The Arctic Ocean is changing rapidly as Earth's climate warms. To document and understand these changing ocean conditions, we developed a rosette that collects high-quality oceanographic data and is deployed from an aircraft through a 30.5 cm diameter hole drilled in the ice (Figure 1). The rosette is modular, with the modules attached vertically on a conducting hydrowire to achieve a small diameter. Typically, three modules with four 4-liter bottles each are positioned above the conductivity-temperature-depth (CTD) module (Figure 2). The rosette package is lowered through the ice and retrieved at speeds up to 40 meters per minute using a small winch mounted in the aircraft. The hydrowire leads from the aircraft to a sheave hung from a tripod on the ice above the hole inside a tent that is heated to prevent water samples and sensors from freezing (Figure 3). The CTD data are acquired and displayed in real time on a laptop computer, and bottles are closed at desired depths electronically. Upon recovery, each water-bottle module is immediately placed in a cooler with bags of snow, which provide thermal stability within ± 2°C of the in situ temperature. The modules are returned to a base camp where they can be sampled and the samples processed under controlled conditions. A wide variety of water samples can be collected. Thus far, we have collected samples for salinity, dissolved oxygen, nutrients, helium isotopes, oxygen isotopes, chlorofluorocarbons, SF6, tritium, CO2, barium, and 129I. The quality of all samples has been excellent. Smethie et al. (2011) provide a detailed description of the rosette and its performance.

Retinoblastoma, the most common primary malignant intraocular tumor of childhood is a great success story in pediatric and ocular oncology. Pathology of retinoblastoma is important to guide the treatment modalities. Differentiated retinoblastoma is commonly seen in younger age group. Since a hundred years, we have been observing two typical true rosettes in retinoblastoma in the form of Flexner-Wintersteiner (FW) and Homer Wright (HW) rosettes and in many occasions pseudorosettes have been documented. In the present case report, a third new type of rosette was identified in a differentiated retinoblastoma which had an unusual anterior segment involvement.

Objetivando melhorar o desenvolvimento in vitro de embriões de pêssegos e nectarinas, de maturação precoce, adicionou-se, em meio básico de cultura, o regulador de crescimento 6-benzilaminopurina (BAP) nas concentrações de 0,5,10,15 e 20µM. Esse meio consistiu na solução salina de Murashige & Skoog acrescido de tiamina, 1mg/l; ácido nicotínico, 0,5mg/l; inositol, 100mg/1, glicina, 250mg/l; glutamina, 500mg/l; asparagina, 250mg/l, ácido giberélico, 0,1mg/l; sacarose, 30g/l, e ágar, 6g/l. A melhor taxa de desenvolvimento dos embriões, após 30dias de cultura, ocorreu nas concentrações de 5 e 10µM de BAP, com uma emissão média de 4,5 brotos por embrião. Nas concentrações de 15 e 20mM de BAP, o número de brotações foi maior, porém com menor aproveitamento, devido à presença dos sintomas indesejáveis de vitrificação. O BAP não eliminou, totalmente, a roseta e o ananismo fisiológico dos embriões. Embryos of early peach and nectarine were cultured on Murashige and Skoog medium containing 3%sacarose,0.6%agar and(in mg/liter) 1 thiamin, 0.5 nicotinic acid, 100 inositol, 250 glycin, 500 glutamin, 250 asparagin, 0.1 giberelic acid, and BAP at concentrations of 0, 5, 10, 15 and 20µM. The frequency of growth was as high as 100% when BAP was included in the medium. The best development of the embryos occurred with BAP rates of 5 and 10µM. At these concentrations of BAP, the mean number of adventitious shoots was 4.5 per embryo. The vitrification ratio was higher at concentrations of 15 and 20µM. The inclusion of BAP, consistently reduced but not fully eliminated lhe rosette and dwarf appearances in vitroplantules.

Testaram-se os efeitos dos períodos de estratificação a frio úmido por 0, 10, 20, 30 e 40 dias, associados a aplicações do ácido giberélico (GA3) nas concentrações de 0, 5, 10, 15 e 20 ppm, na quebra de dormência das sementes do pessegueiro porta-enxerto 'Okinawa'. Os resultados mostraram que os períodos de estratificação de 0, 10 e 20 dias foram insuficientes para a completa quebra de dormência, mesmo quando em associação com o GA3. Apesar da boa emergência verificada, após 20 dias de frio, 20% das plantas desenvolveram-se com formação de rosetas. As melhores respostas foram obtidas aos 30 e 40 dias de estratificação, mesmo na ausência do GA3; a germinação aproximou-se de 100% e nenhuma plântula anômala foi detectada. O GA3 mostrou efeito na germinação das amêndoas, sobretudo nas concentrações maiores, porém insuficiente para eliminar os sintomas de falta de frio das plântulas.The main objective of the present work was to study the effects of stratification period and gibberellic acid (GA3), on emergence of 'Okinawa' peach rootstock seeds. The experiment was conducted with seeds freshly extracted from ripe fruits. The seeds were soaked for 24 hours in aqueous solutions of 0, 5, 10, 15 and 20 ppm GA3 and immediately stratified in a wet cotton substrate for 0, 10, 20, 30 and 40 days. The results showed that for 0, 10 and 20 days, stratification associated with GA3 at 5, 10, 15 and 20 ppm, presented no significant effect on the seed germination and seedling emergence. The best responses were achieved with 30 and 40 days of stratification, even in the absence of GA3. In these treatments, 100% germination and emergence were obtained and no anomalous seedling was detected.

O experimento objetivou eliminar os sintomas de roseta e ananismo em híbridos de pessegueiro (Prunus persica L. Batsch) precoces, provenientes de cultura embrionária. Essas anomalias fisiológicas aparecem em vista dos inibidores de crescimento presentes nos meristemas apicais dos embriões. Dois processos de recuperação de vitroplântulas anômalas foram adotados: (a) eliminação da porção apical logo acima da primeira gema, a partir dos cotilédones; (b) manutenção das vitroplântulas por trinta dias em ambiente de vernalização com temperatura de 5-10 º C. Os melhores resultados foram obtidos quando se eliminou a dominância do meristema apical; as vitroplântulas mantidas em sala de crescimento emitiram, em cinco dias, brotações novas e normais e se desenvolveram rapidamente. Aquelas submetidas à quebra de endodormência, porém,retomaram parcialmente o desenvolvimento, de forma lenta e anormal.Two methods were studied on peach embryo culture in order to recover seedlings presenting rosette and dwarf development. In the first method almost all the epicotyle was eliminated. Only a small portion of the axe just about cotyledons was left. In the second method, the seedlings were maintained under vernalization for thirty days at 5-10°C in a dark room. The results showed that the seedlings submitted to the first method recovered rapidly with normal emission of now shoots. The same was not observed for the seedlings from the second method which developed slowly and still anomalously.

We have reported a marked decrease of lymphocyte sensitivity to prostaglandins (PGs) in patients with untreated cervical carcinoma and other solid neoplasias. Particularly, Ea and ME rosette formation was inhibited only slightly by PGs in all cancer patients studied, as compared with the high inhibition values observed in normal individuals (Clin Immunol Immunopathol, 1981). In the present work, the PG-induced inhibition of Ea and ME rosette formation has been studied in patients with stage 0 cervical carcinoma up to 180 days after hysterectomy. It has been found that in these patients the PG sensitivity increased gradually up to or higher than the levels seen in normal control subjects. However, the PG sensitivity of Ea is restored earlier after surgery, while ME show a delayed recovery. Since the PG sensitivities correlate well with the clinical status of patients, it is suggested that such sensitivity may represent a useful test for following patients with cervical carcinoma.

Observations were carried out in the years 2014-2015 in moor grass meadows, old fields, willow thickets and macroforbs. In the successive study sites, the height of the standing vegetation and soil moisture gradually increased, whilst the light availability decreased. Stable in consecutive seasons, the total number of ramets per generative ramet cluster achieved the lowest values in willow thickets due to mechanical suppression of vegetative growth by the robust unde rground organs of neighbouring plants. Constant during the study period, the share of generative stems decreased in successive study sites, while the percentage of leaf rosettes showed an inverted tendency. The increase of height of generative stems and number of flowers per inflorescence in consecutive Patches, as well as the augmentation of length of flowers in time and space, might trigger an improvement of generative reproduction in a crowded environment. The increase of the number of nodes and dimensions of cauline leaf blades in successive Patches and the augmentation of the number and dimensions of rosette leaves in the time and space might contribute to greater efficacy of light capture in growing shading. Summarizing the plasticity of traits might assure the spread of generative ramet clusters in open habitats, as well as their persistence in crowded sites.

Background During the photosynthesis, two isoforms of the fructose-1,6-bisphosphatase (FBPase), the chloroplastidial (cFBP1) and the cytosolic (cyFBP), catalyse the first irreversible step during the conversion of triose phosphates (TP) to starch or sucrose, respectively. Deficiency in cyFBP and cFBP1 isoforms provokes an imbalance of the starch/sucrose ratio, causing a dramatic effect on plant development when the plastidial enzyme is lacking. Results We study the correlation between the transcriptome and proteome profile in rosettes and roots when cFBP1 or cyFBP genes are disrupted in Arabidopsis thaliana knock-out mutants. By using a 70-mer oligonucleotide microarray representing the genome of Arabidopsis we were able to identify 1067 and 1243 genes whose expressions are altered in the rosettes and roots of the cfbp1 mutant respectively; whilst in rosettes and roots of cyfbp mutant 1068 and 1079 genes are being up- or down-regulated respectively. Quantitative real-time PCR validated 100% of a set of 14 selected genes differentially expressed according to our microarray analysis. Two-dimensional (2-D) gel electrophoresis-based proteomic analysis revealed quantitative differences in 36 and 26 proteins regulated in rosettes and roots of cfbp1, respectively, whereas the 18 and 48 others were regulated in rosettes and roots of cyfbp mutant, respectively. The genes differentially expressed and the proteins more or less abundant revealed changes in protein metabolism, RNA regulation, cell signalling and organization, carbon metabolism, redox regulation, and transport together with biotic and abiotic stress. Notably, a significant set (25%) of the proteins identified were also found to be regulated at a transcriptional level. Conclusion This transcriptomic and proteomic analysis is the first comprehensive and comparative study of the gene/protein re-adjustment that occurs in photosynthetic and non-photosynthetic organs of Arabidopsis mutants lacking FBPase isoforms.
