An accurate mathematical representation of the mechanical behaviour of human skin is essential when simulating deformations occurring in the skin during body movements or clinical procedures. In this study constitutive stress-strain relationships based on experimental data from human skin in vivo were obtained. A series of multiaxial loading experiments were performed on the forearms of four age- and gender matched subjects. The tissue geometry, together with recorded displacements and boundary forces, were combined in an analysis using finite element modelling. A non-linear optimization technique was developed to estimate values for the material parameters of a previously published constitutive law, describing the stress-strain relationship as a non-linear anisotropic membrane. Ten sets of material parameters where estimated from the experiments, showing considerable differences in mechanical behaviour both between individual subjects as well as mirrored body locations on a single subject. The accuracy of applications that simulate large deformations of human skin could be improved by using the parameters found from the in vivo experiments as described in this study.

A method for fitting parameters in a poroviscoelastic (PVE) model of articular cartilage in the mouse is presented. Indentation is performed using two different sized indenters and then these data are fitted using a PVE finite element program and parameter extraction algorithm. Data from a smaller indenter, a 15 mum diameter flat-ended 60 deg cone, is first used to fit the viscoelastic (VE) parameters, on the basis that for this tip size the gel diffusion time (approximate time constant of the poroelastic (PE) response) is of the order of 0.1 s, so that the PE response is negligible. These parameters are then used to fit the data from a second 170 mum diameter flat-ended 60 deg cone for the PE parameters, using the VE parameters extracted from the data from the 15 mum tip. Data from tests on five different mouse tibial plateaus are presented and fitted. Parameter variation studies for the larger indenter show that for this case the VE and PE time responses overlap in time, necessitating the use of both models.

Needles with asymmetric bevel tips naturally bend when they are inserted into soft tissue. In this study, we present an analytical model for the loads developed at the bevel tip during needle-tissue interaction. The model calculates the loads based on the geometry of the bevel edge and gel material properties. The modeled transverse force developed at the tip is compared to forces measured experimentally. The analytical model explains the trends observed in the experiments. In addition to macroscopic studies, we also present microscopic observations of needle-tissue interactions. These results contribute to a mechanics-based model of robotic needle steering, extending previous work on kinematic models.

Microinjection is an effective technique to introduce foreign materials into a biological cell. Although some semi-automatic and fully-automatic microinjection systems have been developed, a full understanding of the mechanical response of biological cells to injection operation remains deficient. In this paper, a new mechanical model based on membrane theory is proposed. This model establishes a relationship between the injection force and the deformation of biological cells with the quasi-static equilibrium equations, which are solved by the Runge-Kutta numerical method. Based on this model, other mechanical responses can also be inferred, such as the effect of the injector radius, the membrane stress and tension distribution, internal cell pressure, and the deformed cell shape. To verify the proposed model, experiments are performed on microinjection of zebrafish embryos at different developmental stages and medaka embryos at the blastula stage. It is demonstrated that the modeling results agree well with the experimental data, which shows that the proposed model can be used to estimate the mechanical properties of cell biomembranes. (In this paper, biomembrane refers to the membrane-like structures enveloping cells.)

In vivo mechanical properties of bulk tissue have not yet been explored sufficiently. One of the major problems researchers face is the lack of agreement between the constitutive models and the standardised methodologies for experimental studies. The object of this study was to obtain bulk modulus of the upper arm under relaxed and controlled contraction that was 25% of the maximum voluntary contraction. A new testing machine was designed to generate constant load on the upper arm and measure the deformation over time. This device is effectively a cuff that applies controllable pressure on a 47-mm wide band of the upper arm. Six different loads (10, 20, 30, 40, 50 and 60 kgf) were applied over a time of up to a maximum of 120 s. The deflection-time curves obtained show strongly non-linear responses of the bulk tissue. The non-linearity manifested by these deflection-time curves is in terms of both time- and load-dependency. A specific mechanical model was developed to represent the creep behaviour of the bulk tissue. The creep behaviour of the upper arm can be simulated by using four Voigt viscoelastic models in series. The three obvious soft tissues of the upper arm, namely skin, fat and muscle, were modelled in series. The effects of blood vessels and connective tissue were also modelled in series with the previous ones. A mechanical model would provide a more controlled method of studying the mechanical properties of the bulk tissue. The purpose of the current research, therefore, was to develop a mechanical model, which would predict the non-linear, viscoelastic behaviour of the human muscular bulk tissue.

The purpose of this study was to design a 'Traveller Friendly Drug Delivery System' for PM-HCl. Conventional promethazine (PM-HCl) tablets are bitter, need to be taken 1 h before symptoms and water is also needed. Taste-masked granules were produced with Eudragit E100 by extrusion, and analyzed with FTIR, DSC, and XRD. Tablets formulated from granules by direct compression using Ac-Di-Sol, Polyplasdone-XL, Primojel and ion-exchanger Tulsion339 and evaluated for mass uniformity, friability, tensile strength, drug content uniformity, water absorption ratio, in-vitro and in-vivo disintegration time and in-vitro dissolution studies. The observed drug-polymer interactions and reduced crystallinity may be reasons for increased dissolution rates. The formulated tablets were disintegrated within 15 s. Tablets (25 mg PM-HCl) with Ac-Di-Sol (4%) showed complete release within 1 min, while marketed conventional tablets (Phenergan; Rhone-Poulec) release 25% during the same period. A preliminary stability studies for the prepared tablets carried at 30 +/- 2 degrees C/60 +/- 5% RH, and 40 +/- 2 degrees C/75 +/- 5%RH for 3 months showed no significant changes in the tablets quality at 30 +/- 2 degrees C/60 +/- 5% RH. However, at 40 +/- 2 degrees C/75 +/- 5%RH marked increase in in-vitro disintegration time, tensile strength and decrease in friability and water absorption ratio was found. The present studies indicate the abilities of Eudragit E 100 for taste masking and improving the dissolution profile of PM-HCl after complexation. In addition, by employing cost effective direct compression method, fast-dissolving tablets of 400 mg total weight with an acceptable quality could be prepared.

An analysis is presented of the deformation of a homogeneous, isotropic, elastic half space subjected to a constant radial strain in a circular area on the boundary. Explicit analytic expressions for the normal and radial displacements and the shear stress on the boundary are used to interpret experiments performed on inflated pig lungs. The boundary strain was induced by inflating or deflating the lung after bonding a flexible disk to the lung surface. The prediction that the surface bulges outward for positive boundary strain and inward for negative strain was observed in the experiments. Poisson's ratio at two transpulmonary pressures was measured, by use of the normal displacement equation evaluated at the surface. A direct estimate of Poisson's ratio was possible because the normal displacement of the surface depended uniquely on the compressibility of the material. Qualitative comparisons between theory and experiment support the use of continuum analyses in evaluating the behavior of the lung parenchyma when subjected to small local distortions.

The deformation occurring in the articular cartilage covering the human femoral head has been measured both when the femoral head is loaded in its natural acetabulum and when the cartilage is loaded with a small indentor. The results indicate that the material response is substantially different in these two situations. In the intact joint the cartilage deformation is substantially greater in older joints, but the response of cartilage to loading with an indentor does not change significantly with age. Theoretical elastic models of the cartilage behavior in these two situations were analyzed. For old cartilage which is idealized as an elastic material the increased deformation which is observed in the intact joint can be attributed to changes in Poisson's ratio, though in the real material increased fluid flux under load is the more probable cause.
