The multi-functional properties of lactic acid bacteria (LAB) on host health have been a popular research topic. The aim of present study was to assess the multi-functional potential of five LAB strains isolated from giant panda. In this study, we analyzed five giant panda LAB strains (Weissella confuse WJ202003 (W3), WJ202009 (W9), WJ202021 (W21), BSP201703 (X3); Lactiplantibacillus plantarum BSGP201683 (G83)) and found that they exhibited rapid growth as well as strong acid production capacity. The five LAB strains possessed high cell surface hydrophobicity to the four tested solvents (xylene, hexadecane, chloroform, ethyl acetate; except strain W9), auto-aggregation ability, co-aggregation ability with three pathogens (Escherichia coli, Enterotoxigenic Escherichia coli, Salmonella), adhesion ability to Caco-2 cell line, and strongly biofilm formation ability, suggesting an adhesion property. As investigated for their antioxidative potential, all the strains showed good tolerance to H2O2, high scavenging ability against 1, 1-diphenyl-2-picrylhydrazyl (DPPH), and hydroxyl (OH−), and reduction ability. Furthermore, the five LAB strains could produce multiple probiotic substances, including exopolysaccharide (EPS), gamma-aminobutyric acid (GABA), bile salt hydrolase (BSH), cellulase (only strain G83), and protease (except strain X3), which was the first to report the production of EPS, GABA, BSH, cellulase, and protease in giant panda-derived LAB strain. Leaf traits reflect the ecological strategy in heterogeneous contexts and are widely used to explore the adaption of plant species to environmental change. However, the knowledge of short-term effect of canopy management on understorey plant leaf traits is still limited. Here, we studied the short-term effect of crown–thinning on the leaf morphological traits of bamboo (Chimonobambusa opienensis), an important understorey plant and staple food for the giant panda (Ailuropoda melanoleuca) of Niba Mountain. Our treatments were two crown–thinnings (spruce plantation, CS, and deciduous broad-leaved forest, CB) and two controls (broad-leaved forest canopy, FC, and the bamboo grove of clearcutting, BC). The results showed that: the CS enhanced the annual leaf length, width, area, and thickness, CB decreased almost all annual leaf traits, and perennial leaf traits in CS and CB were the opposite. The log-transformed allometric relationships of length vs. width, biomass vs. area were significantly positive while those of specific leaf area vs. thickness were significantly negative, which varied largely in treatments and age. The leaf traits and allometric relationships suggested that the CS created a more suitable habitat for bamboo growth. Social impact assessment is a practical requirement for the construction of the national park system, but there is still a lack of comprehensive conceptual frameworks, theoretical explanations, and systematic evaluations. Considering the distinctive characteristics of China's national park system construction, this study improves the existing framework and indicator system for social impact assessment. It conducts a comparative analysis of three typical communities--the inside community, gateway community, and fringe community--surrounding the Giant Panda National Park (Tangjiahe area). Through questionnaire surveys, semi-structured interviews, factor analysis, and One-way ANOVA analysis, we find that the comprehensive perception value of social impacts by the Giant Panda National Park was relatively negative. Respondents' perception values ranked from high to low were social relations, local culture, equity and rights, economy and livelihoods, and knowledge and education. Moreover, except the dimension of economy and livelihoods, there were significant differences in the perception of impacts by the three types of communities. Specifically, the gateway community has the most positive perception of social relations, local culture, and equity and rights; the inside community has the most positive perception of knowledge and education; while the fringe community perceived the most negative impacts in all four dimensions. Furthermore, the primary causes of social impact perception are conservation policies, economic development, and location and transportation. This study suggests that non-economic social impacts should be given more attention, such as equity and rights, and knowledge and education. Also, to ensure that the fringe communities can benefit from the construction and development of national parks, more attention and support from the governments and management agencies are necessary. The host range of feline panleukopenia virus (FPV) is expanding and is a serious threat to both captive and free-range endangered wildlife. The FPV named FPV-am2020 was isolated from fecal samples from four diarrheal captive giant pandas in 2020, and pathogenicity and phylogenetic analysis were conducted in this study. Three-month-old cats challenged with FPV-am2020 experienced 100% mortality. The complete FPV-am2020 sequence was determined and comprised 5277 base pairs (bp), 36.76% GC content, and two open reading frames. According to the phylogenetic analysis of whole genome sequences and VP2 gene sequences, FPV-am2020 was closely related with MG764511.1 (isolated from captive lions in China, 2015), KX685354.1 (isolated from captive tigers in China, 2016), and KX900570.1 (isolated from captive jaguar in China, 1986). Furthermore, the study identified a G299E mutation in VP2 which was a key residue involved in phenotype changes in FPV. Thus, increased surveillance of FPV mutant isolates must be enacted to protect giant pandas against potential viral threats. [ABSTRACT FROM AUTHOR]

The wild giant panda habitat is inaccessible and far away from the main areas of human activity, so environmental pollutants entering the habitat are mainly the result of external migration and spread through the atmospheric advection and diffusion processes and particulate matter deposition. To research the variation, transmission route, chemical characteristics, and source of PM2.5 in the habitat of wild giant pandas, we set up a PM2.5 sampling point near the Shaanxi Foping National Nature Reserve (SFNNR), which is the area with the highest population density of wild giant pandas in the Qinling Mountains. The 12-month average concentration of PM2.5 was 11.3 ± 7.9 μg/m3 from July 2021 to June 2022, and the 12-month average concentration did not exceed the limit value set in the standard. In the results of our analysis of element concentrations, As and Pb were much lower than the limit standard. Si, S, P, and Cl accounted for 99.60% of nonmetallic elements, while the proportion of the six metallic elements, Na, Mg, Al, K, Ca, and Fe, was 96.27%. According to the analysis results of enrichment factor (EF) and pollutant emission sources, there were four sources for the total elements in PM2.5, which were mainly distributed in the areas around the reserve. These included dust, coal combustion, biomass burning, and traffic-related emissions, which contributed 55.10%, 24.78%, 11.91%, and 8.22% of the total element mass in PM2.5, respectively. Additionally, Pb, Cu, Zn, As, Sc, Co, Ga, Mg, and, especially, Se were severely affected by human activities (coal burning, biomass burning, and traffic-related emissions). In the villages and towns around the wild giant panda habitat, the majority of energy for cooking and heating comes from coal and biomass burning, and older vehicles with high emissions are used more frequently. Therefore, to better protect the health of and reduce the impact of environmental pollution on wild giant pandas, we put forward relevant recommendations, including upgrading the energy structure of towns and villages near the habitat to increase the proportion of clean energy, such as photovoltaic power generation, natural gas, etc.; decreasing the combustion of coal and biomass; encouraging the upgrading of agricultural diesel machines and older vehicles used in these areas; and setting limits on vehicle emissions in areas surrounding the habitat. The gut microbiome of the giant panda (Ailuropoda melanoleuca) plays a vital role in nutrient acquisition from its specialized bamboo diet. Giant panda cubs harbour significantly different gut microbiota during their growth and development when feeding on milk before switching to bamboo. The fetal gut is sterile, and following birth, mother‐to‐infant microbial transmission has been implicated as a seeding source for the infant gut microbiota. Details of this transmission in giant pandas remain unclear. In this study, faecal samples were collected from seven panda mother–cub pairs when the cubs were 4–16 months old. Additional samples from the cubs' diet, soil and drinking water, and multiple body sites of the mothers were collected. Bacterial 16S rRNA gene sequencing and shotgun metagenomic sequencing were performed to determine the source and potential transmission routes of the cub gut microbiome. Source tracking analysis showed that maternal vagina, milk and faeces were the primary contributory sources of microbes, shaping the cub gut microbiome. Bacterial species from maternal faeces persisted the longest in the cub gut. Bacterial species in the diet contributed to the microbial community. Metagenomics analysis indicated that the predicted metabolic pathways of the gut microbiome also varied at different growth stages. Gut colonization with bacteria from various body sites of the mothers provides a foundational microbial community that is beneficial in fulfilling the evolving dietary needs of the cubs. With the completion of the draft assembly of the giant panda genome sequence, RNA sequencing technology has been widely used in genetic research on giant pandas. We used RNA‐seq to examine black and white hair follicle samples from adult pandas. By comparison with the giant panda genome, 75 963 SNP loci were labeled, 2426 differentially expressed genes (DEGs) were identified, and 2029 new genes were discovered, among which 631 were functionally annotated. A cluster analysis of the DEGs showed that they were mainly related to the Wnt signaling pathway, ECM–receptor interaction, the p53 signaling pathway, and ribosome processing. The enrichment results showed that there were significant differences in the regulatory networks of hair follicles with different colors during the transitional stage of hair follicle resting growth, which may play a regulatory role in melanin synthesis during growth. Planting bamboo and trees is of great importance in giant panda (Ailuropoda melanoleuca) habitat restoration. In practice, however, simply planting bamboo and trees cannot effectively and rapidly restore lost, degraded, and fragmented habitats. Based on present restoration work in the Qinling Mountains, China, we argue that: (1) Planting should be done on open and suitable forest lands, but the area that could be afforested is relatively small. (2) Forests that have regrown naturally on cut‐over lands are mostly middle‐aged and early secondary stands, which need to be managed, not replanted. (3) Plantations of non‐native larch (Larix spp.) urgently need to be replaced and replanted with native trees and bamboos, but new policies are needed that allow these plantations to be thinned or cut. (4) Bamboo forests in the mid‐high mountain zones need to be assessed to determine if they are disclimaxes of degraded forests. Pilot studies are needed to determine the best need and practices to rehabilitate and restore them. Bamboo is the main food source of the giant panda. To increase bamboo intake in captive giant pandas, we studied factors affecting the bamboo intake. Fourteen healthy captive giant pandas in Dujiangyan Base of China Conservation and Research Center for The Giant Panda ("Dujiangyan Base" for short) were selected as research objects. A bamboo feeding experiment was conducted to study the effects of seasons, bamboo age, slope orientations where bamboo grows and felling-feeding time on bamboo intake of the giant panda. We found that the type of bamboo that captive giant pandas feed on was abundant in spring and summer, but relatively homogeneous in winter. With the increase of bamboo age, the intake of bamboo leaves decreased, while bamboo culms increased. The feed intake of 1-year-old bamboo leaves and 5-year-old bamboo culms reached the highest respectively. The slope orientation also affected the panda's bamboo intake, and the bamboo growing on sunny slopes or semi-sunny slopes was more favored by captive giant pandas. Moreover, the bamboo intake reached the highest when felling-feeding time was less than 24 h. In short, we confirmed that seasons, bamboo age, slope orientations and felling-feeding time were factors affecting bamboo intake for captive giant pandas. Simple Summary: Climate change can have a negative impact on the food sources of wild animals, including giant pandas, who primarily rely on bamboo. To better understand the foraging strategy of giant pandas and aid in their conservation, metabolomic methods were used to analyze fecal metabolites and correlate them with gut microbiota. The results indicate that the sugar content in giant panda feces is higher when they consume bamboo culm (the hollow, jointed stem) with high fiber content and that Streptococcus bacteria are positively correlated with glucose and acetic acid content, both of which are related to fiber. These results suggest that the foraging strategy of giant pandas is based on the nutritional composition of different bamboo parts, and therefore, enriching their habitat with bamboo species is essential to achieve their natural foraging strategy to obtain adequate nutrition. Climate change-induced food shortages pose major threats to wildlife conservation, and the exclusive reliance of giant pandas on bamboo makes them particularly vulnerable. The aim of this study was to provide insight into the reasons for the foraging strategies of giant pandas to selectively forage for different bamboo parts (bamboo shoot, culm, and leaf) during different seasons. This study used a metabolomic approach to analyze the fecal metabolites of giant pandas and conducted a correlation analysis with their gut microbiota. The results indicate that the fecal metabolites of giant pandas differ significantly depending on the bamboo parts they forage on, with higher sugar content observed when they consume bamboo culm with high fiber content. By functional annotation, culm group metabolites were enriched in the galactose metabolic pathway, while shoot group metabolites were enriched in the phenylalanine, tyrosine and tryptophan biosynthesis pathways. Moreover, Streptococcus showed a significant positive correlation with glucose and acetic acid content. Therefore, the foraging strategy of giant pandas is based on the ability to utilize the nutrient content of different bamboo parts. Captive feeding and habitat construction should enrich bamboo species to allow them to express their natural foraging strategies and improve their welfare and reproductive status. Despite retaining a digestive system with carnivorous characteristics, the giant panda has now specialized to eat bamboo. Therefore, research on its feeding habits and foraging strategies has always been an important field in the study of giant panda population ecology. Located in the core distribution area of the giant panda population in the Liangshan mountains, the Meigu Dafengding Nature Reserve acts as the link between the major panda reserves in the Liangshan mountains and the key corridor zone connecting the giant panda populations in the east and west. This study was performed in 2017 and aimed to determine the feeding habits of giant pandas in this region, the heterogeneity of food resources in different seasons, and the selection strategy for foraging patches. To achieve this, we used the line transect method to investigate the giant panda population, collected fresh feces, analyzed the nutritional composition of bamboo samples, and set up a sampling grid of foraging patches in different seasonal distribution areas. It was found that wild giant pandas in the region spend most of the year at lower elevations(1939–3296 m) foraging for Yushania maculata and Yushania ailuropodina, and only move to higher elevations(2844–3770 m) in summer for Bashania fangiana. Their preferred foraging patches(n = 65) had certain topographical features, such as gentle slopes(< 25°) and closer proximity to water sources(< 500 m), and the nutritional quality of bamboo resources in foraging patches was significantly better than in those they avoided or rarely used. The food habits and foraging patch selection strategies of giant pandas in the Liangshan and Qinling mountains are relatively similar, and the results of this study can help provide a scientific basis for the conservation and management of wild giant panda habitats in different mountain regions. The giant panda (Ailuropoda melanoleuca) and the Chinese red panda (Ailurus styani) are distributed in the same region in the mountain forest ecosystem on the eastern edge of the Qinghai Tibet Plateau and share the same food sources. In order to understand how sympatric giant pandas and Chinese red pandas maintain interspecific relationships to achieve stable coexistence, we used species distribution models and diurnal activity rhythms to analyze the spatial and temporal niche characteristics of giant pandas and Chinese red pandas in the Daxiangling Mountain system based on 187 camera traps data. The results show that: (1) In the Daxiangling Mountains, the total area of suitable habitats for giant pandas and Chinese red pandas is 717.61 km2 and 730.00 km2, respectively, accounting for 17.78% and 18.25%, respectively, of the study area. (2) The top five environmental factors contributing to the model of giant panda and Chinese red panda are precipitation seasonality, temperature seasonality, distance to the road, and elevation and vegetation type. (3) The total overlapping area of suitable habitats for giant pandas and Chinese red pandas is 342.23 km2, of which the overlapping area of highly suitable habitats is 98.91 km2. The overlapping index of suitable habitats is 0.472, and the overlapping index of highly suitable habitats is 0.348, which indicates that the two achieve spatial niches are separated to achieve stable coexistence. (4) The overlapping index of the daily activity rhythm of giant panda and Chinese red panda is 0.87, which is significantly different (p <.05). The existence of Chinese red panda will significantly affect the daily activity rhythm of giant panda (p <.001). This research can provide scientific reference for the researches about population and habitat protection of giant pandas and Chinese red pandas, so as to understand the driving mechanism of resource allocation and population dynamics of sympatric species. Roads, acting as barriers, hamper wildlife movements and disrupt habitat connectivity. Bridges and culverts are common structures on roads, and some of them can function to allow wildlife passage. This study investigated the effects of traffic, the surrounding landscape, human disturbance, and bridge and culvert structures on the utilization of bridges and culverts as dedicated passages by wildlife, using motion-activated infrared camera traps along a 64 km road in Giant Panda National Park, Sichuan, China. The results show that both species richness and counts of wildlife recorded at the bridge and culvert were significantly lower than those observed at sites distant from roads. No large-sized wildlife was recorded at the bridges and culverts. Human activities and traffic volume significantly and negatively affect medium-sized wildlife utilization of bridges and culverts. We conclude that bridges and culverts serve as wildlife crossings, but their efficacy is weak. This emphasizes the necessity of retrofitting bridges and culverts via mitigation facilities such as noise and light barriers, and vegetation restoration on both sides of the roads in Giant Panda National Park. Abstract Pancreatic ductal adenocarcinoma (PDAC) is a highly malignant cancer of the digestive system common among humans. However, it is rarely reported in wild animals. In 2018, a giant panda died in the Beijing Zoo. During subsequent histological observation of the pancreas, it was discovered that the glandular epithelial cells had lost the pancreatic acinar structure, tubular areas with obvious structure in the pancreas, and the ductal epithelium was substituted by high columnar mucus cells. Masson staining showed that there were several fibrous tissue proliferative reactions around the ductal adenocarcinoma and immunohistochemical staining revealed that CK7 and CK19 were positively expressed in the pancreatic tissue. Therefore, the pathological diagnosis indicated that the panda had PDAC. In this paper, the panda's living conditions and pathological diagnosis results are examined, with the aim of providing a reference point for the future diagnosis of wild animal tumors.

Abstract Background Indirect interactions between individual solitary mammals, such as the giant panda, are often overlooked because of their nature, yet are important for maintaining the necessary sociality in solitary species. Methods and results Here, we determined the genetic identity of all giant panda individuals in a local population and matched these identities with their associations to determine social network of this solitary animal. Total thirty-five giant panda individuals were found in our field survey, and we constructed genetic and social networks for thirty-three individuals who successfully obtained genetic, age and sex information. The results showed that sex had great impact on both social network and genetic network, and age may have the potential to influence the social network of the giant pandas. Adult males, mostly in the central of the social network, which appeared significantly larger network connections than adult females. Due to the female-biased dispersal pattern of wild giant pandas, male-male pairs showed higher relatedness than female-female ones and multi-generational patrilinear assemblages are expected in the study area. Conclusions The relatedness of individuals has an influence on the formation of community social structure of giant pandas, and indirect interactions among solitary giant pandas potentially function to reduce competition for resources and inbreeding.

Toxoplasma gondii is a worldwide-distributed zoonotic protozoan parasite which causes toxoplasmosis and has a significant effect on public health. In the giant panda (Ailuropoda melanoleuca), toxoplasmosis can cause asymptomatic infections, reproductive disorder and even death, which poses a serious threat to the conservation of this rare protected species. Therefore, serological investigation of T. gondii is essential to understanding its risk to giant pandas, however, there are no specific testing kits for giant pandas. Previous research has used MAT as the reference method for screening T. gondii, to investigate this further, this study focused on the agreement comparing of MAT with ELISA and IHA tests for detecting T. gondii antibodies in 100 blood samples from 55 captive giant pandas in Chengdu, China. The results showed 87.0%, 87.0%, 84.0%, samples were sero-positive for T. gondii using ELISA (kits a, b, c), respectively, while MAT and IHA tests were 84.0% and 9.0% sero-positive, respectively.
