201. Transglycosylation of Stevioside by a Commercial β-Glucanase with Fungal Extracted β-Glucans as Donors

Abstract: Abstract Alternative sweeteners, such as steviol glucosides from the plant Stevia rebaudiana Bertoni, are becoming increasingly popular for the design of next-generation foodstuffs. However, the bitter aftertaste of native steviol glucosides is one of the main reasons behind consumer reluctance towards stevia-containing products. Biocatalysis could be a sustainable solution to this problem, through addition of glucosyl moieties to the molecule. Glycoside hydrolases are enzymes performing transglycosylation reactions, and they can be exploited for such modifications. In the present work, the commercial β-glucanase Finizym 250L® was employed for the transglycosylation of stevioside. After optimization of several reaction parameters, the maximal reaction yield obtained was 19%, with barley β-glucan as the glycosyl donor. With the aim to develop a sustainable process, β-glucan extracts from different fungal sources were prepared. Pulsed Electric Field pretreatment of mycelial biomass resulted in extracts with higher β-glucan content. The extracts were tested as alternative glucosyl donors, reaching up to 15.5% conversion yield, from Pleurotus-extracted β-glucan. Overall, in the present work a novel enzymatic process for the modification of stevioside is proposed, with concomitant valorization of β-glucans extracted from fungal biomass, potentially generated as a byproduct from other applications, in concert with the principles of circular economy. 202. THE BRAND PERSONALITY OF WORLD HERITAGE SITES: TEXT MINING THE PERCEPTIONS OF UNESCO EXPERTS AND VISITORS

Abstract: World Heritage (WH) is acknowledged as a prominent tourism brand. However, research on the intangible relationship between World Heritage sites (WHSs) and outstanding universal values (OUVs) is lacking. This study employed advances in text-mining, using digital textual data, and Brand Personality (BP) lexical methods to explore descriptions of WHSs. Although existing BP dictionaries are useful for measuring the personalities of destinations and attractions, they need further development. This is due to the perception that the current BP dictionary is limited to capturing distinct brand cultural meanings from large digital texts. Additionally, a BP dictionary construction technique is still necessary to enable BP analysis using substantial online text. The necessity to specifically identify the WHSs personality categorization dictionary as microelements of destination that may have significance for affecting visitor preference is still needed. Using 13,741 post-experience visitor-generated TripAdvisor reviews, and 1,121 WH centre website descriptions, a natural and cultural WHSs personality dictionary was developed. Significant differences in the way UNESCO experts and visitors perceive WHSs were explored and practical and managerial implications were discussed.

Abstract: International audience; AIMS: To evaluate the current management and survival of patients with left-sided infective endocarditis (IE) complicated by congestive heart failure (CHF) in the ESC-EORP European Endocarditis (EURO-ENDO) registry. METHODS AND RESULTS: Among the 3116 patients enrolled in this prospective registry, 2449 (mean age: 60 years, 69% male) with left-sided (native or prosthetic) IE were included in this study. Patients with CHF (n~=~698, 28.5%) were older, with more comorbidity and more severe valvular damage (mitro-aortic involvement, vegetations >10\,mm and severe regurgitation/new prosthesis dehiscence) than those without CHF (all p\,≤q\,0.019). Patients with CHF experienced higher 30-day and 1-year mortality than those without (20.5% vs. 9.0% and 36.1% vs. 19.3%, respectively) and CHF remained strongly associated with 30-day (odds ratio[OR] 2.37, 95% confidence interval [CI] [1.73-3.24; p\,10\,mm, severe valvular regurgitation and/or new prosthetic dehiscence, perivalvular complication, and prosthetic IE (OR 0.22, 95% CI 0.12-0.38; p\

205. Evaluation of Basidiomycetes Wild Strains Grown in Agro-Industrial Residues for Their Anti-Tyrosinase and Antioxidant Potential and for the Production of Biocatalysts

Abstract: White-rot basidiomycetes are the only microorganisms with the ability to produce both hydrolytic (cellulases and hemicellulases) and oxidative (ligninolytic) enzymes for degrading cellulose/hemicellulose and lignin. In addition, they produce biologically active natural products with important application in cosmetic formulations, either as pure compounds or as standardized extracts. In the present work, three wild strains of Basidiomycetes fungi (Pleurotus citrinopileatus, Abortiporus biennis and Ganoderma resinaceum) from Greek habitats were grown in agro-industrial residues (oil mill wastewater, and corn cob) and evaluated for their anti-tyrosinase and antioxidant activity and for the production of biotechnologically relevant enzymes. P. citrinopileatus showed the most interesting tyrosinase inhibitory activity, while A. biennis showed the highest DPPH(2,2-diphenyl-1-picryl-hydrazyl) scavenging potential. Corn cobs were the most appropriate carbon source for maximizing the inhibitory effect of fungal biomasses on both activities, while the use of oil mill wastewater selectively increased the anti-tyrosinase potential of P. citrinopileatus culture filtrate. All strains were found to be preferential lignin degraders, similarly to most white-rot fungi. Bioinformatic analyses were performed on the proteome of the strains P. citrinopileatus and A. biennis, focusing on CAZymes with biotechnological relevance, and the results were compared with the enzyme activities of culture supernatants. Overall, all three strains showed strong production of oxidative enzymes for biomass conversion applications.

206. Effect of Chlorination on Microbiological Quality of Effluent of a Full-Scale Wastewater Treatment Plant

Abstract: The evaluation of effluent wastewater quality mainly relies on the assessment of conventional bacterial indicators, such as fecal coliforms and enterococci; however, little is known about opportunistic pathogens, which can resist chlorination and may be transmitted in aquatic environments. In contrast to conventional microbiological methods, high-throughput molecular techniques can provide an accurate evaluation of effluent quality, although a limited number of studies have been performed in this direction. In this work, high-throughput amplicon sequencing was employed to assess the effectiveness of chlorination as a disinfection method for secondary effluents. Common inhabitants of the intestinal tract, such as Bacteroides, Arcobacter and Clostridium, and activated sludge denitrifiers capable of forming biofilms, such as Acidovorax, Pseudomonas and Thauera, were identified in the chlorinated effluent. Chloroflexi with dechlorination capability and the bacteria involved in enhanced biological phosphorus removal, i.e., Candidatus Accumulibacter and Candidatus Competibacter, were also found to resist chlorination. No detection of Escherichia indicates the lack of fecal coliform contamination. Mycobacterium spp. were absent in the chlorinated effluent, whereas toxin-producing cyanobacteria of the genera Anabaena and Microcystis were identified in low abundances. Chlorination significantly affected the filamentous bacteria Nocardioides and Gordonia, whereas Zoogloea proliferated in the disinfected effluent. Moreover, perchlorate/chlorate- and organochlorine-reducing bacteria resisted chlorination.

Abstract: An unbiased, automated and reliable method for analysis of brain lesions in tissue after ischemic stroke is missing. Manual infarct volumetry or by threshold-based semi-automated approaches is laborious, and biased to human error or biased by many false -positive and -negative data, respectively. Thereby, we developed a novel machine learning, atlas-based method for fully automated stroke analysis in mouse brain slices stained with 2% Triphenyltetrazolium-chloride (2% TTC), named “StrokeAnalyst”, which runs on a user-friendly graphical interface. StrokeAnalyst registers subject images on a common spatial domain (a novel mouse TTC- brain atlas of 80 average mathematical images), calculates pixel-based, tissue-intensity statistics (z-scores), applies outlier-detection and machine learning (Random-Forest) models to increase accuracy of lesion detection, and produces volumetry data and detailed neuroanatomical information per lesion. We validated StrokeAnalyst in two separate experimental sets using the filament stroke model. Abstract: World Heritage Sites (WHSs) are perceived as significant tourism brands. However, there is no in-depth analysis of the intangible meaning of World Heritage (WH) cultural attraction types from the visitor's lens. This study employed text mining and a Brand Personality (BP) lexical approach using WH personality categories to identify types of cultural attractions with intangible meanings in WHSs located in seven countries in Asia and the Pacific and Europe. This enabled the WH personality category distribution for each type of WH cultural attraction found on TripAdvisor to be measured. Accordingly, 9,971 visitor-generated reviews were analyzed and fifteen WHS cultural attraction types in the seven countries identified. The relationships between WH personality categories, the seven countries and WH attraction types were analyzed, and attraction types clustered into four groups. Abstract: Places can hold the most significant experiences of peoples' lives, and place attachment rests in these people-place meaningful connections. Traveling abroad for international students is a mobility where changes and mixed emotions entangle to one another. It represents distancing and loosening ties from a familiar place (home) and creating and connecting new ones with another place (host destination). This voluntary relocation is considered a disruption in place attachment and impacts the overall experience of the international student mobility. Nevertheless, the creation of attachment to a place derived from a place disruption remains scarce within international student mobility literature. This case study explores the production of place attachment in international students in the host destination through the conceptual lens of disruptions to attachment via changes in places. Thirty-three in-depth semi-structured interviews were carried out with international students from different nationalities who have finished a Master's degree in Tourism at the University of Girona, Spain. This investigation demonstrates the motives and process of place disruption in international student mobility and the power of a host destination to create new emotional bonds and connections, becoming another home for international students. Abstract: Introduction Working in surgery while pregnant is challenging. Navigating this period safely is of paramount importance. Anecdotal observation suggests that there exists great variation among European nations in regard to maternity leave and radiation safety. The aim of this article was to gain insight into policy patterns and variations across Europe regarding these issues. Methods A series of core question items was distributed to representatives across 12 nations Austria, Belgium, Germany, Greece, Iceland, Italy, Netherlands, Norway, Poland, Republic of Ireland, Spain and the United Kingdom). Results The total number of weeks with full pay ranged from as little as 4 weeks in Belgium to 32 and Iceland. All countries included in this study give the option of additional weeks beyond the initial period, however at reduced pay. Some offer unpaid leave beyond this. Only 5/12 countries had a specific policy on when the pregnant surgeon should come off the on-call rota. Only Austria, Italy and Poland stipulate a requirement for the pregnant clinician to be replaced or be completely exempt in cases involving radiation. Only Germany, Iceland, Norway and Poland highlight the need to limit radiation dose in the first trimester. Beyond this, Germany alone provides written guidance for reduction in gown weight and along with Poland, display arguably the most forward-thinking approach to resting. Conclusion There is a marked range in maternal leave policies across Europe. There also exists a lack of universal guidance on radiation safety for the pregnant urologist. There is urgent need for this void to be addressed.

This chapter, after a short introduction on dry methane reforming and a concise overview of microwave-catalyst thermal interaction in gas–solid catalytic reactors, focuses on the microwave-assisted methane dry reforming reaction discussing relevant catalytic materials and reactor design aspects concerning process scale up. Finally, the potential application of two alternative electricity-based heating techniques, that is Joule heating and induction heating, to dry methane reforming is evaluated at the end of the chapter.

Abstract: We report the results of the WMT 2022 shared task on Quality Estimation, in which the challenge is to predict the quality of the output of neural machine translation systems at the word and sentence levels, without access to reference translations. This edition introduces a few novel aspects and extensions that aim to enable more fine-grained, and explainable quality estimation approaches. We introduce an updated quality annotation scheme using Multidimensional Quality Metrics to obtain sentence- and word-level quality scores for three language pairs. We also extend the Direct Assessments and post-edit data (MLQE-PE) to new language pairs: we present a novel and large dataset on English-Marathi, as well as a zero-shot test set on English-Yoruba. Further, we include an explainability sub-task for all language pairs and present a new format of a critical error detection task for two new language pairs. Participants from 11 different teams submitted altogether 991 systems to different task variants and language pairs.

Abstract: The enzymatic factory of ligninolytic fungi has proven to be a powerful tool in applications regarding the degradation of various types of pollutants. The degradative potential of fungi is mainly due to the production of different types of oxidases, of which laccases is one of the most prominent enzymatic activities. In the present work, crude laccases from the supernatant of Pleurotus citrinopileatus cultures grown in olive oil mill wastewater (OOMW) were immobilized in crosslinked enzyme aggregates (CLEAs), aiming at the development of biocatalysts suitable for the enzymatic treatment of OOMW. The preparation of laccase CLEAs was optimized, resulting in a maximum of 72% residual activity. The resulting CLEAs were shown to be more stable in the presence of solvents and at elevated temperatures compared to the soluble laccase preparation. The removal of the phenolic component of OOMW catalyzed by laccase-CLEAs exceeded 35%, while they were found to retain their activity for at least three cycles of repetitive use. The described CLEAs can be applied for the pretreatment of OOMW, prior to its use for valorization processes, and thus, facilitate its complete biodegradation towards a consolidated process in the context of circular economy.

Abstract: Aims Infective endocarditis (IE) is a life-threatening disease associated with high mortality and morbidity worldwide. We sought to determine how socioeconomic factors might influence its epidemiology, clinical presentation, investigation and management, and outcome, in a large international multicentre registry. Methods and results The EurObservational Programme (EORP) of the European Society of Cardiology EURO-ENDO (European Infective Endocarditis) registry comprises a prospective cohort of 3113 adult patients admitted for IE in 156 hospitals in 40 countries between January 2016 and March 2018. Patients were separated in three groups, according to World Bank economic stratification [group 1: high income (75.6%); group 2: upper-middle income (15.4%); group 3: lower-middle income (9.1%)]. Group 3 patients were younger [median age (interquartile range, IQR): group 1, 66 (53–75) years; group 2, 57 (41–68) years; group 3, 33 (26–43) years; P < 0.001] with a higher frequency of smokers, intravenous drug use, and human immunodeficiency virus infection (all P < 0.001) and presented later [median (IQR) days since symptom onset: group 1, 12 (3–35); group 2, 19 (6–54); group 3, 31 (12–62); P < 0.001] with a higher likelihood of developing congestive heart failure (13.6%, 11.1%, and 22.6%, respectively; P < 0.001) and persistent fever (9.8%, 14.2%, and 27.9%, respectively; P < 0.001). Among 2157 (69.3%) patients with theoretical indication for cardiac surgery, surgery was performed less frequently in group 3 patients (75.5%, 76.8%, and 51.3%, respectively; P < 0.001), who also demonstrated the highest mortality (15.0%, 23.0%, and 23.7%, respectively; P < 0.001). Conclusion Socioeconomic factors influence the clinical profile of patients presenting with IE across the world. Despite younger age, patients from the poorest countries presented with more frequent complications and higher mortality associated with delayed diagnosis and lower use of surgery.

Abstract: β-Galactosidases are key enzymes in the food industry. Apart from the hydrolysis of the saccharide bond of lactose, they also catalyze transgalactosylation reactions, producing galactooligosaccharides (GOS) with prebiotic activity. Here we report the heterologous production in Pichia pastoris of a novel β-galactosidase from the fungus Thermothielavioides terrestris. The enzyme (TtbGal1) was purified and characterized, showing optimal activity at 60 °C and pH 4. TtbGal1 is thermostable, retaining almost full activity for 24 h at 50 °C. It was applied to the production of GOS from defined lactose solutions and acid whey, a liquid waste from the Greek yoghurt industry, reaching yields of 19.4 % and 14.8 %, respectively. HILIC-ESI-QTOF-MS analysis revealed the production of GOS with up to 4 saccharide monomers. The results demonstrate efficient GOS production catalyzed by TtbGal1, valorizing acid whey, a waste with a heavy polluting load from the dairy industry.
