The millennium as an event is a sign of sacred time. It refers to the birth of Christ and raises the question of how Christ shall continue to be embodied in the civilization. The Church should respond to the challenge of the millennium by offering a new vision of human flourishing within the divine order. A new interpretation for the new millennium should be based on popular concepts and represent a convincing crystallization of meaning to help people make sense of things.

The N-terminal domain of HIV-1 integrase contains a pair of His and Cys residues (the HHCC motif) that are conserved among retroviral integrases. Although His and Cys residues are often involved in binding zinc, the HHCC motif does not correspond to any recognized class of zinc binding domain. We have investigated the binding of zinc to HIV-1 integrase protein and find that it binds zinc with a stoichiometry of one zinc per integrase monomer. Analysis of zinc binding to deletion derivatives of integrase locates the binding site to the N-terminal domain. Integrase with a mutation in the HHCC motif does not bind zinc, consistent with coordination of zinc by these residues. The isolated N-terminal domain is disordered in the absence of zinc but, in the presence of zinc, it adopts a secondary structure with a high alpha helical content. Integrase bound by zinc tetramerizes more readily than the apoenzyme and is also more active than the apoenzyme in in vitro integration assays. We conclude that binding of zinc to the HHCC motif stabilizes the folded state of the N-terminal domain of integrase and bound zinc is required for optimal enzymatic activity.

In contrast, Grace Davie, in 'Religion in Britain Since 1945,' notes the continuities of religion through history. Her version of the decline concept is a growing mismatch between statistics on religious practice and on those who profess religious convictions.

A 40-year-old woman with paralysis of the legs, incontinence, and lower back pain was found to have schistosomiasis involving a Schistosoma mansoni infection of the spinal cord and colon. S. mansoni are parasitic worms that lay eggs that can remain in the body for many years. The worms are common in Brazil, where this woman had lived and visited. Magnetic resonance imaging and CT scans showed abnormal tissue and expansion of the spinal cord at the chest level. Biopsies of the lower colon showed clumps of blood and lymph cells around imbedded eggs, typical of schistosomiasis. Blood tests for the worms were positive. The woman was treated with praziquantel and her condition improved, though she experienced a relapse several months later. She was treated with the drug again, for a week, and was able to walk 10 feet without assistance.

IN-(50-212/F185K), a soluble and catalytically active core domain of integrase protein, is formed by the replacement of phenylalanine present at the 185th position by lysine in HIV viral DNA. IN-(50-212/F185K) is a dimer which exists as a monodisperse dimer in solutions. Its stability is probably because of the hydrogen bond formed between Lys185 and Ala105. The catalytic activity of the mutated and unmutated core domain of integrase is less than that of the whole molecule. Integrase is produced by the human immunodeficiency virus type I and helps in the integration of the viral genome with the human DNA.

We have been able to induce a series of lactonisation reactions of unsaturated acids using a carbocationic intermediate formed in situ. The transformation generates five and six membered lactone rings AND forms a new exocyclic C–C bond at the same time, thus removing the need for further derivatisation of typical lactonisation products (such as from iodo lactonisation). Across the ambulatory care network of an integrated health care system, durations of antibiotic therapy prescribed for uncomplicated infections were longer than recommended in 39% of cases.
