This study successfully synthesized oleic acid (OA)-coated Fe3O4 nanoparticles using the coprecipitation method for the needs of biosensor applications, as shown through the characterization of structure and morphology using XRD and TEM. This observation used 0.75 ml of OA (Fe3O4-OA(0.75)) and 1.25 ml of OA (Fe3O4-OA(1.25). A decrease in particle size distribution from 18.55 nm to 16.30 nm was observed, which means a reduction in agglomeration and increased dispersibility. Vibrating Sample Magnetometer (VSM) observations showed that the saturation magnetization of Fe3O4 particles modified with OA decreased from 47.71 emu/g to 45.90 emu/g at Fe3O4-OA(0.75) and decreased again to 42.29 emu/g at Fe3O4-OA(1.25). But they all show soft ferromagnetic properties with very low coercivity, making them suitable for Giant Magnetoresistance (GMR) biosensor applications. A series of concentrations in ethanol solution were performed to evaluate the detection sensitivity of GMR for these pure and oleic acid-modified Fe3O4 samples. Concentrations of 2, 5, 10, 15, 25, and 35 mg/ml were integrated at 2 μL each onto the surface of the GMR chip. A decrease in sensitivity was observed due to the change in saturation. Pure Fe3O4 with a sensitivity of 5.07 mV (mg/ml) decreased to 4.49 mV (mg/ml) and 3.35 mV (mg/ml) after oleic acid coating. Thus, although adding a proportional amount of oleic acid can slightly decrease the saturation magnetization, it can be a valuable method for applications such as GMR biosensors that demand high dispersibility and sensitivity that remains strong.

This research explores the capacity of emerging technologies to enhance well-being. It involves the generation of 2D biophilically-driven geometries to represent human-response-oriented built environments and conducts inter and intra-individual analyses to assess human responses using a range of technologies within the realms of facial micro-expression analysis and EEG biosensor use. The outcomes of this analysis allow for the grading of these geometries in terms of emotional valences, meditation levels, and subjective preferences. These graded geometries can subsequently be employed in specific architectural contexts, such as interior decor, wallpapers, furniture surfaces, or other architectural and interior components. It is an interdisciplinary effort that underscores the importance of incorporating emerging technological means with human-response-oriented design approaches to foster built environments that promote well-being.

Abstract Objectives In the current study, for the first time, we reported a novel HCV molecular diagnostic approach termed reverse transcription loop-mediated isothermal amplification integrated with a gold nanoparticles-based lateral flow biosensor (RT-LAMP-AuNPs-LFB), which we developed for rapid, sensitive, specific, simple, and visual identification of HCV. Methods A set of LAMP primer was designed according to 5’untranslated region (5’UTR) gene from the major HCV genotypes 1b, 2a, 3b, 6a, and 3a, which are prevalent in China. The HCV-RT-LAMP-AuNPs-LFB assay conditions, including HCV-RT-LAMP reaction temperature and time were optimized. The sensitivity, specificity, and selectivity of our assay were evaluated in the current study. The feasibility of HCV-RT-LAMP-AuNPs-LFB was confirmed through clinical serum samples from patients with suspected HCV infections. Results An unique set of HCV-RT-LAMP primers were successfully designed targeting on the 5’UTR gene. The optimal detection process, including crude nucleic acid extraction (approximately 5 min), RT-LAMP reaction (67℃, 30 min), and visual interpretation of AuNPs-LFB results (~ 2 min), could be performed within 40 min without specific instruments. The limit of detection was determined to be 20 copies per test. The HCV-RT-LAMP-AuNPs-LFB assay exhibited high specificity and anti-interference. Conclusions These preliminary results confirmed that the HCV-RT-LAMP-AuNPs-LFB assay is a sensitive, specific, rapid, visual, and cost-saving assay for identification of HCV. This diagnostic approach has great potential value for point-of-care (POC) diagnostic of HCV, especially in resource-challenged regions.

Abstract Background S. pyogenes, is a primary pathogen that leads to pharyngitis and can also trigger severe conditions like necrotizing fasciitis and streptococcal toxic shock syndrome (STSS), often resulting in high mortality rates. Therefore, prompt identification and appropriate treatment of S. pyogenes infections are crucial in preventing the worsening of symptoms and alleviating the disease's impact. Results In this study, a newly developed technique called multiple cross displacement amplification (MCDA) was employed to detect S. pyogenes,specifically targeting the speB gene, at a temperature of 63°C within 30 min. Then, an easily portable and user-friendly nanoparticles-based lateral flow biosensor (LFB) assay was introduced for the rapid analysis of MCDA products in just 2 min. The results indicated that the LFB offers greater objectivity compared to Malachite Green and is simpler than electrophoresis. The MCDA-LFB assay boasts a low detection limit of 200 fg and exhibits no cross-reaction with non-S. pyogenes strains. Among 230 clinical swab throat samples, the MCDA-LFB method identified 27 specimens as positive, demonstrating higher sensitivity compared to 23 samples detected positive by qPCR assay and 18 samples by culture. The only equipment needed for this assay is a portable dry block heater. Moreover, each MCDA-LFB test is cost-effective, priced at approximately $US 5.5. Conclusion The MCDA-LFB assay emerges as a straightforward, specific, sensitive, portable, and user-friendly method for the rapid diagnosis of S. pyogenes in clinical samples.

Abstract Volatile organic compounds (VOCs) have grown due to their crucial role in transitioning from invasive to noninvasive cancer diagnostic methods. This study aimed to assess the feasibility of the metal oxide biosensor platform using urine VOCs for detecting genitourinary cancers. Five different commercially available semiconductor sensors were chosen to detect specific VOCs (methane, iso-butane, hydrogen, ethanol, hydrogen sulfide, ammonia, toluene, butane, propane, trimethylamine, and methyl-mercaptan). Changes in electrical resistance due to temperature variations from the voltage heater were examined to characterize VOC metabolism. Logistic regression and ROC analysis were employed to evaluate potential urine VOCs for genitourinary cancer determination. This study involved 64 participants which were categorized into a cancer and a non-cancer group. The genitourinary cancer (confirmed by tissue pathology) comprised 32 patients, including renal cell carcinoma (3.1%), transitional cell carcinoma (46.9%), and prostate cancer (50%). The non-cancer comprised 32 patients, with 9 healthy subjects and 23 individuals with other genitourinary diseases. Results indicated that VOC sensors for methane, iso-butane, hydrogen, and ethanol, at a voltage heater of 2000 mV, demonstrated a significant predictive capability for genitourinary cancer with P = 0.013. Abstract Early diagnosis of cancer demands sensitive and accurate detection of cancer biomarkers in blood. Carbon dots (CDs) bio-functionalization with antibodies, peptides or aptamers have played significant role in cancer diagnosis and targeted cancer therapy. Herein, a biosensor for detection of cancer biomarker carcinoembryonic antigen (CEA) in blood serum has been designed using CDs bio-functionalized with HRP-conjugated CEA antibody (CUCDs@CEAAb2) as detection probe. CDs were synthesized by upscaling of cow urine, a nitrogen rich biomass waste, by hydrothermal method. Detection probe based on CDs resulted in 3.5 times higher sensitivity as compared to conventional electrochemical sandwich immunoassay. To further improve the sensor performance, hyper-branched polyethylenimine grafted poly amino aniline (PEI-g-PAANI) was used as the sensing interface, which enabled immobilization of higher amount of capture antibody. Detection of CEA in human blood serum coupled with wide linear range (0.5–50 ng/ml), good specificity, stability, reproducibility and low detection limit (10 pg/ml) signified the excellence of CUCDs based CEA immunosensor. CUCDs exhibited excitation wavelength dependent fluorescence property and showed strong blue emission under UV irradiation. MTT assay indicated that the material is not toxic towards human dental pulp stem cells (hDPSCs) and MG63 osteosarcoma cells (cell viability > 90%). The present study demonstrates a methodology for valorization of animal waste to a cost-effective carbon based functional nanomaterial for clinical detection of cancer biomarkers.

In this study, we developed a novel electrochemical biosensor for detecting IL-6 that uses a nano-electrode array (NEA) fabricated via standard CMOS processing. Miniaturizing the electrodes to the nanoscale and arranging them in an array to form an NEA facilitated the creation of a higher electric field magnitude, compared to that available via the use of microelectrodes, that could be used to improve biosensor sensitivity. Additionally, the array configuration of the NEA aided in providing sufficient reaction sites. Each nano-electrode in the NEA was cylindrically shaped, with a radius of 0.1 µm, and a top layer formed by TiN physical vapor deposition. Each NEA biosensor was divided into four independent banks, with each bank including a set of WE, CE and RE. These banks were capable of independently inputting and outputting electrical signals. This design allowed the NEA biosensor to undergo selective modification by CV input. In this study, we discuss and address material and contamination issues associated with CMOS-produced NEAs and their uses as biosensors. To ameliorate these issues, we stored materials and products in a nitrogen-controlled cabinet and conducted pretreatment cleaning on the electrodes. Both steps had a noticeable impact on the cleanliness of the electrode surfaces. These optimized conditions resulted in a remarkable 96.6 % reduction in Rct. The NEA surface was functionalized by electrochemically grafting diazonium salts subsequently immobilized with anti-IL-6 antibodies via EDC/NHS chemistry. The resulting NEA biosensor demonstrated sufficient sensitivity to rapidly distinguish inflammatory conditions and disease severity. This showcases the potential for using NEA devices mass-produced via standard CMOS processing as electrodes for biosensors.

With rapid advances in the development of metabolic pathways and synthetic biology toolkits, a persisting challenge in microbial bioproduction is how to optimally rewire metabolic fluxes and accelerate the concomitant high-throughput phenotype screening. Here we developed a biosensor-assisted titratable CRISPRi high-throughput (BATCH) screening approach that combines a titratable mismatch CRISPR interference and a biosensor mediated screening for high-production phenotypes in Escherichia coli. We first developed a programmable mismatch CRISPRi that could afford multiple levels of interference efficacy with a one-pot sgRNA pool (a total of 16 variants for each target gene) harboring two consecutive random mismatches in the seed region of sgRNA spacers. The mismatch CRISPRi was demonstrated to enable almost a full range of gene knockdown when targeting different positions on genes. As a proof-of-principle demonstration of the BATCH screening system, we designed doubly mismatched sgRNA pools targeting 20 relevant genes in E. coli and optimized a PadR-based p-coumaric acid biosensor with broad dynamic range for the eGFP fluorescence guided high-production screening. Using sgRNA variants for the combinatorial knockdown of pfkA and ptsI, the p-coumaric acid titer was increased by 40.6% to o 1308.6 mg/l from glycerol in shake flasks. To further demonstrate the general applicability of the BATCH screening system, we recruited a HpdR-based butyrate biosensor that facilitated the screening of E. coli strains achieving 19.0% and 25.2% increase of butyrate titer in shake flasks with sgRNA variants targeting sucA and ldhA, respectively. This work reported the establishment of a plug-and-play approach that enables multilevel modulation of metabolic fluxes and high-throughput screening of high-production phenotypes.
