Objectives: The treatment of acute herpangina is inconsistent. We aim to evaluate the effectiveness and safety of interferon α-2b spray versus Ribavirin for this disease. Methods: A randomized, controlled trial was conducted in eight hospitals in China between 2016 and 2018. 668 patients (1–7 years old) were randomized into an experimental group (treated with Interferon α-2b spray) or control group (received Ribavirin Aerosol). Body temperature returning to normal within 72 h and remaining so for 24 h was the primary outcome; release of oral herpes and adverse events were the secondary outcomes. Results: (1) The average age of onset was 2.5 years old. (2) After 72 h treatment, body temperature of 98.5% patients in experimental group and 94.3% in control group returned to normal and remained so for 24 h (P = 0.004). The differences were greater at 48 h treatment (95.2% vs. 85.9%, P < 0.001) and at 24 h (77.5% vs. 66.5%, P = 0.001). (3) The rate of improved oral herpes in the experimental group was higher than that in control group (46.7% vs.37.1%, P = 0.011). No adverse reaction occurred. Conclusions: Local application of recombinant interferon α-2b spray showed better efficacy for acute herpangina in children. It was safe for use.

Abstract Background Surgical resection remains the best option for long-term survival in colorectal cancer (CRC); however, surgery can lead to tumor cell release into the circulation. Previous studies have also shown that surgery can affect cancer cell growth. The role of perioperative factors influencing long-term survival in patients presenting for CRC surgery remains to be investigated. Methods This retrospective single–center cohort study was conducted to collect the clinical data of patients who underwent elective laparoscopic resection for CRC from January 2014 to December 2015, namely clinical manifestations, pathological results, and perioperative characteristics. Survival was estimated using the Kaplan–Meier log-rank test. Univariable and multivariable Cox regression models were used to compare hazard ratios (HR) for death. Results A total of 234 patients were eligible for analysis. In the multivariable Cox model, tumor-node-metastasis (TNM) stage (stage IV: HR 30.63, 95% confidence interval (CI): 3.85–243.65; P = 0.001), lymphovascular invasion (yes: HR 2.07, 95% CI 1.09–3.92; P = 0.027), inhalational anesthesia with isoflurane (HR 1.96, 95% CI 1.19–3.21; P = 0.008), and Klintrup–Makinen (KM) inflammatory cell infiltration grade (low-grade inflammation: HR 2.03, 95% CI 1.20–3.43; P = 0.008) were independent risk factors affecting 5-year overall survival after laparoscopic resection for CRC. Conclusions TNM stage, lymphovascular invasion, isoflurane, and KM grade were independent risk factors affecting CRC prognosis. Sevoflurane and high-grade inflammation may be associated with improved survival in CRC patients undergoing resection.

Summary: The core promoter (CP) of hepatitis B virus (HBV) is critical for HBV replication by controlling the transcription of pregenomic RNA (pgRNA). Host factors regulating the activity of the CP can be identified by different methods. Biotin-based proximity labeling, a powerful method with the capability to capture weak or dynamic interactions, has not yet been used to map proteins interacting with the CP. Here, we established a strategy, based on the newly evolved promiscuous enzyme TurboID, for interrogating host factors regulating the activity of HBV CP. Using this strategy, we identified STAU1 as an important factor involved in the regulation of HBV CP. Mechanistically, STAU1 indirectly binds to CP mediated by TARDBP, and recruits the SAGA transcription coactivator complex to the CP to upregulate its activity. Moreover, STAU1 binds to HBx and enhances the level of HBx by stabilizing it in a ubiquitin-independent manner.

The aged seeds have a significant influence on seed vigor and corn growth. Therefore, it is vital for the planting industry to identify aged seeds. In this study, hyperspectral reflectance imaging (1,000–2,000 nm) was employed for identifying aged maize seeds using seeds harvested in different years. The average spectra of the embryo side, endosperm side, and both sides were extracted. The support vector machine (SVM) algorithm was used to develop classification models based on full spectra to evaluate the potential of hyperspectral imaging for maize seed detection and using the principal component analysis (PCA) and ANOVA to reduce data dimensionality and extract feature wavelengths. The classification models achieved perfect performance using full spectra with an accuracy of 100% for the prediction set. The performance of models established with the first three principal components was similar to full spectrum models, but that of PCA loading models was worse. Compared to other spectra, the two-band ratio (1,987 nm/1,079 nm) selected by ANOVA from embryo-side spectra achieved a better classification accuracy of 95% for the prediction set. The image texture features, including histogram statistics (HS) and gray-level co-occurrence matrix (GLCM), were extracted from the two-band ratio image to establish fusion models. The results demonstrated that the two-band ratio selected from embryo-side spectra combined with image texture features achieved the classification of maize seeds harvested in different years with an accuracy of 97.5% for the prediction set. The overall results indicated that combining the two wavelengths with image texture features could detect aged maize seeds effectively. The proposed method was conducive to the development of multi-spectral detection equipment.

Introduction: The proliferation of vascular smooth muscle cells (VSMCs) is an important physiological and pathological basis for many cardiovascular diseases. Endogenous hydrogen sulfide (H2S), the third gasotransmitter, is found to preserve vascular structure by inhibiting VSMC proliferation. However, the mechanism by which H2S suppresses VSMC proliferation has not been fully clear. Objectives: This study aimed to explore whether H2S persulfidates the transcription factor FOXO1 to inhibit VSMC proliferation. Methods: After the proliferation of VSMC A7r5 cells was induced by endothelin-1 (ET-1), FOXO1 phosphorylation and proliferating cell nuclear antigen (PCNA) expression were detected by Western blotting, the degree of FOXO1 nuclear exclusion and PCNA fluorescent signals in the nucleus were detected by immunofluorescence, and the persulfidation of FOXO1 was measured through a biotin switch assay. Results: The results showed that ET-1 stimulation increased cell proliferation, FOXO1 phosphorylation and FOXO1 nuclear exclusion to the cytoplasm in the cells. However, pretreatment with NaHS, an H2S donor, successfully abolished the ET-1-induced increases in the VSMC proliferation, FOXO1 phosphorylation, and FOXO1 nuclear exclusion to the cytoplasm. Mechanistically, H2S persulfidated the FOXO1 protein in A7r5 and 293T cells, and the thiol reductant DTT reversed this effect. Furthermore, the C457S mutation of FOXO1 abolished the H2S-induced persulfidation of FOXO1 in the cells and the subsequent inhibitory effects on FOXO1 phosphorylation at Ser256, FOXO1 nuclear exclusion to the cytoplasm and cell proliferation. Conclusion: Thus, our findings demonstrated that H2S might inhibit VSMC proliferation by persulfidating FOXO1 at Cys457 and subsequently preventing FOXO1 phosphorylation at Ser256.

Seed vigor is one of the essential contents of agricultural research. The decline of seed vigor is described as an inevitable process. Recent studies have shown that the oxidative damage caused by reactive oxygen species (ROS) is the main reason for the destruction of various chemicals in seeds and eventually evolves into seed death. The traditional vigor tests, such as the seed germination test and TTC staining, are commonly used to assess seed vigor. However, these methods often need a large number of experimental samples, which will bring a waste of seed resources. At present, many new methods that are fast and nondestructive to seeds, such as vibrational spectroscopic techniques, have been used to test seed vigor and have achieved convincing results. This paper is aimed at analyzing the microchanges of seed-vigor decline, summarizing the performance of current seed-vigor test methods, and hoping to provide a new idea for the nondestructive testing of a single seed vigor by combining the physiological alterations of seeds with chemometrics algorithms.

There is currently no way for subway air-conditioning systems to adjust to changes in passenger capacity in terms of air-supply volume and air-supply temperature. In this paper, a study of the setting of the air-supply volume and controlling the temperature in subway trains was conducted. Based on the fact that the air-supply volume in a subway train needs to meet the requirements of cooling load and humidity load, as well as fresh-air volume, the minimum critical air-supply volume curve was determined on the basis of theoretical calculations under different passenger capacities. Under different passenger capacity and air-supply-volume conditions, the PMV (predicted mean vote) distribution of the subway train was simulated and calculated, and the results indicated that increasing the air-supply volume would not be sufficient to achieve PMV = 0. Therefore, in accordance with the theoretical formulation of the PMV, multiple linear regression was used to fit the temperature-control curve for various passenger capacities. A simulation calculation of the subway train was used to assess the uniformity of the flow field and the temperature field within the subway train in accordance with EN14750. It is evident from the results that under the control curve of the air-supply volume and temperature under different passenger capacities, the uniformity of the flow field and the temperature field can meet the standard requirements. By using the air-supply volume and temperature-control curve proposed in this paper, it is possible to adjust the air-supply volume in real-time, control the temperature of the air-conditioning system of the subway train, improve the passenger comfort of the train, and reduce the energy consumption of the air-conditioning system.

Tetramethylpyrazine is an important flavor compound in liquor, which endows liquor with health efficacy. The bacteria that could produce tetramethylpyrazine were isolated from the high-temperature Daqu in Guizhou by using the dilution spread plate method, and were identified by using the methods such as colonial morphology, microscopic observation and molecular biology. Then, their enzyme production characteristics, growth characteristics and tolerance characteristics were studied. The results showed that one strain FSDQ02 producing tetramethylpyrazine was screened from the high-temperature Daqu in Guizhou, which was preliminarily identified as Bacillus amyloliquefaciens. The strain FSDQ02 could secrete protease, amylase and cellulase, and grow well in the range of 30-40°C and pH 5*9, with the tolerance to 6% ethanol and 14% NaCl. Bacillus amyloliquefaciens FSDQ02 isolated from the results of this study had great great application potential in strengthening the production of high-temperature Daqu and liquor. AbstractIn this report, new self-healing hydrogels were obtained through simple one-pot free radical polymerization of acrylamide, phenylboronic acid, and 2-hydroxyethyl acrylate as copolymer monomers, potassium persulfate (KPS) as initiator, and tetramethylethylenediamine (TEMED) as promoter under low-temperature alkaline (pH = 9) conditions. The boronic acid group of phenylboronic acid (PBA) can interact with the hydroxyl group of hydroxyethyl acrylate (HEA) to form dynamic boronic ester bonds instead of the conventional bis acrylate crosslinkers. The effect of PBA content on the mechanical strength of the hydrogels was systematically investigated. The hydrogels exhibited higher stress, elongation, and self-healing properties due to the synergistic effect of H-bonds and boronate-diol complexation. Further studies revealed that the A–H–0.2–P–0.08 hydrogels exhibited excellent resistance to compression fatigue and also showed good biocompatibility through in vitro cytotoxicity tests. Metal based electroreflective devices (MERDs) have gained wide attention in recent years in the fields of smart windows and automotive glass due to their excellent photo-thermal modulation capability in the visible and near-infrared wavelength bands due to their high reflectivity. However, the high porosity due to the island-like growth of silver particles makes the devices susceptible to electrodeposited film breakage during prolonged energization or bleaching. This will seriously affect the effectiveness and stability of the device. In this paper, it is shown that the problem of Ag electrodeposition film fragmentation can be effectively overcome by replacing dimethyl sulfoxide solvent by N-methylpyrrolidone (NMP). The optical constants during film deposition were tested by the spectroscopic ellipsometry technique, and the porosity was calculated (reduced by 21.6%). The results show that MERDs with NMP as the solvent have more and more dispersed silver deposition sites, which leads to lower porosity of the silver deposited film during the deposition process. In addition, the lower turn-on voltage, greater contrast, longer memory effect, and greater coloring efficiency reveal that they are more promising for energy-saving applications. Abstract Background Acute fatty liver of pregnancy (AFLP) is a potentially lethal condition of pregnant women with a high mortality rate. Potential predictors related to postpartum recovery time and prognostic factors of AFLP are still unclear. This study aimed to evaluate potential predictors for prognosis and postpartum recovery time of AFLP. Methods We retrospectively analyzed the clinical data of 76 AFLP patients in our hospital from 2002 to 2017 and investigated potential predictors using univariate analysis and multivariate logistic regression analysis. Abstract Background Experimental and clinical trials have demonstrated the efficiency of bone marrow-derived mesenchymal stromal/stem cells (bMSCs) in the treatment of myocardial infarction. However, after intravenous injection, the ineffective migration of engrafted bMSCs to the hearts remains an obstacle, which has an undesirable impact on the efficiency of cell-based therapy. Therefore, we attempted to identify a marker that could distinguish a subpopulation of bMSCs with a promising migratory capacity. Methods Here, CD51-negative and CD51-positive cells were isolated by flow cytometry from Ter119−CD45−CD31−bMSCs and cultured in specifically modified medium. The proliferation ability of the cells was evaluated by 5-ethynyl-2′-deoxyuridine (EdU) staining or continuously monitored during culture, and the differentiation potential was assessed by culturing the cells in the appropriate conditioned media. Wound healing assays, transwell assays and quantitative polymerase chain reaction (qPCR) were used to measure the migratory ability. The mice were subjected to a sham operation or myocardial infarction (MI) by permanently occluding the coronary artery, and green fluorescent protein (GFP)-labelled cells were transplanted into the mice via intravenous infusion immediately after MI. Heart function was measured by echocardiography; infarct myocardium tissues were detected by triphenyl tetrazolium chloride (TTC) staining. Additionally, immunofluorescence staining was used to verify the characteristics of CD51+bMSCs and inflammatory responses in vivo. Statistical comparisons were performed using a two-tailed Student’s t test. Results In this study, the isolated CD51−bMSCs and CD51+bMSCs, especially the CD51+ cells, presented a favourable proliferative capacity and could differentiate into adipocytes, osteocytes and chondrocytes in vitro. After the cells were transplanted into the MI mice by intravenous injection, the therapeutic efficiency of CD51+bMSCs in improving left ventricular ejection fraction (LVEF) and left ventricular fractional shortening (LVFS) was better than that of CD51−bMSCs. Compared with CD51−bMSCs, CD51+bMSCs preferentially migrated to and were retained in the infarcted hearts at 48 h and 8 days after intravenous injection. Accordingly, the migratory capacity of CD51+bMSCs exceeded that of CD51−bMSCs in vitro, and the former cells expressed higher levels of chemokine receptors or ligands. Interestingly, the retained CD51+bMSCs retained in the myocardium possessed proliferative potential but only differentiated into endothelial cells, smooth muscle cells, fibroblasts or cardiomyocytes. Transplantation of CD51+bMSCs partially attenuated the inflammatory response in the hearts after MI, while the potential for inflammatory suppression was low in CD51−bMSC-treated mice. Conclusions These findings indicated that the CD51-distinguished subpopulation of bMSCs facilitated proliferation and migration both in vitro and in vivo, which provided a novel cell-based strategy to treat acute MI in mice by intravenous injection.
