Objective To explore the feasibility of a modified micro-cannula for intra-cerebral administration of 6-hydroxydopamine(6-OHDA) in order to establish a Parkinsonian mouse model. Methods Twenty female mice(C57BL/6J) were randomly divided into 2 groups with 10 in each: experimental group (new micro-cannula) and control group (regular Hamilton syringe). Following unilateral injection of 6-OHDA, the behavioral test and histological microscopy were performed. Results All mice in experimental group showed the morphology of apomorphine-induced rotational asymmetry, but the same results were found in only 5 mice from control group. Total number of rotations away from the lesion side in 30 minutes showed 6-OHDA-induced motor deficits at day 7 in both groups, which was significantly attenuated at day 28 in control group (P＜0.05). After 28 days of 6-OHDA injection, significant reduction in the number of rotations was observed in control mice as compared with that of experimental group (P＜0.05). As immunohistochemistry showed that the total number of remaining dopaminergic neurons significantly decreased in the substantial nigra of the experimental mice as compared with the control mice(P＜0.05). Moreover, more local astrocytes underwent activation, part of which acquired the properties of neural precursor cells in control mice. Conclusions Intrastriatal injection of 6-OHDA using the new micro-cannula is a feasible technology in the development of a more reliable Parkinsonian model of mice.

Dwarfism is an important trait which is closely related to the efficiency of fruit orchard management and production. However, dwarfing cannot be widely applied in the cultivation of pears, especially Asian pears. Developing varieties with dwarf characteristics is a goal of paramount importance in pear breeding. In the present study, dwarf phenotype pears (DPPs) and arborescent phenotype pears (APPs) were obtained from the offspring of a cross between ‘Aiyuxiang’ and ‘Cuiguan’ pear cultivars, which exhibited dwarfed and arborescent statures, respectively. When compared with APPs, the heights of DPPs showed a 62.8% reduction, and the internode lengths were significantly shorter. Cross-grafting between DPPs and APPs demonstrated that the dwarfed phenotype of DPPs was primarily induced by the aerial portions of the plant, and independent of the root system. Observations of stem tissue sections showed that DPP cells were arranged chaotically with irregular shapes, and the average length was larger than that of the APP cells. A total of 1 401 differently expressed genes (DEGs) in shoot apices between DPPs and APPs were identified by RNA-sequencing (RNA-Seq), and these DEGs were mainly enriched in the ‘phytohormone-related pathways, cell wall metabolism and cell division’ categories. Moreover, 101 DEGs were identified as transcription factors (TFs). In DPPs, several brassinosteroids (BR) signaling and cell cycle-related genes were significantly down-regulated, while genes involved in BR and GA degradation were up-regulated. Comprehensive analysis of RNA-Seq data and stem tissue sections suggested that the dwarfed phenotype of DPPs could be primarily attributed to deficiencies in cell division. Previous work using simple sequence repeat (SSR) markers narrowed the location of the gene responsible for the dwarf phenotype of ‘Le Nain Vert’. Through combined analysis of our transcriptomic data with the SSR results, we identified four genes as promising candidates for the dwarf phenotype, among which, a DELLA gene could be the most promising. The results presented in this study provide a sound foundation for further exploration into the genetic and molecular mechanisms underlying pear dwarfing.

Abstract Background Tuberculosis is a bacterial infection involving multiple organs and systems. Its hematological presentation mainly includes anemia and leukocytosis. Evans syndrome is a rare autoimmune disease characterized by autoimmune hemolytic anemia, immune thrombocytopenia, and neutropenia, with positive results for the direct Coombs test and platelet antibodies. The cooccurrence of tuberculosis and Evans syndrome is rarely reported. Case presentation A 69-year-old female presented with a fever and shortness of breath. Her chest computerized tomography scan showed extensive miliary nodules in the bilateral lung fields. She rapidly developed respiratory failure that required endotracheal intubation and mechanical ventilation. The acid-fast bacilli sputum smear results indicated a grade of 3+. Later on, blood testing revealed hemolytic anemia, a positive direct Coombs test result, and the presence of the platelet antibody IgG. This patient was diagnosed as having disseminated pulmonary tuberculosis and Evans syndrome. She successfully recovered after treatment with antituberculosis drugs and glucocorticoids. Conclusions Tuberculosis can occur together with Evans syndrome. Affected patients should receive both antituberculosis and immunosuppressive drugs.

SEBS-based thermoplastic elastomers modified by PA6 with maleic anhydride grafted copolymer (SEBS-g-MAH or PP-g-MAH) as the compatibilizer were prepared, and the effects of compatibilizer and PA6 on the mechanical and surface properties of blends were investigated in detail. The dosage ratio of SEBS-g-MAH to PA6 at 3:5 is an optimal combination for improving mechanical properties. The maleic anhydride of SEBS-g-MAH can react with the terminal amine of PA6 to form the SEBS-g-PA6 graft copolymer as a bridge at the phase interface. With the increase of PA6, a typical sea-island structure gradually appears in the matrix, and when PA6 is 15 phr, the tensile strength and ultimate elongation both reach the maximum of 18.58 MPa and 1097%, and the corresponding wear rate is only 54.4% of that before modification. Furthermore, the addition of PA6 also significantly reduced the contact angle of the blends to below 90[degrees], transforming the hydrophobic surface into a hydrophilic surface. These outcomes provide an effective strategy for reinforcing and modifying the surface properties of thermoplastic elastomers and benefit the development of SEBS in industrial applications. Abstract Objective To analyze the clinical characteristics and bronchoalveolar lavage fluid pathogens in elderly patients with community‐acquired pneumonia (CAP). Methods This was a retrospective observational epidemiological study using that elderly cases diagnosed with community‐acquired pneumonia receiving treatment at the Affiliated Hospital of North China University of Technology, Tangshan Hongci Hospital and Tangshan Fengnan District Hospital of Traditional Chinese Medicine. A total of 92 cases were divided into two groups according to age. There were 44 patients over 75‐year‐old and 48 patients between 65 and 74‐year‐old. Results Compared with the elderly 65 to 74‐year‐old, the elderly over 75‐year‐old with diabetes are more likely to suffer from CAP (35.42% vs. 63.64%, p = 0.007) and are more likely to have mixed infections (6.25% vs. 22.73%, p = 0.023) or larger lesions (45.83% vs. 68.18%, p = 0.031). Their hospital stays will also be extended (39.58% vs. 63.64%, p = 0.020), and the albumin level (37.51 ± 8.92 vs. 30.93 ± 6.58, p = 0.000), the neutrophils level (9.09(6.26–10.63) vs. 7.18(5.35–9.17),p = 0.026) is significantly lower and the d‐dimer (505.42 ± 197.12 vs. 611.82 ± 195.85, p = 0.011), PCT (0.08 ± 0.04 vs. 0.12 ± 0.07, p = 0.001) levels are significantly higher. Conclusion The clinical symptoms and signs of elderly CAP patients are not so typical, and the infection is more serious. Attention should therefore be paid to elderly patients. Hypoalbuminemia and high d‐dimer can predict the prognosis of patients.

Moth mouthparts, consisting of labial palps and proboscis, not only are the feeding device but also are chemosensory organs for the detection of chemical signals from surrounding environment. Up to now, the chemosensory systems in the mouthpart of moths are largely unknown. Here, we performed systematic analyses of the mouthpart transcriptome of adult Spodoptera frugiperda (Lepidoptera: Noctuidae), a notorious pest that spreads worldwide. A total of 48 chemoreceptors, including 29 odorant receptors (ORs), 9 gustatory receptors (GRs), and 10 ionotropic receptors (IRs), were annotated. Further phylogenetic analyses with these genes and homologs from other insect species determined that specific genes, including ORco, carbon dioxide receptors, pheromone receptor, IR co-receptors, and sugar receptors, were transcribed in the mouthpart of S. frugiperda adults. Subsequently, expression profiling in different chemosensory tissues demonstrated that the annotated ORs and IRs were mainly expressed in S. frugiperda antennae, but one IR was also highly expressed in the mouthparts. In comparison, SfruGRs were mainly expressed in the mouthparts, but 3 GRs were also highly expressed in the antennae or the legs. Further comparison of the mouthpart-biased chemoreceptors using RT-qPCR revealed that the expression of these genes varied significantly between labial palps and proboscises. This study provides the first large-scale description of chemoreceptors in the mouthpart of adult S. frugiperda and provides a foundation for further functional studies of chemoreceptors in the mouthpart of S. frugiperda as well as of other moth species.

BackgroundFLAIR-hyperintense lesions in anti-MOG-associated encephalitis with seizures (FLAMES) has been identified increasingly frequently in recent years. However, this rare MOG antibody disease may coexist with anti-N-methyl-D-aspartate receptor encephalitis (anti-NMDARe), in an overlap syndrome with unknown clinical features and prognosis.MethodsWe report a new case of this overlap syndrome and present a systematic review of similar cases in the literature to provide information on the clinical presentation, MRI features, EGG abnormalities, treatment, and prognosis of patients with this rare syndrome.ResultsA total of 12 patients were analyzed in the study. The most common clinical manifestations of FLAMES overlaid with anti-NMDARe were epilepsy (12/12), headache (11/12), and fever (10/12). Increases in intracranial pressure (median: 262.5 mmH2O, range: 150–380 mmH2O), cerebrospinal fluid (CSF) leukocyte count (median: 128×106/L, range: 1-610×106/L), and protein level (median: 0.48 g/L) were also observed. The median CSF anti-NMDAR antibody titer was 1:10 (1:1–1:32), while the median serum MOG antibody titer was 1:32 (1:10–1:1024). Seven cases exhibited unilateral cortical FLAIR hyperintensity, and five cases (42%) had bilateral cortical FLAIR hyperintensity, including four cases involving the bilateral medial frontal lobes. Of the 12 patients, five showed lesions at other sites (e.g., the brainstem, corpus callosum, or frontal orbital gyrus) before or after the development of cortical encephalitis. EEG showed slow waves in four cases, spike–slow waves in two cases, an epileptiform pattern in one case, and normal waves in two cases. The median number of relapses was two. Over a mean follow-up period of 18.5 months, only one patient experienced residual visual impairment, while the remaining 11 patients had good prognoses.ConclusionFLAMES alone is difficult to distinguish from overlap syndrome based on clinical features. However, FLAMES with bilateral medial frontal lobe involvement suggests the presence of the overlap syndrome.

Chemosensation of tarsi provides moths with the ability to detect chemical signals which are important for food recognition. However, molecular mechanisms underlying the chemosensory roles of tarsi are still unknown. The fall armyworm Spodoptera frugiperda is a serious moth pest that can damage many plants worldwide. In the current study, we conducted transcriptome sequencing with total RNA extracted from S. frugiperda tarsi. Through sequence assembly and gene annotation, 23 odorant receptors 10 gustatory receptors and 10 inotropic receptors (IRs) were identified. Further phylogenetic analysis with these genes and homologs from other insect species indicated specific genes, including ORco, carbon dioxide receptors, fructose receptor, IR co-receptors, and sugar receptors were expressed in the tarsi of S. frugiperda. Expression profiling with RT-qPCR in different tissues of adult S. frugiperda showed that most annotated SfruORs and SfruIRs were mainly expressed in the antennae, and most SfruGRs were mainly expressed in the proboscises. However, SfruOR30, SfruGR9, SfruIR60a, SfruIR64a, SfruIR75d, and SfruIR76b were also highly enriched in the tarsi of S. frugiperda. Especially SfruGR9, the putative fructose receptor, was predominantly expressed in the tarsi, and with its levels significantly higher in the female tarsi than in the male ones. Moreover, SfruIR60a was also found to be expressed with higher levels in the tarsi than in other tissues. This study not only improves our insight into the tarsal chemoreception systems of S. frugiperda but also provides useful information for further functional studies of chemosensory receptors in S. frugiperda tarsi.

BackgroundLignin is a key component of the secondary cell wall of plants, providing mechanical support and facilitating water transport as well as having important impact effects in response to a variety of biological and abiotic stresses.ResultsIn this study, we identified 104 genes from ten enzyme gene families related to lignin biosynthesis in Musa acuminata genome and found the number of MaCOMT gene family was the largest, while MaC3Hs had only two members. MaPALs retained the original members, and the number of Ma4CLs in lignin biosynthesis was significantly less than that of flavonoids. Segmental duplication existed in most gene families, except for MaC3Hs, and tandem duplication was the main way to expand the number of MaCOMTs. Moreover, the expression profiles of lignin biosynthesis genes during fruit development, postharvest ripening stages and under various abiotic and biological stresses were investigated using available RNA-sequencing data to obtain fruit ripening and stress response candidate genes. Finally, a co-expression network of lignin biosynthesis genes was constructed by weighted gene co-expression network analysis to elucidate the lignin biosynthesis genes that might participate in lignin biosynthesis in banana during development and in response to stresses.ConclusionThis study systematically identified the lignin biosynthesis genes in the Musa acuminata genome, providing important candidate genes for further functional analysis. The identification of the major genes involved in lignin biosynthesis in banana provides the basis for the development of strategies to improve new banana varieties tolerant to biological and abiotic stresses with high yield and high quality.

Alport syndrome (AS) is an inherited glomerular basement membrane (GBM) disease leading to end-stage renal disease (ESRD). X-linked AS (XLAS) is caused by pathogenic variants in the COL4A5 gene. Many pathogenic variants causing AS have been detected, but the genetic modifications and pathological alterations leading to ESRD have not been fully characterized. In this study, a novel frameshift variant c.980_983del ATGG in the exon 17 of the COL4A5 gene detected in a patient with XLAS was introduced into a mouse model in by CRISPR/Cas9 system. Through biochemical urinalysis, histopathology, immunofluorescence, and transmission electron microscopy (TEM) detection, the clinical manifestations and pathological alterations of Del-ATGG mice were characterized. From 16 weeks of age, obvious proteinuria was observed and TEM showed typical alterations of XLAS. The pathological changes included glomerular atrophy, increased monocytes in renal interstitial, and the absence of type IV collagen α5. The expression of Col4a5 was significantly decreased in Del-ATGG mouse model. Transcriptomic analysis showed that differentially expressed genes (DEGs) accounted for 17.45% (4,188/24003) of all genes. GO terms indicated that the functions of identified DEGs were associated with cell adhesion, migration, and proliferation, while KEGG terms found enhanced the degradation of ECM, amino acid metabolism, helper T-cell differentiation, various receptor interactions, and several important pathways such as chemokine signaling pathway, NF-kappa B signaling pathway, JAK–STAT signaling pathway. In conclusion, a mouse model with a frameshift variant in the Col4a5 gene has been generated to demonstrate the biochemical, histological, and pathogenic alterations related to AS. Further gene expression profiling and transcriptomic analysis revealed DEGs and enriched pathways potentially related to the disease progression of AS. This Del-ATGG mouse model could be used to further define the genetic modifiers and potential therapeutic targets for XLAS treatment.
