Abstract

Large-scale NaA zeolite membranes in a length of 1000 mm were successfully synthesized for the first time on macroporous tubular stainless steel (MTSS) supports in this work. The influencing factors for membrane synthesis, such as pre-treatment, synthesis time, synthesis temperature, Na2O/SiO2 ratio, synthesis reproducibility, membrane stability and support reusability, were investigated systematically. The experimental results revealed that the high-flux NaA membranes prepared on the reusable MTSS supports exhibits a high total flux of 8.28 kg m−2 h−1 and a separation factor of 11,000 for the dehydration of a 90 wt% ethanol solution through pervaporation. The MTSS tubes can be used for at least 8 synthesis-rinse-synthesis circles, and the as-synthesized membranes still maintain high competitive pervaporation performance. Additionally, 93 pieces of membrane samples were assembled in an industrial plant that can produce approximate 99.8 wt% ethanol from 93 wt% ethanol mixtures with a product flow rate of about 60 kg h−1. Importantly, the membrane showed a high stability for running over a whole month, demonstrating that the MTSS-supported membranes have reliable stability for industrial applications.

Abstract

Background Exosomes are nanoscale membrane vesicles secreted by both normal and cancer cells, and cancer cell-derived exosomes play an important role in the cross-talk between cancer cells and other cellular components in the tumor microenvironment. Mesenchymal stem cells (MSCs) have tropism for tumors and have been used as tumor-tropic vectors for tumor therapy; however, the safety of such therapeutic use of MSCs is unknown. In this study, we investigated the role of glioma cell-derived exosomes in the tumor-like phenotype transformation of human bone marrow mesenchymal stem cells (hBMSCs) and explored the underlying molecular mechanisms. Methods The effect of exosomes from U251 glioma cells on the growth of hBMSCs was evaluated with the CCK-8 assay, KI67 staining, and a cell cycle distribution assessment. The migration and invasion of hBMSCs were evaluated with a Transwell assay. A proteomics and bioinformatics approach, together with Western blotting and reverse transcriptase-polymerase chain reaction, was used to investigate the effect of U251 cell-derived exosomes on the proteome of hBMSCs. Results U251 cell-derived exosomes induced a tumor-like phenotype in hBMSCs by enhancing their proliferation, migration, and invasion and altering the production of proteins involved in the regulation of the cell cycle. Moreover, U251 cell-derived exosomes promoted the production of the metastasis-related proteins MMP-2 and MMP-9, glioma marker GFAP, and CSC markers (CD133 and Nestin). The ten differentially expressed proteins identified participated in several biological processes and exhibited various molecular functions, mainly related to the inactivation of glycolysis. Western blotting showed that U251 cell-derived exosomes upregulated the levels of Glut-1, HK-2, and PKM-2, leading to the induction of glucose consumption and generation of lactate and ATP. Treatment with 2-deoxy-d-glucose significantly reversed these effects of U251 cell-derived exosomes on hBMSCs. Conclusions Our data demonstrate that glioma cell-derived exosomes activate glycolysis in hBMSCs, resulting in their tumor-like phenotype transformation. This suggests that interfering with the interaction between exosomes and hBMSCs in the tumor microenvironment has potential as a therapeutic approach for glioma. Graphical abstract ᅟ

Abstract

An efficient base‐promoted approach for the synthesis of pyrido[1,2‐a]pyrimidinones from ynones and 2‐methylpyrimidin‐4‐ols have been developed via the C−N and C−C formation procedure. Diversely structural pyrido[1,2‐a]pyrimidinones were afforded in up to 95% yield for 29 examples. This reaction featured with advantages such as practical tandem procedure, wide functional group tolerance and metal‐free conditions, which has potential application in synthetic and medicinal chemistry. Abstract

Bing Xin is a prestigious writer in the field of children’s literature in the 20th century. Her beautiful and unique type of writing is named as “Bing Xin Style”. Generally speaking, the Chinese literary circles believe that there are two distinct characteristics in terms of Bing Xin Style, that is to say, “classical vernacular Chinese” and “westernization of Chinese”. To be specific, “classical vernacular Chinese” means one can enjoy the beauty of concise structure of classical Chinese in her vernacular Chinese articles. Meanwhile, “westernization of Chinese” means the Chinese text is imbued with certain features of English grammar. This paper will focus on the Japanese version of Bing Xin’s magnum opus “To My Little Readers”, study the translation methods of “Bing Xin Style” through the use of syllables, level and oblique tones as well as attributives, and then discuss the translation possibility and methods of literary style based on the previous research.

Abstract

Intermediary signals, precociously enhancing GLUT5 transcription in response to perfusion of its substrate, fructose, in the small intestine of neonatal rats, are not known. Because glucose-6-phosphatase (G6Pase), glucose-6-phosphate translocase (G6PT), and fructose-1,6-bisphosphatase (FBPase) expression increases parallel to or precedes that of GLUTS, we investigated the link between these gluconeogenic genes and GLUT5 by using vanadate or tungstate, potent inhibitors of gluconeogenesis. Small intestinal perfusions of 20-d-old rats were performed with fructose alone, fructose + vanadate or tungstate, glucose alone, and glucose + vanadate or tungstate. As expected, fructose, but not glucose nor glucose + inhibitor perfusion, increased GLUT5 mRNA abundance and fructose transport. Fructose perfusion dramatically increased G6Pase mRNA abundance but had no effect on G6Pase activity. In sharp contrast, fructose perfusion did not increase FBPase gene expression but stimulated FBPase activity. Both vanadate and tungstate significantly inhibited G6Pase activity but did not prevent the fructose-induced increases in G6Pase and G6PT gene expression. Perfusion with fructose + vanadate prevented the fructose-induced increases in fructose transport and GLUT5 mRNA abundance, whereas perfusion with fructose + tungstate did not. Interestingly, vanadate, but not tungstate, inhibited the fructose-induced increase in FBPase activity. Thus, vanadate inhibition of fructose-induced increases in FBPase activity paralleled exactly vanadate inhibition of fructose-induced increases in GLUT5 mRNA abundance and activity. Fructose-induced changes in FBPase activity may regulate changes in G LUT5 expression and activity in the small intestine of neonatal rats. The marked increases in intestinal G6Pase and GLUT5 mRNA abundance may be a parallel response to different factors released during fructose perfusion.
