251. MedISure: Towards Assuring Machine Learning-based Medical Image Classifiers using Mixup Boundary Analysis

Abstract: Machine learning (ML) models are becoming integral in healthcare technologies, presenting a critical need for formal assurance to validate their safety, fairness, robustness, and trustworthiness. These models are inherently prone to errors, potentially posing serious risks to patient health and could even cause irreparable harm. Traditional software assurance techniques rely on fixed code and do not directly apply to ML models since these algorithms are adaptable and learn from curated datasets through a training process. However, adapting established principles, such as boundary testing using synthetic test data can effectively bridge this gap. To this end, we present a novel technique called Mix-Up Boundary Analysis (MUBA) that facilitates evaluating image classifiers in terms of prediction fairness. We evaluated MUBA for two important medical imaging tasks -- brain tumour classification and breast cancer classification -- and achieved promising results. This research aims to showcase the importance of adapting traditional assurance principles for assessing ML models to enhance the safety and reliability of healthcare technologies. To facilitate future research, we plan to publicly release our code for MUBA.

252. Diffusion Model Alignment Using Direct Preference Optimization

Abstract: Large language models (LLMs) are fine-tuned using human comparison data with Reinforcement Learning from Human Feedback (RLHF) methods to make them better aligned with users' preferences. In contrast to LLMs, human preference learning has not been widely explored in text-to-image diffusion models; the best existing approach is to fine-tune a pretrained model using carefully curated high quality images and captions to improve visual appeal and text alignment. We propose Diffusion-DPO, a method to align diffusion models to human preferences by directly optimizing on human comparison data. Diffusion-DPO is adapted from the recently developed Direct Preference Optimization (DPO), a simpler alternative to RLHF which directly optimizes a policy that best satisfies human preferences under a classification objective. We re-formulate DPO to account for a diffusion model notion of likelihood, utilizing the evidence lower bound to derive a differentiable objective. Using the Pick-a-Pic dataset of 851K crowdsourced pairwise preferences, we fine-tune the base model of the state-of-the-art Stable Diffusion XL (SDXL)-1.0 model with Diffusion-DPO. Our fine-tuned base model significantly outperforms both base SDXL-1.0 and the larger SDXL-1.0 model consisting of an additional refinement model in human evaluation, improving visual appeal and prompt alignment. We also develop a variant that uses AI feedback and has comparable performance to training on human preferences, opening the door for scaling of diffusion model alignment methods.

Recent advancements in generative AI, exemplified by large language models, hold promise in facilitating the arduous task. However, developing accountable, fair, and inclusive models remains a complicated undertaking. In this perspective, we discuss the trustworthiness of generative AI in the context of automated summarization of medical evidence.

Abstract: Dietary intake data are routinely drawn upon to explore diet-health relationships. However, these data are often subject to measurement error, distorting the true relationships. Beyond measurement error, there are likely complex synergistic and sometimes antagonistic interactions between different dietary components, complicating the relationships between diet and health outcomes. Flexible models are required to capture the nuance that these complex interactions introduce. This complexity makes research on diet-health relationships an appealing candidate for the application of machine learning techniques, and in particular, neural networks. Neural networks are computational models that are able to capture highly complex, nonlinear relationships so long as sufficient data are available. While these models have been applied in many domains, the impacts of measurement error on the performance of predictive modeling has not been systematically investigated. However, dietary intake data are typically collected using self-report methods and are prone to large amounts of measurement error. In this work, we demonstrate the ways in which measurement error erodes the performance of neural networks, and illustrate the care that is required for leveraging these models in the presence of error. We demonstrate the role that sample size and replicate measurements play on model performance, indicate a motivation for the investigation of transformations to additivity, and illustrate the caution required to prevent model overfitting. While the past performance of neural networks across various domains make them an attractive candidate for examining diet-health relationships, our work demonstrates that substantial care and further methodological development are both required to observe increased predictive performance when applying these techniques, compared to more traditional statistical procedures.

255. Rethinking Thorne-\.Zytkow Object Formation: The Fate of X-ray Binary LMC X-4 and Implications for Ultra-long Gamma-ray Bursts

Abstract: We present a start-to-end simulation aimed at studying the long-term fate of high mass X-ray binaries and whether a Thorne-\.Zytkow object (T\.ZO) might ultimately be produced. We analyze results from a 3D hydrodynamical simulation that models the eventual fate of LMC X-4, a compact high mass X-ray binary system, after the primary fills its Roche lobe and engulfs the neutron star companion. We discuss the outcome of this engulfment within the standard paradigm of T\.ZO formation. The post-merger angular momentum content of the stellar core is a key ingredient, as even a small amount of rotation can break spherical symmetry and produce a centrifugally supported accretion disk. Our findings suggest the inspiraling neutron star, upon merging with the core, can accrete efficiently via a disk at high rates ($\approx 10^{-2}M_\odot/{\rm s}$), subsequently collapsing into a black hole and triggering a bright transient with a luminosity and duration typical of an ultra-long gamma-ray burst. We propose that the canonical framework for T\.ZO formation via common envelope needs to be revised, as the significant post-merger accretion feedback will unavoidably unbind the vast majority of the surrounding envelope.

256. Future Lens: Anticipating Subsequent Tokens from a Single Hidden State

Abstract: We conjecture that hidden state vectors corresponding to individual input tokens encode information sufficient to accurately predict several tokens ahead. More concretely, in this paper we ask: Given a hidden (internal) representation of a single token at position $t$ in an input, can we reliably anticipate the tokens that will appear at positions $\geq t + 2$? To test this, we measure linear approximation and causal intervention methods in GPT-J-6B to evaluate the degree to which individual hidden states in the network contain signal rich enough to predict future hidden states and, ultimately, token outputs. We find that, at some layers, we can approximate a model's output with more than 48% accuracy with respect to its prediction of subsequent tokens through a single hidden state. 257. Engineering Waveguide Nonlinear Effective Length via Low Index Thin Films

Abstract: Novel photonic nanowires were fabricated using low-index materials and tested in the near-infrared spectrum to assess their nonlinear optical properties. In this work, we argue the need to redefine the standard nonlinear figure of merit in terms of nonlinear phase shift and optical transmission for a given propagation distance. According to this new metric, our devices largely outperform all established platforms for devices with a linear footprint in the range of 50 to 500 um, which is demonstrated to be an outstanding technological gap. For 85 fs pulses, with carrier wavelength at 1480nm and sub-uW power levels, a spectral broadening exceeding 80% of the initial bandwidth was recorded over a propagation length of just 50 um. Leveraging on CMOS-compatible processes and well-established materials such as silicon, silica, and indium tin oxide, our devices bring great promise for developing alternative all-optical devices with unparalleled nonlinear performances within the aforementioned range.

258. Function Vectors in Large Language Models

Abstract: We report the presence of a simple neural mechanism that represents an input-output function as a vector within autoregressive transformer language models (LMs). Using causal mediation analysis on a diverse range of in-context-learning (ICL) tasks, we find that a small number attention heads transport a compact representation of the demonstrated task, which we call a function vector (FV). FVs are robust to changes in context, i.e., they trigger execution of the task on inputs such as zero-shot and natural text settings that do not resemble the ICL contexts from which they are collected. We test FVs across a range of tasks, models, and layers and find strong causal effects across settings in middle layers. We investigate the internal structure of FVs and find while that they often contain information that encodes the output space of the function, this information alone is not sufficient to reconstruct an FV. Finally, we test semantic vector composition in FVs, and find that to some extent they can be summed to create vectors that trigger new complex tasks. Abstract: Thorne-\.{Z}ytkow objects (T\.{Z}Os), hypothetical merger products in which a neutron star is embedded in a stellar core, are traditionally considered steady-state configurations. Their assembly, especially through dynamical channels, is not well-understood. The predominant focus in the literature has been on the observational signatures related to the evolution and long-term fate of T\.{Z}Os, with their initial formation often treated as a given. However, the foundational calculations supporting the existence of T\.{Z}Os assume non-rotating spherically-symmetric initial conditions that we find to be inconsistent with a binary merger scenario. In this work, we explore the implications of post-merger dynamics in T\.{Z}O formation scenarios with field binary progenitors, specifically the role that angular momentum transport during the common envelope phase plays in constraining possible merger products, using the tools of stellar evolution and three-dimensional hydrodynamics. We also propose an alternative steady-state outcome for these mergers: the thin-envelope T\.{Z}O, an equilibrium solution consisting of a low-mass spherical envelope supported by the accretion disk luminosity of a central stellar-mass black hole. 260. Modulation Instability and Wavenumber Bandgap Breathers in a Time Layered Phononic Lattice

Abstract: We demonstrate the existence of wavenumber bandgap (q-gap) breathers in a time-periodic phononic lattice. These breathers are localized in time and periodic in space, and are the counterparts to the classical breathers found in spatially-periodic systems. We derive an exact condition for modulation instability that leads to the opening of wavenumber bandgaps. The q-gap breathers become more narrow and larger in amplitude as the wavenumber goes further into the bandgap. In the presence of damping, these structures acquire a non-zero, oscillating tail. 261. Model Predictive Planning: Trajectory Planning in Obstruction-Dense Environments for Low-Agility Aircraft

Abstract: We present Model Predictive Planning (MPP), a trajectory planner for low-agility vehicles such as a fixed-wing aircraft to navigate obstacle-laden environments. MPP consists of (1) a multi-path planning procedure that identifies candidate paths, (2) a raytracing procedure that generates linear constraints around these paths to enforce obstacle avoidance, and (3) a convex quadratic program that finds a feasible trajectory within these constraints if one exists. Low-agility aircraft cannot track arbitrary paths, so refining a given path into a trajectory that respects the vehicle's limited maneuverability and avoids obstacles often leads to an infeasible optimization problem. The critical feature of MPP is that it efficiently considers multiple candidate paths during the refinement process, thereby greatly increasing the chance of finding a feasible and trackable trajectory. We demonstrate the effectiveness of MPP on a longitudinal aircraft model.

262. Multimode physics of the unimon circuit

Abstract: We consider a superconducting half-wavelength resonator that is grounded at its both ends and contains a single Josephson junction. Previously this circuit was considered as a unimon qubit in the single-mode approximation where dc-phase-biasing the junction to $\pi$ leads to increased anharmonicity and 99.9% experimentally observed single-qubit gate fidelity. Inspired by the promising first experimental results, we develop here a theoretical and numerical model for the detailed understanding of the multimode physics of the unimon circuit. To this end, first, we consider the high-frequency modes of the unimon circuit and find that even though these modes are at their ground state, they imply a significant renormalization to the Josephson energy. We introduce an efficient method how the relevant modes can be fully taken into account and show that unexcited high-lying modes lead to corrections in the qubit energy and anharmonicity. Interestingly, provided that the junction is offset from the middle of the circuit, we find strong cross-Kerr coupling strengths between a few low-lying modes. This observation paves the way for the utilization of the multimode structure, for example, as several qubits embedded into a single unimon circuit.

263. Characteristics of Gravity Waves in Opposing Phases of the QBO: A Reanalysis Perspective with ERA5

Abstract: ERA5 data for the period of 1979-2019 are used as a basis for investigating the properties of gravity waves as they disperse and propagate upward through the stratosphere during opposing phases of the QBO. Two-sided zonal wavenumber-frequency spectra of vertical velocity in the stratosphere exhibit distinctive gravity wave signatures. Consistent with theory, westward propagating waves tend to be suppressed during the easterly QBO phase and eastward propagating waves tend to be suppressed during the westerly phase. Cospectra of the vertical flux of zonal momentum also show significant asymmetries between eastward and westward propagating waves during opposing QBO phases. Phase speed spectra of the vertical flux of momentum are indicative of a strong dissipation of westward propagating gravity waves during the easterly phase and vice versa; i.e., a selective "wind filtering" of the waves as they propagate upward. The three-dimensional structure of the gravity waves is revealed by compositing. In the absence of a background zonal flow, the waves radiate outward and upward from their respective reference grid points in concentric rings. When a zonal flow is present, the rings are amplified and compressed upstream of the source and attenuated and stretched downstream of it, such that they assume the form of arcs. These results serve to confirm the applicability of the mechanism proposed by Lindzen and Holton (1968) to explain the downward propagation of the QBO. The QBO also influences the spectrum of gravity waves in ERA5 at 100 hPa, below the layer in which wind-filtering occurs.

264. Ensemble Forecasts of Solar Wind Connectivity to 1 Rs using ADAPT-WSA

Abstract: The solar wind which arrives at any location in the solar system is, in principle, relatable to the outflow of solar plasma from a single source location. This source location, itself usually being part of a larger coronal hole, is traceable to 1 Rs along the Sun's magnetic field, in which the entire path from 1 Rs to a location in the heliosphere is referred to as the solar wind connectivity. While not directly measurable, the connectivity between the near-Earth solar wind is of particular importance to space weather. The solar wind solar source region can be obtained by leveraging near-sun magnetic field models and a model of the interplanetary solar wind. In this article we present a method for making an ensemble forecast of the connectivity presented as a probability distribution obtained from a weighted collection of individual forecasts from the combined Air Force Data Assimilative Photospheric Flux Transport - Wang Sheeley Arge (ADAPT-WSA) model. The ADAPT model derives the photospheric magnetic field from synchronic magnetogram data, using flux transport physics and ongoing data assimilation processes. The WSA model uses a coupled set of potential field type models to derive the coronal magnetic field, and an empirical relationship to derive the terminal solar wind speed observed at Earth. Our method produces an arbitrary 2D probability distribution capable of reflecting complex source configurations with minimal assumptions about the distribution structure, prepared in a computationally efficient manner., Comment: Accepted to the journal "Space Weather"

To make progress, astronomers studying exoplanets will need new and innovative technical solutions. Abstract: Vortex fiber nulling (VFN) is a single-aperture interferometric technique for detecting and characterizing exoplanets separated from their host star by less than a diffracted beam width. VFN uses a vortex mask and single mode fiber to selectively reject starlight while coupling off-axis planet light with a simple optical design that can be readily implemented on existing direct imaging instruments that can feed light to an optical fiber. With its axially symmetric coupling region peaking within the inner working angle of conventional coronagraphs, VFN is more efficient at detecting new companions at small separations than conventional direct imaging, thereby increasing the yield of on-going exoplanet search campaigns. We deployed a VFN mode operating in K band ($2.0{-}2.5~\mu$m) on the Keck Planet Imager and Characterizer (KPIC) instrument at the Keck II Telescope. In this paper we present the instrument design of this first on-sky demonstration of VFN and the results from on-sky commissioning, including planet and star throughput measurements and predicted flux-ratio detection limits for close-in companions. The instrument performance is shown to be sufficient for detecting a companion $10^3$ times fainter than a $5^{\mathrm{th}}$ magnitude host star in 1 hour at a separation of 50 mas (1.1$\lambda/D$). This makes the instrument capable of efficiently detecting substellar companions around young stars. 267. Radiomics Boosts Deep Learning Model for IPMN Classification

Abstract: Intraductal Papillary Mucinous Neoplasm (IPMN) cysts are pre-malignant pancreas lesions, and they can progress into pancreatic cancer. Therefore, detecting and stratifying their risk level is of ultimate importance for effective treatment planning and disease control. However, this is a highly challenging task because of the diverse and irregular shape, texture, and size of the IPMN cysts as well as the pancreas. In this study, we propose a novel computer-aided diagnosis pipeline for IPMN risk classification from multi-contrast MRI scans. Our proposed analysis framework includes an efficient volumetric self-adapting segmentation strategy for pancreas delineation, followed by a newly designed deep learning-based classification scheme with a radiomics-based predictive approach. We test our proposed decision-fusion model in multi-center data sets of 246 multi-contrast MRI scans and obtain superior performance to the state of the art (SOTA) in this field. Our ablation studies demonstrate the significance of both radiomics and deep learning modules for achieving the new SOTA performance compared to international guidelines and published studies (81.9\% vs 61.3\% in accuracy). Our findings have important implications for clinical decision-making. Abstract: Most current approaches for protecting privacy in machine learning (ML) assume that models exist in a vacuum. Yet, in reality, these models are part of larger systems that include components for training data filtering, output monitoring, and more. In this work, we introduce privacy side channels: attacks that exploit these system-level components to extract private information at far higher rates than is otherwise possible for standalone models. We propose four categories of side channels that span the entire ML lifecycle (training data filtering, input preprocessing, output post-processing, and query filtering) and allow for enhanced membership inference, data extraction, and even novel threats such as extraction of users' test queries. For example, we show that deduplicating training data before applying differentially-private training creates a side-channel that completely invalidates any provable privacy guarantees. We further show that systems which block language models from regenerating training data can be exploited to exfiltrate private keys contained in the training set--even if the model did not memorize these keys. 269. Retrieving Evidence from EHRs with LLMs: Possibilities and Challenges

Abstract: Unstructured data in Electronic Health Records (EHRs) often contains critical information -- complementary to imaging -- that could inform radiologists' diagnoses. But the large volume of notes often associated with patients together with time constraints renders manually identifying relevant evidence practically infeasible. In this work we propose and evaluate a zero-shot strategy for using LLMs as a mechanism to efficiently retrieve and summarize unstructured evidence in patient EHR relevant to a given query. Our method entails tasking an LLM to infer whether a patient has, or is at risk of, a particular condition on the basis of associated notes; if so, we ask the model to summarize the supporting evidence. Under expert evaluation, we find that this LLM-based approach provides outputs consistently preferred to a pre-LLM information retrieval baseline. Manual evaluation is expensive, so we also propose and validate a method using an LLM to evaluate (other) LLM outputs for this task, allowing us to scale up evaluation. Our findings indicate the promise of LLMs as interfaces to EHR, but also highlight the outstanding challenge posed by "hallucinations". In this setting, however, we show that model confidence in outputs strongly correlates with faithful summaries, offering a practical means to limit confabulations.

Abstract: The FOR-instance dataset (available at https://doi.org/10.5281/zenodo.8287792) addresses the challenge of accurate individual tree segmentation from laser scanning data, crucial for understanding forest ecosystems and sustainable management. Despite the growing need for detailed tree data, automating segmentation and tracking scientific progress remains difficult. Existing methodologies often overfit small datasets and lack comparability, limiting their applicability. Amid the progress triggered by the emergence of deep learning methodologies, standardized benchmarking assumes paramount importance in these research domains. This data paper introduces a benchmarking dataset for dense airborne laser scanning data, aimed at advancing instance and semantic segmentation techniques and promoting progress in 3D forest scene segmentation. The FOR-instance dataset comprises five curated and ML-ready UAV-based laser scanning data collections from diverse global locations, representing various forest types. The laser scanning data were manually annotated into individual trees (instances) and different semantic classes (e.g. stem, woody branches, live branches, terrain, low vegetation). The dataset is divided into development and test subsets, enabling method advancement and evaluation, with specific guidelines for utilization. It supports instance and semantic segmentation, offering adaptability to deep learning frameworks and diverse segmentation strategies, while the inclusion of diameter at breast height data expands its utility to the measurement of a classic tree variable. In conclusion, the FOR-instance dataset contributes to filling a gap in the 3D forest research, enhancing the development and benchmarking of segmentation algorithms for dense airborne laser scanning data.

Abstract: Traditional histochemical staining of post-mortem samples often confronts inferior staining quality due to autolysis caused by delayed fixation of cadaver tissue, and such chemical staining procedures covering large tissue areas demand substantial labor, cost and time. Here, we demonstrate virtual staining of autopsy tissue using a trained neural network to rapidly transform autofluorescence images of label-free autopsy tissue sections into brightfield equivalent images, matching hematoxylin and eosin (H&E) stained versions of the same samples. The trained model can effectively accentuate nuclear, cytoplasmic and extracellular features in new autopsy tissue samples that experienced severe autolysis, such as COVID-19 samples never seen before, where the traditional histochemical staining fails to provide consistent staining quality. This virtual autopsy staining technique provides a rapid and resource-efficient solution to generate artifact-free H&E stains despite severe autolysis and cell death, also reducing labor, cost and infrastructure requirements associated with the standard histochemical staining.

Abstract: PURPOSE: The neutralizing peptibody trebananib prevents angiopoietin-1 and angiopoietin-2 from binding with Tie2 receptors, inhibiting angiogenesis and proliferation. Trebananib was combined with paclitaxel±trastuzumab in the I-SPY2 breast cancer trial. PATIENTS AND METHODS: I-SPY2, a phase II neoadjuvant trial, adaptively randomizes patients with high-risk, early-stage breast cancer to one of several experimental therapies or control based on receptor subtypes as defined by hormone receptor (HR) and HER2 status and MammaPrint risk (MP1, MP2). The primary endpoint is pathologic complete response (pCR). A therapy graduates if/when it achieves 85% Bayesian probability of success in a phase III trial within a given subtype. Patients received weekly paclitaxel (plus trastuzumab if HER2-positive) without (control) or with weekly intravenous trebananib, followed by doxorubicin/cyclophosphamide and surgery. Pathway-specific biomarkers were assessed for response prediction. RESULTS: There were 134 participants randomized to trebananib and 133 to control. Although trebananib did not graduate in any signature [phase III probabilities: Hazard ratio (HR)-negative (78%), HR-negative/HER2-positive (74%), HR-negative/HER2-negative (77%), and MP2 (79%)], it demonstrated high probability of superior pCR rates over control (92%-99%) among these subtypes. Trebananib improved 3-year event-free survival (HR 0.67), with no significant increase in adverse events. Activation levels of the Tie2 receptor and downstream signaling partners predicted trebananib response in HER2-positive disease; high expression of a CD8 T-cell gene signature predicted response in HR-negative/HER2-negative disease. CONCLUSIONS: The angiopoietin (Ang)/Tie2 axis inhibitor trebananib combined with standard neoadjuvant therapy increased estimated pCR rates across HR-negative and MP2 subtypes, with probabilities of superiority >90%. Further study of Ang/Tie2 receptor axis inhibitors in validated, biomarker-predicted sensitive subtypes is warranted.

275. Toddler milk: a scoping review of research on consumption, perceptions, and marketing practices.

Abstract: Toddler milk is an ultra-processed beverage consisting primarily of powdered milk, caloric sweeteners, and vegetable oil. Pediatric health authorities do not support the use of toddler milk, and emerging evidence suggests that toddler-milk marketing practices may mislead consumers. However, studies have not synthesized the extent of toddler-milk marketing practices or how these practices affect parents decisions about whether to serve toddler milk. We aimed to summarize the literature about toddler milk to identify what is known about: (1) parents toddler-milk purchasing and feeding behaviors, (2) toddler-milk marketing, and (3) how marketing practices influence parents beliefs and perceptions about toddler milk. Following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses extension for Scoping Reviews (PRISMA-ScR), we systematically searched 8 databases (PubMed, APA PsycINFO, Scopus, Cochrane Central, Embase, CINAHL, Communication & Mass Media Complete, and Business Source Premier). We identified 45 articles about toddler milk. Studies were conducted in 25 countries across 6 continents. Five types of findings emerged: (1) consumption and feeding behaviors, (2) demographic correlates of toddler-milk purchasing and consumption, (3) misperceptions and beliefs, (4) increased sales, and (5) increased marketing and responses to marketing. The included articles suggested that toddler-milk sales are growing rapidly worldwide. Findings also revealed that toddler-milk packages (eg, labels, branding) resemble infant formula packages and that toddler-milk marketing practices may indirectly advertise infant formula. Purchasing, serving, and consumption of toddler milk were higher in Black and Hispanic populations than in non-Hispanic White populations, and parents with higher educational attainment and income were more likely to offer toddler milk to their children. Findings suggest a need for policies to prevent cross-marketing of toddler milk and infant formula, reduce provision of toddler milk to infants and toddlers, and prevent caregivers from being misled about toddler-milk healthfulness.

Abstract: A wealth of recent research in comparative politics examines how spatial variation in historical conditions shapes modern political outcomes. In an article in the American Political Science Review, Homola, Pereira, and Tavits argue that Germans who live nearer to former Nazi concentration camps are more likely to display out-group intolerance. Clarifying the conceptual foundations of posttreatment bias and reviewing the historical record on postwar state creation in Germany, we argue that state-level differences confound the relationship between distance to camps and out-group intolerance. Using publicly available European Values Survey data and electoral results from 2017, we find no consistent evidence that distance to camps is related to contemporary values. Our findings have implications for literatures on historical persistence, causal inference with spatial data, Holocaust studies, and outgroup tolerance.

Abstract: Individual oncogenic KRAS mutants confer distinct differences in biochemical properties and signaling for reasons that are not well understood. KRAS activity is closely coupled to protein dynamics and is regulated through two interconverting conformations: state 1 (inactive, effector binding deficient) and state 2 (active, effector binding enabled). Here, we use 31P NMR to delineate the differences in state 1 and state 2 populations present in WT and common KRAS oncogenic mutants (G12C, G12D, G12V, G13D, and Q61L) bound to its natural substrate GTP or a commonly used nonhydrolyzable analog GppNHp (guanosine-5-[(β,γ)-imido] triphosphate). Our results show that GppNHp-bound proteins exhibit significant state 1 population, whereas GTP-bound KRAS is primarily (90% or more) in state 2 conformation. This observation suggests that the predominance of state 1 shown here and in other studies is related to GppNHp and is most likely nonexistent in cells. We characterize the impact of this differential conformational equilibrium of oncogenic KRAS on RAF1 kinase effector RAS-binding domain and intrinsic hydrolysis. Through a KRAS G12C drug discovery, we have identified a novel small-molecule inhibitor, BBO-8956, which is effective against both GDP- and GTP-bound KRAS G12C. We show that binding of this inhibitor significantly perturbs state 1-state 2 equilibrium and induces an inactive state 1 conformation in GTP-bound KRAS G12C. In the presence of BBO-8956, RAF1-RAS-binding domain is unable to induce a signaling competent state 2 conformation within the ternary complex, demonstrating the mechanism of action for this novel and active-conformation inhibitor.

Abstract: Achieving health equity in populations with congenital heart disease (CHD) requires recognizing existing disparities throughout the lifespan that negatively and disproportionately impact specific groups of individuals. These disparities occur at individual, institutional, or system levels and often result in increased morbidity and mortality for marginalized or racially minoritized populations (population subgroups (e.g., ethnic, racial, social, religious) with differential power compared to those deemed to hold the majority power in the population). Creating actionable strategies and solutions to address these health disparities in patients with CHD requires critically examining multilevel factors and health policies that continue to drive health inequities, including varying social determinants of health (SDOH), systemic inequities, and structural racism. In this comprehensive review article, we focus on health equity solutions and health policy considerations for minoritized and marginalized populations with CHD throughout their lifespan in the United States. We review unique challenges that these populations may face and strategies for mitigating disparities in lifelong CHD care. We assess ways to deliver culturally competent CHD care and to help lower-health-literacy populations navigate CHD care. Finally, we review system-level health policies that impact reimbursement and research funding, as well as institutional policies that impact leadership diversity and representation in the workforce.

279. Super-Resolution Imaging of Voltages in the Interior of Individual, Vital Mitochondria.

Abstract: We present super-resolution microscopy of isolated functional mitochondria, enabling real-time studies of structure and function (voltages) in response to pharmacological manipulation. Changes in mitochondrial membrane potential as a function of time and position can be imaged in different metabolic states (not possible in whole cells), created by the addition of substrates and inhibitors of the electron transport chain, enabled by the isolation of vital mitochondria. By careful analysis of structure dyes and voltage dyes (lipophilic cations), we demonstrate that most of the fluorescent signal seen from voltage dyes is due to membrane bound dyes, and develop a model for the membrane potential dependence of the fluorescence contrast for the case of super-resolution imaging, and how it relates to membrane potential. This permits direct analysis of mitochondrial structure and function (voltage) of isolated, individual mitochondria as well as submitochondrial structures in the functional, intact state, a major advance in super-resolution studies of living organelles.

Abstract: Non-neovascular or dry age-related macular degeneration (AMD) is a multi-factorial disease with degeneration of the aging retinal-pigmented epithelium (RPE). Lysosomes play a crucial role in RPE health via phagocytosis and autophagy, which are regulated by transcription factor EB/E3 (TFEB/E3). Here, we find that increased AKT2 inhibits PGC-1α to downregulate SIRT5, which we identify as an AKT2 binding partner. Crosstalk between SIRT5 and AKT2 facilitates TFEB-dependent lysosomal function in the RPE. AKT2/SIRT5/TFEB pathway inhibition in the RPE induced lysosome/autophagy signaling abnormalities, disrupted mitochondrial function and induced release of debris contributing to drusen. Accordingly, AKT2 overexpression in the RPE caused a dry AMD-like phenotype in aging Akt2 KI mice, as evident from decline in retinal function. Importantly, we show that induced pluripotent stem cell-derived RPE encoding the major risk variant associated with AMD (complement factor H; CFH Y402H) express increased AKT2, impairing TFEB/TFE3-dependent lysosomal function. Collectively, these findings suggest that targeting the AKT2/SIRT5/TFEB pathway may be an effective therapy to delay the progression of dry AMD.

Abstract: OBJECTIVE: Prenatal exposure to polycyclic aromatic hydrocarbons (PAHs) is associated with adverse birth and developmental outcomes in children. We aimed to describe prenatal PAH exposures in a large, multisite U.S. consortium. METHODS: We measured 12 mono-hydroxylated metabolites (OH-PAHs) of 7 PAHs (naphthalene, fluorene, phenanthrene, pyrene, benzo(c)phenanthrene, chrysene, benz(a)anthracene) in mid-pregnancy urine of 1,892 pregnant individuals from the ECHO PATHWAYS consortium cohorts: CANDLE (n = 988; Memphis), TIDES (n = 664; Minneapolis, Rochester, San Francisco, Seattle) and GAPPS (n = 240; Seattle and Yakima, WA). We described concentrations of 8 OH-PAHs of non-smoking participants (n = 1,695) by site, socioeconomic characteristics, and pregnancy stage (we report intraclass correlation coefficients (ICC) for n = 677 TIDES participants). RESULTS: Exposure to the selected PAHs was ubiquitous at all sites. 2-hydroxynaphthalene had the highest average concentrations at all sites. CANDLE had the highest average concentrations of most metabolites. Among non-smoking participants, we observed some patterns by income, education, and race but these were not consistent and varied by site and metabolite. ICCs of repeated OH-PAH measures from TIDES participants were ≤ 0.51. CONCLUSION: In this geographically-diverse descriptive analysis of U.S. pregnancies, we observed ubiquitous exposure to low molecular weight PAHs, highlighting the importance of better understanding PAH sources and their pediatric health outcomes attributed to early life PAH exposure.

Abstract: Abstract: Travel bans were a globally prevalent policy response to the COVID-19 pandemic. In the United States, travel bans against China and European countries proved a broadly popular mitigation tool among Americans. Why did Americans support COVID-19 travel bans? We fielded two novel survey experiments, surveying 3000 American citizens across five waves (between March 2020 and March 2021). In randomizing the country of origin of those potentially subject to travel ban measures, we find consistent evidence that racial attitudes drive support for travel bans. The strength of this relationship varies across political parties and across hypothetical target countries but is not explained by objective caseloads that change across countries and over the course of the pandemic.

Abstract: OBJECTIVES: The COVID-19 pandemic placed health care personnel (HCP) at risk for stress, anxiety, burnout, and post-traumatic stress disorder (PTSD). To address this, hospitals developed programs to mitigate risk. The objectives of the current study were to measure the availability and use of these programs in a cohort of academic emergency departments (EDs) in the United States early in the pandemic and identify factors associated with program use. METHODS: Cross-sectional survey of ED HCP in 21 academic EDs in 15 states between June and September 2020. Site investigators provided data on the availability of 28 programs grouped into 9 categories. Individual support programs included: financial, workload mitigation, individual COVID-19 testing, emotional (e.g., mental health hotline), and instrumental (e.g., childcare) Clinical work support programs included: COVID-19 team communication (e.g., debriefing critical incident), patient-family communication facilitation, patient services (e.g., social work, ethics consultation), and system-level exposure reduction. Participants provided corresponding data on whether they used the programs. We used generalized linear mixed models clustered on site to measure the association between demographic and facility characteristics and program use. RESULTS: We received 1,541 survey responses (96% response rate) from emergency physicians or advanced practice providers, nurses, and nonclinical staff. Program availability in each of the 9 categories was high (>95% of hospitals). Program use was variable, with clinical work support programs used more frequently (28-50% of eligible HCP across categories) than individual employee support programs (6-13% of eligible HCP across categories). Fifty-seven percent of respondents reported that the COVID-19 pandemic had affected their stress and anxiety, and 12% were at elevated risk for PTSD. Program use did not significantly differ for HCP who reported symptoms of anxiety and/or stress compared to those who did not. CONCLUSIONS: Early in the pandemic, support programs were widely available to ED HCP, but program use was low. Future work will focus on identifying barriers and facilitators to use and specific programs most likely to be effective during periods of highest occupational stress.

Abstract: IntroductionDespite evidence suggesting deleterious effects of cannabis and nicotine tobacco product (NTP) use on white matter integrity, there have been limited studies examining white matter integrity among users of both cannabis and nicotine. Further, updated white matter methodology provides opportunities to investigate use patterns on neurite orientation dispersion and density (NODDI) indices and subtle tissue changes related to the intra- and extra-neurite compartment. We aimed to investigate how cannabis and NTP use among adolescents and young adults interacts to impact the white matter integrity microstructure.Materials and methodsA total of 221 participants between the ages of 16 and 22 completed the Customary Drinking and Drug Use Record (CDDR) to measure substance use, and underwent a magnetic resonance imaging (MRI) session. Participants were divided into NTP-control and NTP groupings and cannabis-control and cannabis groupings (≥26 NTP/cannabis uses in past 6 months). Tract-Based Spatial Statistics (TBSS) and two-way between-subjects ANOVA investigated the effects of NTP use group, cannabis use group, and their interaction on fractional anisotropy (FA) and NODDI indices while controlling for age and biological sex.ResultsNTP use was associated with decreased FA values and increased orientation dispersion in the left anterior capsule. There were no significant effects of cannabis use or the interaction of NTP and cannabis use on white matter outcomes.DiscussionNTP use was associated with altered white matter integrity in an adolescent and young adult sample. Findings suggest that NTP-associated alterations may be linked to altered fiber tract geometry and dispersed neurite structures versus myelination, as well as differential effects of NTP and cannabis use on white matter structure. Future work is needed to investigate how altered white matter is related to downstream behavioral effects from NTP use.

Abstract: Early life substance use, including cannabis and nicotine, may result in deleterious effects on the maturation of brain tissue and gray matter cortical development. The current study employed linear regression models to investigate the main and interactive effects of past-year nicotine and cannabis use on gray matter cortical thickness estimates in 11 bilateral independent frontal cortical regions in 223 16-22-year-olds. As the frontal cortex develops throughout late adolescence and young adulthood, this period becomes crucial for studying the impact of substance use on brain structure. The distinct effects of nicotine and cannabis use status on cortical thickness were found bilaterally, as cannabis and nicotine users both had thinner cortices than non-users. Interactions between nicotine and cannabis were also observed, in which cannabis use was associated with thicker cortices for those with a history of nicotine and tobacco product (NTP) use in three left frontal regions. This study sheds light on the intricate relationship between substance use and brain structure, suggesting a potential modulation of cannabis' impact on cortical thickness by nicotine exposure, and emphasizing the need for further longitudinal research to characterize these interactions and their implications for brain health and development.

294. The Relationship of Circulating Choline and Choline-Related Metabolite Levels with Health Outcomes: A Scoping Review of Genome-Wide Association Studies and Mendelian Randomization Studies.

Abstract: Choline is essential for proper liver, muscle, brain, lipid metabolism, cellular membrane composition, and repair. Understanding genetic determinants of circulating choline metabolites can help identify new determinants of choline metabolism, requirements, and their link to disease endpoints. We conducted a scoping review to identify studies assessing the association of genetic polymorphisms on circulating choline and choline-related metabolite concentrations and subsequent associations with health outcomes. This study follows the Preferred Reporting Items for Systematic Reviews and Meta-Analyses statement scoping review extension. Literature was searched to September 28, 2022, in 4 databases: Embase, MEDLINE, Web of Science, and the Biological Science Index. Studies of any duration in humans were considered. Any genome-wide association study (GWAS) investigating genetic variant associations with circulating choline and/or choline-related metabolites and any Mendelian randomization (MR) study investigating the association of genetically predicted circulating choline and/or choline-related metabolites with any health outcome were considered. Qualitative evidence is presented in summary tables. From 1248 total reviewed articles, 53 were included (GWAS = 27; MR = 26). Forty-two circulating choline-related metabolites were tested in association with genetic variants in GWAS studies, primarily trimethylamine N-oxide, betaine, sphingomyelins, lysophosphatidylcholines, and phosphatidylcholines. MR studies investigated associations between 52 total unique choline metabolites and 66 unique health outcomes. Of these, 47 significant associations were reported between 16 metabolites (primarily choline, lysophosphatidylcholines, phosphatidylcholines, betaine, and sphingomyelins) and 27 health outcomes including cancer, cardiovascular, metabolic, bone, and brain-related outcomes. Some articles reported significant associations between multiple choline types and the same health outcome. Genetically predicted circulating choline and choline-related metabolite concentrations are associated with a wide variety of health outcomes. Further research is needed to assess how genetic variability influences choline metabolism and whether individuals with lower genetically predicted circulating choline and choline-related metabolite concentrations would benefit from a dietary intervention or supplementation.

295. How Does Home Energy Score Affect Home Value and Mortgage Performance?

Abstract: Energy-efficient homes save their occupants money through lower energy bills. These savings might be capitalized into higher home sale prices. They also improve the household’s net cash flow, which might make households better able to pay mortgage debt. The U.S. Department of Energy (DOE)’s Home Energy Score (HES) assigns a 1-10 score to homes and estimates annual energy bills based on modeled energy consumption. In this paper we investigated the relationship between HES metrics and two housing market outcomes: home sale price and mortgage performance. We found that the relationship was only statistically significant in places with a mandatory HES assessment at the time of sale. Using a sample of 26,291 home sales that occurred after HES assessments, we found that a one-point increase in HES in these locations was associated with a 0.5% increase in sale price, and an increase in $100 of estimated annual energy bills was associated with a 0.4% decrease. This magnitude of effect is consistent with estimated magnitudes of home sale premiums for other green or energy-efficient home certifications in the literature. We also found that a one-point increase in HES was associated with a 5.5% reduction in the odds of a loan going 30 days delinquent if the loan originated after the assessment occurred. Similarly, we found that a $100 decrease in estimated annual energy bills was associated with a 2.3% decrease in the odds of a loan going delinquent if it originated after the assessment occurred. Our results suggest that HES provides a valuable signal for housing market transactions in specific situations.

Abstract: BackgroundNitric oxide (NO) has been identified as a signaling molecule generated during β-adrenergic receptor stimulation in the heart. Furthermore, a role for NO in triggering spontaneous Ca2+ release via S-nitrosylation of CaMKIIδ (Ca2+/calmodulin kinase II delta) is emerging. NO donors are routinely used clinically for their cardioprotective effects on the heart, but it is unknown how NO donors modulate the proarrhythmic CaMKII to alter cardiac arrhythmia incidence. We test the role of S-nitrosylation of CaMKIIδ at the Cysteine-273 inhibitory site and cysteine-290 activating site in cardiac Ca2+ handling and arrhythmogenesis before and during β-adrenergic receptor stimulation.MethodsWe measured Ca2+-handling in isolated cardiomyocytes from C57BL/6J wild-type (WT) mice and mice lacking CaMKIIδ expression (CaMKIIδ-KO) or with deletion of the S-nitrosylation site on CaMKIIδ at cysteine-273 or cysteine-290 (CaMKIIδ-C273S and -C290A knock-in mice). Cardiomyocytes were exposed to NO donors, S-nitrosoglutathione (GSNO; 150 μM), sodium nitroprusside (200 μM), and β-adrenergic agonist isoproterenol (100 nmol/L).ResultsBoth WT and CaMKIIδ-KO cardiomyocytes responded to isoproterenol with a full inotropic and lusitropic Ca2+ transient response as well as increased Ca2+ spark frequency. However, the increase in Ca2+ spark frequency was significantly attenuated in CaMKIIδ-KO cardiomyocytes. The protection from isoproterenol-induced Ca2+ sparks and waves was mimicked by GSNO pretreatment in WT cardiomyocytes but lost in CaMKIIδ-C273S cardiomyocytes. When GSNO was applied after isoproterenol, this protection was not observed in WT or CaMKIIδ-C273S but was apparent in CaMKIIδ-C290A. In Langendorff-perfused isolated hearts, GSNO pretreatment limited isoproterenol-induced arrhythmias in WT but not CaMKIIδ-C273S hearts, while GSNO exposure after isoproterenol sustained or exacerbated arrhythmic events.ConclusionsWe conclude that prior S-nitrosylation of CaMKIIδ at cysteine-273 can limit subsequent β-adrenergic receptor-induced arrhythmias, but that S-nitrosylation at cysteine-290 might worsen or sustain β-adrenergic receptor-induced arrhythmias. This has important implications for the administration of NO donors in the clinical setting.

Abstract: Lung adenocarcinoma (LUAD) is the most prevalent subtype of lung cancer and presents clinically with a high degree of biological heterogeneity and distinct clinical outcomes. The current paradigm of LUAD etiology posits alveolar epithelial type II (AT2) cells as the primary cell of origin, while the role of AT1 cells in LUAD oncogenesis remains unknown. Here, we examine oncogenic transformation in mouse Gram-domain containing 2 (Gramd2)+ AT1 cells via oncogenic KRASG12D. Activation of KRASG12D in AT1 cells induces multifocal LUAD, primarily of papillary histology. Furthermore, KRT8+ intermediate cell states were observed in both AT2- and AT1-derived LUAD, but SCGB3A2+, another intermediate cell marker, was primarily associated with AT1 cells, suggesting different mechanisms of tumor evolution. Collectively, our study reveals that Gramd2+ AT1 cells can serve as a cell of origin for LUAD and suggests that distinct subtypes of LUAD based on cell of origin be considered in the development of therapeutics.

Abstract: Flare frequency distributions represent a key approach to addressing one of the largest problems in solar and stellar physics: determining the mechanism that counter-intuitively heats coronae to temperatures that are orders of magnitude hotter than the corresponding photospheres. It is widely accepted that the magnetic field is responsible for the heating, but there are two competing mechanisms that could explain it: nanoflares or Alfv\'en waves. To date, neither can be directly observed. Nanoflares are, by definition, extremely small, but their aggregate energy release could represent a substantial heating mechanism, presuming they are sufficiently abundant. One way to test this presumption is via the flare frequency distribution, which describes how often flares of various energies occur. If the slope of the power law fitting the flare frequency distribution is above a critical threshold, $\alpha=2$ as established in prior literature, then there should be a sufficient abundance of nanoflares to explain coronal heating. We performed $>$600 case studies of solar flares, made possible by an unprecedented number of data analysts via three semesters of an undergraduate physics laboratory course. This allowed us to include two crucial, but nontrivial, analysis methods: pre-flare baseline subtraction and computation of the flare energy, which requires determining flare start and stop times. We aggregated the results of these analyses into a statistical study to determine that $\alpha = 1.63 \pm 0.03$. Abstract: Gravitational lensing by massive objects along the line of sight to the source causes distortions of gravitational wave-signals; such distortions may reveal information about fundamental physics, cosmology and astrophysics. In this work, we have extended the search for lensing signatures to all binary black hole events from the third observing run of the LIGO--Virgo network. We search for repeated signals from strong lensing by 1) performing targeted searches for subthreshold signals, 2) calculating the degree of overlap amongst the intrinsic parameters and sky location of pairs of signals, 3) comparing the similarities of the spectrograms amongst pairs of signals, and 4) performing dual-signal Bayesian analysis that takes into account selection effects and astrophysical knowledge. We also search for distortions to the gravitational waveform caused by 1) frequency-independent phase shifts in strongly lensed images, and 2) frequency-dependent modulation of the amplitude and phase due to point masses. None of these searches yields significant evidence for lensing. Finally, we use the non-detection of gravitational-wave lensing to constrain the lensing rate based on the latest merger-rate estimates and the fraction of dark matter composed of compact objects., Comment: 28 pages, 11 figures
