Adrenal steroids modulate the function of nerve cells. Some, but not all actions of these steroids take place after binding to intracellular receptor systems and translocation of the steroid-receptor complex into the cell nucleus. Studies on the rat brain revealed heterogeneity of receptors. One population of receptor sites is present in abundance in extrahypothalamic limbic brain regions, e.g. neurons of the hippocampus, septum and amygdala. This neuronal receptor system displays a stringent binding specificity towards corticosterone, which is the naturally occurring glucocorticoid of the rat. Focussing the studies on the corticosterone receptor system in hippocampal neurons has provided further insight in the understanding of some of the actions of the steroid. Certain hippocampus-associated behaviors and indices of neurotransmission (serotonin) were disturbed after removal of the adrenals, but selectively restored after replacement with a low dose of corticosterone. The specificity, localization and dose-dependency of the corticosterone action on behavior and neurotransmission corresponds to the properties of its receptor system. The responsiveness to corticosterone is altered after changes in number of receptor sites. Chronic stress or high doses of exogenous corticosterone cause a long-term reduction. Other factors involved in regulation of receptor number are the neurotransmitter serotonin and neuropeptides related to ACTH and vasopressin. These substances restore changes in number of hippocampal corticosterone receptor sites due to aging, endocrine or neural deficiencies. Our results show that the number of corticosterone receptors is a sensitive index for brain functioning. Thus, the receptor system mediates some of the modulatory actions of corticosterone on nerve cell function and it may adjust its capacity under the influence of neural and endocrine factors.

Age-related changes were studied in the concentration of type-I and type-II corticosteroid receptors in the hippocampus of young adult (3 months) and aged (28.5 to 30.5 months) male rats. Using 3H-labelled ligands, in vitro binding of type-I and type-II corticosteroid receptors in the soluble cell fraction (cytosol) revealed an age-related decrease in concentration of both receptor types of 52% and 28%, respectively. Infusion of young and aged male rats for 2 weeks with the ACTH4-9 [adrenocorticotropin4-9] peptide analogue ORG 2766 (0.5 micrograms/0.5 microliter/hr) resulted in only a minor increase (+8%) in the number of type-I receptors in young rats. In the aged animals, however, the type-I receptor concentration was 68% higher than in the vehicle-treated aged animals. In contrast, no effect of the peptide treatment was noted on the concentration of type-II receptors in either young or aged rats. Furthermore, no effect was found for either age or treatment with peptide on the affinity of type-I and type-II receptors for their respective ligands. Binding of 3H-labelled ligands to brain sections of young and aged rats was performed using in vitro autoradiography. Quantitative image analysis of the film showed that in senescence there is a marked reduction in both type-I (62-75%) and type-II (29-56%) receptor concentrations in the hippocampal subregions (CA1, CA2, CA3 and dentate gyrus) as well as in the lateral septum. Treatment of aged rats with ORG 2766 selectively reversed the age-associated reduction in type-I receptors, while the peptide did not affect the type-II receptors.(ABSTRACT TRUNCATED AT 250 WORDS)

Following intrapituitary injection of 3 H-ACTH 4–9 analog, the radioactivity of various brain regions was determined in intact rats and in rats with the pituitary stalk cut one or eight days previously. The regional distribution of radioactivity in the brain was also investigated after intravenous and intrasellar administration. Intrasellar and intrapituitary administration resulted in significantly higher radioactivity levels in the brain than did intravenous injection of an equimolar dose of labeled peptide. Intrapituitary injection resulted in an uptake with clear regional differences and which was highest in the hypothalamus. Twenty four hours after stalk section the uptake of radioactivity in the hypothalamus, but not in other brain regions was markedly depressed. Hypothalamic uptake, however, was restored at eight days after stalk section. The results suggest a significant flow of radioactivity from the pituitary to the brain, particularly to the hypothalamus. Transport to the hypothalamus is presumably partly vascular via the stalk. Transport to other brain areas may occur via the cerebrospinal fluid, but a neural route cannot be excluded.

The present study was designed to localize the effect of hypophysectomy on brain RNA content at a topographical level. For this purpose, a brain dissection method, based on morphological dif-ferences between various brain areas, was developed. In these areas, total cell RNA was measured and expressed as the RNA/DNA ratio. The highest ratio was found in the cortex cerebri (ca. 2.15) and hippocampus (2.21), whereas cerebellum (0.42) and bulbus olfactorius (0.95) showed the lowest values. Hypophysectomy reduced the RNA content mainly of brain-stem structures (ca.–24%), i.e., the thalamus, hypothalamus, mesencephalon, and medulla oblongata. Nevertheless, a small but significant reduction was found in the rostral cortex. In view of previous studies, it is suggested that this disturbed RNA metabolism in brain-stem structures could have a bearing on the impaired avoidance performance of hypophysectomized rats.

The effects of corticosteroids were studied on the concanavalin A (Con A)-induced mitogenesis of peripheral blood T-lymphocytes obtained from intact and adrenalectomized (ADX) Wistar rats. One week of adrenalectomy reduced the proliferative response of T-cells by 65% compared with sham-operated controls. Substitution of ADX rats with subcutaneously implanted 12.5-mg corticosterone (Cort) pellets, which resulted in low circulating Cort levels (17 +/- 3 nM), restored the reduced proliferative capacity to that of sham-ADX animals. In contrast, T-lymphocyte proliferation was nearly absent in ADX rats substituted with high circulating Cort levels (173 +/- 15 nM; 100-mg Cort pellet). In vitro, Cort suppressed the mitogenic response of T-lymphocytes from ADX and sham-ADX animals. The glucocorticoid antagonist RU-486 (500 nM) completely blocked this suppressive effect. However, a 10 times lower concentration of RU-486 reversed the effects of a low (10 nM) Cort concentration from suppression to stimulation. It is concluded that high Cort concentrations in vivo and in vitro suppressed T-lymphocyte mitogenesis but that low concentrations in vivo were stimulatory, whereas this stimulation in vitro occurred only in the presence of antiglucocorticoids. These opposing effects of Cort emphasize a bimodal regulatory role of this hormone in immune regulation that may be mediated by different corticosteroid receptors.

The cell nucleus is highly organized. Many nuclear functions are localized in discrete domains, suggesting that compartmentalization is an important aspect of the regulation and coordination of nuclear functions. We investigated the subnuclear distribution of the glucocorticoid receptor, a hormone-dependent transcription factor. By immunofluorescent labeling and confocal microscopy we found that after stimulation with the agonist dexamethasone the glucocorticoid receptor is concentrated in 1,000-2,000 clusters in the nucleoplasm. This distribution was observed in several cell types and with three different antibodies against the glucocorticoid receptor. A similar subnuclear distribution of glucocorticoid receptors was found after treatment of cells with the antagonist RU486, suggesting that the association of the glucocorticoid receptor in clusters does not require transformation of the receptor to a state that is able to activate transcription. By dual labeling we found that most dexamethasone-induced receptor clusters do not colocalize with sites of pre-mRNA synthesis. We also show that RNA polymerase II is localized in a large number of clusters in the nucleus. Glucocorticoid receptor clusters did not significantly colocalize with these RNA polymerase II clusters or with domains containing the splicing factor SC-35. Taken together, these results suggest that most clustered glucocorticoid receptor molecules are not directly involved in activation of transcription.

Ultradian release of glucocorticoids is thought to be essential for homeostasis and health. Furthermore, deviation from this pulsatile release pattern is considered to compromise resilience to stress-related disease, even after hormone levels have normalised. In the present study, we investigate how constant exposure to different concentrations of corticosterone affects diurnal and ultradian pulsatility. The rate of recovery in pulsatile hypothalamic-pituitary-adrenal (HPA) activity after withdrawal of exogenous corticosterone is also examined. Finally, the behavioural and neuroendocrine responsiveness to an audiogenic stressor is studied. Adrenally intact male rats were subcutaneously implanted with vehicle, 40% or 100% corticosterone pellets for 7 days. The continuous release of corticosterone from these implants abolished diurnal and ultradian corticosterone variation, as measured with high-frequency automated blood sampling. Pellet removal on post-surgery day 8 allowed rapid recovery of endogenous rhythms in animals previously exposed to daily average concentrations (40%) but not after exposure to high concentrations (100%) of corticosterone. Behavioural and neuroendocrine responsiveness to stress was distinctly different between the treatment groups. Audiogenic stimulation 1 day after pellet removal resulted in a similar corticosterone response in animals previously exposed to 40% corticosterone or vehicle. The 40% pellet group, however, showed less and shorter behavioural activity (i.e. locomotion, risk assessment) to noise stress compared to 100% corticosterone and vehicle-treated animals. In conclusion, unlike the animals impanted with 100% corticosterone, we find that basal HPA axis activity in the 40% group, which had mean daily levels of circulating corticosterone in the physiological range, rapidly reverts to the characteristic pulsatile pattern of corticosterone secretion. Upon reinstatement of the ultradian rhythm, and despite the fact that these animals did not differ from controls in their response to noise stress, they did show substantial changes in their behavioural response to stress.

In Vitro cytosol binding, receptor autoradiography with radiolabelled corticosteroid analogs, and immunocytochemistry with monoclonal antibodies have revealed the presence of two receptor systems for corticosteroids in rat and hamster brains. The type I receptor is found mainly in the hippocampal region, and in the hamster it binds cortisol (F) and corticosterone (B) with similar affinity while in the rat (a species which unlike the hamster secretes solely B) the type I receptor shows high affinity to B and not to F. The type II receptor is more widely distributed in the brain and it binds to F (hamster) or B (rat) with affinity 4–6-fold lower than to the type I. in vivo , the hamster type I and II retain F much more than B while those in the rat show the opposite. In conclusion, the present study clearly indicates species-specificity in type I and type II receptor systems in these animals. Furthermore, the type I receptor displays in vivo stringent preference for retention of the animal's predominantly circulating corticosteroid (F in hamster, in B in rat).

Central administration of neurotensin (NT) has been shown to activate the hypothalamic-pituitary-adrenal axis, an effect which seems dependent upon the release of corticotropin-releasing factor. In this study, we describe the distribution of NT binding sites in the hypothalamus using film and emulsion receptor autoradiography. Among the 125I-NT-labelled hypothalamic nuclei, relatively high densities of neurotensin binding sites were detected over the paraventricular nucleus. Silver grains on emulsion-coated slides overlaid indiscriminately cell bodies and surrounding processes of magnocellular and parvocellular parts of the nucleus. Two newly developed NT receptor antagonists, SR 48692 and its analog SR 48450, competed for 125I-NT binding to hypothalamic tissue sections and membrane preparations with Ki values in the nanomolar range. Moreover, intracerebrally injected SR 48450 was able to block the NT-induced hypothalamic-pituitary-adrenal axis activation in freely moving rats, whereas its administration alone did not significantly affect basal plasma levels of adrenocorticotropin and corticosterone. These data provide anatomical substrate for a potential neurotensin action at the hypothalamus in the hypothalamic-pituitary-adrenal axis activation and highlight the use of new non-peptide NT receptor antagonists to characterize the effects of NT on neuroendrocrine functions.

We report here that with a direct method for measurement of cardiovascular parameters in conscious rats, intracerebroventricular administration of the mineralocorticoid receptor (MR) antagonist RU-28318 (100 ng) reduces the blood pressure, heart rate, and the corticosterone response to a brief restraint stress, provided the rats were previously subjected to a daily 30-min exposure to 32 degrees C for 2 wk. The daily exposure to warming and restraint stress are applied identically to the training procedure required for indirect blood pressure measurement using the tail-cuff method. The basal arterial pressure is not affected by the MR antagonist. The effect of the MR antagonist on the stress-induced pressor response develops with a delay of several hours in the normotensive rats. The corticosterone response to daily warming and stress is also attenuated by the intracerebroventricular infusion of MR antagonist but with shorter onset and shorter duration. The findings suggest that conditioning to daily warming and stress imposes mineralocorticoid dependency of the pressor response, which involves MR functioning in brain.

The ontogeny of the Type 2 glucocorticoid receptor (GR) in the rat brain was examined using a monoclonal antibody raised against the rat liver GR. Marked changes both in the intensity and in the localization of GR immunoreactivity (GR-ir) were found to occur as a function of age and brain area examined. First, GR-ir was high perinatally and decreased to a low intensity of immunostaining around postnatal day 12 (pnd 12). Thereafter, GR-ir increased to a moderate intensity, which resembled adult levels by pnd 20 in most brain areas. Second, in some regions, such as the hippocampal CA3-4 pyramidal cell fields and the suprachiasmatic nucleus of the hypothalamus, GR-ir was only clearly present during the first postnatal week. Third, in the hippocampus, GR-ir localization showed a distinctive developmental trend towards greater compactness within the CA1-2 pyramidal cell fields and a greater restriction of immunoreactive staining to these cell fields with exclusion of the adjoining areas. Fourth, adrenalectomy reduced overall GR-immunopositive staining, which could be reversed by administration of the selective glucocorticoid agonist, RU 28362. Our results suggest that during ontogeny the glucocorticoid receptor system displays considerable plasticity. Such plasticity may provide a basis for understanding the role of glucocorticoids during brain development.

Corticosterone (CORT) binds to two receptor systems in rat brain: the type I CORT-preferring receptor (CR) and the type II glucocorticoid receptor (GR). Discrimination between the two receptor types can be achieved with the 'pure' synthetic glucocorticoid RU 28362. In this study, we show that the binding capacity of GR in the rat hippocampus exhibits a strikingly different response from CR to adrenalectomy (ADX), chronic steroid replacement, hypophysectomy (HYPOX) and during circadian variation. Under those experimental conditions neither receptor site showed changes in binding affinity. After ADX, CR number remained relatively constant for a period of 13 days, while GR capacity increased by 133%, a level which was reached 5 days post-surgery. CR capacity showed circadian variation, since CR number was 65% higher in the evening than in the morning. GR capacities at those two time points were not significantly different. Replacement with subcutaneous CORT implants (100-mg pellets) for 7 days following ADX rats did not affect CR number, but caused a 38% decrease in GR number compared to control animals (cholesterol-treated, 7-day-ADX rats). On the other hand, dexamethasone (DEX) implants (5-, 15-, 25-mg pellets) elicited a dose-dependent increase in CR capacity (up to 99%) and a dose-dependent decrease in GR capacity (40-44%). Finally, 2 weeks after HYPOX, CR and GR numbers were increased by 60 and 38%, respectively. We conclude that the type II GR capacity responds in an autoregulatory manner to changes in circulating plasma glucocorticoid levels, while type I CR does not.

The rat brain contains two receptor systems for corticosterone: the type-I corticosterone-preferring receptor and the classical type-II glucocorticoid receptor. The two receptor populations can be distinguished in binding studies with the 'pure' synthetic glucocorticoid 11β,17β-dihydroxy-6-methyl-17α (1-propynyl)-androsta-1,4,6-trione-3-one (RU 28362). In-vitro autoradiography and quantitative image analysis showed that the type-I receptor was localized almost exclusively in the hippocampus, whereas the type-II receptor extended throughout the brain, with the highest levels in the nucleus paraventricularis, nucleus supraopticus and in the thalamic, amygdaloid, hippocampal and septal regions. Unoccupied type-I and type-II receptor sites, as measured in vitro by cytosol binding of 3H-labelled steroids, displayed a large difference in the rate of appearance after adrenalectomy. The availability of type-I receptors exhibited a marked increase, reaching maximal levels within 4–7 h, and then remained constant until 2 weeks after adrenalectomy. The availability of type-II receptors did not change considerably during the first 24 h after adrenalectomy, but displayed a large increase in capacity during the subsequent 2 weeks. After adrenocortical activation as a consequence of exposure to a novel environment, plasma concentrations of corticosterone increased to reach a peak of 811 nmol/l after 30 min and attained the basal concentration (43 nmol/l) after 240 min. During this time, occupation of type-I receptors increased from 77·8% at 0 min to 97% at 30–60 min and then declined to 84·8% after 240 min. Occupation of the type-II receptors was 28·1% at 0 min, 74·5% after 30 min and 32·8% after 240 min. Injection of dexamethasone (25 μg/100 g body wt) at 08.00 h resulted in suppression of basal plasma concentrations of corticosterone and prevented the circadian-driven rise in circulating corticosterone. Occupation of type-I receptors did not change considerably as a result of injection of dexamethasone, but occupation of type-II receptors was markedly increased till 16.00 h compared with that after injection of vehicle. It was concluded that the type-I and type-II receptors are not only localized differently in the rat brain, but also exhibit a striking difference in occupation after manipulation of the pituitary-adrenocortical system. The data further support the concept of a type-I receptor-mediated tonic activating influence and a type-II receptor-mediated feedback action of corticosterone on brain function. J. Endocr. Quantitative in vitro autoradiography, cytosol receptor assay in punched brain tissue, and immunocytochemistry have revealed that the glucocorticoid receptor is present in the rat supraoptic nucleus (SON). Based on its binding characteristics the receptor appears to be the type II glucocorticoid receptor. With the use of a monoclonal antibody against purified liver glucocorticoid receptor, immunostaining was found in magnocellular neurosecretory neurons in the SON, but not in magnocellular neurons in the paraventricular nucleus. Immunoreactive cells seem to be concentrated in ventral parts of the SON where vasopressin cells were previously shown to be located. One to 2 weeks after bilateral adrenalectomy, there was a substantial decrease in glucocorticoid receptor immunostaining in magnocellular as well as other types of neurons in various brain regions. Administration of synthetic glucocorticoids (RU 28362 or dexamethasone) induced a robust increase in the intensity of immunostaining in cell nuclei of neurosecretory cells. The presence of glucocorticoid receptors in the SON suggests that glucocorticoids may affect vasopressin synthesis or/and secretion through a direct action on magnocellular neurons.

In mice with a homozygous or heterozygous deficiency for ApoE as well as in wild-type animals we established synaptic responsiveness in the hippocampal CA1 area following stimulation of the Schaffer/commissural fibers. The maximal population spike amplitude was significantly larger in wild-type animals than in mice lacking the ApoE gene, whereas the facilitation in population spike amplitude after paired pulse stimulation was most pronounced in homozygous mutant mice. Primed burst stimulation induced a lasting increase in population spike amplitude of all three groups. Apart from a more pronounced initial potentiation in the homozygous mutants, primed burst potentiation was comparable in all groups. Subsequent theta burst stimulation resulted in a long-term enhanced synaptic responsiveness which was impaired in heterozygous animals. The data show that both homo- and heterozygous ApoE mutant mice display altered synaptic plasticity in the hippocampal CA1 area.

The role of brain mineralocorticoid receptor (MR) sites in the pathogenesis of mineralocorticoid hypertension was studied after an intracerebroventricular injection of the MR antagonist RU-28318. Male Wistar rats received subcutaneously implanted deoxycorticosterone acetate (DOCA) pellets and were maintained on 0.9% saline as drinking solution. Under these conditions hypertension developed in approximately 5 wk as assessed in conscious rats by means of the tail-cuff technique. During the development of this hypertension (after 3 wk of DOCA-salt treatment) a single intracerebroventricular injection of the specific MR antagonist RU-28318 reduced systolic blood pressure (SBP) as measured with the tail-cuff method. A decrease in SBP was observed 2–24 h after this intracerebroventricular injection, with the lowest SBP values occurring at 8 h. In these animals (3 wk after DOCA implantation) continuous direct blood pressure recording via chronic cannulation revealed, on the day of the intracerebroventricular injection of RU-28318, a slight increase in arterial pressure during the light phase, followed by a decrease during the dark phase. In the established hypertensive rats (5 wk after DOCA RU-28318 on the arterial pressure or heart rate was detectable. It is concluded that central MR blockade during the development of the DOCA-salt hypertension reduces blood pressure within 24 h assessed with 1) the indirect method at certain time points after exposure to warming and stress and 2) the direct method during the dark phase of the diurnal cycle.

β-Endorphin (β-LPH61–91) is a well known endogenous opioid ligand. It and related peptides have recently been implicated in the control of adaptive behaviour. Smaller β-endorphin fragments appeared to be more active moieties than the parent molecule in a number of behavioural situations1. Their effects seemed to occur independently of interaction with opiate receptor sites in the brain. Moreover, elimination of the opiate-like properties of γ-endorphin (β-LPH61–77) by removing the N-terminal amino acid yielded des-tyrosine-γ-endorphin (β-LPH62–77, dTγE) which had greater behavioural activity than γ-endorphin2,3. The CNS effects of dTγE resembled those of neuroleptic drugs in several test systems, α-Endorphin (β-LPH61–76) exerted effects opposite to those of dTγE and in some aspects its activity was comparable to that of psychostimulant drugs4. This opposition of effects suggests that a balance between γ- and α-type endorphins is involved in the control of brain function5. We report here that either γ-endorphin and dTγE or α-endorphin and des-tyrosine-α-endorphin (β-LPH62–769 dTαE) can be formed preferentially from β-endorphin by enzymes associated with an enriched synaptosomal plasma membrane fraction from brain. It is suggested that these enzymes have a role in brain homeostatic mechanisms by regulating the generation of these substances.

A single intracerebroventricular injection of 10 ng of the mineralocorticoid aldosterone in normotensive male Wistar rats induced a transient increase in blood pressure. Corticosterone produced this response only at a 100-fold higher dose. In contrast, a decrease in systolic blood pressure occurred following the intracerebroventricular administration of a selective glucocorticoid agonist (Ru 28362; 10 ng). The intracerebroventricular administration in normotensive and adrenalectomized, corticosterone-replaced, male rats of an antimineralocorticoid (Ru 28318; 10 ng) decreased systolic blood pressure, while an antiglucocorticoid (Ru 38486; 10 ng) caused an increase. These data suggest that adrenal steroids affect central mechanisms underlying cardiovascular regulation by binding to central mineralo- and glucocorticoid receptors.
