This study is a follow-up of a previously preregistered experiment on the effects of being exposed to an authoritarian populist discourse on moral value threat and tolerance. Supporting our initial hypothesis, in the previous study we observed that an authoritarian populist discourse elicits more value threat compared to a moderate discourse, even when controlling for discourse-participant political orientation matching. We also hypothesized that those participants exposed to the authoritarian populist discourse become less tolerant to alternative political options. This hypothesis was not supported by the data as the analysis of variance did not yield a statistically significant main effect of the type of discourse on tolerance. However, we find an interaction with the variable of satisfaction with democracy. The latter moderated the relationship between the condition and political tolerance, such as tolerance for those satisfied with democracy tended to decrease in the authoritarian populist discourse, while the trend was reversed for those dissatisfied. In the present study we aim to replicate this findings, while also extending them in two main ways: - First, we seek to understand which kind of values are threatened by the populist authoritarian discourse. While it was hypothesized that values would be threatened by this discourse regardless of political orientation, the fact that the value threat scale was content free opens up interesting questions. On one hand it could be that people feel their own personal and ingroup values threatened in all cases, regardless of sharing political orientation with the leader or not. This would mean that the leader elicits a sense of being attacked to those likeminded, while more directly atacking those with opposing values. However, another possibility is that those that share a similar political orientation with the leader especifically perceive their democratic values under attack. This would imply that people share the general outlook of the leader but disagree on his methods. To provide an answer to these questions we will look at this more closely by measuring three types of value threat: threat to personal values, threat to democratic values and threat to social values. Aditionally, as we are mostly interested in the nuances of value threat for those in favour of the political leader, we modify the procedure so that the political orientation of participants and the leader will always match and its effects will be added in the models as a covariable. - Second, aside from replicating the interaction effect of the experimental condition and satisfaction with democracy on tolerance found in the previous Study, we also seek to add a measure that captures agreement with specific intolerant actions. Accordingly, we will include Voelkel's et al. (2023) scale of support for undemocratic practices.

Objective: Perampanel (PER) is indicated as adjunctive antiseizure medication (ASM) in adolescents and adults with epilepsy. Data from clinical trials show good efficacy and tolerability, while limited information is available on the routine clinical use of PER, especially when used as only add-on treatment. Methods: We performed an observational, retrospective, multicenter study on people with focal or generalized epilepsy aged >12 years, consecutively recruited from 52 Italian epilepsy centers. All patients received PER as the only add-on treatment to a background ASM according to standard clinical practice. Retention rate, seizure frequency, and adverse events were recorded at 3, 6, and 12months after PER introduction. Subanalyses by early or late use of PER and by concomitant ASM were also conducted. Results: Five hundred and three patients were included (age 36.5 ± 19.9 years). Eighty-one percent had focal epilepsy. Overall, the retention rate was very high in the whole group (89% at 12months) according with efficacy measures. No major differences were observed in the subanalyses, although patients who used PER as early add-on, as compared with late add-on, more often reached early seizure freedom at 3-month follow-up (66% vs 53%, P=.05). Treatment-emergent adverse events occurred in 25%, far less commonly than in PER randomized trials. Significance: This study confirms the good efficacy and safety of PER for focal or generalized epilepsy in real-life conditions. We provide robust data about its effectiveness as only add-on treatment even in patients with a long-standing history of epilepsy and previously treated with many ASMs.

BACKGROUND: Fibroblast growth factor receptors (FGFRs) are frequently altered in cancers and present a potential therapeutic avenue. However, the type and prevalence of FGFR alterations in infiltrating gliomas (IGs) needs further investigation. OBJECTIVE: To understand the prevalence/type of FGFR alterations in IGs. METHODS: We reviewed clinicopathologic and genomic alterations of FGFR-mutant gliomas in a cohort of 387 patients. Tumors were examined by DNA next-generation sequencing for somatic mutations with a panel interrogating 205-genes. For comparison, cBioPortal databases were queried to identify FGFR-altered IGs. RESULTS: Fourteen patients (3.6%) with FGFR-mutant tumors were identified including 11 glioblastomas, Isocitrate dehydrogenase (IDH) – wildtype (GBM-IDH-WT), 2 oligodendrogliomas, and 1 astrocytoma IDH-mutant. FGFR-altered IGs showed endocrinoid capillaries, microvascular proliferation, necrosis, oligodendroglioma-like cells, fibrin thrombi, microcalcifications, and nodular growth. FGFR3 was the most commonly altered FGFR gene (64.3%). The most common additional mutations in FGFR-altered IGs were TERTp, CDKN2A/B, PTEN, CDK4, MDM2, and TP53. FGFR3 alterations were only observed in GBM-IDH-WT. EGFR alterations were rarely identified in FGFR3-altered gliomas. CONCLUSIONS: Histologic features correlate with FGFR alterations in IGs. FGFR3-TACC3 fusion and FGFR3 amplification are the most common FGFR alterations in IGs. FGFR alterations are a rare, but potentially viable, therapeutic target in asubset of IGs.

BACKGROUND: The prevalence of obesity in adolescents has increased worldwide for recent years. Body mass index (BMI)/age, waist circumference (WC), and waist-to-height Ratio (WHtR) are commonly used for assessment of nutritional status. Daily intake of fruits and vegetables and eating time of fruits inversely associated with obesity. AIM: This study aimed to investigate the associations of daily intake of fruits and vegetables and eating time of fruits with obesity in female adolescents at the Gontor Islamic boarding school for girls. MATERIALS AND METHODS: This cross-sectional study recruited 245 female adolescents from the Gontor Islamic Boarding School for girls, which were selected with a purposive sampling technique. They were eligible to follow this study if aged 12–16 years old and were healthy. Data of daily intake of fruits and vegetables were collected using the modified semi quantitative food frequency questionnaire. Anthropometric data consisted of body weight, height, and WC were measured using the standard measurements. Associations of individual variables with anthropometric parameters were analyzed using the Chi-square test. The multiple logistic regression tests were, then, used to analyze associations of independent variables and confounding factors together with BMI/age, WC, and WHtR with a 95% significant level. RESULTS: The prevalence of overweight and obese was 31 and 2% among female adolescents, respectively. Intake of fruits and vegetables associated with BMI/age (p = 0.001), WC (p = 0.039), and WHtR (p = 0.024). Eating time of fruits had no associations with BMI/age (p = 0.207), WC (p = 0.551), and WHtR (p = 0.132). Inadequate intake of fruits and vegetables inversely associated with BMI/age (OR = 2.60; p = 0.002), WC (OR = 1.86; p = 0.047), and WHtR (OR = 1.92; p = 0.039). CONCLUSION: Daily intake of fruits and vegetables inversely associates with BMI/age, WC, and WHtR but not for eating time of fruits among female adolescents in the Gontor Islamic Boarding School for girls.
