Haas [sup.1] , [...], The emergence and global spread of SARS-CoV-2 has resulted in the urgent need for an in-depth understanding of molecular functions of viral proteins and their interactions with the host proteome. Several individual omics studies have extended our knowledge of COVID-19 pathophysiology.sup.1-10. Integration of such datasets to obtain a holistic view of virus-host interactions and to define the pathogenic properties of SARS-CoV-2 is limited by the heterogeneity of the experimental systems. Here we report a concurrent multi-omics study of SARS-CoV-2 and SARS-CoV. Using state-of-the-art proteomics, we profiled the interactomes of both viruses, as well as their influence on the transcriptome, proteome, ubiquitinome and phosphoproteome of a lung-derived human cell line. Projecting these data onto the global network of cellular interactions revealed crosstalk between the perturbations taking place upon infection with SARS-CoV-2 and SARS-CoV at different levels and enabled identification of distinct and common molecular mechanisms of these closely related coronaviruses. The TGF-[beta] pathway, known for its involvement in tissue fibrosis, was specifically dysregulated by SARS-CoV-2 ORF8 and autophagy was specifically dysregulated by SARS-CoV-2 ORF3. The extensive dataset (available at https://covinet.innatelab.org) highlights many hotspots that could be targeted by existing drugs and may be used to guide rational design of virus- and host-directed therapies, which we exemplify by identifying inhibitors of kinases and matrix metalloproteases with potent antiviral effects against SARS-CoV-2. Multi-omics profiling of effects of SARS-CoV-2 and SARS-CoV on A549, a lung-derived human cell line, produces a dataset enabling identification of common and virus-specific mechanisms of infection.

Polymer‐based pyroelectric thin films are crucial functional materials at the core of flexible and lightweight electronic devices, such as wearable monitoring sensors, energy harvesters, and infrared detectors. Nevertheless, the pyroelectric properties of the polymer films, such as poly(vinylidene fluoride‐trifluoroethylene) (P(VDF‐TrFE)), vanish when the surrounding temperature exceeds the ferroelectric‐to‐paraelectric transition temperature, and thus limits their pyroelectric performance to a low‐temperature range. Herein, to mitigate this issue by employing a new class of P(VDF‐TrFE) copolymer which has a low TrFE molar content is proposed. Fine‐tuning of the structure through thermal annealing in a vacuum environment significantly favors robust and highly polarized polymer films with a large area. Electric poling combined with an optimal annealing temperature (110–120 °C) gives highly ordered ferroelectric crystalline domains in the polymer films. Consequently, this remarkably broadens the temperature range (roughly up to 140 °C) for which the polymer film still presents high pyroelectric properties (pyroelectric coefficient 50 µC (m2K)−1). Symmetry is a fundamental property of many natural systems, which is observable through signals. In most out-of-equilibrium complex dynamic systems, the observed signals are asymmetric. However, for certain operating modes, some systems have demonstrated a resurgence of symmetry in their signals. Research has naturally focused on examining time invariance to quantify this symmetry. Measures based on the statistical and harmonic properties of signals have been proposed, but most of them focused on harmonic distortion without explicitly measuring symmetry. This paper introduces a new mathematical framework based on group theory for analyzing signal symmetry beyond time invariance. It presents new indicators to evaluate different types of symmetry in non-stochastic symmetric signals. Both periodic and non-periodic symmetric signals are analyzed to formalize the problem. The study raises critical questions about the completeness of symmetry in signals and proposes a new classification for periodic and non-periodic signals that goes beyond the traditional classification based on Fourier coefficients. Furthermore, new measures such as "symmetrometry" and "distorsymmetry" are introduced to quantify symmetry. These measures outperform traditional indicators like Total Harmonic Distortion (THD) and provide a more accurate measurement of symmetry in complex signals from applications where duty cycle plays a major role. Introduction: Mammals are the main hosts for Brucella sp., agents of worldwide zoonosis. Marine cetaceans and pinnipeds can be infected by Brucella ceti and B. pinnipedialis, respectively. Besides classical bacteriological typing, molecular approaches such as MLVA, MLSA, and whole-genome sequencing (WGS) can differentiate these species but are cumbersome to perform. Methods: We compared the DNA and genome sequences of 12 strains isolated from nine marine mammals, with highly zoonotic B. melitensis, B. abortus, and B. suis, and the publicly available genomes of B. ceti and B. pinnipedialis. In silico pipelines were used to detect the antimicrobial resistance (AMR), plasmid, and virulence genes (VGs) by screening six open-source and one home-made library. Results and discussion: Our results show that easier-to-use HRM-PCR, Bruceladder, and Suis-ladder can separate marine Brucella sp., and the results are fully concordant with other molecular methods, such as WGS. However, the restriction fragment length polymorphism (RFLP) method cannot discriminate between B. pinnipedialis and B. ceti B1-94-like isolates. MLVA-16 results divided the investigated strains into three clades according to their preferred host, which was confirmed in WGS. In silico analysis did not find any AMR and plasmid genes, suggesting antimicrobial susceptibility of marine Brucella, while the presence of the VGs btpA gene was variable dependent on the clade. Conclusion: The HRM-PCR and Suis-ladder are quick, easy, and cost-effective methods to identify marine Brucella sp. Moreover, in silico genome analyses can give useful insights into the genetic virulence and pathogenicity potential of marine Brucella strains. [ABSTRACT FROM AUTHOR]

ABSTRACT Background Methods Results Conclusion While some labor interventions are essential in preventing maternal and neonatal morbidity, there is little evidence to support systematic early augmentation of labor (EAL). Our objective was to assess the association between EAL and cesarean delivery rate, postpartum hemorrhage and adverse neonatal outcomes.Population‐based study using data from the 2016 French Perinatal Survey. Women with a singleton cephalic fetus, delivering at term after a spontaneous labor were included. “EAL” was defined by artificial rupture of the membranes (AROM) and/or oxytocin within 1 h of admission and/or duration between interventions of less than 1 h. Women without EAL were women without labor augmentation or without EAL. The primary endpoint, cesarean delivery and the secondary endpoints were compared between women with and without EAL using univariate analysis. A multivariable logistic regression was adjusted on the suspected confounders and a propensity score approach was then performed.Among the 7196 women included, 1524 (21.2%) had EAL. Cesarean delivery rates were significantly higher in the EAL group compared with the no EAL group, 8.40% versus 6.15% (p < 0.01). EAL was associated with cesarean delivery in the multivariable analysis aOR 1.45 95% CI [1.15–1.82] and in the cohort matched on the propensity score, OR 1.56 [1.17–2.07]. EAL was not associated with severe postpartum hemorrhage, low 5‐min Apgar score, low neonatal cord pH or transfer to NICU.EAL is frequent, involving one in five spontaneous laboring women in France. Optical second-harmonic generation (SHG) is achieved using adsorbed gold nanoparticles (AuNPs) with an average diameter of 16 nm at the aqueous solution–air interface in reflection. A detailed analysis of the depth profile of the SHG intensity detected shows that two contributions appear in the overall signal, one arising from the aqueous solution–air interface that is sensitive to the AuNP surface excess and one arising from the bulk aqueous phase. The latter is an incoherent signal also known as hyper-Rayleigh scattering (HRS). The results agree with those of an analysis involving Gaussian beam propagation optics and a Langmuir-like isotherm. Discrepancies are revealed for the largest AuNP concentrations used and indicate a new route for the design of soft metasurfaces. [ABSTRACT FROM AUTHOR]

Abstract Background A novel Brucella strain closely related to Brucella (B.) melitensis biovar (bv) 3 was found in Croatian cattle during testing within a brucellosis eradication programme. Case presentation Standardised serological, brucellin skin test, bacteriological and molecular diagnostic screening for Brucella infection led to positive detection in one dairy cattle herd. Three isolates from that herd were identified to species level using the Bruce ladder method. Initially, two strains were typed as B. melitensis and one as B. abortus, but multiplex PCR based on IS711 and the Suis ladder showed that all of them to belong to B. melitensis, and the combination of whole-genome and multi-locus sequencing as well as Multi-Locus Variable numbers of tandem repeats Analysis (MLVA) highlighted a strong proximity within the phylogenetic branch of B. melitensis strains previously isolated from Croatia, Albania, Kosovo and Bosnia and Herzegovina. Two isolates were determined to be B. melitensis bv. 3, while the third showed a unique phylogenetic profile, growth profile on dyes and bacteriophage typing results. This isolate contained the 609-bp omp31 sequence, but not the 723-bp omp31 sequence present in the two isolates of B. melitensis bv. 3. Conclusions Identification of a novel Brucella variant in this geographic region is predictable given the historic endemicity of brucellosis. The emergence of a new variant may reflect a combination of high prevalence among domestic ruminants and humans as well as weak eradication strategies. The zoonotic potential, reservoirs and transmission pathways of this and other Brucella variants should be explored.

Uniaxial tensile tests were carried out on nickel sheets containing only few grains across the thickness thus presenting a so-called multicrystalline state. Pristine sheets used as reference and substrates covered by a nickel layer deposited by physical vapor deposition (magnetron sputtering) were studied. Results attest that the surface treatment affects the plastic deformation mechanisms by playing the role of a blocking barrier to dislocation movement which leads to a significant improvement of mechanical performances. The surface restructuring of the nickel sheets is efficient to stop dislocation escape through free surfaces and allows a mechanical improvement of the well-known strong mechanical softening of the multicrystals.

Summary: Targeting immune checkpoints, such as Programmed cell Death 1 (PD1), has improved survival in cancer patients by restoring antitumor immune responses. Most patients, however, relapse or are refractory to immune checkpoint blocking therapies. Neuropilin-1 (NRP1) is a transmembrane glycoprotein required for nervous system and angiogenesis embryonic development, also expressed in immune cells. We hypothesized that NRP1 could be an immune checkpoint co-receptor modulating CD8+ T cells activity in the context of the antitumor immune response. Here, we show that NRP1 is recruited in the cytolytic synapse of PD1+CD8+ T cells, cooperates and enhances PD-1 activity. In mice, CD8+ T cells specific deletion of Nrp1 improves anti-PD1 antibody antitumor immune responses. Likewise, in human metastatic melanoma, the expression of NRP1 in tumor infiltrating CD8+ T cells predicts poor outcome of patients treated with anti-PD1. NRP1 is a promising target to overcome resistance to anti-PD1 therapies.
