Phospholipases A2 (PLA2) are enzymes that trigger the degradation cascade of the arachidonic acid, leading to the formation of pro-inflammatory eicosanoids. The selective inhibition of PLA2s is crucial in the search for a more efficient anti-inflammatory drug with fewer side effects than the drugs currently used. Hence, we studied the influences caused by two pyrazolonic inhibitors: dipyrone (DIP) and 1-phenyl-3-methyl-5-pyrazolone (PMP) on the kinetic behavior of PLA2 from Crotalus adamanteus venom. Molecular modeling results, by DFT and MM approaches, showed that DIP is strongly associated to the active site of PLA2 through three hydrogen bonds, whereas PMP is associated to the enzyme just through hydrophobic interactions. In addition, only PMP presents an intramolecular hydrogen bond that make difficult the formation of more efficient interactions with PLA2. These results help in the understanding of the experimental observations. Experimentally, the results showed that PLA2 from C. adamanteus present a typical Michaelian behavior. In addition, the calculated kinetic parameters showed that, in the presence of DIP or PMP, the maximum enzymatic velocity (VMAX) value was kept constant, whereas the Michaelis constant (KM) values increased and the inhibition constant (KI) decreased, indicating competitive inhibition. These results show that the phenyl-pyrazolonic structures might help in the development and design of new drugs able to selectively inhibit PLA2. © 2008 Wiley Periodicals, Inc. The purpose of the current study was to investigate the effect of 10 weeks of strength training on the flexibility of sedentary middle-aged women. Twenty women were randomly assigned to either a strength training group (n = 10; age, 37 +/- 1.7 years; body mass, 65.2 +/- 10.7 kg; height, 157.7 +/- 10.8 cm; and body mass index, 25.72 +/- 3.3 kg x m(-2)) or a control group (n = 10; age, 36.9 +/- 1.2 years; body mass, 64.54 +/- 10.18 kg; height, 158.1 +/- 8.9 cm; and body mass index, 26.07 +/- 2.8 kg x m(-2)). The strength training program was a total body session performed in a circuit fashion and consisted of 7 exercises performed for 3 circuits of 8 to 12 repetitions maximum (RM), except for the abdominal exercise which was performed for 15 to 20 RM. Flexibility measurements were taken for 10 articulation movements pre and post training: shoulder flexion and extension, shoulder horizontal adduction and abduction, elbow flexion, hip flexion and extension, knee flexion, and trunk flexion and extension. Pre and post training, 10 RM strength significantly increased (p < 0.05). Of the movements examined, only shoulder horizontal adduction, hip flexion and extension, and trunk flexion and extension demonstrated significant increases (p < 0.05). Neither elbow nor knee flexion showed a significant change with weight training. The control group showed no significant change in any of the flexibility measures determined. The purpose of this study was to characterize the anthropometric aspects and also the body composition of the athletes who have taken part at the Amazon Adult Male Cup of Clubs, in the year of 2002. Besides defining the athletes' position per game, it was considered the characteristics of the best player per game in each position. Therefore, some measures were taken: body mass, stature, width of the sail, hand size, wrist width, lower extremity length, sitting height, forearm circumference, abdominal circumference, % fat, fat mass, lean body mass. For the results analysis it was used score Z, considering the results of the best player and the average obtained by the others players of the same position. The results have shown that the best players and the others players of distinct positions presented values lower than the suggested average for the players of this sport, except for the left and right wings and the background central, in fat percentage. Despite the scarcity of data about this sport, these can be taken as initial parameters, and more studies should be developed aiming to characterize not only adult athletes, but also athletes of different age levels, what later on will serve as reference parameters for the improvement of athletes that practice this modality of sport, in Brazilian north region. The arterial vascularization of agoutis’ penis ( Dasyprocta prymnolopha) were analysed using ten male adults from ‘Núcleo de Estudos e Preservação de Animais Silvestres da Universidade Federal do Piauí’ (FUFPI/IBAMA n° 02/99). Among the total number of specimens, six animals had natural death and were members of the research collection of the Laboratory of Anatomy, and four were killed after anaesthesia. Stained bi-centrifugated-Cis-I-4 latex was injected in arterial vessels responsible for penis vascularization throughout the abdominal portion of aorta. The samples were fixed in 10% formaldehyde solution and arteries were dissected. The penile artery is originated as a branch of internal pudendal artery. At the level of ischiatic arch, the penile artery project two branches, the penile dorsal and the deep arteries; those arteries irrigates the penile dorsal surface and the corpus cavernosum penis. The penile dorsal arteries have an independent course up to the glans penis. Based on the conditions of this work a remarkable similarity regarding the distribution of vessels destined to the agouti penis when compared to other domestic, wild and lagomorph rodents as rabbits. Aim: to evaluate the degree of implementation of the Program for the Control of Tuberculosis in Primary Health Care of a capital of the Brazilian Northeast. Method: this is a quantitative and evaluative study, with an emphasis on the degree of program implementation. 96 nurses from 48 primary health care units in the city of Natal/RN will participate according to the results of the sample calculation using the Barbetta formula, considering the 54 units in the municipality. The data will be collected through a structured questionnaire in three sections: professional characterization, structure dimension, and process dimension. The analysis will be done by descriptive statistics and the percentages identified will allow classifying the degree of implantation in: not implanted (0% to 25%), incipient implantation (26% to 50%), partially implanted (51% to 75%) or totally implanted (76% to 100%). Expected results: provide managers and professionals with data that will assist in making decisions that impact on improvements to the Tuberculosis Control Program. [ABSTRACT FROM AUTHOR]

A technique for improving the performance of an OSNR monitor based on a polarisation nulling method with the downhill simplex algorithm is demonstrated. Establishing a compromise between accuracy and acquisition time, the monitor has been calibrated to 0.72 dB/390 ms and 0.98 dB/320 ms, over a range of nearly 21 dB. The objective of the study was to determine whether Parkia platycephala families with superior morphophysiological traits perform consistently across four stages of early development (plantlets, seedlings, and juveniles at 34 and 60 months), as a means to assess the possibility of conducting preselection. For 21 morphophysiological traits, the following genetic parameters were estimated: heritability, coefficients of variation (genetic, relative, and environmental), accuracy, genetic and phenotypic correlation, percentage of coincidence (PC) of the best families, effective population size, and inbreeding in three progeny tests conducted under laboratory, nursery, and field conditions. Families of P. platycephala showed significant phenotypic and genotypic differences. The traits evaluated show genetic control. The percentage of coincidence between the best families was more accurate when the selection intensity was greater than 50% (PC ≥ 52%). Based on the evaluated traits, a pre-selection strategy at the juvenile stage for P. platycephala was researched to effectively identify superior families. [ABSTRACT FROM AUTHOR]

This study aimed to determine the most appropriate conditions of temperature and substrate for germination and performance of A. niopoides seedlings. The design was completely randomized in 10x7 factorial arrangement (eight constant temperatures of 5, 10, 15, 20, 25, 30, 35, and 40°C and two alternating temperatures of 20-30 and 25-35°C, and seven substrates, that are paper towel, vermiculite, sand, blotting paper, coir dust, sugarcane bagasse, and tropstrato®) with four replicates of 25 seeds. The following variables were analyzed: (1) germination percentage; (2) germination speed index; (3) shoot length and primary root; and (4) dry weight of shoot and of root system. There was a significant interaction among the tested factors for all variables. The A. niopoides seeds germinate under a wide temperature range, with minimum and maximum limit at 10 and 40°C temperatures, respectively. Alternating temperatures of 20-30 and 25-35°C with sand substrate are ideal conditions for germination and performance of A. niopoides seedlings. [ABSTRACT FROM AUTHOR]

Objective: to identify and analyze the available evidence on the strategies used in the studies evaluating health interventions at school. Method: this is an integrative review searching in LILACS, CINAHL, CUIDEN, ScienceDirect, and PubMed. From the pre-defined inclusion and exclusion criteria, there were 121 articles chosen to compose the sample. Results: english studies (97.5%), with a quantitative approach (80.2%), related to the interventions carried out in the Region of the Americas (54.6%) and the European Region (23.1%) predominated. For the most part, they are interventions as programs (70.2%), interested in evaluating results (73.5%) from the value judgment (83.4%). Prevalence of interventions focused on efficacy, effects or impact, and activities carried out on interventions were focused on physical activity, healthy eating, sexual and reproductive health, mental health, and use of tobacco, alcohol, and other drugs. They are worked through activities of clinical monitoring, health promotion and disease prevention. Conclusion: the evidence indicates that the evaluations of health interventions in the school focus the results produced in programs through the judgment of value. The Coronary artery disease (CAD) is considered the first cause of death in the world. In Brazil, by the year of 1998, CAD's were responsible for 27,4% of death known cases. Scientific researches have demonstrated a possible relation between anthropometric indicators: Body mass index (BMI), waist to hip circumference (WRC) and waist circumference (WC) with CAD. In Manaus (AM), however, there is no data that can be associated with CAD. As a result, this study aimed to verify anthropometric characteristics; Body mass index (BMI), waist to hip ratio (WRC) and waist circumference (WC) of patients with coronary artery disease (CAD). The survey has been developed at the Amazonas Heart Institute, with 30 male patients, from 26 to 50 years old, who presented CAD. The results obtained are stated as follows; BMI (32,6±5,7) was classified as obesity level 1, WRC (0,97±0,06) presented a level beyond the desired (RCQ<0,95). Regarding CC, the value obtained was (107,5±15,0), classified as action level 2 (>102), thus associated with cardiovascular high risk level. As a result, the anthropometric indicators BMI, WRC and WC, from the studied subjects, were all beyond the expected average considering health parameters. [ABSTRACT FROM AUTHOR]

FDA approved agents for the treatment of chronic GVHD including ruxolitinib and belumosudil are effective steroid-sparing agents, with overall response rates (ORR) of 76% and 65% respectively. Ruxolitinib and belumosudil are well tolerated with different primary targets. Little data is available on the use of combination ruxolitinib and belumosudil. This is a single center, retrospective analysis of 20 treatment-refractory patients with chronic GVHD treated with combination ruxolitinib and belumosudil. The ORR including complete response (CR) and partial response (PR) at any time was 55% (11/20). Among responding patients, other immunosuppressive agents were tapered or discontinued in all patients. None of the patients developed EBV or CMV reactivation requiring treatment. 4 patients (20%) developed pneumonia and 2 patients (10%) developed viral URI. Cytopenias were not exacerbated. No patients had graft failure or relapsed disease. The combination is tolerable, delays the need for alternative therapies, and facilitates tapering of corticosteroids.

Objectives: The control chart is a classic statistical technique in epidemiology for identifying trends, patterns, or alerts. One meaningful use is monitoring and tracking Infant Mortality Rates, which is a priority both domestically and for the World Health Organization, as it reflects the effectiveness of public policies and the progress of nations. This study aims to evaluate the applicability and performance of this technique in Brazilian cities with different population sizes using infant mortality data., Results: In this article, we evaluate the effectiveness of the statistical process control chart in the context of Brazilian cities. We present three categories of city groups, divided based on population size and classified according to the quality of the analyses when subjected to the control method: consistent, interpretable, and inconsistent. In cities with a large population, the data in these contexts show a lower noise level and reliable results. However, in intermediate and small-sized cities, the technique becomes limited in detecting deviations from expected behaviors, resulting in reduced reliability of the generated patterns and alerts., (© 2024. Chimeric antigen receptor T-cell (CAR-T) therapy has significantly impacted treatment algorithms and clinical outcomes for a variety of patients with hematologic malignancies over the past decade. The field of cellular immunotherapy is currently experiencing a rapid expansion of the number of patients eligible for CAR-T therapies as approvals are being seen in earlier lines of therapy. With the expanded patients eligible for these therapies, more treatment centers will be necessary to keep up with demand. High-mountain water bodies represent critical components of their ecosystems, serving as vital freshwater reservoirs, environmental regulators, and sentinels of climate change. To understand the environmental dynamics of these regions, comprehensive analyses of lakes across spatial and temporal scales are necessary. While remote sensing offers a powerful tool for lake monitoring, applications in high-mountain terrain present unique challenges. The Ancash and Cuzco regions of the Peruvian Andes exemplify these challenges. These regions harbor numerous high-mountain lakes, which are crucial for fresh water supply and environmental regulation. This paper presents an exploratory examination of remote sensing techniques for lake monitoring in the Ancash and Cuzco regions of the Peruvian Andes. The study compares three deep learning models for lake segmentation: the well-established DeepWaterMapV2 and WatNet models and the adapted WaterSegDiff model, which is based on a combination of diffusion and transformation mechanisms specifically conditioned for lake segmentation. In addition, the Normalized Difference Water Index (NDWI) with Otsu thresholding is used for comparison purposes. To capture lakes across these regions, a new dataset was created with Landsat-8 multispectral imagery (bands 2-7) from 2013 to 2023. Quantitative and qualitative analyses were performed using metrics such as Mean Intersection over Union (MIoU), Pixel Accuracy (PA), and F1 Score. The results achieved indicate equivalent performance of DeepWaterMapV2 and WatNet encoder-decoder architectures, achieving adequate lake segmentation despite the challenging geographical and atmospheric conditions inherent in high-mountain environments. In the qualitative analysis, the behavior of the WaterSegDiff model was considered promising for the proposed application. Considering that WatNet is less computationally complex, with 3.4 million parameters, this architecture becomes the most pertinent to implement. Additionally, a detailed temporal analysis of Lake Singrenacocha in the Vilcanota Mountains was conducted, pointing out the more significant behavior of the WatNet model.

Abstract: Chimeric antigen receptor (CAR) T-cell therapy has revolutionized treatment for relapsed/refractory B-cell non-Hodgkin lymphoma (NHL). Robust biomarkers and a complete understanding of CAR T-cell function in the postinfusion phase remain limited. Here, we used a 37-color spectral flow cytometry panel to perform high dimensional single-cell analysis of postinfusion samples in 26 patients treated with CD28 costimulatory domain containing commercial CAR T cells for NHL and focused on computationally gated CD8+ CAR T cells. We found that the presence of postinfusion Programmed cell death protein 1 (PD-1)+ CD8+ CAR T cells at the day 14 time point highly correlated with the ability to achieve complete response (CR) by 6 months. Further analysis identified multiple subtypes of CD8+ PD-1+ CAR T cells, including PD-1+ T cell factor 1 (TCF1)+ stem-like CAR T cells and PD-1+ T-cell immunoglobulin and mucin-domain containing-3 (TIM3)+ effector-like CAR T cells that correlated with improved clinical outcomes such as response and progression-free survival. Additionally, we identified a subset of PD-1+ CD8+ CAR+ T cells with effector-like function that was increased in patients who achieved a CR and was associated with grade 3 or higher immune effector cell-associated neurotoxicity syndrome.
