252. Limited functional and metabolic improvements in hypertrophic and healthy rat heart overexpressing the skeletal muscle isoform of SERCA1 by adenoviral gene transfer in vivo

Abstract

Adenoviral gene transfer of sarco(endo)plasmic reticulum [Ca.sup.2+]-ATPase (SERCA)2a to the hypertrophic heart in vivo has been consistently reported to lead to enhanced myocardial contractility. It is unknown if the faster skeletal muscle isoform, SERCA1, expressed in the whole heart in early failure, leads to similar improvements and whether metabolic requirements are maintained during an adrenergic challenge. In this study, Ad.cmv.SERCA1 was delivered in vivo to aortic banded and sham-operated Sprague-Dawley rat hearts. The total SERCA content increased 34%. At 48-72 h posttransfer, echocardiograms were acquired, hearts were excised and retrograded perfused, and hemodynamics were measured parallel to NMR measures of the phosphocreatine (PCr)-to-ATP ratio (PCr/ATP) and energy substrate selection at basal and high workloads (isoproterenol). In the Langendorff mode, the rate-pressure product was enhanced 27% with SERCA1 in hypertrophic hearts and 10% in shams. The adrenergic response to isoproterenol was significantly potentiated in both groups with SERCA1. [sup.31]P NMR analysis of PCr/ATP revealed that the ratio remained low in the hypertrophic group with SERCA 1 overexpression and was not further compromised with adrenergic challenge. [sup.13]C NMR analysis revealed fat and carbohydrate oxidation were unaffected at basal with SERCA1 expression; however, there was a shift from fats to carbohydrates at higher workloads with SERCA1 in both groups. Transport of NADH-reducing equivalents into the mitochondria via the [alpha]-ketoglutamate-malate transporter was not affected by either SERCA1 overexpression or adrenergic challenge in both groups. Echocardiograms revealed an important distinction between in vivo versus ex vivo data. In contrast to previous SERCA2a studies, the echocardiogram data revealed that SERCA1 expression compromised function (fractional shortening) in the hypertrophic group. Shams were unaffected. Abstract

A field experiment was conducted to study the effects of different ratios of chemical fertilizer nitrogen substitution by biogas slurry on yield, growth characteristics(development progress, plant height dynamic, population dynamic and dry matter accumulation dynamic), quality and heavy metal content of winter wheat. The results showed that winter wheat yield increased significantly(P-2, 18.14% higher than the treatment with total chemical fertilizer nitrogen(Ncf). There were no significant differences between the yields of 100%Nbs and Ncf(P>0.05). The 1000-kernels weight presented a significant declining trend(PPPPP>0.05). The contents of these grain heavy metal of each treatment were lower than the maximum levels of contaminants in foods. When the ratio of chemical fertilizer nitrogen substitution by biogas slurry raised above 50%, because of the prolonged growth duration and the increased basal internode length could lead to lodge and yield reduction, and because of greater use of biogas slurry could lead to the risk of secondary pollution. Therefore, in our preliminary study, in this experiment, the optimal ratio of chemical fertilizer nitrogen substitution by biogas slurry was 50%.

270. Preparation and electrochemical performances of ZnMoO4-ZnFe2O4 composite electrode materials.

Abstract

As a typical transition metal oxide anode material, ZnMoO4 can deliver a higher theoretical capacity of 951.6 mAh g−1 due to its variable oxidation state and alloying reaction. However, lower conductivity and huge volumetric expansion restrict its further development. Herein, the modified sol–gel method is applied to synthesize a series of (1-x)ZnMoO4-xZnFe2O4 (x = 0.1, 0.2, 0.3, 0.4, and 0.5) composites. The effects of compound proportion of ZnFe2O4 on the crystal phase structure and morphology for ZnMoO4 have been investigated in detail. All the composites have been conducted on electrochemical measurements, and the results illustrate that the composites display better electrochemical performance than that of ZnMoO4. Especially for 0.6ZnMoO4-0.4ZnFe2O4, it can deliver a capacity of 550.7 mAh g−1 after 500 cycles at a current density of 200 mA g−1. Moreover, it also presents the higher capacity at higher current density. The improvement of electrochemical performance should be attributed to the synergistic effect of ZnFe2O4 and ZnMoO4. Abstract

This article explores the two‐interval, two‐alternative forced‐choice (2I2AFC) paradigm in signal detection theory (SDT), which can also be called the paired A‐Not A with AB and BA pairs in the sensory and consumer fields. The article describes how to estimate the parameters and their variances from the psychometric function of the method. Tables and R codes were developed and are provided for the estimations. The performance of the method is evaluated. It shows that the paired A‐Not A with AB and BA pairs is more effective and powerful than the conventional A‐Not A and the A‐Not A with reminder (A‐Not AR) tests. An extension of the method, that is, the ratings of the paired A‐Not A with AB and BA pairs is also explored. A facial shine case study applying this method is provided. Practical applications: As one of the paired versions of sensory discrimination methods, the paired A‐Not A with AB and BA pairs has wide potential applications to detect and measure sensory difference, especially to detect and measure before‐after treatment effects of products.
