Abstract

It is a great pleasure to offer this tribute to Professor Leslie M. Beebe and to help celebrate her vital contribution to the field of cross-cultural pragmatics (CCP). There is no denying that Professor Beebe stands as a distinguished scholar in the field. Anyone who wishes to conduct research on second language (L2) pragmatics would not be able to begin without learning of or citing her work. Among her contributions to the field, I would first like to ponder her groundbreaking call for serious attention to L2 pragmatics as early as the mid-1980s, a time when pragmatics was a neglected area in second language acquisition (SLA) research and L2 pedagogy. In a number of scholarly papers and at conferences, she suggested that “the social rules of speaking” are “basics, not frosting on the cake” (Beebe, 1995, p. 4). Professor Beebe is acknowledged among SLA researchers as one of the earliest linguists who was deeply concerned with cross-cultural misunderstandings (which often lead to unfortunate and offensive cultural stereotyping) resulting from a lack of pragmatic competence., Studies in Applied Linguistics and TESOL, Vol. 8 No. 2 (2008)

259. Addition of Rifampicin to Bolton Broth to Inhibit Extended-Spectrum β-Lactamase-Producing Escherichia coli for the Detection of Campylobacter.

Abstract

Exponential growth of extended-spectrum β-lactamase (ESBL)-producing Escherichia coli in Campylobacter media has become a common problem for the detection of Campylobacter in chicken meats. We investigated the minimum inhibitory concentration of 40 ESBL-producing E. coli isolates from meats obtained from various countries against antibacterial agents in Bolton broth (cefoperazone, vancomycin, and trimethoprim). All ESBL-producing E. coli strains were resistant to cefoperazone and vancomycin, whereas 50% of them were resistant to trimethoprim and grew in Bolton broth. We found that 20 μg/mL of rifampicin inhibited the growth of trimethoprim-resistant E. coli strains. Hence, we added 20 μg/mL of rifampicin to Bolton broth to improve the isolation of Campylobacter from chicken carcass rinses. The isolation rate of Campylobacter was significantly higher in the modified broth (44 out of 58, 75.9%, P < 0.05) than in the normal broth (0 out of 58, 0%). Furthermore, the number of agar plates with non- Campylobacter spp. was much lower after enrichment in the modified broth (4 out of 58, 6.9%, P < 0.05) than in the normal broth (58 out of 58, 100%). Abstract

In this letter, we demonstrated a high performance organic thin-film transistor using thermally evaporated amorphous phase MoOx as a hole injection layer between metal electrodes and organic semiconductor. The fabricated organic thin-film transistors showed the field-effect mobility of 7 cm $^{{2}}$ /Vs in linear and saturation regimes and an ON/OFF current ratio of $10^{{7}}$ . The MoOx hole injection layers significantly reduced the injection barrier from metal electrode, resulting in the improvement of ohmic contact properties of a synthesized thiophene-rich heteroacene, dibenzothiopheno [6,5-b:6’,5’-f] thieno [3,2-b] thiophene p-type organic semiconductor, as compared with those with single metals. Furthermore, high performance organic thin-film transistors can be successfully realized with Al electrode, which is not suitable for p-type organic semiconductors due to its low work function by introducing a 75-nm-thick MoOx hole injection layer. 293. Optimization of ethanol extraction of antioxidative phenolic compounds from torrefied oak wood ( Quercus serrata) using response surface methodology.

Abstract

A torrefaction treatment process followed by ethanol extraction was applied for extracting antioxidant components from oak wood. Response surface methodology (RSM) was applied to optimize the ethanol extraction conditions of antioxidant compounds from torrefied oak wood (severity factor Ro = 4.23). Response values were assessed such as total polyphenol content, total flavonoid content, and DPPH radical scavenging activity. Optimal extraction conditions were found as follows: ethanol concentration 69.15 %, extraction temperature 71.60 °C and processing time 70.15 min for total polyphenol content; ethanol concentration 66.93 %, extraction temperature 69.52 °C and time 66.09 min for total flavonoids content; ethanol concentration 68.18 %, extraction temperature 51.77 °C and time 74.22 min for DPPH radical scavenging activity. The experimental values agreed with those predicted within a 95 % confidence interval indicating the suitability of RSM in optimizing the ethanol extraction of antioxidant compounds from the torrefied oak wood. Abstract

In this study, the environmental adaptive metabolic processes were investigated using a psychrotrophic polar bacterium Bacillus pumilus PAMC 23174 in response to various temperatures and nutrients, especially in regard to the synthesis of fatty acids. Fatty acid methyl ester analysis was performed using gas chromatography-mass spectrometry and we found that a sensitive changes in iso-branched fatty acid (iso-15:0) synthesis occurred when adjusting the nutritional ratio of branched chain fatty acids (anteiso/iso) with different temperatures, resulting in a change in the balance of anteiso- and iso-form fatty acids. We also observed that this Arctic bacterium preferred amino acid leucine for the synthesis of fatty acids. The increased and decreased synthesis of iso-form fatty acids in response to different temperatures and leucine preference, changes the fatty acid ratio in bacteria, which further affects the membrane fluidity and it is also directly correlated with survival of bacteria in an extreme environment. Hence, this study suggests that B. pumilus PAMC 23174 is a potential model organism for the analysis of the unique ecological adaptations of polar bacteria in changing and the extreme environments. 295. Chromohalobacter is a Causing Agent for the Production of Organic Acids and Putrescine during Fermentation of Ganjang, a Korean Traditional Soy Sauce.

Abstract

Ganjang, a Korean traditional fermented soy sauce, is prepared by soaking doenjang-meju (fermented soybeans) in approximately 20% (w/v) solar salt solution. The metabolites and bacterial communities during ganjang fermentation were simultaneously investigated to gain a better understanding of the roles of the microbial population. The bacterial community analysis based on denaturing gradient gel electrophoresis of 16S rRNA gene sequences showed that initially, the genus Cobetia was predominant (0 to 10 d), followed by Bacillus (5 to 74 d), and eventually, Chromohalobacter became predominant until the end of the fermentation process (74 to 374 d). Metabolite analysis using ¹H-NMR showed that carbon compounds, such as fructose, galactose, glucose, and glycerol, probably released from doenjang-meju, increased rapidly during the early fermentation period (0 to 42 d). After removal of doenjang-meju from the ganjang solution (42 d), the initial carbon compounds remained nearly constant without the increase of fermentation products. At this point, Bacillus species, which probably originated from doenjang-meju, were predominant, suggesting that Bacillus is not mainly responsible for ganjang fermentation. Fermentation products including acetate, lactate, α-aminobutyrate, γ- aminobutyrate, and putrescine increased quickly with the rapid decrease of the initial carbon compounds, while Chromohalobacter, probably derived from the solar salts, was predominant. Multivariate redundancy analysis indicated that the Chromohalobacter population was closely correlated with the production of the organic acids and putrescine during the ganjang fermentation. These results may suggest that Chromohalobacter is a causing agent responsible for the production of organic acids and putrescine during ganjang fermentation and that the solar salts, not doenjang-meju, is an important microbial source for ganjang fermentation.
