Exploring high‐performance catalysts for hydrogen storage in magnesium hydride (MgH2) is crucial but still a challenge. Herein, Nb2CTx with controllable surface termination groups is developed as an efficient catalyst and the bifunctional modulation (adsorption energy and work function) of different surface termination groups (F, O, OH, or defects) is explored. First, compared to F and O, the introduction of OH on the surface or the direct removal of functional groups both leads to a significant increase in the adsorption of H by Nb2CTx. Second, compared to the surface bare, OH‐rich Nb2CTx has a lower work function, making it easier for hydrogen to enter Mg/MgH2 from the Nb2CTx surface or escape from the Mg/MgH2 surface into the Nb2CTx, thus facilitating the hydrogen ad/desorption properties of MgH2, i.e., the rate‐determining step (RDS) shifts from penetration to diffusion. The Nb2CTx‐KOH‐catalyzed MgH2 with optimal surface termination groups, therefore exhibits a release of 6.56 wt.% H2 in 5 min at 250 °C, and 6.46 wt.% H2 uptake within 5 min at 150 °C. The dehydrogenation and hydrogen uptake activation energies show 49.5% and 60.1% enhancements over pristine MgH2. As one of the most important economic traits for domestic animal husbandry, skeletal muscle is regulated by an intricate molecular network. Adenosine deaminase acting on RNA (ADAR1) involves various physiological processes and diseases, such as innate immunity and the development of lung adenocarcinoma, breast cancer, gastric cancer, etc. However, its role in skeletal muscle growth requires further clarification. Here, we explored the functions of ADAR1 in the myogenic process of goat skeletal muscle satellite cells (MuSCs). The ADAR1 transcripts were noticeably enriched in goat visceral tissues compared to skeletal muscle. Additionally, its levels in slow oxidative muscles like the psoas major and minor muscles were higher than in the fast oxidative glycolytic and fast glycolytic muscles. Among the two common isoforms from ADAR1, p110 is more abundant than p150. Moreover, overexpressing ADAR1 enhanced the proliferation and myogenic differentiation of MuSCs. The mRNA-seq performed on MuSCs' knockdown of ADAR1 obtained 146 differentially expressed genes (DEGs), 87 upregulated and 59 downregulated. These DEGs were concentrated in muscle development and process pathways, such as the MAPK and cAMP signaling pathways. Furthermore, many DEGs as the key nodes defined by protein–protein interaction networks (PPI), including STAT3, MYH3/8, TGFβ2, and ACTN4, were closely related to the myogenic process. Finally, RNA immunoprecipitation combined with qPCR (RIP-qPCR) showed that ADAR1 binds to PAX7 and MyoD mRNA. This study indicates that ADAR1 promotes the myogenic development of goat MuSCs, which provides a useful scientific reference for further exploring the ADAR1-related regulatory networks underlying mammal skeletal muscle growth. Skeletal muscle development is spotlighted in mammals since it closely relates to animal health and economic benefits to the breeding industry. Researchers have successfully unveiled many regulatory factors and mechanisms involving myogenesis. However, the effect of N6-methyladenosine (m6A) modification, especially demethylase and its regulated genes, on muscle development remains to be further explored. Here, we found that the typical demethylase FTO (fat mass- and obesity-associated protein) was highly enriched in goats' longissimus dorsi (LD) muscles. In addition, the level of m6A modification on transcripts was negatively regulated by FTO during the proliferation of goat skeletal muscle satellite cells (MuSCs). Moreover, a deficiency of FTO in MuSCs significantly retarded their proliferation and promoted the expression of dystrophin-associated protein 1 (DAG1). m6A modifications of DAG1 mRNA were efficiently altered by FTO. Intriguingly, the results of DAG1 levels and its m6A enrichment from FB23-2 (FTO demethylase inhibitor)-treated cells were consistent with those of the FTO knockdown, indicating that the regulation of FTO on DAG1 depended on m6A modification. Further experiments showed that interfering FTO improved m6A modification at site DAG1-122, recognized by Insulin-like growth factor 2 mRNA-binding protein 1 (IGF2BP1) and consequently stabilized DAG1 transcripts. Our study suggests that FTO promotes the proliferation of MuSCs by regulating the expression of DAG1 through m6A modification. Introduction: Limited studies report anxiety and depression prevalence and their correlations with prognosis in acute myeloid leukemia (AML). Even worse, their risk factors for AML remained unclear. This study aimed to investigate the prevalence, risk factors, and prognostic value of anxiety and depression in AML patients. Methods: Totally, 132 de novo AML patients, 60 non-malignant hematological disease patients (as disease controls), and 60 healthy controls were enrolled. Anxiety and depression status were evaluated by the Hospital Anxiety and Depression Scale (HADS) in all participants. Results: HADS-anxiety score (8.2 ± 3.2 vs. 6.1 ± 2.9 vs. 4.7 ± 2.8), anxiety rate (48.5% vs. 25.0% vs. 10.0%), HADS-depression score (7.8 ± 3.0 vs. 5.8 ± 3.0 vs. 4.0 ± 2.8), and depression rate (43.2% vs. 23.3% vs. 8.3%) were highest in AML patients, followed by disease controls, and the lowest in healthy controls (all p < 0.001). Multivariate logistic regression analysis identified that factors independently associated with anxiety included male (p = 0.002, odds ratio [OR] = 0.240), smoking (p = 0.043, OR = 2.474), education duration (p = 0.024, OR = 0.889), and NCCN high-risk stratification (p = 0.008, OR = 2.347), while those independently associated with depression were age (p = 0.005, OR = 1.055), single/divorced/widowed status (p = 0.014, OR = 3.149), NCCN high-risk stratification (p = 0.002, OR = 3.077), and white blood cell (WBC) (p < 0.001, OR = 1.062). Additionally, depression was correlated with shorter accumulating event-free survival (p = 0.012) and overall survival (p = 0.041) in AML patients, whereas anxiety was not. Conclusions: Anxiety and depression are prevalent, among which depression is associated with poor survival profile, but anxiety is not; moreover, age, male, education, single/divorced/widowed status, smoking, NCCN high-risk stratification, and WBC were independent related factors of anxiety and depression in AML patients. Glacial lake area change is an indicator of global climate change. The topography of Tianshan Mountains is complex, and the glacial lakes are widely distributed, so their variation characteristics and response mechanism to climate change need to be further explored. Based on the Google Earth Engine (GEE) platform, this paper studied the changes of glacial lakes in the Tianshan Mountains in the past 20 years, and analyzed the driving forces in combination with precipitation and temperature. The results showed that: (1) There were 1532 glacial lakes in Tianshan Mountains in 2020, with an area of 164.02 km². The distribution of glacial lakes in the Tianshan Mountains was characterized by more in the west and less in the east, and glacial lake area with an annual average growth rate of 3.47% and 0.63% in the east and west, respectively. (2) The glacial lakes at lower altitudes (<3600 m) in the study area generally showed an expanding trend, and the glacial lakes with an area <0.1 km2 showed a more obvious change. (3) The results of the geographic detector showed that temperature and glaciers were the main driving factors affecting the change of the glacial lakes in Tianshan Mountains. From 2000 to 2020, the precipitation in the eastern Tianshan Mountains increased significantly, while the temperature showed an insignificant trend of decrease. The glacial lake area in this region continued to expand under the stable and sufficient glacial meltwater and runoff recharge. Research results can provide scientific basis for the rational development of water resources and the risk assessment of glacial lake outburst disaster in the Tianshan Mountains. Simple Summary: Investigating the proliferation and differentiation of skeletal muscle satellite cells (MuSCs) in vitro is crucial for elucidating the underlying mechanisms of skeletal muscle development in goats. Real-time quantitative PCR (RT-qPCR) is a widely utilized technique for quantifying the expression levels of target genes. A significant challenge associated with this method is the identification of optimal reference genes for accurate normalization. In this study, we evaluated ten candidate reference genes for the standardization of gene expression. Our results indicated that RPL14 and RPS15A constituted the most stable reference gene combination during the proliferation and differentiation of goat MuSCs. The process of skeletal muscle development is intricate and involves the regulation of a diverse array of genes. Accurate gene expression profiles are crucial for studying muscle development, making it essential to choose the right reference genes for real-time quantitative PCR (RT-qPCR). In the present study, eight candidate reference genes were identified from our previous transcriptome sequencing analysis of caprine skeletal muscle satellite cells (MuSCs), and two traditional reference genes (ACTB and GAPDH) were assessed. The quantitative levels of the candidate reference genes were determined through the RT-qPCR technique, while the stability of their expression was evaluated utilizing the GeNorm, NormFinder, BestKeeper, and RefFinder programs. Furthermore, the chosen reference genes were utilized for the normalization of the gene expression levels of PCNA and Myf5. It was determined that conventional reference genes, including ACTB and GAPDH, were not appropriate for normalizing target gene expression. Conversely, RPL14 and RPS15A, identified through RNA sequencing analysis, exhibited minimal variability and were identified as the optimal reference genes for normalizing gene expression during the proliferation and differentiation of goat MuSCs. This study presents novel life cycle assessment (LCA) findings on hydrochar production from Saudi-Arabia-based date palm fronds biomass waste using hydrothermal carbonization (HTC). The LCA procedure incorporated normalization, weighting, and improvement assessment. The system boundary encompassed water consumption and energy requirements within a lab setting representing a gate-to-gate process. The OpenLCA 1.11.0 software with the European Life Cycle Database 3.2 (ELCD 3.2) was utilized for the study and we employed the ReCiPe Midpoint (H) 2016 and Environmental Footprint 3.0 (EF 3.0) impact assessment methods. The results indicated that fossil fuel usage represented the most significant impact category with the HTC and drying processes identified as major contributors. It was also observed that the HTC process exerted far greater detrimental impacts on the environment than the biomass grinding process. The overwhelming impact of fossil fuel resources could be mitigated by optimizing the batches of biomass or hydrochar samples in each operation, which could alleviate fossil fuel consumption by up to 94%. The findings emphasize the need for targeted interventions to mitigate the environmental burden and contribute to sustainable hydrochar production.

L-shaped concrete-filled steel tubular (CFST) columns represent a valuable structural element with an L-shaped cross-section, primarily employed in the corner columns of framed structures. These columns offer several advantages, including space efficiency by avoiding column protrusion, robust mechanical properties, high load-bearing capacity, ductility, and efficient utilization of internal building space. This article presents the outcomes of an experimental investigation into the seismic behavior of L-shaped CFST columns. The experimental study investigated the seismic performance of nine L-shaped CFST columns while considering different cross-sectional dimensions, their combinations, and varying levels of confinement. The results obtained from this study indicate that L-shaped CFST columns possess favorable seismic performance characteristics. However, there exists the potential for significant improvement by modifying certain parameters. Enhancements in seismic performance were observed when increasing the cross-sectional dimensions of the column and the length of its legs. The use of steel tubes and the provision of adequate confinement also demonstrated notable benefits. Moreover, the better arrangement of steel tubes within the column positively influenced seismic performance. These findings can potentially inform and enhance the design of L-shaped CFST columns, rendering them more resilient to seismic forces.

ObjectivesMetagenomic next-generation sequencing (mNGS) technology is helpful for the early diagnosis of infective endocarditis, especially culture-negative infective endocarditis, which may guide clinical treatment. The purpose of this study was to compare the presence of culture-negative infective endocarditis pathogens versus culture-positive ones, and whether mNGS test results could influence treatment regimens for patients with routine culture-negative infective endocarditis.MethodsThe present study enrolled patients diagnosed with infective endocarditis and tested for mNGS in the First Affiliated Hospital of Zhengzhou University from February 2019 to February 2022 continuously. According to the culture results, patients were divided into culture-negative group (Group CN, n=18) and culture-positive group (Group CP, n=32). The baseline characteristics, clinical data, pathogens, 30 day mortality and treatment regimen of 50 patients with infective endocarditis were recorded and analyzed.ResultsExcept for higher levels of PCT in the Group CN [0.33 (0.16-2.74) ng/ml vs. 0.23 (0.12-0.49) ng/ml, P=0.042], there were no significant differences in the basic clinical data and laboratory examinations between the two groups (all P>0.05). The aortic valve and mitral valve were the most involved valves in patients with infective endocarditis (aortic valve involved: Group CN 10, Group CP 16; mitral valve involved: Group CN 8, Group CP 21; P>0.05) while 9 patients had multiple valves involved (Group CN 2, Group CP 7; P>0.05). The detection rate of non-streptococci infections in the Group CN was significantly higher than that in the Group CP (9/18 vs. 3/32, P=0.004). There was no significant difference in patients with heart failure hospitalization and all-cause death at 30 days after discharge (3 in Group CN vs. 4 in Group CP, P>0.05). It is worth noting that 10 patients with culture-negative infective endocarditis had their antibiotic regimen optimized after the blood mNGS.ConclusionsCulture-negative infective endocarditis should be tested for mNGS for early diagnosis and to guide clinical antibiotic regimen.

ObjectiveTo identify the gene subtypes related to immune cells of cholangiocarcinoma and construct an immune score model to predict the immunotherapy efficacy and prognosis for cholangiocarcinoma.MethodsBased on principal component analysis (PCA) algorithm, The Cancer Genome Atlas (TCGA)-cholangiocarcinoma, GSE107943 and E-MTAB-6389 datasets were combined as Joint data. Immune genes were downloaded from ImmPort. Univariate Cox survival analysis filtered prognostically associated immune genes, which would identify immune-related subtypes of cholangiocarcinoma. Least absolute shrinkage and selection operator (LASSO) further screened immune genes with prognosis values, and tumor immune score was calculated for patients with cholangiocarcinoma after the combination of the three datasets. Kaplan-Meier curve analysis determined the optimal cut-off value, which was applied for dividing cholangiocarcinoma patients into low and high immune score group. To explore the differences in tumor microenvironment and immunotherapy between immune cell-related subtypes and immune score groups of cholangiocarcinoma.Results34 prognostic immune genes and three immunocell-related subtypes with statistically significant prognosis (IC1, IC2 and IC3) were identified. Among them, IC1 and IC3 showed higher immune cell infiltration, and IC3 may be more suitable for immunotherapy and chemotherapy. 10 immune genes with prognostic significance were screened by LASSO regression analysis, and a tumor immune score model was constructed. Kaplan-Meier (KM) and receiver operating characteristic (ROC) analysis showed that RiskScore had excellent prognostic prediction ability. Immunohistochemical analysis showed that 6 gene (NLRX1, AKT1, CSRP1, LEP, MUC4 and SEMA4B) of 10 genes were abnormal expressions between cancer and paracancer tissue. Immune cells infiltration in high immune score group was generally increased, and it was more suitable for chemotherapy. In GSE112366-Crohn’s disease dataset, 6 of 10 immune genes had expression differences between Crohn’s disease and healthy control. The area under ROC obtained 0.671 based on 10-immune gene signature. Moreover, the model had a sound performance in Crohn’s disease.ConclusionThe prediction of tumor immune score model in predicting immune microenvironment, immunotherapy and chemotherapy in patients with cholangiocarcinoma has shown its potential for indicating the effect of immunotherapy on patients with cholangiocarcinoma.

High temperature thermal-mechanical stability of tribological thin coatings is extremely important to a large number of applications in modern industries. DC magnetron sputtering of single metallic element (Cr, Si) and alloy (Ni:Cr) targets formed transition metal nitrides film coatings, CrSiN and CrNiN onto M2 steel. High temperature in-situ synchrotron X-ray diffraction, in the range 25 °C–700 °C, obtained experimental data for a range of structural and mechanical properties. Furthermore, experimental room temperature Nanoindentation measurements, made before and after the in-situ heating cycle, provided corresponding hardness and shear modulus results. The structural results identified microstructure and phase transformation changes, while the mechanical results identified microstrain, hardness, elastic modulus and deformation resistance properties of the coatings. Density functional theory (DFT) and quasi-harmonic approximation (QHA) modelled the high temperature thermal and mechanical properties such as: Young's modulus, shear modulus and thermal expansion coefficients (populated up to 1200 °C). Estimates of hardness are made by correlating the bulk phase hardness and shear modulus, of the CrN and Ni phases, as a function of temperature. Results indicate that Si doping enhances the hardness of the CrN framework, increasing from 29 to 36 GPa and improves the coatings elastic modulus, and resistance to deformation. However the addition of Ni reduced these properties. Furthermore, formation of (Cr,Si)N and Ni(Cr) solid solutions is inferred from DFT, Rietveld and lattice constant analysis.
