Abstract Although tendon-driven continuum manipulators have been extensively researched, how to realize tip contact force sensing in a more general and efficient way without increasing the diameter is still a challenge. Rather than use a complex modeling approach, this paper proposes a general tip contact force-sensing method based on a recurrent neural network that takes the tendons’ position and tension as the input of a recurrent neural network and the tip contact force of the continuum manipulator as the output and fits this static model by means of machine learning so that it may be used as a real-time contact force estimator. We also designed and built a corresponding three-degree-of-freedom contact force data acquisition platform based on the structure of a continuum manipulator designed in our previous studies. After obtaining training data, we built and compared the performances of a multi-layer perceptron-based contact force estimator as a baseline and three typical recurrent neural network-based contact force estimators through TensorFlow framework to verify the feasibility of this method. We also proposed a manually decoupled sub-estimators algorithm and evaluated the advantages and disadvantages of those two methods.

Abstract Background It has been suggested that patent foramen ovale (PFO) contributes to the majority of cryptogenic stroke cases in young people, however, the direct link is still undetermined. Here we analyzed the correlation between PFO and brain ischemia lesions in a cohort of cases that were long-term residents in the plateau to provide solid evidence to support the causal relation between PFO and brain ischemia lesion or cryptogenic stroke. Methods Long-term residents with young age from Qinghai Plateau were recruited and separated by PFO positivity. Brain MRI was used to image 100 PFO positive cases and 100 healthy controls. The diameter of PFO was measured by echocardiography. The location, number and anterior/posterior circulation of ischemia lesions were also evaluated. The correlation between PFO (including positivity and diameter) and brain ischemia lesion (including positivity and other characteristics) was analyzed by chi-square test. Further, the chi-square test for the trend test was used to analyze the linear correlation between these groups. Results We found a strong correlation between the positivity of PFO and brain ischemia lesion, with 71% of PFO cases showing the presence of brain ischemia lesions, and only 19% for healthy controls (p

Cervical angina has been defined as chest pain that resembles true cardiac angina but originates from the disorders of the cervical spine. Thus, physicians and spine surgeons alike should raise awareness of this unusual condition for diagnosis and treatment. Particularly when neurologic signs and symptoms are present, there should be a strong suspicion for cervical angina in any patient with inadequately explained noncardiac chest pain. Cervical angina can be diagnosed according to negative cardiac workups, positive neurologic examination, and cervical radiographic findings (herniated disk, spinal cord compression, or foraminal encroachment). However, the mechanisms of pain production in cervical angina remain unclear. Previous studies attributed the pain to cervical nerve root compression, cervical sympathetic afferent fibers, referred pain, or lesions of the posterior horn of the spinal cord. Conservative treatments, which include neck collar fixation, head traction, and nonsteroidal anti-inflammatory drugs, have been determined to be successful in most patients with cervical angina. But when conservative treatment fails, anterior cervical surgery with complete decompression of the spinal cord and/or nerve root has been identified to effectively relieve cervical angina symptoms.

Abstract Background Our recent studies have identified that the red nucleus (RN) dual-directionally modulates the development and maintenance of mononeuropathic pain through secreting proinflammatory and anti-inflammatory cytokines. Here, we further explored the action of red nucleus IL-33 in the early development of mononeuropathic pain. Methods In this study, male rats with spared nerve injury (SNI) were used as mononeuropathic pain model. Immunohistochemistry, Western blotting, and behavioral testing were used to assess the expressions, cellular distributions, and actions of red nucleus IL-33 and its related downstream signaling molecules. Results IL-33 and its receptor ST2 were constitutively expressed in the RN in naive rats. After SNI, both IL-33 and ST2 were upregulated significantly at 3 days and peaked at 1 week post-injury, especially in RN neurons, oligodendrocytes, and microglia. Blockade of red nucleus IL-33 with anti-IL-33 neutralizing antibody attenuated SNI-induced mononeuropathic pain, while intrarubral administration of exogenous IL-33 evoked mechanical hypersensitivity in naive rats. Red nucleus IL-33 generated an algesic effect in the early development of SNI-induced mononeuropathic pain through activating NF-κB, ERK, p38 MAPK, and JAK2/STAT3, suppression of NF-κB, ERK, p38 MAPK, and JAK2/STAT3 with corresponding inhibitors markedly attenuated SNI-induced mononeuropathic pain or IL-33-evoked mechanical hypersensitivity in naive rats. Red nucleus IL-33 contributed to SNI-induced mononeuropathic pain by stimulating TNF-α expression, which could be abolished by administration of inhibitors against ERK, p38 MAPK, and JAK2/STAT3, but not NF-κB. Conclusions These results suggest that red nucleus IL-33 facilitates the early development of mononeuropathic pain through activating NF-κB, ERK, p38 MAPK, and JAK2/STAT3. IL-33 mediates algesic effect partly by inducing TNF-α through activating ERK, p38 MAPK and JAK2/STAT3.

Abstract Background Wiedemann-Rautenstrauch syndrome (WRS) is a rare autosomal recessive neonatal progeroid disorder characterized by prenatal and postnatal growth retardation, short stature, a progeroid appearance, hypotonia, and mental impairment. Case presentation A 6-year-old patient, who initially presented with multiple postnatal abnormalities, facial dysplasia, micrognathia, skull appearance, hallux valgus, and congenital dislocation of the hip, was recruited in this study. The patient was initially diagnosed with progeria. The mother of the patient had abnormal fetal development during her second pregnancy check-up, and the clinical phenotype of the fetus was similar to that of the patient. Whole-exome sequencing (WES) of the patient was performed, and POLR3B compound heterozygous variants—c.2191G > C:p.E731Q and c.3046G > A:p.V1016M—were identified in the patient. Using Sanger sequencing, we found that the phenotypes and genotypes were segregated within the pedigree. These two variants are novel and not found in the gnomAD and 1000 Genomes databases. The two mutation sites are highly conserved between humans and zebrafish. Conclusions Our study not only identified a novel WRS-associated gene, POLR3B, but also broadened the mutational and phenotypic spectra of POLR3B. Furthermore, WES may be useful for identifying rare disease-related genetic variants.

Aiming at the application research of polar code in optical communication system, this paper proposes the combination of polar coding modulation and optical fiber Module Division Multiplexing (MDM) system. We construct an MDM system based on bit interleaved polar coding modulation, and study the performance of Maximum Likelihood (ML) detection algorithm. The simulation results show that the addition of polar code greatly improves the system performance. Compared with the MDM system without coding, the MDM system with bit interleaved polar coding modulation improves the system performance by about 8 dB under the same Mode Dependent Loss (MDL) . The paper also shows the performance of ML detection algorithm and Minimum Mean Square Error (MMSE) detection algorithm in MDM system. The impact of different mode loss on system performance is also studied. Compared with MMSE detection algorithm, ML detection algorithm improves the bit error performance by about 3 dB.

Background: Significant survival benefit of adjuvant imatinib therapy has been observed in gastrointestinal stromal tumor (GIST). However, the impact of neoadjuvant imatinib on prognosis of GIST remains unclear. This meta-analysis aimed to compare the prognostic impact between upfront surgery and neoadjuvant imatinib plus surgery on GIST.Methods: A comprehensive literature search was performed to identify eligible studies up to 30 Sep 2021, through PubMed, Embase, Web of Science, and Cochrane Library. Studies compared the impact of upfront surgery and neoadjuvant imatinib plus surgery on disease-free (DFS) or overall survival (OS) in patients with GIST were selected.Results: Seven eligible studies with 17,171 patients were included. The reduction rates of tumor size in rectal and mixed site GIST were 33% and 29.8%, respectively. Neoadjuvant imatinib was not significantly associated with DFS compared with no-neoadjuvant therapy in rectal GIST (HR: 0.71, 95% CI: 0.35–1.41). The OS of rectal GIST was significantly improved by neoadjuvant imatinib compared with no-neoadjuvant therapy (HR: 0.36, 95% CI: 0.17–0.75).Conclusion: Neoadjuvant imatinib therapy contributed to tumor shrinkage and R0 resection of rectal GIST. Neoadjuvant imatinib plus surgery significantly improved overall survival of rectal GIST in comparison with upfront surgery.

Cancer stem cells drive tumor initiation, progression, and recurrence, which compromise the effectiveness of anti-tumor drugs. Here, we report that demethylzeylasteral (DML), a triterpene anti-tumor compound, suppressed tumorigenesis of liver cancer stem cells (LCSCs) by interfering with lactylation of a metabolic stress-related histone. Using RNA sequencing (RNA-seq) and gas chromatography-mass spectrometric (GC-MS) analysis, we showed that the glycolysis metabolic pathway contributed to the anti-tumor effects of DML, and then focused on lactate downstream regulation as the molecular target. Mechanistically, DML opposed the progress of hepatocellular carcinoma (HCC), which was efficiently facilitated by the increase in H3 histone lactylation. Two histone modification sites: H3K9la and H3K56la, which were found to promote tumorigenesis, were inhibited by DML. In addition, we used a nude mouse tumor xenograft model to confirm that the anti-liver cancer effects of DML are mediated by regulating H3 lactylation in vivo. Our findings demonstrate that DML suppresses the tumorigenicity induced by LCSCs by inhibiting H3 histone lactylation, thus implicating DML as a potential candidate for the supplementary treatment of hepatocellular carcinoma.

Gifted children and normal controls can be distinguished by analyzing the structural connectivity (SC) extracted from MRI data. Previous studies have improved classification accuracy by extracting several features of the brain regions. However, the limited size of the database may lead to degradation when training deep neural networks as classification models. To this end, we propose to use a data augmentation method by adding artificial samples generated using graph empirical mode decomposition (GEMD). We decompose the training samples by GEMD to obtain the intrinsic mode functions (IMFs). Then, the IMFs are randomly recombined to generate the new artificial samples. After that, we use the original training samples and the new artificial samples to enlarge the training set. To evaluate the proposed method, we use a deep neural network architecture called BrainNetCNN to classify the SCs of MRI data with and without data augmentation. The results show that the data augmentation with GEMD can improve the average classification performance from 55.7 to 78%, while we get a state-of-the-art classification accuracy of 93.3% by using GEMD in some cases. Our results demonstrate that the proposed GEMD augmentation method can effectively increase the limited number of samples in the gifted children dataset, improving the classification accuracy. We also found that the classification accuracy is improved when specific features extracted from brain regions are used, achieving 93.1% for some feature selection methods.

The economic value of a mineral resource is highly dependent on the accuracy of grade estimations. Accurate predictions of mineral grades can help businesses decide whether to invest in a mining project and optimize mining operations to maximize the resource. Conventional methods of predicting gold resources are both costly and time-consuming. However, advances in machine learning and processing power are making it possible for mineral estimation to become more efficient and effective. This work introduces a novel approach for predicting the distribution of mineral grades within a deposit. The approach integrates machine learning and optimization techniques. Specifically, the authors propose an approach that integrates the random forest (RF) and k-nearest neighbor (kNN) algorithms with the marine predators optimization algorithm (MPA). The RFKNN_MPA approach uses log normalization to reduce the impact of extreme values and improve the accuracy of the machine learning models. Data segmentation and the MPA algorithm are used to create statistically equivalent subsets of the dataset for use in training and testing. Drill hole locations and rock types are used to create each model. The suggested technique’s performance indices are superior to the others, with a higher R-squared coefficient of 59.7%, a higher R-value of 77%, and lower MSE and RMSE values of 0.17 and 0.44, respectively. The RFKNN_MPA algorithm outperforms geostatistical and conventional machine-learning techniques for estimating mineral orebody grades. The introduced approach offers a novel solution to a problem with practical applications in the mining sector.

Electrocatalytic nitrate reduction into ammonia is promising for its restricted activity and selectivity in wastewater treatment, however, it remains challenging. In this work, Co3O4 nanosheet electrodes with rich oxygen vacancies (OVs) (Co3O4−x/NF) are prepared and then applied as efficient catalysts for selective electrocatalytic reduction of nitrate to ammonia. The resulting Co3O4−x/NF electrodes exhibit high NO3−-N removal efficiency and NH4+-N selectivity, at 93.7% and 85.4%, respectively. X-Ray photoelectron spectroscopy (XPS) and electron paramagnetic resonance spectra (EPR) results clearly reveal the formation of OVs in Co3O4−x/NF. The electrochemical characterization results confirm that OVs can effectively improve electron transfer as well as the electrochemically active area. The Co2+/Co3+ ratio of Co3O4−x/NF increases after the electrocatalytic reduction of nitrate, highlighting the crucial role played by Co2+ in mediating ammonia production via the Co2+/Co3+ cycle. These findings offer valuable guidelines for the development of more efficient and sustainable approaches for nitrate-contaminated wastewater treatment and ammonia synthesis.

MicroRNAs (miRNAs) play important roles in insect growth and development, but they were poorly studied in insects. In this study, a total of 883 miRNAs were detected from the early embryo (EE), late larva (LL), early pupa (EP), late pupa (LP), and early adult (EA) of Tribolium castaneum by microarray assay. Further analysis identified 179 differentially expressed unique miRNAs (DEmiRNAs) during these developmental stages. Of the DEmiRNAs, 102 DEmiRNAs exhibited stage-specific expression patterns during development, including 53 specifically highly expressed miRNAs and 20 lowly expressed miRNAs in EE, 19 highly expressed miRNAs in LL, 5 weakly expressed miRNAs in EP, and 5 abundantly expressed miRNAs in EA. These miRNAs were predicted to target 747, 265, 472, 234, and 121 genes, respectively. GO enrichment analysis indicates that the targets were enriched by protein phosphorylation, calcium ion binding, sequence-specific DNA binding transcription factor activity, and cytoplasm. An RNA interference-mediated knockdown of the DEmiRNAs tca-miR-6-3p, tca-miR-9a-3p, tca-miR-9d-3p, tca-miR-11-3p, and tca-miR-13a-3p led to defects in metamorphosis and wing development of T. castaneum. This study has completed the identification and characterization of development-related miRNAs in T. castaneum, and will enable us to investigate their roles in the growth and development of insect.
