loss through TPO. Scaling of gate length not only decreases the program/erase speed but also leads to worse lateral charge migration. Scaling of spacer length is critical for the interference of adjacent cells and should be carefully optimized according to specific cell operation conditions. The gate stack shape is also found to be an important factor affecting the lateral charge migration. Our results provide guidance for high density and high reliability 3D CTM integration.

Abstract

To improve the limitation of applying traditional fault diagnosis method to the diagnosis of multi-concurrent vibrant faults of turbo-generator set, a novel diagnosis approach integrating the wavelet transform with neural network is proposed. The effective eigenvectors are acquired by binary discrete wavelet transform and the fault modes are classified by neural network. The back-propagation (BP) algorithm is used to fulfill the neural network structure and parameter initialization. By means of choosing enough practical samples to verify the wavelet neural network (WNN) and the information representing the faults is inputted into the trained WNN, and according to the output result the type of fault can be determined. Actual applications show that the proposed method can effectively diagnose the multi-concurrent vibrant faults of turbo-generator sets and the diagnosis result is correct. The method can be generalized to other devices' fault diagnosis.

Abstract

The potential application of anti-inflammatory and analgesic compounds in medication and therapeutic care have become of increasing interest. We purified and characterized two novel analgesic and anti-inflammatory peptides, VQ-5 and AQ-5, from the coelomic fluid of the earthworm ( Eisenia foetida ). Their primary structures were determined as VSSVQ and AMADQ, respectively. Both peptides, especially AQ-5, exhibited analgesic activity in mouse models of persistent neuropathic pain and inflammation. AQ-5 also inhibited tumor necrosis factor alpha and cyclooxygenase-2 production. The mitogen-activated protein kinase signaling pathway, which is involved in analgesic and anti-inflammatory functions, was inhibited by AQ-5. Thus, the analgesic and anti-inflammatory effects of these peptides, especially AQ-5, demonstrated their potential as candidates for the development of novel analgesic medicines.
