Abstract

Anthelmintic resistance has become a serious problem worldwide especially for small ruminants but it is also rising for cattle. The sustainability to maintain the efficacy of anthelmintics is an important objective. Furthermore, reduction of anthelmintic use is desired to assure safe and high quality food. Therefore targeted selective treatment (TST) systems should be developed. Through TST the use of anthelmintics can be reduced and selection pressure on sensible endoparasite isolates decreases. The current project aims at 1st the investigation of the current efficacy of macrocyclic lac-tone anthelmintics in first season grazing cattle in Northern Germany and 2nd to com-paratively investigate different approaches to TST such as body weight, body condition-ing scores (BCS) and egg output (EPG).

Abstract

Helminth infections have large negative impacts on production efficiency in ruminant farming systems worldwide, and their effective management is essential if livestock production is to increase to meet future human needs for dietary protein. The control of helminths relies heavily on routine use of chemotherapeutics, but this approach is unsustainable as resistance to anthelmintic drugs is widespread and increasing. At the same time, infection patterns are being altered by changes in climate, land-use and farming practices. Future farms will need to adopt more efficient, robust and sustainable control methods, integrating ongoing scientific advances. Here, we present a vision of helminth control in farmed ruminants by 2030, bringing to bear progress in: (1) diagnostic tools, (2) innovative control approaches based on vaccines and selective breeding, (3) anthelmintics, by sustainable use of existing products and potentially new compounds, and (4) rational integration of future control practices. In this review, we identify the technical advances that we believe will place new tools in the hands of animal health decision makers in 2030, to enhance their options for control and allow them to achieve a more integrated and sustainable approach to helminth control in support of animal welfare and production. Abstract

Grazing goats are naturally infected with gastrointestinal nematodes which are an important cause of diseases and economic losses. Targeted selective treatment (TST) is a possibility to avoid spreading of anthelmintic resistances and to reduce the use of anthelmintics. The aim of this study is the development of an goat-milk ELISA for the detection of infection with T. circumcincta in order to provide a monitoring tool for herd infection levels. Therefore a group of kids were artificially infected, blood and faecal samples were taken over a period of 10 weeks. Additional a herd of milk goats,kept on pasture at the experimental farm were used to obtain milk, blood and faecal samples regularly.

Abstract

In grazing cattle, infections with gastrointestinal nematodes pose some of the most important health threats and subclinical infections result in considerable production losses. While there is little doubt that climate change will affect grazing ruminants directly, mean temperature increases of ∼3 °C and longer drought stress periods in summer may also influence the free-living stages of parasitic nematodes. Hostile climatic conditions reduce the number of L3s on pasture and therefore the refugium, which is expected to result in a higher selection pressure, accelerating development of resistance against anthelmintic drugs. The aim of the current experiments was to investigate the effects of drought stress and different temperature/humidity ranges over time on the survival and fitness of Cooperia oncophora L3s and their distribution in grass and soil under controlled conditions using a climate chamber. Grass containers inoculated with L3s were analysed after 1–6 weeks using descriptive statistics as well as linear models. A large proportion of L3s was recovered from soil where fitness was also better preserved than on grass. Numbers and fitness of recovered L3s declined with duration in the climate chamber under both temperature profiles. However, the results of the linear models confirmed that higher temperatures (20–33 °C versus 17–22.6 °C) significantly impaired survival, distribution and fitness of L3s. Application of drought stress, known as another important factor, had a surprisingly smaller impact than its duration or higher temperatures. The climate chamber enabled exclusion of confounding factors and therefore accurate interpretation of the investigated climatic aspects. The obtained results highlight the relative importance of those factors, and will help to design better models for the population dynamics of L3s on pasture in the future. Additionally, the outcomes of these investigations may offer explanations regarding interdependencies of development of anthelmintic resistance and the presence of hot/dry weather conditions. 231. Anthelmintic resistance to ivermectin and moxidectin in gastrointestinal nematodes of cattle in Europe.

Abstract

Anthelmintic resistance has been increasingly reported in cattle worldwide over the last decade, although reports from Europe are more limited. The objective of the present study was to evaluate the efficacy of injectable formulations of ivermectin and moxidectin at 0.2 mg per kg bodyweight against naturally acquired gastro-intestinal nematodes in cattle. A total of 753 animals on 40 farms were enrolled in Germany (12 farms), the UK (10 farms), Italy (10 farms), and France (8 farms). Animals were selected based on pre-treatment faecal egg counts and were allocated to one of the two treatment groups. Each treatment group consisted of between 7 and 10 animals. A post-treatment faecal egg count was performed 14 days (±2 days) after treatment. The observed percentage reduction was calculated for each treatment group based on the arithmetic mean faecal egg count before and after treatment. The resistance status was evaluated based on the reduction in arithmetic mean faecal egg count and both the lower and upper 95% confidence limits. A decreased efficacy was observed in half or more of the farms in Germany, France and the UK. For moxidectin, resistance was confirmed on 3 farms in France, and on 1 farm in Germany and the UK. For ivermectin, resistance was confirmed on 3 farms in the UK, and on 1 farm in Germany and France. The remaining farms with decreased efficacy were classified as having an inconclusive resistance status based on the available data. After treatment Cooperia spp. larvae were most frequently identified, though Ostertagia ostertagi was also found, in particular within the UK and Germany. The present study reports lower than expected efficacy for ivermectin and moxidectin (based on the reduction in egg excretion after treatment) on European cattle farms, with confirmed anthelmintic resistance on 12.5% of the farms. Abstract

Background: In Europe, canine babesiosis is most frequently caused by Babesia canis and Babesia vogeli, and occasionally by Babesia gibsoni.. In Germany, B. canis is recognized as endemic. The aims of this study were to assess how often Babesia spp. infections were diagnosed in a commercial laboratory in samples from dogs from Germany, and to evaluate potential risk factors for infection. Methods: The database of the LABOKLIN laboratory was screened for Babesia spp.-positive polymerase chain reaction (PCR) tests for dogs for the period January 2007–December 2020. Sequencing was performed for positive tests from 2018 and 2019. Binary logistic regression analysis was performed to determine the effects of sex, season, and year of testing. Questionnaires were sent to the submitting veterinarians to obtain information on travel abroad, tick infestation, and ectoparasite prophylaxis of the respective dogs. Fisher's exact test was used to calculate statistical significance and P < 0.05 was considered statistically significant. Results: In total, 659 out of 20,914 dogs (3.2%) tested positive for Babesia spp. by PCR. Of 172 sequenced samples, B. canis was identified in 156, B. vogeli in nine, B. gibsoni in five, and B. vulpes in two. Season had a statistically significant impact on test results when summer/winter (1.6% tested positive) was compared to spring/autumn (4.7%), with peaks in April (5.2%) and October (7.4%) [P < 0.001, odds ratio (OR) = 3.16]. Sex (male 3.5%, female 2.8%; P = 0.012, OR = 1.49) and age (< 7 years old 4.0%, ≥ 7 years old 2.3%; P < 0.001, OR = 1.76) of the tested dogs also had a statistically significant effect. A statistically significant impact was demonstrated for observed tick attachment (P < 0.001, OR = 7.62) and lack of ectoparasite prophylaxis (P = 0.001, OR = 3.03). The frequency of positive Babesia spp. tests did not significantly differ between the 659 dogs that had never left Germany and the 1506 dogs with known stays abroad (P = 0.088). Conclusions: The possibility of canine infection with B. canis needs to be especially taken into consideration in spring and autumn in Germany as the activity of the tick Dermacentor reticulatus, a potential vector for canine babesiosis, is highest in these seasons. Travel and importation of dogs are considered major factors associated with canine babesiosis in Germany.
