Abstract

We have measured directly the residual energy of cosmic ray muons crossing the MACRO detector at the Gran Sasso Laboratory. For this measurement we have used a transition radiation detector consisting of three identical modules, each of about 12 m(2) area, operating in the energy region from 100 GeV to 1 TeV, The results presented here were obtained with the first module collecting data for more than two years. The average single muon energy is found to be 320 +/- 4 (stat.) +/- 11 (syst.) GeV in the rock depth range 3000-6500 hg/cm(2). The results are in agreement with calculations of the energy loss of muons in the rock above the detector. (C) 1999 Elsevier Science B.V.

Abstract

The contemporaneous measurement of TeV muons in deep underground laboratories and of the e.m. component at the surface allows checking the hadron interaction models and the propagation codes used in EAS experiments in a primary energy range 10–50 TeV in which the primary spectra are measured by direct experiments. First results of such measurements between MACRO and EAS-TOP at the Gran Sasso laboratory, in this energy range, are here reported.

Abstract

Presently, the only effective treatment for celiac disease is a life-long gluten-free diet. In this work, we used a new mixture of selected sourdough lactobacilli and fungal proteases to eliminate the toxicity of wheat flour during long-time fermentation. Immunological (R5 antibody-based sandwich and competitive enzyme-linked immunosorbent assay [ELISA] and R5 antibody-based Western blot), two-dimensional electrophoresis, and mass spectrometry (matrix-assisted laser desorption ionization-time of flight, strong-cation-exchange-liquid chromatography/capillary liquid chromatography-electrospray ionization-quadrupole-time of flight [SCX-LC/CapLC-ESI-Q-TOF], and high-pressure liquid chromatography-electrospray ionization-ion trap mass spectrometry) analyses were used to determine the gluten concentration. Assays based on the proliferation of peripheral blood mononuclear cells (PBMCs) and gamma interferon production by PBMCs and intestinal T-cell lines (iTCLs) from 12 celiac disease patients were used to determine the protein toxicity of the pepsin-trypsin digests from fermented wheat dough (sourdough). As determined by R5-based sandwich and competitive ELISAs, the residual concentration of gluten in sourdough was 12 ppm. Albumins, globulins, and gliadins were completely hydrolyzed, while ca. 20% of glutenins persisted. Low-molecular-weight epitopes were not detectable by SCX-LC/CapLC-ESI-Q-TOF mass spectrometry and R5-based Western blot analyses. The kinetics of the hydrolysis of the 33-mer by lactobacilli were highly efficient. All proteins extracted from sourdough activated PBMCs and induced gamma interferon production at levels comparable to the negative control. None of the iTCLs demonstrated immunoreactivity towards pepsin-trypsin digests. Bread making was standardized to show the suitability of the detoxified wheat flour. Food processing by selected sourdough lactobacilli and fungal proteases may be considered an efficient approach to eliminate gluten toxicity.

Abstract

""The full coverage extensive air shower detector ARGO-YBJ, located in Tibet at 4300 m of altitude, has monitored the sky at gamma ray energy E>0.6TeV in the declination band from -10° to 70°. In 424 days the Crab Nebula and Mrk421 have been detected with a significance, respectively, of 7.0 and 8.0 standard deviations. The analysis of the cosmic ray background in the same sky band, has revealed the existence of a significant excess of the cosmic ray flux in two localized regions of angular size 10°30°, confirming previous indications. The origin of such excesses is still unexplained. Concerning the gamma ray sources, the measured spectrum of the Crab Nebula is dN\\\/dE=(3.7±0.8)×10-11E-2.67±0.25photonscm-2s-1TeV-1, in agreement with other experiments. During 2008 the observed Mrk421 flux was highly variable, with the strongest flares in MarchJune, in good correlation with X-ray data. One of the most intense flares occurred in the first half of June and has been deeply studied by different detectors in the energy range from optical to 100 MeV gamma rays, but only partially up to TeV energies, since the moonlight hampered the Cherenkov telescope measurements during the second and most intense part of the emission. Our data complete these observations, with the detection of a signal of intensity of about 7 Crab units on June 1113, with a statistical significance of 4.2 standard deviations. The observed flux is consistent with a prediction made in the framework of the Synchrotron Self-Compton model, in which the flare is caused by a rapid acceleration of leptons in the jet. ""

Abstract

The analysis of the e.m. component of EAS and of the deep underground muons detected in coincidence by EAS-TOP and MACRO has been performed to study the primary cosmic ray composition in the energy range 5 · 1014 − 1 · 1016eV, around the knee of the primary spectrum. New results are presented with improved statistics, mostly above the knee of the energy spectrum.

Abstract

This Technic Note is an integration of the previous study: "Climatic chambm for plant physiology: a new project concept". This note gives details regarding the planning of the lamp arrangement and dmonseates how by mixing, in appropriate quantities, the radiative range of various types of lamps can give apparentely contrasting results: - to maximize radiation in maximum absorption range of chlorophylls and carotenes; -to minimize heat emission in the climatic chamber. With nine day-light fluorescent tubes , four sun-light metallic halide and spotlights nine red-blue fluorescent tubes, for a total of 562 W m^-2(electric), mounted on the ceiling of a 2 m high chamber with 4 m^2 surface area, on the chamber floor about 130 W m^-2 "total Solar radiation equivalent" was obtained. This means the power emitted in the bands of clorophylls and carotenoids absorption, from a total solar radiation (black body of 5500 K) of about 130 W m^-2. This radiation is sufficient to grow a large number of plant species. In the lamp arrangement there are seven other light fixtures, for fluorescent tubes, defined "auxiliary" because tubes can be inserted which either integrate active radiation on the photoreceptors or produce particular spectral ranges. In the above cited work, fluorescent tubes producing in the ultraviolet B range were mounted in these auxiliary fixtures. Less thermal energy emitted in the climatic chamber means that it is possible to use a less powerful conditioning system and, thus, have lower costs of set-up and management. The efficiency of the lighting system is demonstrated by the fact that during 15 days of experimentation on 18-month-old, potted poplar plants (Populus Alba), symptoms of insufficient light were not detected.

217. An analytical method for the characterization of microclimates for the conservation of wooden Cultural Heritage objects

Abstract

Most of the museum conservation standards are based on "static" criteria, specifying "acceptable" or "optimal" ranges for Temperature and Relative Humidity for the welfare of the artefacts to be conserved; no clear specifications are usually given for their evolution in time. Such approach appears unsatisfactory for artefacts made of wood, which not only is highly hygroscopic, but also conserves a "memory" of the climate to which it has been previously exposed. The work reported in this paper is carried out in the framework of studies aiming towards more effective specifications. It proposes a dynamic method for characterizing real microclimates from the point of view of the conservation of wooden artefacts belonging to the Cultural Heritage; such characterization considering the environmental parameters which may cause biological decay and/or mechanical damage caused, at equilibrium or during transients, by shrinkage/swelling and the connected distortions and/or stresses. The proposed method is based on analytical algorithms, to be applied to recorded or specified climatic data. Firstly the variation in time of typical EMC (Equilibrium Moisture Content) of wood is computed, then the resulting time series is processed in order to compute 1 DISTAF University of Florence 2 CNR-IBIMET Laboratory for Monitoring of Agroecosystems - Sassari. 2 the number, duration and size of periods during which EMC remains stable or changes. Computation is performed using several values of the two parameters ? (defining the stability criterion) and Tmin (defining the minimum duration for any period to be considered significant). The proposed method itself can not identify if given environmental conditions may in general be harmful for the wooden artworks; however, on the basis of the characterization that it provides, experts in conservation may express objective judgments/evaluations about its suitability for individual artworks. In this paper two very different climatic conditions have also been analysed using the proposed method: one is the rather stable indoor environment of a monumental hall in a Middle Age building, located in the centre of Florence (Italy); the second one is a quite unstable outdoor environment, also in the Florence area. The results of the analysis highlight objectively strong differences between the two environments; however, the proposed method is still at an initial stage, and needs to be reviewed and refined in order to provide more synthetic and significant indexes.

227. Proteolysis by sourdough lactic acid bacteria: effects on wheat flour protein fractions and gliadin-peptides involved in human cereal intolerance

Abstract

Sourdough lactic acid bacteria were preliminarily screened for proteolytic activity by using a digest of albumin and globulin polypeptides as a substrate. Based on their hydrolysis profile patterns, Lactobacillus alimentarius 15M, Lactobacillus brevis 14G, Lactobacillus sanfranciscensis 7A, and Lactobacillus hilgardii 51B were selected and used in sourdough fermentation. A fractionated method of protein extraction and subsequent two-dimensional electrophoresis were used to estimate proteolysis in sourdoughs. Compared to a chemically acidified (pH 4.4) dough, 37 to 42 polypeptides, distributed over a wide range of pIs and molecular masses, were hydrolyzed by L. alimentarius 15M, L. brevis 14G, and L. sanfranciscensis 7A. Albumin, globulin, and gliadinfractions were hydrolyzed, while glutenins were not degraded. The concentrations of free amino acids, especially proline and glutamic and aspartic acids, also increased in sourdoughs. Compared to the chemically acidified dough, proteolysis by lactobacilli positively influenced the softening of the dough during fermentation, as determined by rheological analyses. Enzyme preparations of the selected lactobacilli which contained proteinase or peptidase enzymes showed hydrolysis of the 31-43 fragment of A-gliadin, a toxic peptide for celiac patients. A toxic peptic-tryptic (PT) digest of gliadins was used for in vitro agglutination tests on K 562 (S) subclone cells of human myelagenous leukemia origin. The lowest concentration of PT digest that agglutinated 100% of the total cells was 0.218 g/liter. Hydrolysis of the PT digest by proteolytic enzymes of L. alimentarius 15M and L. brevis 14G completely prevented agglutination of the K 562 (S) cells by the PT digest at a concentration of 0.875 g/liter. Considerable inhibitory effects by other strains and at higher concentrations of the PT digest were also found. The mixture of peptides produced by enzyme preparations of selected lactobacilli showed a decreased agglutination of K 562 (S) cells with respect to the whole 31- 43 fragment of A-gliadin.
