Background Epidemiological studies have reported lower risk of cardiovascular disease with moderate coffee consumption. In addition, emerging evidence indicates that consumption of coffee beverages enriched in chlorogenic acids (CGAs) may influence blood pressure and endothelial function, suggesting that the beneficial cardiovascular effect of coffee may relate to its CGA content. Objectives We conducted a double-blind randomized crossover trial to test the effect of acute consumption of a decaffeinated green coffee extract (DGCE), rich in CGAs, on endothelial function in healthy subjects. Methods We compared 3 different doses of DGCE (302, 604, and 906 mg, respectively) with a placebo. Endothelial function was defined as the percentage change in the internal diameter of the brachial artery in response to flow-mediated dilation (%FMD). In addition, we followed the plasma concentration-time profiles of 25 systemic CGA metabolites over 24 h after DGCE consumption and we explored the relation between systemic concentrations of CGAs and the effect on %FMD. Results The DGCE formulations containing different amounts of CGAs resulted in dose-proportional increases in overall total polyphenol concentrations. The systemic appearance of total CGAs was biphasic, in agreement with previous results suggesting 2 sites of absorption in the gastrointestinal tract. Compared with the placebo group, a significant FMD increase (>1%) was observed 8.5, 10, and 24 h after consumption of 302 mg DGCE (∼156.4 mg CGAs). The differences with placebo observed in the other 2 groups were not statistically significant. Evaluation of the relation between phenolic exposure and %FMD showed a positive tendency toward a larger effect at higher concentrations and different behavior of CGA metabolites depending on the conjugated chemical position. Conclusions We demonstrated an acute improvement in %FMD over time after ingestion of a DGCE, explained at least partly by the presence in the blood circulation of CGAs and their metabolites. To gain comprehensive insight into the interactions of key coffee odorants, like the Strecker aldehydes, acetaldehyde, propanal, methylpropanal, 2- and 3-methylbutanal, and methional, and the nonvolatile fraction of coffee, an untargeted metabolomics approach was applied. Ultra performance liquid chromatography (UPLC)-time of flight (TOF)-mass spectrometry (ESI-) profiling followed by statistical data analysis revealed a marker substance for a coffee beverage spiked with acetaldehyde with an accurate mass of 217.0703 [M - H]-. This compound could be identified as a reaction product of quinic acid (QA) and acetaldehyde linked by acetalization at the cis-diol function of QA. Consequently, the acetalization of aldehydes, QA, 5-O-caffeoyl quinic acid (CQA), and quinic acid γ-lactone (QAL) was investigated by means of model reactions, followed by synthesis, isolation, and structure elucidation via UPLC-TOF-MS and 1D and 2D NMR techniques. UHPLC-MS/MSMRM screening and the quantification of aldehyde adducts in coffee beverages revealed the presence of QA/acetaldehyde, -/propanal, -/methylpropanal, and -/methional reaction products and CQA/acetaldehyde, -/propanal, -/methylpropanal, -/2- and 3-methylbutanal, and -/methional and QAL/acetaldehyde adducts for the first time, in concentrations of 12-270 μg/L for QA/aldehydes, 5-225 μg/L for CQA/aldehydes, and 62-173 μg/L for QAL/acetaldehyde. The transition of vegetative meristems to a reproductive state followed by floral development and flowering is a complex process regulated by many regulatory pathways that integrate (epi) genetic and environmental factors to guarantee the reproductive success in angiosperms. In woody and perennial species, such as coffee trees, the extended life cycle reflects adaptive strategies, i.e., two-year cycle, sequential flowering and bud latency during the winter, that ensure reproductive success in a variable environment. The dynamics of coffee flowering integrate multiple stimuli to induce morpho-physiological changes, especially during the reproductive phase. We discussed diverse stimuli which are directly and indirectly associated with molecular pathways related to the reproductive meristem transition and floral development until anthesis. Here, we propose to redefine some concepts regarding coffee floral development, such as: i) a summary of genes possibly involved in the flowering pathways; ii) a new classification of buds by position in the node (B1 - B4) to avoid misunderstandings with the uneven developmental stages; iii) comparative ABC model to Arabidopsis and tomato to allow evolutionary discussions of whorls development in Coffea sp. From this, we expect to collaborate to drive future studies towards coffee breeding and production, for example, improving floral bud activation, controlling the flowering time, anthesis synchronization to produce more homogeneous ripening fruits, reducing abortion or early flower openings, and increasing fruitification. Moreover, we think that the explored relationship between physiological and molecular approaches during the coffee flowering may be extended to understand other tropical perennial species, especially those with asynchronous flowering behavior, and to understand evolutive aspects in Rubiaceae.

The production of sustainable coffee (Coffea sp.) has become an alternative to add value to coffee production, reaching new world markets and increasing the farmer's profitability. Biostimulants have been presented as a tool to increase coffee production. The hypothesis is that the use of biostimulants improves the germination rate and the development of coffee seedlings. This study aims to compare and evaluate the application of biostimulants in the initial development of coffee seedlings. An experiment was carried out in Monte Carmelo, Minas Geras. Applications of biostimulants (i) 4-indole-3-ylbutyric acid + gibberellic acid + cinetina, AIA+AG+C; (ii) foliar fertilizer + algae, F+A; (iii) vegetal extracts + gibberellic acid, EV+AA were carried out on coffee seedlings. After 94 days of applying the biostimulants, monitored the length of the aerial part, the number of leaves, stem diameter, root biomass (pivoting, auxiliary and total). Results showed that the application of F+A promoted the greatest length of the aerial part, the number of leaves, and biomass of the roots. The application of AIA+AG+C showed the same performance as F+A in the number of leaves of the coffee seedlings. Given the results, we concluded that the application of biostimulants based on mixed foliar fertilizer + algae demonstrated to be a better alternative to increase the development of the aerial and root part of coffee seedlings. Effluents from wet processing of coffee industries pollute water bodies since the wastewater is released to the water bodies' vicinity. The wastes water from these industries affects the surface and groundwater. The study was initiated to treat wastewater effluent coming from the coffee industry in the specific area by using an anaerobic mixed culture of Pseudomonas fluorescence and Escherichia coli bacteria. The treatment reduced BOD5, COD, and TS from its initial concentration of 320.26 mg/l, 1261 mg/l, and 3545 mg/l to 58.37 mg/l, 152 mg/l, and 1198 mg/l, respectively. The study's treatment had reduced the BOD5 load up to 81.67%, the COD load up to 87.94%, and TS load up to 33.79% from the initial load using optimization of the treatment method. Therefore, it can be concluded that mixed culture bacteria of P. fluorescence and E. coli as a new effective treatment having the potential for BOD5, COD, and TS reduction from the effluent. The mixed culture of microorganisms can reduce the contaminants that made the approach cost-effective, time-saving compared with other results found by other studies without mixed culture. The microorganisms included in the study can be applied for the treatment of effluent containing multiple contaminants.

Background: Foodborne diseases are caused by acquired pathogenic bacteria such as Salmonella enteritidis. It causes an intestinal imbalance and the microbial toxins found in the gastrointestinal tract induce symptoms such as diarrhea. Coffee contains active ingredients such as antioxidants and is used as an anti-inflammatory agent by reducing pro-inflammatory cytokine levels in the body. Aim: The purpose of this study was to determine the interaction between Lampung’s robusta coffee and tissue damage in chickens infected by S. enteritidis. Methods: This study used first-day-old Isa brown layer chickens (n = 60), which were divided into five treatment groups. The negative control group consisted of healthy and normal chickens, whereas the positive control group consisted of chickens infected with S. enteritidis bacteria at a concentration of 108 CFU/ml. Groups T1, T2, and T3 were given coffee extract with doses of 500 mg/kg BW (low dose), 1,000 mg/kg BW (moderate dose), and 1,500 mg/kg BW (high dose), respectively, and then infected with S. enteritidis bacteria at a concentration of 108 CFU/ml. The coffee extract and bacteria were given orally via a feeding tube at a volume of 0.5 ml per chick. The extract was given for 14 days (from day 3 to day 16), and the bacteria were given on days 16 and 17. On day 18, the chickens were necropsied. Due to the importance in the socioeconomic role of coffee culture for the region, the study aimed to map the coffee culture of the Caparaó Capixaba Territory and to evaluate the dynamics of coffee cultivation in the period from 2007 to 2015. Photos from the orthophotomosaic of the state of Espírito Santo, cut to the Territory, with spatial resolution of 1 m for 2007 and 0.25 m for 2015, on the cartographic scale of 1: 35,000. The photointerpretation was performed using a computational application on a scale of 1: 2,000, with error checking using the Kappa Index. To position the cultivation area to the type of coffee between arabica and conilon, the zoning of agroclimatic suitability, restriction and inaptitude was carried out, and later involving the altitude, according to criteria adopted by the state agency. From the zoning point of view, the photointerpretation of the 2007 and 2015 cultivation area was interpolated, making it possible to compare them. It was found that the total occupation area was 16.37% (2007), 15.82% (2015) with 8.78% of common area. Regarding the total planting area of conilon and arabica, there was a reduction in the cultivation of both species of 11.51 and 7.10%, respectively. Geotechnology was of great relevance to the present work, in the activity of planning and administration of agribusiness. Em virtude da importância no papel socioeconômico da cultura cafeeira para a região, o estudo teve como objetivo mapear a cafeicultura do Território do Caparaó Capixaba e avaliar a dinâmica de ocupação do cultivo de café no período de 2007 a 2015. Foram utilizadas fotos do ortofotomosaico do estado do Espírito Santo, recorte para o Território, com resolução espacial de 1 m para 2007 e 0,25 m para 2015, na escala cartográfica de 1:35.000. A fotointerpretação foi realizada aplicativo computacional na escala de 1:2.000, com checagem do erro pelo Índice Kappa. A fim de posicionar a área de cultivo ao tipo de café entre arábica e conilon, foi realizado o zoneamento de aptidão, restrição e inaptidão Agroclimática, e posteriormente envolvendo a altitude, conforme critérios adotados pelo órgão estadual. De pose do zoneamento, foi interpolada a fotointerpretação da área de cultivo de 2007 e 2015, possibilitando sua comparação. Detectou-se que a área total de ocupação do foi de 16,37% (2007), 15,82% (2015), sendo 8,78% de área comum. Em relação a área de plantio total de conilon e arábica, verificou-se redução no cultivo de ambas as espécies de 14,59 e 6,72 km², respectivamente. A geotecnologia foi de grande relevância para o presente trabalho, na atividade de planejamento e administração do agronegócio.

Background The chemical compounds in coffee are important indicators of quality. Its composition varies according to several factors related to the planting and processing of coffee. Thus, this study proposed to use near-infrared spectroscopy (NIR) associated with partial least squares (PLS) regression to estimate quickly some chemical properties (moisture content, soluble solids, and total and reducing sugars) in intact green coffee samples. For this, 250 samples produced in Brazil were analyzed in the laboratory by the standard method and also had their spectra recorded. Results The calibration models were developed using PLS regression with cross-validation and tested in a validation set. The models were elaborated using original spectra and preprocessed by five different mathematical methods. These models were compared in relation to the coefficient of determination, root mean square error of cross-validation (RMSECV), root mean square error of test set validation (RMSEP), and ratio of performance to deviation (RPD) and demonstrated different predictive capabilities for the chemical properties of coffee. The best model was obtained to predict grain moisture and the worst performance was observed for the soluble solids model. The highest determination coefficients obtained for the samples in the validation set were equal to 0.810, 0.516, 0.694 and 0.781 for moisture, soluble solids, total sugar, and reducing sugars, respectively. Conclusion The statistics associated with these models indicate that NIR technology has the potential to be applied routinely to predict the chemical properties of green coffee, and in particular, for moisture analysis. However, the soluble solid and total sugar content did not show high correlations with the spectroscopic data and need to be improved. © 2020 Society of Chemical Industry.

Background External preference mapping is a powerful tool to explain consumer preference or rejection. Combining the technique for order of preference by similarity to ideal solution (TOPSIS) multicriteria analysis with rapid descriptive techniques can improve preference map (PREFMAP) results. This study was conducted to compare the PREFMAPs generated with rapid descriptive flash profile (FP), check-all-that-apply (CATA), and Napping® versus PREFMAPs constructed with FP-TOPSIS, CATA-TOPSIS, and Napping-TOPSIS. Results Only 38.46%, 63.66%, and 42% of sensory attributes initially generated by FP, CATA, and Napping techniques respectively were considered for the determination of their weight W and allocation as positive or negative in the TOPSIS technique. The PREFMAPs constructed with FP-TOPSIS, CATA-TOPSIS, and Napping-TOPSIS presented a better explanation of the preference and rejection than the PREFMAPs directly generated with rapid sensory techniques. The results of the multiple factor analysis and coefficient Rv indicated similarities in the sensory vocabularies used after the TOPSIS technique. Conclusion The combination of the TOPSIS technique with rapid sensory techniques is a reliable alternative for the construction of PREFMAPs in order to identify the sensory attributes responsible for preference and rejection of food products. © 2020 Society of Chemical Industry.

Transplant experiments, functional genomics and population genomics are complementary tools to efficiently characterize heritable phenotypic traits and to decipher the genetic bases of adaptive traits. Using an integrative approach combining phenotypic assessment in common garden, transcriptomics and landscape genomics, we investigated leaf adaptive traits in Coffea mauritiana, a forest tree endemic to Reunion Island. Eight populations of C. mauritiana originating from sites with contrasted environmental conditions were sampled in common garden to assessseveral leaf morphological traits, to analyze the leaf transcriptome and leaf cuticular wax composition. The relative alkane content of cuticular waxes was significantly correlated with major climatic gradients, paving the way for further transcriptome-based analyses. The expression pattern of cuticle biosynthetic genes was consistent with a modulation of alkane accumulation across the population studied, supporting the hypothesis that the composition of cuticular wax is involved in the local adaptation of C. mauritiana. Association tests in landscape genomics performed using RNA-seq-derived single-nucleotide polymorphisms revealed that genes associated with cell wall remodeling also likely play an adaptive role. By combining these different approaches, this study efficiently identified local adaptation processes in a non-model species. Our results provide the first evidence for local adaptation in trees endemic to Reunion Island and highlight the importance of cuticle composition for the adaptation of trees to the high evaporative demand in warm climates.

Growing water restrictions associated with climate changes constitute daunting challenges to crop performance. This study unveils the impacts of moderate (MWD) or severe (SWD) water deficit, and their interaction with air [CO2], on the photosynthetic apparatus of Coffea canephora cv. Conilon Clone 153 (CL153) and C. arabica cv. Icatu. Seven year-old potted plants grown under 380 (aCO2) or 700μLL−1 (eCO2) [CO2] gradually reached predawn water potentials between −1.6 to −2.1MPa (MWD), and below −3.5MPa (SWD). Under drought, stomata closure was chiefly related to ABA rise. Increasing drought severity progressively affected gas exchange and fluorescence parameters in both genotypes, with non-stomatal limitations becoming gradually dominating, especially regarding the photochemical and biochemical components of CL153 SWD plants. In contrast, Icatu plants were highly tolerant to SWD, with minor, if any, negative impacts on the potential photosynthetic functioning and components (e.g., Amax, Fv/Fm, electron carriers, photosystems (PSs) and RuBisCO activities). Besides, drought-stressed Icatu plants displayed increased abundance of a large set of proteins associated with the photosynthetic apparatus (photosystems, light harvesting complexes, cyclic electron flow, RuBisCO activase) regardless of [CO2]. Single eCO2 did not promote stomatal and photosynthetic down-regulation in both genotypes. Instead, eCO2 increased photosynthetic performance, moderately reinforced photochemical (PSs activity, electron carriers) and biochemical (RuBisCO, Ru5PK) components, whereas photoprotective mechanisms and protein abundance remained mostly unaffected. In both genotypes, under MWD, eCO2 superimposition delayed stress severity and promoted photosynthetic functioning with lower energy dissipation and PSII impacts, whereas stomatal closure was decoupled from increases in ABA. In SWD plants most impacts on the photosynthetic performance were reduced by eCO2, especially in the moderately drought affected CL153 genotype, although maintaining RuBisCO as the most sensitive component, deserving special breeder’s attention to improve coffee sustainability under future climate scenarios.

The wild diversity of the Coffea genus is large, which is valuable for coffee plant breeding programs intended to obtain higher sensory quality varieties; but a biochemical characterization of accessions, already available in coffee germplasm banks, is still needed. In this work, a quantitative determination of caffeine, trigonelline and 5-caffeoylquinic acid (5-CQA), in coffee leaves of 80 different Coffea spp. accessions from the Coffee Germplasm Bank located in Huatusco, Veracruz, Mexico was done. These metabolic variables might be useful as quality-related selection markers in future coffee breeding programs. Fresh coffee leaves from each accession were processed and metabolites determined by UPLC-MS/MS analyses. Multidimensional Scaling (MDS), Hierarchical Cluster Analysis (HCA) and Analysis of Variance (AOV) revealed that levels of caffeine and trigonelline in leaves significantly varied among accessions, from 0.00 to 2.04% (w/w) and from 0.42 to 1.82% (w/w), respectively. While 5-CQA ranged from 0.00 to 0.66%. In C. liberica no caffeine was detected, demonstrating that coffee leaves have the potential to be used in the genetic profiling of Coffea spp. These results could be useful in formulating a proposal for intraspecific crosses in C. arabica, for the generation of F1 hybrids with good cup quality attributes, while in C. canephora they could be helpful for enhancing sensory quality traits of Robusta coffees.

BACKGROUND Natural environments within agricultural landscapes have been recognized as reservoirs of biodiversity and, therefore, providers of fundamental ecosystem services to human beings. Bees are the main providers of pollination and thus contribute to the production of food consumed worldwide. In this work, we evaluated the distribution of bees in an agricultural landscape of coffee plantation before and after coffee flowering. We aimed at understanding how richness, abundance and composition of bee communities vary among the different vegetation types within and around the coffee crops. RESULTS A total of 638 bees were collected - 312 in the dry season and 326 in the rainy season - totaling 85 species. The sampling methods collected different species, which provided complementary sampling. Only Euglossa leucotricha and Eulaema nigrita were recurrent in both seasons and vegetation types. There was no temporal difference in richness or abundance; however, both varied in relation to the vegetation type and were higher in the coffee-native transition area. Diverging from richness or abundance, the composition of the communities differed regarding season and vegetation types. CONCLUSION We reinforce the importance of maintaining native vegetation in areas surrounding coffee plantations since the crop poorly hosts pollinators when it is not flowering. Natural and semi-natural areas may act as reservoirs of floral visitors, thus maintaining potential cross-pollination services available to coffee production. © 2020 Society of Chemical Industry.

During the Cold War, coffee became a strategically important crop in the global contest between the United States and the Soviet Union. The economies of many US allies in Latin America depended upon coffee. In the Cold War context, then, the coffee leaf rust ( Hemileia vastatrix) became a geopolitical problem. Coffee experts in Latin America, which produced most of the world's coffee, began to prepare for an outbreak. In the 1950s, they built a global network of coffee experts. This network was sustained by US-led Cold War programs that promoted technical collaboration across the Global South, such as Harry Truman's Point Four programs. We explore the network's growth and evolution through one of its central figures, the American plant pathologist Frederick L. Wellman. This network has survived the end of the Cold War and evolved to reflect the new geopolitical context.

The coffee berry borer (CBB) is one of the main coffee pests in the world including Colombia. This pest is difficult to manage because of its cryptic habits and the continuous availability of coffee fruits. Among the new management strategies being tested is the use of volatile compounds as insect repellents. In this work, the behavioral response of female adult CBBs to terpenes previously identified in the CBB-repellent plant species Lantana camara was evaluated. α-Terpinene, (R)-limonene, farnesene and β-caryophyllene terpenes were tested via a Y-tube olfactometer in which ripe coffee fruits were accompanied by terpenes at concentrations between 25 and 200 ppm. Only β-caryophyllene induced a significant and consistent CBB repellent effect at all tested doses. The protective effect of microencapsulated β-caryophyllene was then determined under laboratory conditions by incorporating the terpene in a colloidosome-gel system at 2.8 × 105 ng/h in the middle of coffee fruits with adult CBBs. The coffee fruits in turn presented a decrease in fruit infestation. Furthermore, the protection of coffee fruits when β-caryophyllene gels were hung in coffee trees was evaluated in the field; infestations were artificially induced by the use of raisins (CBB-infested old coffee fruits) placed on the ground. Compared with unprotected trees, the trees treated with caryophyllene gels exhibited a 33 to 45% lower degree of infestation. Taken together, the results show that β-caryophyllene is a promising compound for an integrated pest management (IPM) program in commercial coffee plantations.

BACKGROUND: Coffee and tea are the major contributors of caffeine in the diet. Evidence points to the premise that caffeine may benefit cognition. OBJECTIVE: We examined the associations of habitual regular coffee or tea and caffeine intake with cognitive function whilst additionally accounting for genetic variation in caffeine metabolism. METHODS: We included white participants aged 37–73 y from the UK Biobank who provided biological samples and completed touchscreen questionnaires regarding sociodemographic factors, medical history, lifestyle, and diet. Habitual caffeine-containing coffee and tea intake was self-reported in cups/day and used to estimate caffeine intake. Between 97,369 and 445,786 participants with data also completed ≥1 of 7 self-administered cognitive functioning tests using a touchscreen system (2006–2010) or on home computers (2014). Multivariable regressions were used to examine the association between coffee, tea, or caffeine intake and cognition test scores. We also tested interactions between coffee, tea, or caffeine intake and a genetic-based caffeine-metabolism score (CMS) on cognitive function. RESULTS: After multivariable adjustment, reaction time, Pairs Matching, Trail Making test B, and symbol digit substitution, performance significantly decreased with consumption of 1 or more cups of coffee (all tests P-trend < 0.0001). Tea consumption was associated with poor performance on all tests (P-trend < 0.0001). No statistically significant CMS × tea, CMS × coffee, or CMS × caffeine interactions were observed. CONCLUSIONS: Our findings, based on the participants of the UK Biobank, provide little support for habitual consumption of regular coffee or tea and caffeine in improving cognitive function. On the contrary, we observed decrements in performance with intakes of these beverages which may be a result of confounding. Whether habitual caffeine intake affects cognitive function therefore remains to be tested.

Aceh is one of the biggest coffee producers in Indonesia’s province. The coffee farmers separate the coffee beans with the flesh of the fruit for processing the seeds, while the coffee fruit is considered as waste used as animal feed. Based on existing research, the coffee flesh produced contains polyphenol compounds as antibacterial. A very promising sea product from Aceh is seaweed. Brown seaweed (Sargassum sp.) is very common in the west coast of Aceh province. One of the derivatives is alginate. In this study, a combination of polyphenols extracts from coffee flesh and alginates from seaweed Sargassum sp. used as a basic ingredient in making face masks. Tests on organoleptic observations, pH testing, testing time of dried preparations, and testing of bacterial activity have been carried out in this study. In addition, variations in the mixture preparation based on the particle size of alginate powder have also been carried out in this study. The results obtained indicate that the yield produced from coffee fruit extracts is 5.86%, the highest yield of alginate is by potassium carbonate of 40.77%, the acidity of the combination is 4.5 - 6.5. For preparations, drying time has an average estimated time of approximately 6 minutes. Bacterial activity test showed that K2CO3 extracting agent concentration of 2% was strong against bacterial growth with a clean area of ​​14 mm. With these results, it can be seen the combination is able to inhibit the rate of bacterial growth, so it is recommended for alternative ingredients in the manufacture of cosmetics pharmaceutical industry masks.

Green tea and coffee contain various bioactive compounds (e.g., polyphenols), and their consumption has been proposed to decrease the risk of cardiovascular diseases. Here, we investigated the associations between the consumption of green tea and that of coffee and the prevalence of coronary artery disease (CAD) in Japanese patients. The study group was 612 patients who underwent coronary angiography at Tokyo Medical Center between July 2008 and February 2017. CAD was confirmed in 388 of the patients: one-vessel disease (1-VD, n=166); two-vessel disease (2-VD, n=112); three-vessel disease (3-VD, n=110). Myocardial infarction (MI) was found in 138 patients. After adjustment for well-known atherosclerotic risk factors and other dietary habits, greater green tea consumption was significantly inversely associated with CAD prevalence (p for trend=0.044), and the patients who drank3 cups/d had a lower prevalence of CAD compared to those who drank1 cup/d (odds ratio [OR]: 0.54, 95% CI: 0.30-0.98). Greater green tea consumption (3 cups/d) was also associated with a decreased prevalence of 3-VD (OR: 0.49, 95% CI: 0.24-0.98, p-trend=0.047) and MI (OR: 0.51, 95% CI: 0.27-0.97, p-trend=0.037). In contrast, coffee consumption was not associated with CAD or MI. In subgroup analyses, the inverse association between green tea consumption and CAD or MI was found in the high intake groups of vegetables or fruits but not in the low intake groups of vegetables or fruits. These results suggest a beneficial effect of green tea consumption, especially with a diet rich in vegetables and fruits, against coronary atherosclerosis in Japanese.

Coffee is one of the most consumed hot beverages worldwide and is highly regarded because of its stimulating effect despite having a pronounced bitterness. Even though numerous bitter ingredients have been identified, the detailed molecular basis for coffee's bitterness is not well understood except for caffeine, which activates five human bitter taste receptors. We elucidated the contribution of other bitter coffee constituents in addition to caffeine with functional calcium imaging experiments using mammalian cells expressing the cDNAs of human bitter taste receptors, sensory experiments, and in silico modeling approaches. We identified two human bitter taste receptors, TAS2R43 and TAS2R46, that responded to the bitter substance mozambioside with much higher sensitivity than to caffeine. Further, the structurally related bitter substances bengalensol, cafestol, and kahweol also activated the same pair of bitter taste receptors much more potently than the prototypical coffee bitter substance caffeine. However, for kahweol, a potent but weak activator of TAS2R43 and TAS2R46, we observed an inhibitory effect when simultaneously applied together with mozambioside to TAS2R43 expressing cells. Molecular modeling experiments showed overlapping binding sites in the receptor's ligand binding cavity that suggest that the partial agonist kahweol might be useful to reduce the overall bitterness of coffee-containing beverages. Taken together, we found that the bitterness of coffee is determined by a complex interaction of multiple bitter compounds with several human bitter taste receptors.

Coffee husk, eucalyptus, and pine residues were torrefied at 290 °C in a screw reactor, during 5, 10, 15 or 20 min. The effects of feedstock type and torrefaction process parameters (holding time) on their energy characteristics were investigated. Raw and torrefied biomasses were then submitted successively to leaching and to white and brown rot fungi, to mimic storage conditions. Mass loss after leaching step, water content and weight loss due to fungal deterioration after 2, 4, 8, 12, 16 weeks were recorded. The chemical composition and high heating value (HHV) of the torrefied samples were measured to determine the alterations compared to raw biomass during their storage. Increasing torrefaction residence time improves the decay resistance of the biomasses. Variation of carbon content (%wt., dry basis) and HHV (kJ/kg, dry basis) were observed during native and torrefied biomasses fungal degradations. Carbon contents and HHV values of raw and torrefied biomasses decreased during Trametes versicolor exposure [49.65% > C > 44.07% and 19.71 kJ/kg > HHV > 17.19 kJ/kg, results from results from all tests combined.], whereas they increased during exposure to Coniophora puteana [46.15%

The Coffea genus includes about 124 species, of which Coffea arabica L. and Coffea canephora Pierre ex A. Froehner are the most commercially important, supplying 63 and 37% of the world coffee production, respectively. C. arabica L. is associated with better cup quality, but presents serious problems with pests and diseases, potentiated by climate change. The development of new varieties of C. arabica L. requires the inclusion of genetic diversity, looking for incorporating new genes to tolerate or resist the most important pests and diseases, maintaining or improving yield and cup quality of the new materials. The technology of diversity arrays based on sequencing (DArTseq™) allows the rapid identification of the single nucleotide polymorphism (SNP) for a genomic approach of genetic diversity. The production of high-quality coffee is being challenged by changing climates in coffee-growing regions. The coffee beans from the upper and lower canopy at different development stages of the same plants were analyzed to investigate the impact of the microenvironment on gene expression and coffee quality. Compared with coffee beans from the upper canopy, lower canopy beans displayed more intense aroma with higher caffeine, trigonelline, and sucrose contents, associated with greater gene expression in the representative metabolic pathways. Global gene expression indicated a longer ripening in the lower canopy, resulting from higher expression of genes relating to growth inhibition and suppression of chlorophyll degradation during early bean ripening. Selection of genotypes or environments that enhance expression of the genes slowing bean development may produce higher quality coffee beans, allowing coffee production in a broader range of available future environments., The lower canopy was found to produce better quality coffee with slower maturity. Candidate genes associated with slow bean maturity and improved coffee quality were characterized.

Insect and mite pests are damaging stressors that are threatening the cultivation of tea plants, which result in enormous crop loss. Over the years, the effectiveness of synthetic pesticides has allowed for its prominent application as a control strategy. However, the adverse effects of synthetic pesticides in terms of pesticide residue, environmental contamination and insect pest resistance have necessitated the need for alternative strategies. Meanwhile, microbial pesticides have been applied to tackle the damaging activities of the insect and mite pests of tea plants, and their performances were scientifically adjudged appreciable and environmental friendly. Herein, entomopathogenic microbes that were effective against tea geometrid (Ectropis obliqua Prout), tea green leafhopper (Empoasca onukii Matsuda), paraguay tea ampul (Gyropsylla spegazziniana), tea mosquito bug (Helopeltis theivora Waterhouse) and red spider mite (Oligonychus coffea Nietner) have been reviewed. The current findings revealed that microbial pesticides were effective and showed promising performances against these pests. Overall, this review has provided the basic and integrative information on the integrated pest management (IPM) tool(s) that can be utilized towards successful control of the aforementioned insect and mite pests.

Untargeted liquid chromatography/mass spectrometry (LC/MS) flavoromics analysis was carried out on 18 coffee brews ranging in Specialty Coffee Association (SCA) cup scores. Six compounds highly predictive of low cup score were isolated from coffee using multidimensional preparative LC/MS and further evaluated by sensory recombination analysis with certified SCA quality graders. No. of bitstreams: 0 Previous issue date: 2020-07-01 We developed models for simulating trends over time as functions of the thermal index and models for estimating the levels of infestation of the coffee leaf miner and coffee berry borer and the severity of disease for coffee leaf rust and cercospora, the main phytosanitary problems in coffee crops around the world. We used historical series of climatic data and levels of pest infestation and disease severity in Coffea arabica for high and low yields for seven locations in the two main coffee-producing regions in the state of Minas Gerais in Brazil, Sul de Minas Gerais and Cerrado Mineiro. We conducted two analyses: (a) we simulated the trends of the progress of diseases and pests over time using non-linear models. We only used the thermal index because air temperature is commonly measured by farmers in the regions. (b) We estimated the levels of pest infestation and disease severity using multiple linear regression, with the levels of diseases and pests as dependent variables and accumulated degree days (DD), coffee foliage (LF) estimated by DD and the number of nodes (NN) estimated by DD as independent variables. We used DD and LF = f (DD) and NN = f (DD) to predict diseases and pests with accuracy. MAPEs were 19.6, 5.7, 9.5, and 15.8% for rust, cercospora, leaf miner, and berry borer, respectively, for Sul de Minas Gerais. Newly developed magnetic carbon particles prepared from coffee grounds were used as the sorbent for the magnetic solid-phase extraction of eight phthalic acid esters (PAEs) from plastic bottled water prior to their analysis by GC-MS. The method, which uses coffee-ground particles coated with iron oxide, was validated, and exhibited linearities for the eight PAEs, with coefficients of determination above 0.998 in the 0.005 to 0.1 mg/L concentration range. Limits of detection and limits of quantification of 0.00003 to 0.002 mg/L and 0.0001 to 0.005 mg/L, respectively, were achieved, with recoveries (%) ranging between 77% and 120%, and relative standard deviations for intra- and interday precisions below 16.3% at three fortification levels. No PAE residues were detected when the developed and validated method was applied to 10 real plastic bottled water samples. Taken together, the developed magnetic solid-phase extraction method is a useful tool for monitoring phthalate esters in aqueous samples. PRACTICAL APPLICATION: The development of a new, inexpensive, and efficient magnetic sorption material derived from spent coffee grounds, and its ability to determine phthalate esters in aqueous solutions was described by GC-MS/MS. The developed magnetic solid-phase extraction method is a useful tool for monitoring phthalate esters in aqueous samples.

Angiogenesis is a process of forming new capillaries found in the blood vessel system. Angiogenesis plays an important role in tumor growth and metastasis which includes several biological processes. Cancer cells require a supply of nutrients through the blood vessels to grow and develop so that the cancer cells secrete growth substances to stimulate the formation of new blood vessels to meet their nutritional supply. Robusta coffee beans (C.robusta) contain compounds that function as strong antioxidants namely Chlorogenic acid which is a polyphenol group compound that is thought to have antiangiogenesis effects. This study aims to look at the antiangiogenesis potential of robusta coffee bean ethanol extract (C.robusta) with the Chorio Allantoic Membrane (CAM) method and determine the concentration of robusta coffee bean extract (C.robusta) which has the potential as an antiangiogenesis. Antiangiogenesis test using the Chorio Allantoic Membrane (CAM) method using 9-day-old chicken eggs. In this study eggs were divided into 4 groups, group I was the untreated group, groups II, III, IV were groups that were given ethanol extract of robusta coffee beans (C.robusta) with concentrations of 6000 μg/ml, 5500 µg/ml respectively. and 5000 µg/ml. The eggs are re-incubated for 72 hours. In this study, a macroscopic semi-quantitative score was observed. The score data obtained were analyzed statistically by the One Way Anova test. Based on the Saphiro Wilk normality test results, the results of observations of antiangiogenesis in ethanol extracts of Robusta (Coffea robusta) concentrations of 6000 μg/ml, 5500 μg/ml, and 5000 μg/ml obtained a significance value of 0.248 (p>0.05). Continuing the One Way Anova test in the antiangiogenesis test obtained a significance value of 0.007 (p0.05 which means that there were no differences between groups in inhibiting angiogenesis. Based on the results of the study it can be concluded that the ethanol extract of robusta coffee beans (C. robusta) 6000 μg/ml, 5500 μg/ml, and 5000 μg/ml has antiangiogenesis effect on Chorio Allantoic Membrane (CAM) of chicken embryo.

Green (unroasted) coffee is one of the most traded agricultural commodities in the world. The Arabica (Coffea arabica L.) and Robusta (Coffea canephora Pierre ex A. Froehner) species are the two main types of coffees for commercial production. In general, Arabica coffee is known to have better quality in terms of sensory characteristics; thus, it has a higher market value than Robusta coffee. Accurate differentiation of green beans of the two species is, therefore, of commercial interest in the coffee industry. Using the newly developed single nucleotide polymorphism (SNP) markers, we analyzed a total of 80 single green bean samples, representing 20 Arabica cultivars and four Robusta accessions. Reliable SNP fingerprints were generated for all tested samples. Unambiguous differentiation between Robusta and Arabica coffees was achieved using multivariate analysis and assignment test. The SNP marker panel and the genotyping protocol are sufficiently robust to detect admixture of green coffee in a high-throughput fashion. Moreover, the multilocus SNP approach can differentiate every single bean within Robusta and 55% of Arabica samples. This advantage, together with the single-bean sensitivity, suggests a significant potential for practical application of this technology in the coffee industry.

One of the therapeutic approaches for decreasing postprandial hyperglycemia is to retard absorption of glucose by the inhibition of carbohydrate hydrolyzing enzymes, α-amylase, and α-glucosidases, in the digestive organs. Coffee consumption has been reported to beneficial effects for controlling calorie and cardiovascular diseases, however, the clear efficacy and mode of action are yet to be proved well. Therefore, in this study we evaluated in- vitro rat intestinal α-glucosidases and porcine α-amylase inhibitory activities as well as in vivo (Sprague-Dawley rat model) blood glucose lowering effects of selected coffee extracts. The water extracted Sumatra coffee (SWE) showed strong α-glucosidase inhibitory activity (IC50, 4.39 mg/mL) in a dose-dependent manner followed by Ethiopian water extract (EWE) (IC50, 4.97) and Guatemala water extract (GWE) (IC50, 5.19). Excepted for GWE all the coffee types significantly reduced the plasma glucose level at 0.5 hour after oral intake (0.5 g/kg-body weight) in sucrose and starch-loaded SD rats. In sucrose loading test SWE (p p max) of EWE administration group were decreased by about 18% (from 222.3 ± 16.0 to 182.5 ± 15.4, p p in vitro and in vivo studies therefore could provide the biochemical rationale for the benefit of coffee-based dietary supplement and the basis for further clinical study.

The use of vacuum cycles for the cold extraction of coffee is a new process that leads to a significant reduction in process time of Cold Brew compared to conventional methods. This research aimed at specifying the necessary parameters for producing a consumer-accepted cold brew coffee by applying vacuum cycles. This was achieved by investigating the effect of the number of cycles and of the applied pressure (vacuum) on the physicochemical characteristics of the cold brew coffee, i.e., total dissolved solids (TDS%), pH, acidity, phenol and caffeine content and color. Furthermore, sensory evaluation took place by members of the Specialty Coffee Association of America (SCAA) to specify parameters such as coffee blend, coffee/water ratio, total water hardness and grind size and secondly to determine the optimal pressure and number of cycles for a tasty final beverage. The sensory and physiochemical characteristics of cold extraction coffee were investigated by Principal Component Analysis (PCA). It became evident that coffee extraction by applying two vacuum cycles at 205 mbar pressure produced the lowest intensity of physiochemical properties (caffeine, phenols, acidity, TDS% and pH), and the highest score of sensory characteristics (fragrance, body, acidity, flavor, balance, and aftertaste). Caffeine and phenol concentration of the optimal beverage were 26.66 ± 1.56 mg/g coffee and 23.36 ± 0.79 mg gallic acid/g coffee respectively. The physiochemical characteristics were also compared to a beverage of hot extraction of the same blend and ratio of coffee to water.

Terpenoids are a class of compounds that are found in all living organisms. In plants, some terpenoids are part of primary metabolism, but most terpenes found in plants are classified as specialized metabolites, encoded by terpene synthases (TPS). It is not obvious how to assign the putative product of a given TPS using bioinformatics tools. Phylogenetic analyses easily assign TPS into families; however members of the same TPS family can synthetize more than one terpenoid-and, in many biotechnological applications, researchers are more interested in the product of a given TPS rather than its phylogenetic profile. Automated protein annotation can be used to classify TPS based on their products, despite the family they belong to. Here, we implement an automated bioinformatics method, search_TPS, to identify TPS proteins that synthesize mono, sesqui and diterpenes in Angiosperms. We verified the applicability of the method by classifying wet lab validated TPS and applying it to find TPS proteins in Coffea arabica, C. canephora, C. eugenioides, and Quillaja saponaria. Search_TPS is a computational tool based on PERL scripts that carries out a series of HMMER searches against a curated database of TPS profile hidden Markov models. Coffea karyotype organization and evolution has been uncovered by classical cytogenetics and cytogenomics. We revisit these discoveries and present new karyotype data. Coffea possesses ~ 124 species, including C. arabica and C. canephora responsible for commercial coffee production. We reviewed the Coffea cytogenetics, from the first chromosome counting, encompassing the karyotype characterization, chromosome DNA content, and mapping of chromosome portions and DNA sequences, until the integration with genomics. We also showed new data about Coffea karyotype. The 2n chromosome number evidenced the diploidy of almost all Coffea, and the C. arabica tetraploidy, as well as the polyploidy of other hybrids. Since then, other genomic similarities and divergences among the Coffea have been shown by karyotype morphology, nuclear and chromosomal C-value, AT and GC rich chromosome portions, and repetitive sequence and gene mapping. These cytogenomic data allowed us to know and understand the phylogenetic relations in Coffea, as well as their ploidy level and genomic origin, highlighting the relatively recent allopolyploidy. In addition to the euploidy, the role of the mobile elements in Coffea diversification is increasingly more evident, and the comparative analysis of their structure and distribution on the genome of different species is in the spotlight for future research. An integrative look at all these data is fundamental for a deeper understanding of Coffea karyotype evolution, including the key role of polyploidy in C. arabica origin. The 'Híbrido de Timor', a recent natural allotriploid, is also in the spotlight for its potential as a source of resistance genes and model for plant polyploidy research. Considering this, we also present some unprecedented results about the exciting evolutionary history of these polyploid Coffea.

While coffee beans have been studied for many years, researchers are showing a growing interest in coffee leaves and by-products, but little information is currently available on coffee species other than Coffea arabica and Coffea canephora. The aim of this work was to perform a targeted and untargeted metabolomics study on Coffea arabica, Coffea canephora and Coffea anthonyi. The application of the recent high-resolution mass spectrometry-based metabolomics tools allowed us to gain a clear overview of the main differences among the coffee species. The results showed that the leaves and fruits of Coffea anthonyi had a different metabolite profile when compared to the two other species. In Coffea anthonyi, caffeine levels were found in lower concentrations while caffeoylquinic acid and mangiferin-related compounds were found in higher concentrations. A large number of specialized metabolites can be found in Coffea anthonyi tissues, making this species a valid candidate for innovative healthcare products made with coffee extracts.

Coffea canephora (2n = 2x = 22 chromosomes) is a species with extensive genetic diversity and desirable agronomic traits for coffee breeding programs. However, obtaining a new coffee cultivar through conventional breeding techniques may require more than 30 years of crossing cycles and selection, which hampers the effort of keeping up with market demands and rapidly proposing more resilient to climate change varieties. Although, the application of modern biotechnology tools such as precision genetic engineering technologies may enable a faster cultivar development process. Therefore, we aimed to validate the CRISPR/Cas9 system to generate mutations on a selected genotype of C. canephora, the clone 14. Embryogenic calli and a multiplex binary vector containing two sgRNAs targeting different exons of the CcPDS gene were used. The sgRNAs were under the C. canephora U6 promoter regulation. The target gene encodes phytoene desaturase, an enzyme essential for photosynthesis involved in β-carotene biosynthesis. Somatic seedlings and embryos with albino, variegated and green phenotypes regenerated after Agrobacterium tumefaciens-mediated genetic transformation were analyzed by verifying the insertion of the Cas9 gene and later by sequencing the sgRNAs target regions in the genome of Robusta modified seedlings. Among them, 77% had the expected mutations, and of which, 50% of them had at least one target with a homozygous mutation. The genotype, temperature of co-cultivation with the bacteria, and light intensity used for subsequent embryo regeneration appeared to strongly influence the successful regeneration of plants with a mutated CcPDS gene in the Coffea genus.

RESUMO Hypothenemus hampei, é uma importante praga do café no Brasil e no mundo. A maioria dos trabalhos referentes à associação entre este inseto e fungos teve como alvo a parte aérea de plantas de Coffea canephora. Estudos de campo foram realizados em plantios de C. canephora, em Ouro Preto do Oeste, Rondônia, para levantar a micota associada à broca e frutos de café. Brocas e galerias foram obtidas a partir da dissecção de frutos de C. canephora brocados e não brocados. Os resultados mostraram que existe uma grande diversidade de fungos associados aos insetos. Nas brocas do solo foram encontrados 110 isolados (10 gêneros) e na parte aérea 91 isolados (10 gêneros). Em relação aos frutos brocados (galerias) foram registrados 5 gêneros no solo e 2 na parte aérea e nos frutos não brocados estes valores foram 6 e 5, respectivamente. O "ambiente" solo mostrou maior riqueza que a "parte aérea", tanto para brocas como para frutos brocados. Em relação aos frutos não brocados esta diferença não foi significativa revelando a ação da broca na dispersão destes microrganismos entre os dois ambientes. Este trabalho oferece uma descrição da micota associada à H. hampei em C. canephora e mostra a influência do ambiente na sua diversidade. ABSTRACT Hypothenemus hampei is an important pest of coffee in Brazil and the world. The majority of thestudies concerning the association between this insect and fungus were lead in the aerial part of plants of Coffea canephora. Field studies were carried out on C. canephora, in Ouro Preto do Oeste, Rondônia, to determine the mycobiota associated with coffee berry borer and coffee fruits. Bored and unbored fruits were collected from plants of C. canephora and, borers and galleries were obtained from the disection of the infested fruits. The results that there is a great diversity of fungus associated to the insects. In the borers of the soil there were registered 110 isolates (10 genera) and in the aerial part there were found 91 isolates (5 genera). In relation to the bored fruits (galleries) there were registered 5 genera in the soil and 2 in the aerial parts of plants, and for unbored fruits these values were 6 and 5, respectively. The "soil environment" showed greater diversity than the "aerial part" as much borers as bored fruits. In relation to unbored fruits this difference was not significant, showing the action of the borer in the dispersion of these microorganisms between these two environments. This work offers a description of mycota associated to H. hampei in C. canephora and shows the influence of the environment in the diversity of this mycota.

Prior to recommending insecticides to treat the coffee berry borer (CBB) Hypothenemus hampei, it is valuable to know the mortality and repellency of these insecticides against adult insects or their impact on reproductive output. However, currently available methods assess adult mortality only, limiting the selection of novel insecticides with a different mode of action. In this work, different experimental methods were examined to identify the diverse effects on the CBB under laboratory conditions. For this, green coffee fruits (GFs) were collected and disinfected by immersion in sodium hypochlorite solution followed by UV light irradiation. In parallel, CBB adults from a colony were disinfected by immersion in sodium hypochlorite solution. To assess fruit protection (preinfestation), the fruits were placed in plastic boxes, and the insecticides were applied. Then, the CBB adults were released at a rate of two CBBs per GF. The GFs were left under controlled conditions to evaluate CBB infestation and survival after 1, 7, 15, and 21 days. To evaluate insecticide efficacy after CBB infestation (postinfestation), CBB adults were released to the GFs in a 2:1 ratio for 3 h at 21 °C. Infested fruits showing CBB adults with their abdomens partially exposed were selected and placed in 96-well racks, and the CBBs boring into the fruits were treated directly. After 20 days, the fruits were dissected, and the CBB biological stages inside each fruit were recorded. The GFs served as substrates that mimic natural conditions to evaluate toxic, chemical, and biological insecticides against the CBB.
