Twelve years have passed since the early outlooks of applying genomic selection (GS) to forest tree breeding, initially based on deterministic simulations, soon followed by empirical reports. Given its solid projections for causing a paradigm shift in tree breeding practice in the years to come, GS went from a hot, somewhat hyped, topic to a fast-moving area of applied research and operational implementation worldwide. The hype cycle curve of emerging technologies introduced by Gartner Inc. in 1995, models the path a technology takes in terms of expectations of its value through time. Starting with a sudden and excessively positive "peak of inflated expectations" at its introduction, a technology that survives the "valley of disappointment" moves into maturity to climb the "slope of enlightenment", to eventually reach the "plateau of productivity". Following the pioneering steps of GS in animal breeding, we have surpassed the initial phases of the Gartner hype cycle and we are now climbing the slope of enlightenment towards a wide application of GS in forest tree breeding. By merging modern high-throughput DNA typing, time-proven quantitative genetics and mixed-model analysis, GS moved the focus away from the questionable concept of dissecting a complex, polygenic trait in its individual components for breeding advancement. Instead of trying to find the needle in a haystack, i.e., the "magic" gene in the complex and fluid genome, GS more efficiently and humbly "buys the whole haystack" of genomic effects to predict complex phenotypes, similarly to an exchange-traded fund that more efficiently "buys the whole market". Tens of studies have now been published in forest trees showing that GS matches or surpasses the performance of phenotypic selection for growth and wood properties traits, enhancing the rate of genetic gain per unit time by increasing selection intensity, radically reducing generation interval and improving the accuracy of breeding values. Breeder-friendly and cost-effective SNP (single nucleotide polymorphism) genotyping platforms are now available for all mainstream plantation forest trees, but methods based on low-pass whole genome sequencing with imputation might further reduce genotyping costs. In this perspective, I provide answers to why GS will soon become the most efficient and effective way to carry out advanced tree breeding, and outline a simple pilot demonstration project that tree breeders can propose in their organization. While the fundamental properties of GS in tree breeding are now solidly established, strategic, logistics and financial aspects for the optimized adoption of GS are now the focus of attentions towards the plateau of productivity in the cycle, when this new breeding method will become fully established into routine tree improvement. Rice production is often affected by biotic and abiotic stressors. The breeding of resistant cultivars is a cost-cutting and environmentally friendly strategy to maintain a sustainable high production level. An elite water-saving and drought-resistant rice (WDR), Hanhui3, is susceptible to blast and bacterial blight (BB). This study was conducted to introgress three resistance genes (Pi2, xa5, and Xa23) for blast and BB into Hanhui3, using marker-assisted selection (MAS) for the foreground selection and a whole-genome single-nucleotide polymorphism (SNP) array for the background selection. As revealed by the whole-genome SNP array, the recurrent parent genome (RPG) recovery of the improved NIL was 94.2%. The resistance levels to blast and BB of the improved NIL and its derived hybrids were higher than that of the controls. In addition, the improved NIL and its derived hybrids retained the desired agronomic traits from Hanhui3, such as yield. The improved NIL could be useful to enhance resistance against biotic stressors and produce stable grain yields in Oryza sativa subspecies indica rice breeding programs. [ABSTRACT FROM AUTHOR]

Oilseed rape (Brassica napus) has significant heterosis and Ogura CMS is a major way to use it. Ogura CMS has the advantages of complete and stable male sterility and easy-to-breed maintainers. Therefore, to breed better restorers has become an important goal for this system. Incidentally, clubroot is a soil-borne disease that is difficult to control by fungicidal chemicals, and it has been the main disease of oilseed rape in recent years in China, severely restricting the development of the oilseed rape industry. At present, the most effective method for controlling clubroot disease is to cultivate resistant varieties. One Ogura CMS restorer line (R2163) has shown much better combining ability, but lacks the clubroot disease resistance. This study was carried out to improve R2163 through marker-assisted backcross breeding (MABB). The resistant locus PbBa8.1 was introduced into the restorer R2163, and we then selected R2163R with clubroot disease resistance. Using the new restorer R2163R as the male parent and the sterile lines 116A and Z11A as the female parent, the improved, new resistant hybrids Kenyouza 741R and Huayouza 706R performed well, providing strong resistance and good agronomic traits. Powdery mildew of wheat (Triticum aestivum), caused by Blumeria graminis f.sp. tritici (Bgt), is a destructive disease that seriously threatens the yield and quality of its host. Identifying resistance genes is the most attractive and e􀀀ective strategy for developing disease-resistant cultivars and controlling this disease. In this study, a wheat breeding line Yannong 99102-06188 (YN99102), an elite derivative line from the same breeding process as the famous wheat cultivar Yannong 999, showed high resistance to powdery mildew at the whole growth stages. Genetic analysis was carried out using Bgt isolate E09 and a population of YN99102 crossed with a susceptible parent Jinhe 13-205 (JH13-205). The result indicated that a single recessive gene, tentatively designated pmYN99102, conferred seedling resistance to the Bgt isolate E09. Using bulked segregant exome capture sequencing (BSE-Seq), pmYN99102 was physically located to a ∼33.7Mb (691.0-724.7Mb) interval on the chromosome arm 2BL, and this interval was further locked in a 1.5cM genetic interval using molecular markers, which was aligned to a 9.0Mb physical interval (699.2-708.2Mb). Based on the analysis of physical location, origin, resistant spectrum, and inherited pattern, pmYN99102 di􀀀ered from those of the reported powdery mildew (Pm) resistance genes on 2BL, suggesting pmYN99102 is most likely a new Pm gene/allele in the targeted interval. To transfer pmYN99102 to di􀀀erent genetic backgrounds using marker-assisted selection (MAS), 18 closely linked markers were tested for their availability in di􀀀erent genetic backgrounds for MAS, and all markers expect for YTU103-97 can be used in MAS for tracking pmYN99102 when it transferred into those susceptible cultivars. The hull-less pumpkin (Cucurbita pepo) seed does not require de-hulling before use for human consumption, as a result highly preferred by the oil, nut, and baking industries. In hull-less seeds, a single recessive gene is responsible for the absence of outer thick seed coat layers; however, the genomic region and gene(s) controlling the trait are unclear to date. In this study, four crosses attempted to derive F2 and backcross populations confirmed the single recessive gene inheritance of hull-less seed trait in populations adapted to the sub-tropical climate. The candidate genomic region for hull-less seed trait was identified through the BSA-QTLseq approach using bulks of F2: 3 progenies from a cross of HP111 (hulled) and HLP36 (hull-less). A novel genomic region on chromosome 12 ranging from 1.80 to 3.86Mb was associated with the hull-less seed trait. The re-sequencing data identified a total of 396 SNPs within this region and eight were successfully converted into polymorphic KASP assays. The genotyping of segregating F2 (n = 160) with polymorphic KASP assays resulted in a 40.3cM partial linkage map and identified Cp_3430407 (10cM) and Cp_3498687 (16.1cM) as flanking markers for hull-less locus (Cphl-1). These flankingmarkers correspond to the 68.28 kb region in the reference genome, and the marker, Cp_3430407 successfully predicted the genotype in 93.33% of the C. pepo hull-less germplasm lines, thus can be used for marker-assisted selection in parents polymorphic for the hull-less seed trait. The Cphl-1-linked genomic region (2.06Mb) encompasses a total of 182 genes, including secondary cell wall and lignin biosynthesis-related transcriptional factors viz., "NAC" (Cp4.1LG12g04350) and "MYB" (Cp4.1LG12g03120). These genes were differentially expressed in the seeds of hulled and hull-less genotypes, and therefore could be the potential candidate genes governing the hull-less seed trait in pumpkin. Fish diseases have a "stress-related" nature, whereas fish exposure to stressors will increase their susceptibility to infections. It was also noted that fish exposure to biotic and abiotic stressors would exaggerate the disease signs, elicit high mortalities, and cause severe economic losses. Motile aeromonad septicemia (MAS) is a major bacterial disease affecting a variety of finfish species throughout the globe and is caused by Aeromonas hydrophila. Herein, we have evaluated the impacts of ammonia-N stress and/or Nile tilapia challenge with pathogenic A. hydrophila on the clinical picture of MAS disease. Clinical signs, postmortem lesions, histoarchitectural changes, and gene transcription analysis were studied. Fish experimentally infected with A. hydrophila were exophthalmic and showed darkened skin. Moreover, opercular hyperemia, petechial hemorrhages, and gill congestion alongside dermal ulcerations were noticed in ammonia-exposed fish. On the other side, fish exposed to both stressors exhibited exophthalmia, corneal opacity, severe dropsy, and hemorrhagic dermal ulcerations. At the tissue levels, the histopathological lesions were exaggerated in the fish group exposed to ammonia stress and challenged with A. hydrophila than fish group exposed to each one alone. At the molecular levels, the mRNA expression analysis reveals significant upregulation of inflammatory cytokines such as interleukin-1 beta, CXC chemokine , and tumor necrosis factor-alpha in the kidney tissues of Nile tilapia exposed to ammonia and/or challenged with A. hydrophila. In a similar trend, the mRNA expression values of heat shock protein 70 (HSP70), oxidative stress related genes (SOD and CAT) and apoptosis-related genes (caspase 3 , BAX , and cytochrome P450) were also increased in the hepatic tissues of fish exposed to singular or dual stressors. Interestingly, the highest expression levels of the above-mentioned genes were found in the fish group exposed to both stressors. Taken together, these findings indicate the occurrence of severe inflammatory and apoptotic changes in fish exposed to ammonia and infected with A. hydrophila more than each one alone. In contrast, there was a significant decrease in the expression values of the antioxidant enzyme glutathione-S-transferase (GST) in stressed fish, suggesting the occurrence of oxidative stress. We hereby demonstrate that 1H detected 15N‐1H heteronuclear multiple‐quantum spectroscopy can be carried out at a magic angle spinning frequency of 150 kHz. While the 15N‐1H multiple‐quantum coherences can be directly excited from the dipolar order created by the method of adiabatic demagnetization in the rotating frame, it is technically more advantageous to acquire the chemical shift evolution of the heteronuclear multiple‐quantum coherence by two separate chemical shift evolution periods for 1H and 15N. We also show that the heteronuclear multiple‐quantum correlation spectrum can be obtained by shearing the corresponding heteronuclear single‐quantum correlation spectrum. [ABSTRACT FROM AUTHOR]

Wheat gene Yr39 confers durable high-temperature adult-plant (HTAP) resistance to stripe rust caused by Puccinia striiformis f. sp. tritici, one of the most destructive diseases of wheat worldwide. The objective of this study was to transfer Yr39 into four Chinese elite wheat cultivars. Backcross inbred line populations were developed from four Chinese wheat cultivars Chuanmai 42 (CM42), Bainong Aikang 58 (AK58), Han 6172 (H6172) and Zhengmai 9023 (ZM9023) crossed with a Yr39 single-gene line. The F1, BC1F1 and BC1F6 lines were genotyped using resistance gene analogs polymorphism (RGAP) markers Xwgp36, Xwgp44 and Xwgp43, which are closely linked to Yr39. Progeny lines selected with the markers for Yr39 were evaluated in the field for stripe rust resistance and agronomic traits including plant height, tiller numbers, spike grain numbers and thousand-grain weight. Eleven lines were selected with stripe rust resistance and desirable agronomic traits. These lines with production potential can be used for further testing in various wheat production regions and as germplasm resources for breeding new wheat cultivars with durable stripe rust resistance, high yield, and adaptation to different production environments. A finite-time adaptive tracking control algorithm based on non-singular terminal sliding mode (NTSM) is proposed to solve the consensus problem of second-order multi-agent system (MAS) with disturbance. Firstly, a finite-time disturbance observer based on NTSM is introduced to estimate and compensate the disturbance. Then, a NTSM adaptive controller is designed to enhance the robustness of the system, and improve the response speed and tracking accuracy. A proof of the finite-time convergence of the method is given by using Lyapunov theory. Finally, simulation experiments are carried out to demonstrate the effectiveness of the proposed method. Recently, a candidate gene (CJt020762) for MALE STERILITY 1 (MS1) in Cryptomeria japonica has been identified, which made it possible to perform accurate selection of trees with mutant alleles (ms1–1 or ms1–2). Marker-assisted selection (MAS) is an effective method for drastically reducing the time required for a breeding cycle; however, a larger sample size for selection increases the labor and cost of analysis. In this study, firstly, we developed an efficient and low-cost marker selection method using bulk DNA extracted from a mixture of needle tissues from several individual trees. The time required for the extraction of bulk DNA, the accuracy of target peak identification in fragment analysis, and the numbers of samples required to identify trees with ms1 were compared for 3-, 5-, 7-, and 10-individual mixed bulk DNA samples. The results showed that MAS using 10-individual mixed bulk DNA samples was the most efficient and lowest cost for selecting trees with ms1. The accuracy of genotyping using 10-individual mixed bulk DNA samples was verified by conducting a blind test consisting of sample preparation, extraction of bulk DNA, and genotyping under blind conditions (i.e., all researchers were unaware of the correct genotype of the samples). Next, we tried selecting trees with ms1 from 866 breeding materials by MAS using 10-individual mixed bulk DNA samples. We successfully selected nine previously untested trees that were heterozygous for MS1. Finally, we showed that the use of bulk DNA in MAS enabled significant reductions in labor and cost by comparing the approaches using bulk DNA samples with single-individual (non-bulk) DNA samples, although it should be noted that the efficiency of selection depends on the proportion of samples with a target allele. Introduction: Maximal aerobic speed (MAS), usually measured by cardiopulmonary exercise testing (CPET) on a treadmill, is gaining popularity in soccer to determine aerobic performance. Several field tests are used to estimate MAS, although, gold standard methods are still not clarified. Therefore, this work aims 1) to compare two different CPET based methods to assess MAS and 2) to investigate the convergent validity of two common field tests to estimate MAS in soccer.Methods: Thirteen trained male soccer players completed an CPET on a treadmill to determine two VO2-kinetic based definitions of MAS (MASPlateau = speed at onset of VO2-plateau = gold standard; MAS30s = first speed of 30-s-interval of VO2max), the Université de Montreal Track Test (UMTT; VUMTT = speed of the last stage), and a 1500-m-time trial (1500-m-TT; V1500m = average speed). MASPlateau, MAS30s, VUMTT, and V1500m were compared using ANOVA. Additionally, limits of agreement analysis (LoA), Pearson’s r, and ICC were calculated between tests.Results: MAS30s, VUMTT, and V1500m significantly overestimated MASPlateau by 0.99 km/h (ES = 1.61; p < 0.01), 1.61 km/h (ES = 2.03; p < 0.01) and 1.68 km/h (ES = 1.77; p < 0.01), respectively, with large LoA (-0.21 ≤ LoA≤3.55), however with large-to-very large correlations (0.65 ≤ r ≤ 0.87; p ≤ 0.02; 0.51 ≤ ICC≤ 0.85; p ≤ 0.03).Discussion: The overestimation and large LoA of MASPlateau by all estimates indicate that 1) a uniform definition of MAS is needed and 2) the UMTT and a 1500-m-TT seem questionable for estimating MAS for trained soccer players on an individual basis, while regression equations might be suitable on a team level. The results of the present work contribute to the clarification of acquisition of MAS in soccer.

BackgroundMacrophage activation syndrome (MAS) is a potentially life-threatening complication of various inflammatory disorders, including multisystem inflammatory syndrome in children (MIS-C). MIS-C refractory to treatment should raise suspicion of MAS, which can be fatal if a definitive diagnosis is delayed. Unfortunately, there is a lack of data on MAS in children with MIS-C.ObjectiveOur study aims to analyze the risk factors for the development of MAS in MIS-C, its clinical course and response to treatment, and identify predictive factors for pediatric intensive care.Material and methodsWe analyzed data from the Polish MIS-C registry of the MultiOrgan Inflammatory Syndromes COVID-19 Related Study. Patients were diagnosed according to the WHO MIS-C definition and treated according to national guidelines (Polish Pediatric Society) based on international consensus. MAS definition was based on 2016 Classification Criteria for Macrophage Activation Syndrome Complicating Systemic Juvenile Idiopathic Arthritis.ResultsTwo-hundred and seventy four children met the study inclusion criteria. Fifty-nine patients fulfilled MAS classification criteria, nine of which required admission to the pediatric intensive care unit (PICU). MIS-C patients with MAS were significantly older than patients without MAS (median 11.2 vs. 8.1 years). Multivariable analysis showed that age, symptoms characteristic of atypical Kawasaki disease, and skin erosions were significant factors associated with MAS in MIS-C patients. Analysis of laboratory parameters showed that on admission, MIS-C patients with MAS had significantly lower median lymphocyte and platelet counts, albumin and sodium levels, and higher median levels of C-reactive protein, procalcitonin, ferritin, D-dimers, triglycerides, serum creatinine, urea, and γ-glutamyl transpeptidase, and neutrophil count. Multivariate analysis showed that higher procalcitonin, ferritin, and fibrinogen levels at admission were predictive of MAS. Only elevated troponin level was a factor indicating a requirement of PICU hospitalization for children with MAS. MIS-C patients fulfilling MAS criteria were treated more often with intravenous immunoglobulins and steroids than children without MAS. Children with MAS more often required mechanical ventilation. None of the patients required biological agents.ConclusionsThe clinical course of MAS in MIS-C seems milder, treatment less aggressive, and the prognosis better than expected based on the current knowledge on MAS complicating other rheumatological diseases.

Abstract Background Maize rough dwarf disease (MRDD), a widespread disease caused by four pathogenic viruses, severely reduces maize yield and grain quality. Resistance against MRDD is a complex trait that controlled by many quantitative trait loci (QTL) and easily influenced by environmental conditions. So far, many studies have reported numbers of resistant QTL, however, only one QTL have been cloned, so it is especially important to map and clone more genes that confer resistance to MRDD. Results In the study, a major quantitative trait locus (QTL) qMrdd2, which confers resistance to MRDD, was identified and fine mapped. qMrdd2, located on chromosome 2, was consistently identified in a 15-Mb interval between the simple sequence repeat (SSR) markers D184 and D1600 by using a recombinant inbred line (RIL) population derived from a cross between resistant (“80007”) and susceptible (“80044”) inbred lines. Using a recombinant-derived progeny test strategy, qMrdd2 was delineated to an interval of 577 kb flanked by markers N31 and N42. We further demonstrated that qMrdd2 is an incompletely dominant resistance locus for MRDD that reduced the disease severity index by 20.4%. Conclusions A major resistance QTL (qMrdd2) have been identified and successfully refined into 577 kb region. This locus will be valuable for improving maize variety resistance to MRDD via marker-assisted selection (MAS).

In the containment control problem of multi-agent systems (MASs), the convergence of followers is always a potential threat to the security of system operations. From the perspective of system topology, the inherently non-linear properties of the algebraic connectivity of the follower2follower (F2F) network, combined with the influence of the leader2follower (L2F) topology on the system, make it difficult to design the convergence positions of the followers through mere mathematical analysis. Therefore, in the background of temporary networking tasks for large-scale systems, to achieve the goal of forecasting the performance of the whole system when networking is only completed with local information, this paper investigates the application and effectiveness of recurrent neural networks (RNNs) in the containment control system performance identification, thus improving the efficiency of system networking while ensuring system security. Two types of identification models based on two types of neural networks (NNs), MLP and standard RNN are developed, according to the range of information required for performance identification. Evaluation of the models is carried out by means of the coefficient of determination (R2) as well as the root-mean-square error (RMSE). The results show that each model may produce a better forecasting accuracy than the other models in specific cases, with models based on the standard RNN possessing smaller errors. With the proposed method, model identification can be achieved, but in-depth development of the model in further studies is still necessary to the extent the accuracy of the model.

The pomegranate (Punica granatum L.) is one of the fruit species with the oldest cultural history. There are many traits to determine the quality of pomegranate fruits. Among them, soft-seeded feature of pomegranate fruit is important trait for the market value of the fruit. For this reason, the demand for pomegranate varieties with soft seeds has been increasing, especially in recent years. In this study, molecular markers associated with seed hardness were developed to distinguish pomegranate cultivars with soft-seeded feature based on genomic DNA at the early stages of the pomegranate breeding process. For this purpose, pomegranate genotypes and/or cultivars from the population involved in reciprocal crosses of hard-seeded Ernar, medium-hard-seeded Hicaznar, and soft-seeded Fellahyemez cultivars were grouped as soft-seeded or hard-seeded. Further, leaf samples were collected from individuals belonging to each group. Then, the genomic DNA was isolated from each plant separately, and equal amount of genomic DNA from individuals with the similar seed hardness were mixed for bulked segregant analysis (BSA). The bulked genomic DNAs of opposite characters were analyzed by polymerase chain reaction (PCR) using random decamer primers to develop random amplified polymorphic DNA (RAPD) markers associated with soft-seeded or hard-seeded pomegranates. A total of three RAPD markers were determined to distinguish the individuals having soft- or hard-seeded pomegranate genotypes and/or cultivars. As a result of the comparison of the DNA sequences of these RAPD markers, insertion-deletions (inDels) primers were designed to developed and validate a PCR assay to distinguish the soft- and hard-seeded pomegranate genotypes/cultivars from each other. The molecular markers developed in this study will enable us to distinguish soft-seeded pomegranate types easily in a short time at the early stages of the pomegranate breeding programs.

Vitality is the vigor newborn animals exhibit during the first hours of life. It can be assessed by a numerical score, in which variables, such as heart rate, respiratory rate, mucous membranes’ coloration, time the offspring took to stand up, and meconium staining, are monitored. Vitality can be affected by several factors, and therapies are used to increase it. This manuscript aims to review and analyze pharmacological and physical therapies used to increase vitality in newborn farm animals, as well as to understand the factors affecting this vitality, such as hypoxia, depletion of glycogen, birth weight, dystocia, neurodevelopment, hypothermia, and finally, the physiological mechanism to achieve thermostability. It has been concluded that assessing vitality immediately after birth is essential to determine the newborn’s health and identify those that need medical intervention to minimize the deleterious effect of intrapartum asphyxia. Vitality assessment should be conducted by trained personnel and adequate equipment. Evaluating vitality could reduce long-term neonatal morbidity and mortality in domestic animals, even if it is sometimes difficult with the current organization of some farms. This review highlights the importance of increasing the number of stock people during the expected days of parturitions to reduce long-term neonatal morbidity and mortality, and thus, improve the farm’s performance.

Improving the spectral sensitivity and resolution of biological solids is one of the long-standing problems in nuclear magnetic resonance (NMR) spectroscopy. In this report, we introduce low-power supercycled variants of two-pulse phase-modulated (TPPM) sequence for heteronuclear decoupling. The utility of the sequence is shown by improvements in the transverse relaxation time of observed nuclei (with 1H decoupling) with its application to different samples (uniformly 13C, 15N, 2H-labeled GB1 back-exchanged with 25% H 2 O and 75% D 2 O, uniformly 13C, 15N, 2H-labeled human derived Asyn fibril back-exchanged with 100% H 2 O and uniformly 13C, 15N -labeled human derived Asyn fibril) at fast MAS using low radiofrequency (RF) fields. To understand the effect of spinning speed, the transverse relaxation time is monitored under different spinning frequencies. In comparison to existing heteronuclear decoupling sequences, the supercycled TPPM (sTPPM) sequence significantly improves the spectral sensitivity and resolution and is robust towards B 1 inhomogeneity and decoupler offset. Peanuts (Arachis hypogaea L.) with high oleic acid content have extended shelf life and several health benefits. Oleic, linoleic, and palmitic acid contents in peanuts are regulated by ahFAD2A and ahFAD2B mutant alleles. In the present study, ahFAD2A and ahFAD2B mutant alleles from SunOleic 95R were introgressed into two popular peanut cultivars, GG-7 and TKG19A, followed by markers-assisted selection (MAS) and backcrossing (MABC). A total of 22 MAS and three MABC derived lines were developed with increased oleic acid (78–80%) compared to those of GG 7 (40%) and TKG 19A (50%). Peanut kernel mineral and vitamin composition remained unchanged, while potassium content was altered in high oleic ingression lines. Two introgression lines, HOMS Nos. 37 and 113 had over 10% higher pooled pod yield than respective best check varieties. More than 70% recurrent parent genome recovery was observed in HOMS-37 and HOMS-113 through recombination breeding. However, the absence of recombination in the vicinity of the target locus resulted in its precise introgression along with ample background genome recovery. Convenience food products tend to alter their quality and texture while stored. Texture‐giving food components are often starch‐rich ingredients, such as pasta or rice. Starch transforms depending on time, temperature and water content, which alters the properties of products. Monitoring these transformations, which are associated with a change in mobility of the starch chain segments, could optimize the quality of food products containing multiple ingredients. In order to do so, we applied a simple and efficient in situ 13C solid‐state magic angle spinning (MAS) NMR approach, based on two different polarization transfer schemes, cross polarization (CP) and insensitive nuclei enhanced by polarization transfer (INEPT). The efficiency of the CP and INEPT transfer depends strongly on the mobility of chain segments—the time scale of reorientation of the CH‐bond and the order parameter. Rigid crystalline or amorphous starch chains give rise to CP peaks, whereas mobile gelatinized starch chains appear as INEPT peaks. Comparing 13C solid‐state MAS NMR experiments based on CP and INEPT allows insight into the progress of gelatinization, and other starch transformations, by reporting on both rigid and mobile starch chains simultaneously with atomic resolution by the 13C chemical shift. In conjunction with 1H solid‐state MAS NMR, complementary information about other food components present at low concentration, such as lipids and protein, can be obtained. The genus Aeromonas has been recognised as an important pathogenic species in aquaculture that causes motile Aeromonas septicaemia (MAS). This study was conducted on 224 samples were collected from tilapia (n = 56), water (n = 56), the sediment of the fish farm soil (n= 56), and stool of patients suffering from gastroenteritis (n = 56). A total of 87 positive Aeromonas spp. isolates were recovered from 224 samples with an overall prevalence of 38.8%, distribution of them based on their sources was 55.3% (31/56) from diseased tilapia fish, 21.4% (12/56) from water, 30.3% (17/56) from soil, and 48.2% (27/56) from stool swab of diarrheic patients. The Aeromonas species showed different level of antibiotics susceptibility based on the inhibition zone observed around the antibiotics disc. Aeromonas spp. were all resistant to ampicillin, amoxacillin and pencillin g with an average zone of inhibition of 10mm, 8mm and 9mm respectively but highly susceptible to moxfloxacin, norfloxacin and ciprofloxacin with an average zone of inhibition of 21 mm, 17 mm and 24 mm respectively while they were randomly susceptible to cefotaxime, gentamycin and streptomycin. Therefore, moxfloxacin, norfloxacin and ciprofloxacin are effective drugs that can be used in the treatment of Aeromonas associated infections. PCR using 16S rRNA was used to confirm A. hydrophila positive isolates. Due to Aeromonas is a hard stretched and elastic organism there is a need for antibiotic sensetivity test before the treatment since some strains of the Aeromonas species were randomly susceptible to the some of the antibiotics. Bacterial cold water disease (BCWD) is an important disease in rainbow trout aquaculture. Previously, we have identified and validated two major QTL (quantitative trait loci) for BCWD resistance, located on chromosomes Omy08 and Omy25, in the odd-year Troutlodge May spawning population. We also demonstrated that marker-assisted selection (MAS) for BCWD resistance using the favorable haplotypes associated with the two major QTL is feasible. However, each favorable haplotype spans a large genomic region of 1.3–1.6 Mb. Recombination events within the haplotype regions will result in new haplotypes associated with BCWD resistance, which will reduce the accuracy of MAS for BCWD resistance over time. The objectives of this study were 1) to identify additional SNPs (single nucleotide polymorphisms) associated with BCWD resistance using whole-genome sequencing (WGS); 2) to validate the SNPs associated with BCWD resistance using family-based association mapping; 3) to refine the haplotypes associated with BCWD resistance; and 4) to evaluate MAS for BCWD resistance using the refined QTL haplotypes. Four consecutive generations of the Troutlodge May spawning population were evaluated for BCWD resistance. Parents and offspring were sequenced as individuals and in pools based on their BCWD phenotypes. Over 12 million SNPs were identified by mapping the sequences from the individuals and pools to the reference genome. SNPs with significantly different allele frequencies between the two BCWD phenotype groups were selected to develop SNP assays for family-based association mapping in three consecutive generations of the Troutlodge May spawning population. Among the 78 SNPs derived from WGS, 77 SNPs were associated with BCWD resistance in at least one of the three consecutive generations. The additional SNPs associated with BCWD resistance allowed us to reduce the physical sizes of haplotypes associated with BCWD resistance to less than 0.5 Mb. We also demonstrated that the refined QTL haplotypes can be used for MAS in the Troutlodge May spawning population. Hessian fly [ Mayetiola destructor (Say)] is a major pest of wheat (Triticum aestivum L.) throughout the United States and in several other countries. A highly effective and economically feasible way to control Hessian fly is with resistant cultivars. To date, over 37 Hessian fly resistance genes have been discovered and their approximate locations mapped. Resistance breeding is still limited, though, by the genes' effectiveness against predominant Hessian fly biotypes in a given production area, genetic markers that are developed for low-throughput marker systems, poorly adapted donor germplasm, and/or the inadequacy of closely linked DNA markers to track effective resistance genes in diverse genetic backgrounds. The purposes of this study were to determine the location of the Hessian fly resistance gene in the cultivar "Kelse" (PI 653842) and to develop and validate Kompetitive Allele Specific PCR (KASP) markers for the resistance locus. A mapping population was genotyped and screened for Hessian fly resistance. The resulting linkage map created from 2,089 Single Nucleotide Polymorphism SNP markers placed the resistance locus on the chromosome 6B short arm, near where H34 has been reported. Three flanking SNPs near the resistance locus were converted to KASP assays which were then validated by fine-mapping and testing a large panel of breeding lines from hard and soft wheat germplasm adapted to the Pacific Northwest. The KASP markers presented here are tightly linked to the resistance locus and can be used for marker-assisted selection by breeders working on Hessian fly resistance and allow confirmation of this Hessian fly resistance gene in diverse germplasm. Boll weight (BW) is a key determinant of yield component traits in cotton, and understanding the genetic mechanism of BW could contribute to the progress of cotton fiber yield. Although many yield-related quantitative trait loci (QTLs) responsible for BW have been determined, knowledge of the genes controlling cotton yield remains limited. Here, association mapping based on 25,169 single-nucleotide polymorphisms (SNPs) and 2,315 insertions/deletions (InDels) was conducted to identify high-quality QTLs responsible for BW in a global collection of 290 diverse accessions, and BW was measured in nine different environments. A total of 19 significant markers were detected, and 225 candidate genes within a 400 kb region (± 200 kb surrounding each locus) were predicted. Of them, two major QTLs with highly phenotypic variation explanation on chromosomes A08 and D13 were identified among multiple environments. Furthermore, we found that two novel candidate genes (Ghir_A08G009110 and Ghir_D13G023010) were associated with BW and that Ghir_D13G023010 was involved in artificial selection during cotton breeding by population genetic analysis. The transcription level analyses showed that these two genes were significantly differentially expressed between high-BW accession and low-BW accession during the ovule development stage. Thus, these results reveal valuable information for clarifying the genetic basics of the control of BW, which are useful for increasing yield by molecular marker-assisted selection (MAS) breeding in cotton. Although solid‐state nuclear magnetic resonance (NMR) is a versatile analytical tool to study polymorphs and phase transitions of pharmaceutical molecules and products, this work summarizes examples of spontaneous and unexpected (and unwanted) structural rearrangements and phase transitions (amorphous‐to‐crystalline and crystalline‐to‐crystalline) under magic angle spinning (MAS) conditions, some of them clearly being due to the pressure experienced by the samples. It is widely known that such changes can often be detected by X‐ray powder diffraction (XRPD); here, the capability of solid‐state NMR experiments with a special focus on 1H‐13C frequency‐switched Lee–Goldburg heteronuclear correlation (FSLG HETCOR)/MAS NMR experiments to detect even subtle changes on a molecular level not observable by conventional 1D NMR experiments or XRPD is presented. Furthermore, it is shown that a polymorphic impurity combined with MAS can induce a crystalline‐to‐crystalline phase transition. This showcases that solid‐state NMR is not always noninvasive and such changes upon MAS should be considered in particular when compounds are studied over longer time spans. [ABSTRACT FROM AUTHOR]

Although few in number, studies on epigenome of the heart of COVID-19 patients show that epigenetic signatures such as DNA methylation are significantly altered, leading to changes in expression of several genes. It contributes to pathogenic cardiac phenotypes of COVID-19, e.g., low heart rate, myocardial edema, and myofibrillar disarray. DNA methylation studies reveal changes which likely contribute to cardiac disease through unknown mechanisms. The incidence of severe COVID-19 disease, including hospitalization, requiring respiratory support, morbidity, and mortality, is disproportionately higher in individuals with co-morbidities. This poses unprecedented strains on the global healthcare system. While their underlying conditions make patients more susceptible to severe COVID-19 disease, strained healthcare systems, lack of adequate support, or sedentary lifestyles from ongoing lockdowns have proved detrimental to their underlying health conditions, thus pushing them to severe risk of congenital heart disease (CHD) itself. Prophylactic vaccines against COVID-19 have ushered new hope for CHD. A common connection between COVID-19 and CHD is SARS-CoV-2's host receptor ACE2, because ACE2 regulates and protects organs, including the heart, in various ways. ACE2 is a common therapeutic target against cardiovascular disease and COVID-19 which damages organs. Current and advanced breeding tools are being used to improve economically important horticultural crops to meet the consumers' needs and preferences. Genotyping-by-sequencing (GBS) is an extremely useful tool in the investigation and analysis of the genetic diversity of different cultivars. Based on a broad range of genetic backgrounds like single nucleotide polymorphism (SNPs), GBS is known as a novel technique to facilitate the detection of quantitative trait loci (QTL) regions robustly linked with interested traits compared to genome-wide association study (GWAS) and QTL. GBS has gained popularity among breeders in recent years and it is also employed in cucumber breeding programs. Cucumbers (C. sativus L.) are monoecious, gynoecious and some of them are parthenocarpic species. Cucumber is one of the most economical and essential crops in the Cucurbitaceae family. For time immemorial, cucumber has been produced and consumed all over the world like other cucurbits. To a large extent, cultivated cucurbits are beneficial to human health for providing necessary minerals and fibers. Therefore, this review portrays the current status of advances made by using GBS and its combination with other tools in various studies of cucumber such as the use of GBS and single nucleotide polymorphism (SNP) markers, GBS and GWAS, also with QTL and marker-assisted selection (MAS) are applied to display and detect explicit genetic architecture complex traits in crops and chromosome rearrangements. Cucumber breeding programs have undoubtedly benefited from genotyping-by-sequencing. Using the GBS method, research discovered lots of new candidate genes that control various traits including spine color, fruit stalk-end color, and disease resistance in cucumber lines. Cultivated modern maize (Zea mays L.) originated through the continuous process of domestication from its wild progenitors. Today, maize is considered as the most important cereal crop which is extensively cultivated in all parts of the world. Maize shows remarkable genotypic and phenotypic diversity which makes it an ideal model species for crop genetic research. However, intensive breeding and artificial selection of desired agronomic traits greatly narrow down the genetic bases of maize. This reduction in genetic diversity among cultivated maize led to increase the chance of more attack of biotic stress as climate changes hampering the maize grain production globally. Maize germplasm requires to integrate both durable multiple-diseases and multiple insect-pathogen resistance through tapping the unexplored resources of maize landraces. Revisiting the landraces seed banks will provide effective opportunities to transfer the resistant genes into the modern cultivars. Here, we describe the maize domestication process and discuss the unique genes from wild progenitors which potentially can be utilized for disease resistant in maize. We also focus on the genetics and disease resistance mechanism of various genes against maize biotic stresses and then considered the different molecular breeding tools for gene transfer and advanced high resolution mapping for gene pyramiding in maize lines. At last, we provide an insight for targeting identified key genes through CRISPR/Cas9 genome editing system to enhance the maize resilience towards biotic stress. [ABSTRACT FROM AUTHOR]

Powdery mildew of wheat, caused by Blumeria graminis f. sp. tritici (Bgt), is a devastating disease that seriously reduces yield and quality worldwide. Utilization of plant resistance genes is an attractive and effective strategy for controlling this disease. Among the reported powdery mildew (Pm) resistance genes, Pm2 exhibits a diverse resistance spectrum among its multiple alleles. It has been widely used in China for resistance breeding for powdery mildew. To mine more Pm2 alleles and clarify their distribution, we screened 33 wheat cultivars/breeding lines carrying Pm2 alleles from 641 wheat genotypes using diagnostic and Pm2 -linked markers. To further investigate the relationships within the Pm2 alleles, we compared their resistance spectra, polymorphism of marker alleles and gene sequences, and found that they have identical marker alleles and gene sequences but diverse resistance spectra. In addition, the diagnostic kompetitive allele-specific PCR (KASP) marker, YTU-KASP-Pm2 , was developed and was shown to detect all the Pm2 alleles in the different genetic backgrounds. These findings provide valuable information for the distribution and rational use of Pm2 alleles, push forward their marker-assisted breeding (MAS), and hence improve the control of wheat powdery mildew. A type of boron nitride–magnesium aluminum silicate (BN‐MAS) composite ceramics was fabricated by hot‐press sintering at different sintering temperatures. The relationship between the sintering temperature and microstructure was investigated by analyzing the interaction between hexagonal boron nitride (h‐BN) and the MAS phase. The main MAS phase in the composite ceramics is the α‐cordierite phase at a sintering temperature of 1300°C. At temperatures above 1400°C, the inhibitory effect of h‐BN on the crystallization of the MAS system is significant, and MAS mainly exists in the form of an amorphous phase. The composite sintered at 1700°C exhibited the highest bending strength of 218MPa. h‐BN and MAS were co‐enhanced. MAS can be used as an effective liquid‐phase sintering aid to assist in the sintering of h‐BN, whereas h‐BN can absorb the fracture energy of the composite ceramics through the pull‐out and bridging effect of the particles. Política exterior y diplomacia son, en el marco de los estudios de las Relaciones Internacionales en general, dos categorías que gozan de gran protagonismo. En términos prácticos, poseen una vinculación estrecha, simbiótica e incluso, persiste la idea que ambas significan lo mismo. No obstante, desde una perspectiva teórico-conceptual, la política exterior implica, en líneas generales, los objetivos del Estado en el plano internacional, mientras que la diplomacia refleja los medios para concretarlos. En base a ello, el presente escrito procura identificar y reflexionar sobre los ejes centrales que atravesó la dinámica de la política exterior y la diplomacia de Bolivia durante los tres gobiernos de Evo Morales (2006-2019). Asimismo, se realiza una breve proyección sobre este binomio tan particular a partir del regreso del MAS al poder, tras el triunfo de Luis Arce y David Choquehuanca el 18 de octubre de 2020.

Macrophage activation syndrome (MAS) is also associated with the severity of the disease in SARS-CoV-2-infected patients. Neopterin is a macrophage activation marker produced by monocytes and macrophages upon activation by interferon-gamma (IFN-γ). Neopterin is a well-established marker in a variety of diseases, and recent evidence indicates that it could be helpful in early prediction of the severity of COVID-19 disease and serve as a prognostic marker.
