Abstract

Nutrients sensing is crucial for fundamental metabolism and physiological functions, and it is also an essential component for maintaining body homeostasis. Traditionally, basic taste receptors exist in oral cavity to sense sour, sweet, bitter, umami, salty and et al. Recent studies indicate that gut can sense the composition of nutrients by activating relevant taste receptors, thereby exerting specific direct or indirect effects. Gut taste receptors, also named as intestinal nutrition receptors, including at least bitter, sweet and umami receptors, have been considered to be activated by certain nutrients and participate in important intestinal physiological activities such as eating behavior, intestinal motility, nutrient absorption and metabolism. Additionally, gut taste receptors can regulate appetite and body weight, as well as maintain homeostasis via targeting hormone secretion or regulating the gut microbiota. On the other hand, malfunction of gut taste receptors may lead to digestive disorders, and then result in obesity, type 2 diabetes and gastrointestinal diseases. At present, researchers have confirmed that the brain-gut axis may play indispensable roles in these diseases via the secretion of brain-gut peptides, but the mechanism is still not clear. In this review, we summarize the current observation of knowledge in gut taste systems in order to shed light on revealing their important nutritional functions and promoting clinical implications. 215. Immunomodulatory effects of umbilical mesenchymal stem cell-derived exosomes on CD4+ T cells in patients with primary Sjögren's syndrome.

Abstract

Background: Primary Sjögren's syndrome (pSS) is an autoimmune disease that leads to the destruction of exocrine glands and multisystem lesions. Abnormal proliferation, apoptosis, and differentiation of CD4+ T cells are key factors in the pathogenesis of pSS. Autophagy is one of the important mechanisms to maintain immune homeostasis and function of CD4+ T cells. Human umbilical cord mesenchymal stem cell-derived exosomes (UCMSC-Exos) may simulate the immunoregulation of MSCs while avoiding the risks of MSCs treatment. However, whether UCMSC-Exos can regulate the functions of CD4+ T cells in pSS, and whether the effects via the autophagy pathway remains unclear. Methods: The study analyzed retrospectively the peripheral blood lymphocyte subsets in pSS patients, and explored the relationship between lymphocyte subsets and disease activity. Next, peripheral blood CD4+ T cells were sorted using immunomagnetic beads. The proliferation, apoptosis, differentiation, and inflammatory factors of CD4+ T cells were determined using flow cytometry. Autophagosomes of CD4+ T cells were detected using transmission electron microscopy, autophagy-related proteins and genes were detected using western blotting or RT-qPCR. Results: The study demonstrated that the peripheral blood CD4+ T cells decreased in pSS patients, and negatively correlated with disease activity. UCMSC-Exos inhibited excessive proliferation and apoptosis of CD4+ T cells in pSS patients, blocked them in the G0/G1 phase, inhibited them from entering the S phase, reduced the Th17 cell ratio, elevated the Treg ratio, inhibited IFN-γ, TNF-α, IL-6, IL-17A, and IL-17F secretion, and promoted IL-10 and TGF-β secretion. UCMSC-Exos reduced the elevated autophagy levels in the peripheral blood CD4+ T cells of patients with pSS. Furthermore, UCMSC-Exos regulated CD4+ T cell proliferation and early apoptosis, inhibited Th17 cell differentiation, promoted Treg cell differentiation, and restored the Th17/Treg balance in pSS patients through the autophagy pathway. Conclusions: The study indicated that UCMSC-Exos exerts an immunomodulatory effect on the CD4+ T cells, and maybe as a new treatment for pSS. [ABSTRACT FROM AUTHOR]

Abstract

Significant challenges exist in inspecting thick composite laminates for manufacturing defects and operational damage. This is due to acoustic attenuation and impedance mismatch at the interface between the different composite layers. An innovative concept for enhancing ultrasonic testing of such composite laminates is introduced in this study. The proposed solution exploits the ability of acoustic metamaterials to cloak virgin composite. Herein, we show that by incorporating carefully designed metamaterials in a pulse-echo ultrasonic testing setup, the position and size of a delamination in a thick hybrid composite laminate can be determined accurately. Abstract

Ionic liquid (IL) electrolytes have been widely used in high‐voltage Li‐based dual ion batteries (DIBs) due to their strong resistance against oxidation. However, their applications in Zn‐based DIBs are restricted because the high charge density of divalent Zn2+ aggravates the ionic interactions in ionic networks and leads to insufficient Zn2+ mobility. Herein, we introduce a hydrofluoroether diluent into a Zn‐based IL electrolyte to break down the larger ionic aggregates into smaller ones with weakened ionic interactions. This unique solvation structure reduces the Stokes radius of Zn2+ from 1.3 nm to 0.97 nm and increases its diffusion coefficient by ∼30 times while retaining the high oxidation stability, enabling the charge/discharge cycling of Zn/graphite DIBs at a high rate of 20 C. Moreover, the enhanced mobility of Zn2+ also promotes compact Zn deposition, which allows the operation of anode‐free Zn/graphite DIBs with 90 % capacity retention after 100 cycles. Abstract

Allicin is the main active ingredient in freshly-crushed garlic and some other allium plants, and its anticancer effect on cancers of digestive system has been confirmed in many studies. The aim of this review is to summarize epidemiological studies and in vitro and in vivo investigations on the anticancer effects of allicin and its secondary metabolites, as well as their biological functions. In epidemiological studies of esophageal cancer, liver cancer, pancreatic cancer, and biliary tract cancer, the anticancer effect of garlic has been confirmed consistently. However, the results obtained from epidemiological studies in gastric cancer and colon cancer are inconsistent. In vitro studies demonstrated that allicin and its secondary metabolites play an antitumor role by inhibiting tumor cell proliferation, inducing apoptosis, controlling tumor invasion and metastasis, decreasing angiogenesis, suppressing Helicobacter pylori, enhancing the efficacy of chemotherapeutic drugs, and reducing the damage caused by chemotherapeutic drugs. In vivo studies further demonstrate that allicin and its secondary metabolites inhibit cancers of the digestive system. This review describes the mechanisms against cancers of digestive system and therapeutic potential of allicin and its secondary metabolites.

Abstract

Magnesium (Mg) metal secondary batteries have attracted much attention for their high safety and high energy density characteristics. However, the significant issues of the cathode/electrolyte interphase (CEI) in Mg batteries are still being ignored. In this work, a significant CEI layer on the typical Mo6S8 cathode surface has been unprecedentedly constructed through the oxidation of the chloride‐free magnesium tetrakis(hexafluoroisopropyloxy)borate (Mg[B(hfip)4]2) salt under a proper charge cut‐off voltage condition. The CEI has been identified to contain BxOy effective species originating from the oxidation of [B(hfip)4]− anion. It is confirmed that the BxOy species is beneficial to the desolvation of solvated Mg2+, speeding up the interfacial Mg2+ transfer kinetics, thereby improving the Mg2+‐storage capability of Mo6S8 host. Abstract

With the development of geo-positioning technologies, the volumes of spatio-textual data are growing rapidly. These data are available in many applications such as location-based services and geo-social networks, and thus, various types of geo-social keyword queries have been intensively studied in the literature. A top-k geo-social keyword (TkGSK) query retrieves the objects by considering spatial, social, and textual constraints between the query and objects. Moreover, the result diversification is becoming a common practice to enhance the quality of the query result. Motivated by these, in this paper, we study the problem of diversified top-k geo-social keyword (DkGSK) query, which considers not only the relevance but also the diversity of the result. We first prove that this problem is NP-hard, and then, we present an exact algorithm with several effective pruning strategies. Also, we develop an approximate algorithm with proved approximation ratio to support the D k GSK query. Considerable experiments on real data sets demonstrate the effectiveness and efficiency of our proposed algorithms.

246. Lactobacillus acidophilussupernatant alleviates osteoporosis by upregulating colonic SERT expression

Abstract

Aims:To investigate the involvement of serotonin transporter (SERT) in colonic epithelial cells in the anti-osteoporosis role of Lactobacillus acidophilus(LA) supernatant (LAS). Methods:The abundance of fecal LA and bone mineral density (BMD) in patients with osteoporosis (OP) or severe osteoporosis were assessed. The protective role of LA in osteoporosis and the expression of SERT and relative signaling were evaluated. Results:Abundance of fecal LA was decreased in patients with severe OP and was positively correlated with BMD. Supplementing LAS to mice alleviated senile osteoporosis. In vitro, NOD2/RIP2/NF-κB signaling was inhibited by LAS due to increased SERT expression. Conclusion:LAS alleviates OP in mice by producing protective metabolites and upregulating SERT expression and represents a promising therapeutic agent.
