The effect of wheat heat damage was estimated using hard red winter (HRW) wheat varieties grown in Oklahoma, and analyses were performed on wheat kernels, flour, and isolated starch. Wheat kernels were analyzed by photoacoustic spectroscopy (PAS), flour was analyzed by DSC, capillary zone electrophoresis (CZE) and rheometry, while starch was analyzed by DSC. DSC data showed little difference in the temperatures of starch gelatinization. The ΔH values increased at 50 °C storage temperature. PAS data revealed differences in sensitivities to heat between the three cultivars. The pericap of cultivar Jagger was about five times more heat resistant than the pericarp of the other two cultivars. CZE maps showed that gluten proteins from cultivar 2137 was more affected by the 60 days and 50 °C treatment than the other two cultivars. Non-linear steady shearing showed that all heat-treated flours had lower viscosity, which suggests a deleterious effect on the potential performance of yeasted baked products. Viscoelastic properties of the Jagger untreated suspension sample showed strong gluten. The linear and non-linear viscoelastic behaviours of Jagger cultivar were less altered by heat treatment than were the other cultivars. [Copyright &y& Elsevier]

We present high spatial resolution, near-infrared images in J, H and K of the nucleus of NGC 3227, obtained with the Adaptive Optics Bonnette on the Canada–France–Hawaii Telescope. The ∼0.15 arcsec (17 pc) resolution allows structures to be probed in the core region. Dust obscuration becomes significantly less pronounced at longer wavelengths, revealing the true geometry of the core region. We are able to identify two main features in our maps: (i) a spiralling association of knots with a counterpart in a Hubble Space Telescope F606W image; and (ii) a smaller-scale annulus, orthogonal to the spiral of knots. The fall armyworm (FAW), Spodoptera frugiperda, is a major pest native to the Americas that has recently invaded the Old World. Point mutations in the target‐site proteins acetylcholinesterase‐1 (ace‐1), voltage‐gated sodium channel (VGSC) and ryanodine receptor (RyR) have been identified in S. frugiperda as major resistance mechanisms to organophosphate, pyrethroid and diamide insecticides respectively. Mutations in the adenosine triphosphate‐binding cassette transporter C2 gene (ABCC2) have also been identified to confer resistance to Cry1F protein. In this study, we applied a whole‐genome sequencing (WGS) approach to identify point mutations in the target‐site genes in 150 FAW individuals collected from China, Malawi, Uganda and Brazil. This approach revealed three amino acid substitutions (A201S, G227A and F290V) of S. frugiperda ace‐1, which are known to be associated with organophosphate resistance. The Brazilian population had all three ace‐1 point mutations and the 227A allele (mean frequency = 0.54) was the most common. Populations from China, Malawi and Uganda harbored two of the three ace‐1 point mutations (A201S and F290V) with the 290V allele (0.47–0.58) as the dominant allele. Point mutations in VGSC (T929I, L932F and L1014F) and RyR (I4790M and G4946E) were not detected in any of the 150 individuals. A novel 12‐bp insertion mutation in exon 15 of the ABCC2 gene was identified in some of the Brazilian individuals but absent in the invasive populations. Our results not only demonstrate robustness of the WGS‐based genomic approach for detection of resistance mutations, but also provide insights for improvement of resistance management tactics in S. frugiperda. The Bemisia cassava whitefly complex includes species that cause severe crop damage through vectoring cassava viruses in eastern Africa. Currently, this whitefly complex is divided into species and subgroups (SG) based on very limited molecular markers that do not allow clear definition of species and population structure. Based on 14,358 genome-wide SNPs from 62 Bemisia cassava whitefly individuals belonging to sub-Saharan African species (SSA1, SSA2 and SSA4), and using a well-curated mtCOI gene database, we show clear incongruities in previous taxonomic approaches underpinned by effects from pseudogenes. We show that the SSA4 species is nested within SSA2, and that populations of the SSA1 species comprise well-defined south-eastern (Madagascar, Tanzania) and north-western (Nigeria, Democratic Republic of Congo, Burundi) putative sub-species. Signatures of allopatric incipient speciation, and the presence of a 'hybrid zone' separating the two putative sub-species were also detected. These findings provide insights into the evolution and molecular ecology of a highly cryptic hemipteran insect complex in African, and allow the systematic use of genomic data to be incorporated in the development of management strategies for this cassava pest. Whilst the benefit of direct‐acting antiviral agents (DAAs) in achieving sustained virological response (SVR) is now well‐accepted, their impact on liver function, particularly in relation to achievement of SVR, has not been well documented. We studied 2394 patients with chronic HCV infection, 1276 receiving DAAs and 1118 interferon‐based therapy. Liver function was assessed by the albumin‐bilirubin (ALBI) score or grade. Overall survival according to SVR status and baseline ALBI grade was examined. We also studied time to first decompensation according to ALBI grade, as well as longitudinal changes in ALBI score over time according to SVR. Among the patients receiving DAAs, 89% achieved SVR (Japan = 99%, UK = 78%). Amongst the decompensated patients in the UK cohort, three distinct risk groups according to ALBI grade at baseline were observed. The UK patients receiving DAAs, who had predominantly decompensated disease, showed clear evidence of improvement of liver function detectable within 2 years of the start of treatment, especially in those achieving SVR. These early changes in liver function were very similar to those observed in the first 2‐3 years after interferon‐based therapy. DAAs improve liver function especially in those with decompensated disease who achieve SVR. Experience with interferon‐based therapy suggests that failure to achieve SVR is associated with long‐term decline in liver function and, in contrast, patients who do achieve SVR can expect long‐term disease improvement and subsequent stabilization of liver function. The chloroplast genomes of Euglena gracilis and Spinacia oleracea were investigated in their compositional heterogeneity, by using several different experimental approaches. Euglena chloroplast DNA has a dG + dC content of 28%. Preparations averaging 20 x 10[sup6] in molecular weight exhibit a gross heterogeneity in their elution profiles from hydroxyapatite and in their buoyant densities because the rRNA genes have a high rG + rC content. Finer analysis by melting, buoyant density of restriction fragments and micrococcal nuclease degradation have revealed an extended compositional heterogeneity. From micrococcal nuclease digestion data, approximately 30 % of the chloroplast genome is as low as 12 % in its dG + dC content, whereas 10% is higher than 60% dG + dC. Since the average dG + dC content of large restriction endonuclease fragments varied to a lesser extent, most of dA + dT-rich sequences must occur in short stretches interspersed with dG + dC-rich stretches. Spinach chloroplast DNA (dG + dC = 36.5%) did not exhibit any gross compositional heterogeneity in its hydroxyapatite elution or in its buoyant density profile. But the higher resolution methods of melting, buoyant densities of restriction fragments and micrococcal nuclease degradation revealed a high degree of heterogeneity which appears to be due to interspersion of short DNA stretches of different base composition. The patient underwent needle electrode examination and magnetic resonance imaging (MRI). It adds that the heterozygous mutation G294V in TARDBP was considered causative in the patient.

BACKGROUND: Moringa oleifera (MO) kernel oil is categorized as a high‐oleic oil that resembles olive oil. However, unlike olive trees, MO trees are largely present in most subtropical and tropical countries. In these countries, therefore, the benefits of oleic acid can be obtained at a cheaper price through the consumption of MO kernel oil. This study reports on the effect of different extraction methods on oxidative properties of MO kernel oil during storage for 140 days at 13, 25, and 37 °C. RESULTS: All aqueous enzymatic extraction (AEE)‐based methods generally resulted in oil with better oxidative properties and higher tocopherol retention than the use of solvent. Prior to AEE, boiling pre‐treatment deactivated the hydrolytic enzymes and preserved the oil's quality. In contrast, high‐pressure processing (HPP) pre‐treatment accelerated hydrolytic reaction and resulted in an increase in free fatty acids after 140 days at all temperatures. No significant changes were detected in the oils' iodine values and fatty acid composition. The tocopherol content decreased significantly at both 13 and 25 °C after 60 days in the oil from SE method, and after 120 days in oils from AEE‐based methods. CONCLUSION: These findings are significant in highlighting the extraction methods resulting in crude MO kernel oil with greatest oxidative stability in the storage conditions tested. Subsequently, the suitable storage condition of the oil prior to refining can be determined. Objective: The Tripartite Influence Model posits that social agents emphasize a thin ideal for women and a muscularity ideal for men. There is a gap in the literature of how sociocultural body ideal internalization affects overall disordered eating symptoms in men and specifically drive for muscularity in women. Method: The sample consisted of 1,929 participants (44.2% men) who completed online surveys. It was predicted that internalization would be a stronger predictor for overall disordered eating in women and for muscularity in men. Results: Women with high internalization had increased disordered eating symptoms in comparison to men, whereas for men, increased internalization was linked to more drive for muscularity, as compared with women. Conclusions: It is important to emphasize, however, that men still exhibited higher rates of disordered eating symptoms when they had increased internalization and women exhibited an increased drive for muscularity when they had higher internalization. Objective: This study aims to assess whether ultrasound-guided injection of platelet-rich plasma can safely and effectively treat symptoms associated with acetabular hip labral tears. Design: Institutional review board approval was gained for a prospective study of eight patients (N = 8), who have previously failed conservative management, to receive ultrasound-guided injection of platelet-rich plasma at the site of hip labrum tear.We assessed pain reduction and functional ability at baseline and then 2, 6, and 8 wks after injection, using the visual analog scale and Harris Hip Score, respectively. Results: Statistically significant differences in Harris Hip Score were seen 2 wks (86.5 ± 10.8, P < 0.01), 6 wks (88.0 ± 10.7) P < 0.01), and 8 wks (92.1 ± 11.6, P < 0.01) after injection as compared with baseline (76.0 ± 13.4). Corresponding improvementswere seen in visual analog scale 2 wks (1.0, P < 0.01 at rest, 2.5, P < 0.01 with activity), 6 (0.9, P < 0.01 at rest, 2.3, P < 0.01 with activity), and 8 wks (0.5, P < 0.01 at rest, 1.3, P < 0.01 with activity) compared with baseline (3.8 at rest, 5.4 with activity). Conclusions: Ultrasound-guided injection of platelet-rich plasma holds promise as an emerging, minimally invasive technique toward symptom relief, reducing pain, and improving function in patients with hip labral tears.
