Abstract

The purpose of this research is to investigate the role of information & communication technology (ICT) and e-governance in health sector of Pakistan, where these facilities are at initial stage of development. This study utilizes primary data collected from 170 patients in both public and private hospitals of district Peshawar Pakistan. This is an exploratory study and descriptive statistics are being used for the analysis of the data. The result of the study shows that the use of e-governance in health sector is at a rudimentary stage in the study area. Few people use ICT for medical purposes but the overall level is low. There is need of awareness. In addition, the facilitating conditions for the adoption of e-governance in health sector include infrastructure, electricity, proper user interface and data privacy & confidentiality. The government needs to initiate practical steps for the implementation of e-governance in health sector. This will improve the efficiency, transparency, and accountability of the system. Moreover, it will also speed up the service delivery system. The study is valuable as it explored the role of ICT and e-governance in health sector of Pakistan for the first time. Therefore, this study contributed to literature in a positive way.

257. Combinatorial Metabolic Engineering of Escherichia colifor Enhanced L-Cysteine Production: Insights into Crucial Regulatory Modes and Optimization of Carbon-Sulfur Metabolism and Cofactor Availability

Abstract

Microbial production of valuable compounds can be enhanced by various metabolic strategies. This study proposed combinatorial metabolic engineering to develop an effective Escherichia colicell factory dedicated to L-cysteine production. First, the crucial regulatory modes that control L-cysteine levels were investigated to guide metabolic modifications. A two-stage fermentation was achieved by employing multi-copy gene expression, improving the balance between production and growth. Subsequently, carbon flux distribution was further optimized by modifying the C1 unit metabolism and the glycolytic pathway. The modifications of sulfur assimilation demonstrated superior performance of thiosulfate utilization pathways in enhancing L-cysteine titer. Furthermore, the studies focusing on cofactor availability and preference emphasized the vital role of synergistic enhancement of sulfur-carbon metabolism in L-cysteine overproduction. In a 5 L bioreactor, the strain BW15-3/pED accumulated 12.6 g/L of L-cysteine. This work presented an effective metabolic engineering strategy for the development of L-cysteine-producing strains.

Abstract

Background: The Gene Regulatory Network (GRN) is a model for studying the function and behavior of genes by treating the genome as a whole, which can reveal the gene expression mechanism. However, due to the dynamics, nonlinearity, and complexity of gene expression data, it is a challenging task to construct a GRN precisely. And in the circulating cooling water system, the Slime-Forming Bacteria (SFB) is one of the bacteria that helps to form dirt. In order to explore the microbial fouling mechanism of SFB, constructing a GRN for the fouling-forming genes of SFB is significant. Objective: Propose an effective GRN construction method and construct a GRN for the foulingforming genes of SFB. Methods: In this paper, a combination method of Long Short-Term Memory Network (LSTM) and Mean Impact Value (MIV) was applied for GRN reconstruction. Firstly, LSTM was employed to establish a gene expression prediction model. To improve the performance of LSTM, a Particle Swarm Optimization (PSO) was introduced to optimize the weight and learning rate. Then, the MIV was used to infer the regulation among genes. In view of the fouling-forming problem of SFB, we have designed electromagnetic field experiments and transcriptome sequencing experiments to locate the fouling-forming genes and obtain gene expression data. Results: In order to test the proposed approach, the proposed method was applied to three datasets: a simulated dataset and two real biology datasets. By comparing with other methods, the experimental results indicate that the proposed method has higher modeling accuracy and it can be used to effectively construct a GRN. And at last, a GRN for fouling-forming genes of SFB was constructed using the proposed approach. Conclusion: The experiments indicated that the proposed approach can reconstruct a GRN precisely, and compared with other approaches, the proposed approach performs better in extracting the regulations among genes.

【Abstract】Objective: To investigate the clinical features and treatment strategy of dissymmetric bilateral frontal contusion, and to summarize our experience in treat-ing these patients by minimally invasive surgery. Methods: Over the past 3 years, we have treated a total of 31 patients with dissymmetric bilateral frontal contusion using endoscopy-assisted unilateral cerebral falx incision. Other 30 patients treated by routine bilateral approaches within the same period were taken as control. Results: Seventeen cases (54.8%) in the unilateral operation group survived and were in good condition, 8 (25.8%) had moderate disability, 4 (12.9%) had severe disability, 1 (3.2%) was in vegetative state, and 1 (3.2%) died. Compared with the control group, the Glasgow Out-come Scale score was not significantly different in the uni-lateral operation group, but the operation time, blood trans-fusion volume, the length of hospital stay, the incidences of mental disorder and olfactory nerve injury were greatly reduced in the unilateral operation group. Conclusions: Endoscopy-assisted unilateral cerebral falx incision can shorten the operation time, reduce surgical trauma and complications in treatment of patients with dis-symmetric bilateral frontal contusion. It can obviously di-minish the chance of delayed intracerebral hematoma and subsequently minimize the incidences of subfalcial and centrencephalic herniation.
