Objective To determine the effects of ethanol on the cell proliferation, migration and invasion in human colorenctal cancer HT29 and SW480 cells and investigate the underlying mechanisms. Methods The effect of ethanol on the proliferation of HT29 and SW480 cells was detected by CCK-8 assay. After the IC50 value was obtained by the relevant analysis, and low concentration ethanol exposure models (1/10 IC50 value) were established. The cell cycle distribution was measured by flow cytometry, cell migration and invasion were detected by cell scratch test and Transwell chamber assay respectively, and the cell ROS production were detected using flow cytometry. The protein levels of epithelial-mesenchymal transition (EMT) related proteins, such as E-cadherin, N-cadherin and vimentin were detected by Western blotting. The production of acetaldehyde in the cells after ethanol treatment was detected with acetaldehyde detection kit. Results The IC50 values of ethanol to HT29 and SW480 cells were 436 and 424 mmol/L, respectively. With the increase of ethanol concentration, the proportion of cells at G1 stage was increased. Compared with the control group, the cell scratch test showed that the percentages of wound closure of HT29 and SW480 cells were increased significantly after ethanol treatment in each concentration group (P < 0.05). Transwell chamber test indicated that ethanol treatment significantly increased the number of HT29 and SW480 cells permeating without and with matrigel (P < 0.05). Further experiments showed that ethanol significantly enhanced the intracellular ROS level of HT29 and SW480 cells (P < 0.05). Western blotting displayed that ethanol decreased the expression level of E-cadherin, while increased those of N-cadherin and vimentin (P < 0.05). And as the concentration of ethanol increasing, the acetaldehyde content was elevated in the cells. Conclusion Ethanol may enhance the invasion and migration of colorectal cancer cells through ROS-mediated EMT pathway.

This project takes XX unit as the analysis object to study the first-order vibration wind speed of wind turbine under shutdown vibration. The vortex-induced vibration response is calculated and the displacement under resonant wind speed is calculated by fluid-solid coupling method. The above analysis techniques and methods are forward-looking and advanced in vibration characteristics of domestic wind turbines. On the basis of fluid dynamics analysis, the suppression measures are studied emphatically. Through the optimization and comparison of different inhibition schemes, the effect of inhibition measures was quantitatively analyzed. The analysis shows that the vibration characteristics of wind turbine with high flexible tower has its own particularity. The first order vibration probability and vibration displacement can be effectively reduced by pneumatic method and external measures.

Background: Spinal fracture is a common traumatic condition in orthopaedics, accounting for 5%–6% of total body fractures, and is a high‐risk factor for venous thromboembolism (VTE), which seriously affects patient prognosis. Aim: The aim of this study was to determine the impact of VTE prophylaxis on the prognosis of patients with spinal fractures in intensive care units (ICUs) and to provide a scientific basis for clinical treatment and nursing. Design: A retrospective study of patients with spinal fractures from the multicenter eICU Collaborative Research Database. Method: The outcomes of this study were ICU mortality and in‐hospital mortality. Patients were divided into the VTE prophylaxis (VP) and no VTE prophylaxis (NVP) groups according to whether they had undergone VTE prophylaxis during their ICU admission. The association between groups and outcomes were analysed using Kaplan–Meier (KM) survival curve, log‐rank test and the Cox proportional‐hazards regression model. Results: This study included 1146 patients with spinal fractures: 330 in the VP group and 816 in the NVP group. KM survival curves and log‐rank tests revealed that both ICU and in‐hospital survival probabilities in the VP group were significantly higher than in the NVP group. After the Cox model was adjusted for all covariates, the hazard ratio for ICU mortality in the VP group was 0.38 (0.19–0.75); the corresponding value for in‐hospital mortality in the VP group was 0.38 (0.21–0.68). Conclusions: VTE prophylaxis is associated with reduced ICU and in‐hospital mortality in patients with spinal fractures in ICUs. More research is necessary to further define specific strategies and optimal timing for VTE prophylaxis. Relevance to clinical practice: This study provides the basis that VTE prophylaxis may be associated with improved prognosis in patients with spinal fractures in ICUs. The new insulated single-sided composite shear wall (NISCSW) composition involves setting a precast wall panel on one side and an insulation panel on the other side, with a middle cavity for casting concrete. To investigate the seismic performance of NISCSW under different shear spans and axial compression ratios, eight specimens are made, including six composite and two cast-in-place walls. The shear span ratio is controlled at 1.2 and 1.9, and the axial compression ratio is controlled at 0.1, 0.3, and 0.4. The specimens are subjected to quasistatic tests to analyze failure modes, hysteresis characteristics, stiffness degradation, displacement ductility, and energy dissipation capacity and to compare the seismic performance of the composite and cast-in-place walls. Results show that for each composite specimen, under the same axial compression ratio, the large shear span ratio specimen has a lower ultimate bearing capacity and faster stiffness degradation but better ductility and postyield energy dissipation capacity. Under the same shear span ratio, the high axial compression ratio specimen had a higher ultimate bearing capacity, slightly worse ductility, and similar stiffness degradation and energy dissipation capacity compared to other specimens. Compared with the cast-in-place specimen with the same axial compression ratio, the composite specimen failure mode and hysteresis characteristics are similar, and the ductility and energy dissipation capacity are comparable to the cast-in-place shear wall specimen, indicating that NISCSW has similar seismic performance to the cast-in-place shear wall under conditions of a large shear span ratio and high axial compression ratio. Based on the test results, the program ABAQUS is used to simulate the specimens. Compared with the test results, the simulated specimen failure mode is consistent with the test results, and the hysteresis and skeleton curves are consistent with the test curve, indicating that the model is correct, reliable, and can be verified with test results. Traditional eye drops are administered via topical instillation. However, frequent dosing is needed due to their relatively rapid precorneal removal and low ocular bioavailability. To address these issues, stearoyl L-carnitine-modified nanoemulsions (SC-NEs) were fabricated. The physicochemical properties of SC-NEs in terms of size, morphology, zeta potential, encapsulation efficiency, and in vitro drug release behavior were characterized. The cellular uptake and mechanisms of SC-NEs were comprehensively studied in human corneal epithelial cells and the stearoyl L-carnitine ratio in SC-NEs was optimized. The optimized SC-NEs could target the novel organic cation/carnitine transporter 2 (OCTN2) and amino acid transporter B (0 +) (ATB0,+) on the corneal epithelium, which led to superior corneal permeation, ocular surface retention ability, ocular bioavailability. Furthermore, SC-NEs showed excellent in vivo anti-inflammatory efficacy in a rabbit model of endotoxin-induced uveitis. The ocular safety test indicated that the SC-NEs were biocompatible. In general, the current study demonstrated that OCTN2 and ATB0,+-targeted nanoemulsions were promising ophthalmologic drug delivery systems that can improve ocular drug bioavailability and boost the therapeutic effects of drugs for eye diseases. The Northern China Domain is located between the Central Asian Orogenic Belts to the north and the Kunlun–Qinling belt to the south, and it comprises the North China, Alxa and Tarim blocks. The relationships among the Northern China domain and the southern tectonic elements such as the Qaidam Basin/Terrane are debated because of the major modification by crustal deformation in the late Mesozoic–Cenozoic. To address this issue, we conducted a palaeomagnetic and high-precision radiometric dating study of Triassic volcanic rocks and Middle Jurassic strata in the Qaidam Terrane. Our objective was to determine the relationship between the Qaidam Terrane with the Tarim Block and the North China Block (NCB) during the late Palaeozoic and early Mesozoic. Four volcanic samples yielded zircon U-Pb ages of 236–243 Ma. The characteristic remanent magnetizations (Middle Triassic: D  = 40.2°, I  = 54.6°, α95 = 3.4°; Middle Jurassic: D  = 27.4°, I  = 48.0°, α95 = 7.9°) passed the fold and reversal tests, and yielded Middle Triassic and Middle Jurassic palaeopole positions at 57.6° N, 178.2° E, A 95 = 4.0° and 65.8° N, 197.6° E, A 95 = 7.8°, respectively. Based on these new poles, combined with other reliable data, we compared the apparent polar wander path (APWP) of the Qaidam Terrane with those of the NCB and Tarim Block. The results show that, from the Carboniferous through Early Cretaceous, the APWP of the Qaidam Terrane resembles that of the Tarim Block, but it is quite different from that of the NCB. Combined with other reported evidence, we conclude that the Qaidam Terrane was an independent dynamic unit during the late Palaeozoic until its connection with the Tarim Block, which was followed by continuous eastward motion. During this process, the connection between the Qaidam Terrane and the NCB–Alxa blocks occurred in the Middle Triassic, and subsequently the Qaidam Terrane underwent multiple tectonic responses to collisions with the Qiangtang Terrane, Lhasa Terrane and the India Plate, before the formation of its modern tectonic configuration. Hepatocellular carcinoma (HCC) does not respond well to current treatments, even immune checkpoint inhibitors. PD‐L1 (programmed cell death ligand 1 or CD274 molecule)‐mediated immune escape of tumor cells may be a key factor affecting the efficacy of immune checkpoint inhibitor (ICI) therapy. However, the regulatory mechanisms of PD‐L1 expression and immune escape require further exploration. Here, we observed that DDX1 (DEAD‐box helicase 1) was overexpressed in HCC tissues and associated with poor prognosis in patients with HCC. Additionally, DDX1 expression correlated negatively with CD8+ T cell frequency. DDX1 overexpression significantly increased interferon gamma (IFN‐γ)‐mediated PD‐L1 expression in HCC cell lines. DDX1 overexpression decreased IFN‐γ and granzyme B production in CD8+ T cells and inhibited CD8+ T cell cytotoxic function in vitro and in vivo. In conclusion, DDX1 plays an essential role in developing the immune escape microenvironment, rendering it a potential predictor of ICI therapy efficacy in HCC. [ABSTRACT FROM AUTHOR]

Objective To investigate the fatigue status and occurrence of systemic exertion intolerance disease (SEID) in soldiers from a unit of special operations force. Methods A total of 260 officers and soldiers from a SPF unit were selected in June 2018, and surveyed for fatigue level with multidimensional fatigue inventory 20 (MFI-20). According to the SEID diagnostic criteria recommended by the Institute of Medicine (IOM) in 2015, the occurrence and influencing factors of SEID were assessed using a questionnaire. Results The total score of MFI of the participants was 54.49±11.13, and the scores of general fatigue, physical fatigue, reduced activity, reduced motivation and mental fatigue were 13.63±3.48, 11.75±3.48, 9.72±2.33, 9.00±2.52 and 10.39±3.74, respectively. About 34.27% of the participants suffered from deep fatigue for longer than half a year. The incidences of SEID-related symptoms were 76.00% for unrefreshing sleep, 71.54% for substantial reduction or impairment in the ability, 46.18% for cognitive impairment, 26.52% for orthostatic intolerance and 22.09% for post-exertional malaise in order. According to the diagnostic criteria recommended by IOM, the incidence of SEID was 12.61%. The daily training time was closely correlated to fatigue level (r=0.223, P < 0.01) and SEID incidence (P=0.039). There were no differences in fatigue score and SEID incidence among the participants of different durations of military service, posts and other factors. Conclusion The fatigue score and incidence of SEID are quite high in the participants. The daily training time is closely correlated to fatigue level and SEID incidence.
