Mycoplasma mycoides subsp. mycoides (Mmm) is the etiological agent of contagious bovine pleuropneumonia (CBPP), one of the major diseases affecting cattle in sub-Saharan Africa. Some evidences suggest that the immune system of the host (cattle) plays an important role in the pathogenic mechanism of CBPP, but the factors involved in the process remain largely unknown. The present study aimed to investigate the cell response of bovine polymorphonuclear neutrophils (PMNs) after Mmm in vitro exposure using one step RT-qPCR and Western blotting. Data obtained indicate that gene and protein expression levels of some pro-inflammatory factors already change upon 30 min of PMNs exposure to Mmm. Of note, mRNA expression level in Mmm exposed PMNs increased in a time-dependent manner and for all time points investigated; targets expression was also detected by Western blotting in Mmm exposed PMNs only. These data demonstrate that when bovine PMN cells are triggered by Mmm, they undergo molecular changes, upregulating mRNA and protein expression of specific pro-inflammatory factors. These results provide additional information on host-pathogen interaction during CBPP infection.

The current longitudinal study examined factors (sex, physical function, response to novelty, ability to adapt to a shift in light/dark cycle, brain connectivity), which might predict the emergence of impaired memory during aging. Male and female Fisher 344 rats were tested at 6, 12, and 18 months of age. Impaired spatial memory developed in middle-age (12 months), particularly in males, and the propensity for impairment increased with advanced age. A reduced response to novelty was observed over the course of aging, which is inconsistent with cross-sectional studies. This divergence likely resulted from differences in the history of environmental enrichment/impoverishment for cross-sectional and longitudinal studies. Animals that exhibited lower level exploration of the inner region on the open field test exhibited better memory at 12 months. Furthermore, males that exhibited a longer latency to enter a novel environment at 6 months, exhibited better memory at 12 months. For females, memory at 12 months was correlated with the ability to behaviorally adapt to a shift in light/dark cycle. Functional magnetic resonance imaging of the brain, conducted at 12 months, indicated that the decline in memory was associated with altered functional connectivity within different memory systems, most notably between the hippocampus and multiple regions such as the retrosplenial cortex, thalamus, striatum, and amygdala. Overall, some factors, specifically response to novelty at an early age and the capacity to adapt to shifts in light cycle, predicted spatial memory in middle-age, and spatial memory is associated with corresponding changes in brain connectivity. We discuss similarities and differences related to previous longitudinal and cross-sectional studies, as well as the role of sex differences in providing a theoretical framework to guide future longitudinal research on the trajectory of cognitive decline. In addition to demonstrating the power of longitudinal studies, these data highlight the importance of middle-age for identifying potential predictive indicators of sexual dimorphism in the trajectory in brain and cognitive aging.

Bacteria of the genus Brucella cause brucellosis, an infectious disease common to humans as well as to terrestrial and aquatic mammals. Since 1994 several cases of Brucella spp. infection have been reported in marine mammals worldwide. While sero-epidemiological data suggest that Brucella spp. infection is widespread globally, detecting Brucella spp.-associated antigens by immunohistochemistry (IHC) in tissues from infected animals is often troublesome. The present study was aimed at investigating, by means of IHC based upon the utilization of an anti-Brucella LPS monoclonal antibody (MAb), the central nervous system (CNS) immunoreactivity shown by B. ceti-infected, neurobrucellosis-affected striped dolphins. The aforementioned MAb, previously characterized by means of ELISA and Western Blotting techniques, was able to immunohistochemically detect smooth brucellae both within the CNS from B. ceti-infected striped dolphins and within a range of tissues from Brucella spp.-infected domestic ruminants. In conclusion, the results of the present study are of relevance both from the B. ceti infection's diagnostic and pathogenetic standpoints.

Introduction: Excessive intake of sodium is linked to cardiovascular disease. Activation of the renin-angiotensin-aldosterone system is the most common mechanism underlying an increase in sodium seeking or sodium appetite. Numerous brain regions are known to be excited or inhibited during the activation of sodium appetite. Objective: To determine whether new brain regions linked to chronically elevated sodium intake could be uncovered using novel brain imaging techniques. Material and Methods: Male SD rats received either a subcutaneous pellet of the aldosterone precursor deoxycorticosterone acetate (DOCA, 150 mg/60 day release) or sham surgery (n=6-8/group). Following surgery, all animals had access to both normal water and a 1% NaCl, 0.2% KCl solution. Between 11-13 days post-surgery the average daily sodium intake in the DOCA group was 196±21 versus 21±4 ml in the control group. On day 14 post-surgery all animals were given an intraperitoneal injection of manganese chloride (25 mg/kg). Following 24 hrs of recovery, while lightly anesthetized, the brain of each animal was imaged on a 4.7 Tesla MRI using a multi-slice spin echo sequence: TE=15ms, TR=350ms, 22 slices, 256x256 resolution. MEMRI enabled mapping of signal enhancement or depression in the brain associated with neuronal activity marked by paramagnetic Mn2+ accumulation. Results: Preliminary results suggest an overall decrease in activity within many brain regions of interest in the DOCA/salt animals versus controls. El objetivo del presente trabajo es analizar el texto de la Ley de Acceso a la Información nº 27.275 que se sancionó en el año 2016. En ese sentido, nuestra intención es describir el contenido del texto legal, haciendo hincapié en los aspectos más fundamentales de la normativa, pero también se revisará el proceso legislativo que tuvo la mencionada norma. Es decir, se analizará el texto finalmente sancionado y las diferentes posturas que se desarrollaron en la discusión parlamentaria que originaron diferentes dictámenes y opiniones por parte de distintos legisladores. Asimismo, también nos referiremos a opiniones que varios colectivos relacionados con la comunicación y el derecho han mani­festado en torno a la ley de acceso a la información. Para realizar la descripción, consideramos necesario primero analizar conceptualmente el derecho de acceso a la información, entendiendo que dicho concepto no es operativo sólo para quienes se desempeñan en los medios de comunicación social, sino que su ejercicio pleno a través de la garantía estatal redunda en un mayor y mejor ejercicio de la ciudadanía.

Abstract Introduction Pyrovalerone (4‐methyl‐β‐keto‐prolintane) is a synthetic cathinone (beta‐keto‐amphetamine) derivative. Cathinones are a concern as drugs of abuse, as related street drugs such as methylenedioxypyrovalerone have garnered significant attention. The primary mechanism of action of cathinones is to inhibit reuptake transporters (dopamine and norepinephrine) in reward centers of the central nervous system. Methods We measured bioenergetic, behavioral, and molecular responses to pyrovalerone (nM‐µM) in zebrafish to evaluate its potential for neurotoxicity and neurological impairment. Results Pyrovalerone did not induce any mortality in zebrafish larvae over a 3‐ and 24‐hr period; however, seizures were prevalent at the highest dose tested (100 µM). Oxidative phosphorylation was not affected in the embryos, and there was no change in superoxide dismutase 1 expression. Following a 3‐hr treatment to pyrovalerone (1–100 µM), larval zebrafish (6d) showed a dose‐dependent decrease (70%–90%) in total distance moved in a visual motor response (VMR) test. We interrogated potential mechanisms related to the hypoactivity, focusing on the expression of dopamine‐related transcripts as cathinones can modulate the dopamine system. Pyrovalerone decreased the expression levels of dopamine receptor D1 (~60%) in larval zebrafish but did not affect the expression of tyrosine hydroxylase, dopamine active transporter, or any other dopamine receptor subunit examined, suggesting that pyrovalerone may regulate the expression of dopamine receptors in a specific manner. Discussion Further studies using zebrafish are expected to reveal new insight into molecular mechanisms and behavioral responses to cathinone derivates, and zebrafish may be a useful model for understanding the relationship between the dopamine system and bath salts.

The Forma Urbis Romae (F.U.) was a 3rd-century-AD monumental map of ancient Rome consisting of 151 rectangular marble slabs. Several efforts have been made to reconstruct it from its current incomplete and fragmentary condition. In this paper, we explore the potential of an archaeometric approach to serve this purpose. Almost a hundred F.U. fragments have been characterized, particularly focusing on cathodoluminescence (CL) microscopy and stable isotopes (δ18O and δ13C). Different statistical methods have been used to quantify the similarity between samples. The central assumption is that samples from a given slab share similar CL colors and isotopic ratios. The assumption has been verified for samples from single fragments and then it has been used to check ten debated reconstruction hypotheses. The measured isotopic ratios confirm the Proconnesian nature of the F.U. marble, except for a fragment. Beyond provenance, the results cast doubts on four out of the ten checked reconstruction hypotheses and support the other six. The reconstruction of the F.U. remains a fascinating challenge, and both isotopic and CL analyses have demonstrated their potential to tackle it. Further research could extend the presented methodology to a higher number of samples. The innovative use of CL to reconstruct a fragmented artwork could be applied to other projects.

The dopamine transporter (DAT), a regulator of dopamine homeostasis in the brain, and sigma-1 receptor (σ1R), an endoplasmic reticulum membrane protein, are both implicated in drug addiction. In this work, the authors investigate how σ1R modulates DAT response to methamphetamine.

Objectives: Puerto Rico (PR), is the fifth highest jurisdiction of the United States of America (US) with respect to HIV prevalence and the leading in cervical cancer incidence. This cross-sectional study describes the prevalence and correlates of cervical HPV infection among a clinic-based sample of 302 women living with HIV/AIDS in PR. Methods: Data collection included questionnaires, blood and cervical samples. Multivariable logistic regression models were used to estimate the magnitude of association (adjusted Prevalence odds ratio [aPOR]) between HPV cervical infection and other covariates. Results: Mean age of participants was 40.3 years (± 10.3 SD). The prevalence of HPV infection was 50.3%; 41.1% for low-risk types and 29.5% for high-risk types. Having ⥠10 lifetime sexual partners (aPOR = 2.10, 95% CI:1.02â4.29), an abnormal Pap (aPOR = 3.58, 95% CI:1.93â6.62), active genital warts (aPOR = 3.45, 95% CI:1.60â7.42), and CD4 counts ⤠200 (aPOR = 4.24, 95% CI: 1.67â10.78) were positively associated with any cervical HPV infection. Similar results were observed for HR HPV infection. Conclusions: A high burden of HPV co-infection exists among women living with HIV/AIDS in this population. Given the high incidence of HIV in PR and the higher risk of cervical cancer among women living with HIV/AIDS, HPV vaccination should be promoted in this population. Hexagonal neo-formed crystallites have been observed in thin section of different medieval and post-medieval lead-glazed ceramics. Although they are clearly visible in thin section using plane polarized light, their plate shape makes them barely seen on the polished cross sections. Basal sections have never been found on the polished sections and only few transversal very thin sections could be seen. In this case, the morphology resembles acicular and it is not possible to analyze them properly by SEM–EDX because the crystals are very thin and the glaze surrounding is analyzed as well. Micro-Raman microscopy was carried out directly on the polished thin sections. This technique allows specific areas as small as 1 μm in diameter to be analyzed and it is able to characterize inclusions that are not found on the glaze surface. However, the wavenumber features observed cannot be assigned to a specific compound. The thickness of the crystallites (a few hundred nanometers) seems to be responsible for the low sensitivity of the Raman instrumentation. 15 × 15 μm2 micro-X-ray diffraction patterns using synchrotron radiation (SR-μXRD) in transmission geometry were obtained from the crystals using the same thin section preparation. SR-μXRD was able to localize the crystallites and avoid the overlapping signals corresponding to other mineral phases. In this way, the hexagonal crystallites present in the glaze have been unambiguously identified as hematite crystallites. Finally, some replications were made under laboratory-controlled conditions to determine the firing conditions in the formation of those crystallites. Celiac disease is an autoimmune enteropathy that affects the small intestine and is caused by the gliadin fraction of wheat gluten and other alcohol-soluble proteins (prolamines) of barley and rye in genetically predisposed subjects. Nowadays, the only recognized cure for this pathology is the use of gluten-free (GF) products. Although the market for GF products has grown in the last few years, their improvement is still a challenge for the food industry. With regard to GF bakery products, some of them can result in an unappetizing taste with a high fat content. These inconveniences can be minimized or solved by using sourdough fermentation. Sourdough is a stable culture of lactic acid bacteria (LAB) and yeast in a mixture of flour and water. In particular, GF sourdough represents a rich source of naturally occurring LAB and wild yeasts, which play important roles in food fermentation. This is mainly due to their potential in improving functional, technological, and probiotics properties, contributing to safe food with a better taste. Moreover, exopolysaccharide (EPS)-producing strains can also contribute to improving the sensory and rheological quality of the different GF products. This study aims to isolate and characterize the microbial pool from a GF sourdough made with rice flour and to evaluate the ability of selected LAB strains to produce exopolysaccharides (EPS). The yeast and LAB were isolated in selective agar media. A total of 220 isolates (110 yeasts and 110 bacteria) were obtained. They were taxonomically identified and characterized using both traditional and molecular approaches. Sequencing analysis of the D1/D2 domain of the 26S rRNA and the 16S rRNA genes, for yeasts and bacteria, respectively, revealed the occurrence of Saccharomyces cerevisiae as predominant yeasts and Lactiplantibacillus plantarum and Pediococcus pentosaceus as dominant LAB. Regarding the EPS production, 25 strains were able to produce dextran-like EPS with a maximum yield of 2 g/L. This study allowed the isolation and selection of promising EPS-producing starter cultures for the different types of flour sourdough, in particular, GF sourdough.

Introduction: This study investigates the interactions between histaminergic system and glucocorticoid-induced leucin zipper (GILZ) in the inflammatory process and glucocorticoid modulation in lung fibrosis. Methods: Wild-type (WT) and GILZ Knock-Out (KO) mice were treated with bleomycin (0.05 IU) or saline, delivered by intra-tracheal injection. After surgery, mice received a continuous infusion of JNJ7777120 (JNJ, 2 mg/kg b.wt.) or vehicle for 21 days. Lung function was studied by measuring airway resistance to air insufflation through the analysis of pressure at airway opening (PAO). Lung samples were collected to evaluate the expression of histamine H4R, Anx-A1, and p65-NF-kB, the activity of myeloperoxidase (MPO), and the production of pro-inflammatory cytokines. Results: Airway fibrosis and remodeling were assessed by measuring TGF-β production and α-SMA deposition. JNJ reduces PAO in WT but not in GILZ KO mice (from 22 ± 1 mm to 15 ± 0.5 and from 24 ± 1.5 to 19 ± 0.5 respectively), MPO activity (from 204 ± 3.13 pmol/mg to 73.88 ± 2.63 in WT and from 221 ± 4.46 pmol/mg to 107 ± 5.54 in GILZ KO), the inflammatory response, TGF-β production, and α-SMA deposition in comparison to WT and GILZ KO vehicle groups. Conclusion: In conclusion, the role of H4R and GILZ in relation to glucocorticoids could pave the way for innovative therapies to counteract pulmonary fibrosis. [ABSTRACT FROM AUTHOR]
