Glioblastoma (GBM) is a type of tumor that is highly lethal despite maximal therapy. Standard therapeutic approaches provide modest improvement in progression-free and overall survival, necessitating the investigation of novel therapies. Oncologic therapy has recently experienced a rapid evolution toward "targeted therapy", with drugs directed against specific targets which play essential roles in the proliferation, survival, and invasiveness of GBM cells, including numerous molecules involved in signal transduction pathways. Inhibitors of these molecules have already entered or are undergoing clinical trials. However, significant challenges in their development remain because several preclinical and clinical studies present conflicting results. In this article, we will provide an up-to-date review of the current targeted therapies in GBM.

Autophagy is the spontaneous degradation of intracellular proteins and organelles in response to nutrient deprivation. The phagocytosis of iron oxide nanoparticles (IONPs) results in intracellular degradation that can be exploited for use in cancer treatment. Non-invasive magnetic control has emerged as an important technology, with breakthroughs achieved in areas such as magneto-thermal therapy and drug delivery. This study aimed to regulate autophagy in mouse B-lymphoma cells (A20) through the incorporation of IONPs–quantum dots (QDs). We hypothesized that with the application of an external magnetic field after phagocytosis of IONPs–QDs, autophagy of intracellular IONPs–QDs could be regulated in a non-invasive manner and subsequently modulate the regulation of inflammatory responses. The potential of this approach as a cancer treatment method was explored. The application of IONPs and an external magnetic force enabled the non-invasive regulation of cell autophagy and modulation of the self-regulatory function of cells. The combination of non-invasive magnetic fields and nanotechnology could provide a new approach to cancer treatment.

A series of three-dimensional numerical simulations were performed to understand the thermal-solutal capillary-buoyancy flow of Ge1-xSix melts during Czochralski crystal growth with a rotating crystal or crucible. The crystal and crucible rotation Reynolds numbers in this work are 0∼3.5 × 103 (0∼4.4 rpm) and 0∼−2.4 × 103 (0∼−1.5 rpm), respectively. Simulation results show that if the thermal capillary Reynolds number is relatively low, the flow will be steady and axisymmetric, even though the crystal or crucible rotates at a constant rate. The critical thermal capillary Reynolds number for the initiation of the three-dimensional oscillatory flow is larger than that of pure fluids. As the crystal or crucible rotation rate increases, the critical thermal capillary Reynolds number first increases and then decreases. The dominant flow pattern after the flow destabilization is azimuthal traveling waves. Furthermore, a reversed evolution from the oscillatory spoke pattern to traveling waves appears in the melt. Once the crystal or crucible rotation rate is relatively large, the traveling waves respectively evolve to rotating waves at the crystal rotation and a spindle-like pattern at the crucible rotation. In addition, the maximum amplitude of solute concentration oscillation on the free surface initially decreases, but finally rises with the crystal or crucible rotation rate increasing.
