232. Saikosaponin-A Exhibits Antipancreatic Cancer Activity by Targeting the EGFR/PI3K/Akt Pathway

Abstract

Background: A diagnosis of pancreatic cancer is pretty grim. Saikosaponin-A (SSA) is a Chinese herbal extract with anticancer activity. However, the therapeutic effect of SSA on pancreatic cancer remains elusive. Aim: The study aims to evaluate the antitumor effects of SSA on pancreatic cancer cells in vitro and in vivo. Methods: After treatment with SSA, cell viability was measured using the CCK-8 assay, DAPI staining was performed to analyze the effect on nuclear morphology, propidium iodide (PI) staining was used to detect the cell cycle, and Annexin V/PI double staining was conducted to analyze apoptosis. Then, the expression of apoptosis-related proteins and EGFR/PI3K/Akt pathway-related proteins was determined using western blotting. The binding of SSA to EGFR was analyzed by performing molecular docking. The mouse pancreatic cancer model was established by subcutaneously injecting pancreatic cancer cells, and after 30 days of SSA gavage, the tumor volume was calculated. Tumor tissue sections were subjected to Ki67 immunohistochemical staining and HE staining. Results: SSA inhibited the proliferation of pancreatic cancer cells. As the concentration of SSA increased, the proportions of BxPC-3 and MIA PaCa-2 cells in the G0/G1 phase increased, the proportions of early and late apoptotic cells also increased, and the apoptosis rate gradually increased. Apoptosis inhibitor experiments indicated that SSA promoted the activation of caspase 3 to induce apoptosis in pancreatic cancer cells. In addition, SSA treatment significantly reduced the levels of phosphorylated EGFR, Akt, and PI3K in the two cell lines. Molecular docking results showed that SSA may have potential binding sites in EGFR. Results of the xenograft experiment confirmed the antitumor effects of SSA, as evidenced by the decreased tumor weight and downregulated expression of Ki67. Conclusion: The results revealed that SSA exerted inhibitory effects on pancreatic cancer cells. These effects may be related to the inactivation of the EGFR/PI3K/Akt signalling pathway.

235. Evaluation of Chito-Oligosaccharide (COS) in Vitro and in Vivo: Permeability Characterization in Caco-2 Cells Monolayer and Pharmacokinetics Properties in Rats.

Abstract

Chito-oligosaccharide (COS) had shown a variety of biological activities and potential biomedical implications. The present study investigated the pharmacokinetics, bioavailability, and in vitro absorption of COS with degrees of polymerization (DPs) 2–7 and explored the influence of DPs on them. From Caco-2 cell permeation studies, COS were low permeability compounds with no directional effects, suggesting a low in vivo absorption mediated by facilitation diffusion and paracellular absorption. After an intragastrical administration to rats, COS2 showed the highest systemic exposure in six oligosaccharides. The bioavailability of COS2–7 was 7.33%, 6.11%, 4.67%, 4.13%, 4.02%, 0.99%, respectively. Differences in bioavailability for each COS correlated to structural variations, with high DPs contributing to a decrease in bioavailability. In conclusion, COS could be absorbed by the intestinal tract both in vitro and in vivo. The very low oral bioavailability of COS could be due to low permeability. DPs can affect absorption and bioavailability of COS2–7. This study provided evidence for the absorption characteristics of COS2–7 to help us better understanding the pharmacological actions. Abstract

• The flame-retardant nanofiber membrane was prepared by electrospinning. • The membrane and carbon fiber fabric alternating layered to prepare composite. • Nanofiber membrane interlayer charring improved composite's flame retardancy. • Nanofiber membrane interlayer reinforcement enhanced mechanical properties. A novel flame-retardant method based on the intercalation of nanofiber flame-retardant membrane has been studied for composite laminates. Compared to traditional additive flame-retardant method, this method is more efficient, does not affect the composite forming process, and can improve the mechanical properties of composite. Nanofiber flame-retardant membranes (FPm) are prepared from polyether sulfone (PES) and pentaerythritol phosphate ester (PEPA) by electrospinning technique and inserted in the interlayer of carbon fiber reinforced vinyl ester resin (CF/VER) composites. The FPm can improve the flame-retardancy and smoke suppression of the CF/VER composites by promoting the charring of interlayer structure and form a barrier layer. The thickness of the FPm and the flame-retardant performance of the composites are positive correlation. At a FPm's thickness of 50 μm, the limiting oxygen index (LOI) and UL-94 ratings of the CF/VER composites reached 33.5 % and V-1, respectively. The total heat release (THR) and total smoke release (TSP) are reduced by 26.4 % and 30.4 %, respectively. Due to the interlayer enhancement effect of FPm, the mechanical strength and interlaminar fracture toughness of CF/VER composites are also improved. Especially, when the thickness of FPm is 30 μm, the mode I interlaminar fracture toughness (G IC) of the CF/VER composite increased by 17.4 %. while the flexural strength, interlayer shear strength (ILSS), mode II interlaminar fracture toughness (G IIC) and storage modulus (E') are all enhanced.
