Objective To investigate whether the interferon-γ (IFN-γ) gene (CA)n repeat polymorphism is associated with susceptibility to endometriosis.Design Case-control study. Setting University Department of Obstetrics and Gynaecology. Population Women with (n = 622) and without (n = 442) endometriosis. Methods Genotyping was performed by fluorescent polymerasechain reaction (PCR) and gene-scan analysis. Main outcome measures Genotype distribution and allele frequency of the dinucleotide (CA)n repeat polymorphism in the IFN-γ gene. Results Seven alleles (12-18 repeats) of the IFN-γ gene (CA)n repeat polymorphism were found. In both patients with endometriosis and controls the most common allele was composed of 13 repeats, followed by an allele of 15 repeats, and then by an allele of 12 repeats. Patients with endometriosis had a significantly higher incidence of genotypes with alleles composed of fewer repeats (12-13 repeats), compared with the controls (92.0 versus 84.4%, respectively, P < 001). Conclusions Our results suggest that the (CA)n repeat polymorphism in the IFN-γ gene may be associated with a risk of endometriosis in the South Korean population. [ABSTRACT FROM AUTHOR]

Human peripheral blood lymphocytes (PBLs) electroporated with RNA encoding anti-Her-2/neu-specific chimeric immune receptor (CIR) have been reported to elicit potent immune responses against SKOV3 tumors in a nude mouse model. However, CIR-electroporated PBL (CIR-PBL) did not proliferate, and the cell number rapidly decreased in the absence of exogenous interleukin-2 (IL-2). In this study, PBLs electroporated with both CIR and IL-2 RNA (CIR/IL-2-PBL) were studied to determine whether antitumor effects could be improved by adoptive immunotherapy. CIR and IL-2 were expressed in CIR/IL-2-PBL at levels similar to PBLs electroporated, with IL-2 RNA (IL-2-PBL) or CIR-PBL. Transfer of IL-2 RNA induced proliferation and prolonged survival of PBLs in vitro. In a xenograft model, both IL-2-PBL and CIR/IL-2-PBL showed significantly higher antitumor effects than CIR-PBL. The number of tumor-infiltrating natural killer (NK) cells was significantly increased in IL-2-PBL and CIR/IL-2-PBL. After NK cell depletion, IL-2-PBL showed significantly lower antitumor effects than CIR/IL-2-PBL. These results suggest that transfer of IL-2 RNA to CIR-PBL can promote NK cell infiltration of tumors and prolong survival of infused PBLs in vivo. Hepatitis B virus (HBV) infections are endemic in Korea. The aims of this study were to determine the prevalence of HBsAg positivity in Korea and to evaluate the changes in intrafamilial transmission after introduction of HBV vaccination in 1983. This study was based on the 2001 Korea National Health and Nutrition Examination Survey. A total of 2512 study subjects, aged 10-29 years, were selected from across Korea using a stratified multi-stage probability sampling design. To identify the changes in intrafamilial transmission after the introduction of the HBV vaccination programme, 1850 subjects with parental serological markers were selected. These subjects were then grouped into two birth cohorts (cohort 1: born before 1983; cohort 2: born after 1983). Appropriate sampling weights were used for all analyses. The weighted age-specific prevalence of HBsAg was 4.9% in participants in their 20s and 1.9% in adolescents ; the combined weighted prevalence was 3.2%. Of subjects with HBsAg positivity in either parent, 17.5% were HBsAg-seropositive. Of subjects with two HBsAg-negative parents, 1.5% were HBsAg-seropositive. The HBsAg positivity rate of offspring with HBsAg-positive mothers was higher than those with HBsAg-positive fathers (27.3% vs. 4.8%, P<0.001). The weighted HBsAg positivity rate of offspring with HBsAg-negative mothers was 2.3% for cohort 1 and 0.4% for cohort 2 (P<0.01), and for those offspring with HBsAg-positive mothers it was also significantly decreased compared to cohorts 1 and 2 (40.2% vs. 16.4%, P<0.01). However, the weighted HBsAg positivity rate of offspring with HBsAg-positive mothers was still high. Objective:To investigate whether drugs targeting peripheral cannabinoid-1 (CB1) receptor ameliorate adiposity comparable to central CB1-receptor antagonist or not.Measurements:Receptor binding assay and functional assay in vitro. Pharmacokinetic parameters in mice, brain uptake clearance of compounds in rats and antagonism on the CB1-agonist-induced hypothermia in mice. Diet consumption, body weight changes, hepatic gene expression of sterol-regulatory element-binding protein-1 (SREBP-1) and plasma/tissue concentrations of compounds in HF diet-induced obese (HF-DIO) mice after acute and chronic treatment.Results:Compound-1, an SR141716A derivative, is a peripheral CB1-receptor-selective antagonist that is 10 times less potent than SR141716A in in vitro evaluations. Although the plasma concentrations of Compound-1 are five times higher than those of SR141716A, its potency is still 10 times lower than that of SR141716A in reducing the consumption of normal or HF diet by mice. Through evaluations of brain uptake and the effect on CB1-agonist-induced hypothermia, it was verified that the blood–brain barrier (BBB) penetration of Compound-1 is much lower than that of SR141716A. In HF-DIO mice, chronic treatment by Compound-1 showed dose-dependent antiobesity activities, while its brain distribution was very low as compared with that of SR141716A. Compound-1’s effective doses for antiobesity activity were just over 30 mg kg−1. However, Compound-1 completely suppressed the elevated hepatic SREBP-1 expression even at 10 mg kg−1.Conclusion:These results suggest that (1) central CB1 receptors mediate anorectic response of CB1-receptor antagonists and (2) peripheral modulations, including SREBP-1 expression, are not major mechanisms in the antiobesity effects of CB1-receptor antagonists. The article discusses a study that uses genotypic and phenotypic analyses to 12 syndromic craniosynostosis patients in Korea. Syndromic craniosynostosis is hereditary and linked with several gene mutations including FGFR1, FGFR2, FGFR3, in which the most diverse is FGFR2. It has shown that the most common skull deformity among the patients is brachycephaly and their facial morphology was very different. Moreover, 11 missinse mutations of FGFR1 and FGFR3 were also discovered in 12 patients.

The current gene transfer technology for single chain (scFv)-based chimeric immune receptor (CIR) has relied on retrovirus and lentivirus vectors which require a long time to obtain sufficient number of transduced cells and stably incorporate into genome. To ameliorate these limitations, we applied RNA electroporation to human peripheral blood lymphocytes (PBLs) activated with anti-CD3 antibody and interleukin-2 (IL-2) for 3 days and assessed that PBL transiently expressing anti-Her-2/neu CIR (CIR-PBL) containing signaling portion of CD28 and CD3ζ could elicit strong cytotoxicity in vitro and antitumor responses in vivo. The CIR-PBL expressed high level of CIR in CD4+, CD8+ and CD56+ cells. Her-2/neu-specific stimulation induced secretion of type-I cytokines including interferon-γ (IFN-γ), IL-8 and granulocyte-macrophage colony-stimulating factor, and IFN-γ secretion was mainly mediated by CD8+ T cells. CIR-PBL specifically killed SKOV3 cell line expressing Her-2/neu. Adoptive transfer of CIR-PBL in SKOV3 xenograft model led to significant inhibition of tumor growth compared with transfer of mock-transduced PBL and showed higher inhibition than those with Herceptin, humanized monoclonal antibody specific for Her-2/neu. Tetragonal lead zirconate titanate (PZT) films with different orientations and 200 nm film thicknesses were prepared on platinized silicon substrates. Types of substrate and control of thermal processes, such as layer-by-layer and one-crystallization heat treatments, result in highly (111) or (100)-oriented PZT films. The piezoelectric, dielectric, and ferroelectric properties of polycrystalline PZT films have been investigated as a function of preferred orientation. The property difference between (111) and (100)-oriented films appears to be induced by the effect of ferroelastic domain existence (90° domain in tetragonal composition). From a modified phenomenological equation, the higher electrostrictive coefficient value of 5.6 × 10−2 m4/C2 for (100)-oriented PZT may be responsible for the larger piezoelectric coefficients in (100)-oriented polycrystalline PZT films of 44 pm/V in comparison to (111)-oriented PZT films with about 3.1 × 10−2 m4/C2 of Q33 and 40 pm/V of d33, f. It was also observed that two (100)-oriented films prepared by different heat treatments showed different values in piezoelectric, dielectric, and ferroelectric properties even though only (100) orientation was characterized for both cases. This process-induced difference may also play an important role in determining both intrinsic and extrinsic contribution to the properties, even though these parameters seem to be more responsible for extrinsic components, such as domain wall motion. To find differentially expressed protein spots using two-dimensional electrophoresis proteomic analysis, we took blood serum samples from 40 purebred Yorkshire pigs at 12, 18, 24, and 30 weeks. Each growth stage contained 10 male pigs having half-sib pedigrees. With the pooled serum samples, two interesting spots, differentially expressed in the growth stages, were identified using MALDI-TOF-TOF MS/MS analysis as haptoglobin alpha 1S (Hp) and apolipoprotein A-IV (APOA4) gene products. The Hp was down-regulated from 12 to 30 weeks, and APOA4 was not expressed much before 18 weeks but was highly expressed in the late growth stages. There may be an inverse relationship between the Hp and APOA4 genes. Four segments for the Hp and APOA4 genes were successfully amplified with sizes around 500 bp. The porcine Hp and APOA4 genes were screened in the 40 purebred Yorkshire pigs and a random cross population (90 pigs), resulting in the location of 6 single nucleotide polymorphisms (SNPs) in the coding regions. The mutations resulted in amino acid changes in segments of Hp627, Hp742, and APOA41203. Further investigation of the function of the Hp and APOA4 genes with SNPs will be necessary to understand fully the different expression profiles and association studies. [ABSTRACT FROM AUTHOR]

Abstract: Mesoporous carbon (MC) with high surface area and large pore volume was synthesized using mesophase pitch as a carbon precursor and nanosized MgO as an additive. The maximum surface area, largest pore volume and highest mesoporous ratio of as-prepared MC were up to 1400m2/g, 2.8cm3/g and 89%, respectively. The mesoporous structures (3–40nm) of MC were directly observed under SEM and TEM. The adsorption capacity and adsorption rate of MC to vitamin B12 (VB), chicken egg white albumin (CEWA) and bovine serum albumin (BSA) were proportional to the mesopore volume and average pore size. MC (PM4-OC) exhibited the maximum adsorption capacity to the typical biomolecules, 486, 140 and 176mg/g for VB, CEWA and BSA, respectively. In contrast, Maxsorbs (commercial activated carbons) with a surprising surface area gave a very low adsorption to such biomolecules. The research indicates that MC may be potential in the selective adsorption and separation of biomolecules, based on a molecule sieve effect. [Copyright &y& Elsevier]

The characterization and quantification of anthocyanins in grape cultivars of Oll-Meoru ( Vitis coignetiae× Vitis labrusca), Neut-Meoru ( Vitis coignetiae× Vitis labrusca), Muscal Bailey A. ( Vitis labruscana), and Campbell Early ( Vitis labrusca× V. vinifera) cultivated in Korea were carried out by partial purification through XAD-7 column chromatography followed by C-18 HPLC/diode array detector (DAD), HPLC/MS, and HPLC/MS/MS analyses. The column oven temperature during the reverse phase C-18 HPLC greatly affected the separation of individual anthocyanins. The result showed that the optimum column oven temperature was 35 °C. Sixteen different anthocyanins (11 nonacylated and 5 acylated anthocyanins) were identified in the grape juices. Oll-Meoru, Neut-Meoru, and Muscat Bailey A (MBA) grape juices contained only nonacylated anthocyanins. Oll-Meoru and Neut-Meoru grape juices had same anthocyanins, but their proportions were considerably different. Peonidin 3,5-diglucoside and malvidin 3,5-diglucoside were the major anthocyanins in Oll-Meoru grape juice. Delphinidin 3-glucoside was, however, the major anthocyanin in Neut-Meoru grape juice. Peonidin 3-glucoside and malvidin 3-glucoside were the most abundant anthocyanins in Muscal Bailey A grape juice. Campbell Early grape juice contained both nonacylated and acylated anthocyanins. Cyanidin 3-( p-coumaroyl)glucoside-5-glucoside and peonidin 3-( p-coumaroyl)glucoside-5-glucoside were the most abundant anthocyanins in Campbell Early grape juice. Total anthocyanin contents were greatly different in different grape jucies, with the highest in Neut-Meoru juice (1043.5 μg/mL), followed by Oll-Meoru (997.7 μg/mL), MBA (390.2 μg/mL), and Campbell Early (183.9 μg/mL) juices. The total anthocyanin content in Neut-Meoru grape juice was 5.67 times higher than that in Campbell Early grape juice. To investigate the genetic characteristics of the Korean porcine reproductive and respiratory syndrome virus (PRRSV), we determined the complete sequence of the nucleocapsid protein gene (ORF7) from 105 PRRSV isolates from all nine Korean prefectures during the years 2003 through 2006. These sequences were then analyzed along with the published ORF7 sequences for two Korean PRRS viruses (PL97-1/1997 and LMY/2002) and 36 non-Korean viruses. The ORF7 nucleotide sequence identities among the 107 Korean PRRS viruses ranged from 86.2 to 100%, corresponding to 85.4 to 100% identity at the amino acid level. All of the Korean isolates examined belonged to the North American genotype. The ORF7 gene sequence from the North American prototype virus (VR-2332) and its derived vaccine virus (Ingelvac PRRS MLV) was 90.0–100% identical to the various ORF7 sequences of the Korean isolates, with corresponding amino acid identities from 91.0 to 100%. In the phylogenetic tree obtained by neighbor-joining analysis, all of the Korean PRRSVs were divided into four groups. Our ORF7 sequence data also revealed no correlations between the date or place of collection and the distribution of PRRSV in Korea. North American genotype PRRSVs may have been introduced into Korean swine herds some time ago; these viruses apparently radiated nationwide within a relatively short period of time. Within the North American genotype PRRSVs from around the world, the Korean PRRSVs did not emerge as a single independent clade overall, and their immediate relationships with the PRRSVs from other countries could not be determined. Objective: To investigate the possible association between polymorphisms [the -2510A/G promoter polymorphism (rs1024611) and the Cys35Cys coding polymorphism (rs4586) in exon 2] of the chemokine (C-C motif) ligand 2 (CCL2) gene and knee osteoarthritis (OA) in a Korean population. Methods: DNA was obtained from 153 Korean primary knee OA patients and 270 healthy controls. CCL2 genomic variants (-2510A/G and Cys35Cys polymorphisms) were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). In additional, the effect of -2510A/G on CCL2 transcription was examined, using a luciferase reporter gene construct transfected into HMC-1 cells. Results: The -2510A/G promoter polymorphism was associated with OA [genotype frequency, p = 0.041; allele frequency, p = 0.017, odds ratio (OR) = 1.45, 95% confidence interval (CI) = 1.07-1.96]. Significant association was observed between the G carrier of the -2510A/G promoter polymorphism and primary knee OA patients (p = 0.021, OR = 2.25, 95% CI = 1.12-4.52). The G carrier of the -2510A/G promoter polymorphism was also associated with both clinically subtyped OA patients (OA patients with functionally poor index and radiographically severe OA patients). However, no significant difference was found in the Cys35Cys polymorphism. Haplotype frequency analysis revealed a significant difference (χ2 = 8.98, p = 0.030). The CCL2 serum level of subjects with the G carrier (290.0±87.5 pg/mL) of the -2510A/G promoter polymorphism was statistically higher than that of subjects with the non-G carrier (161.5±48.3 pg/mL). The luciferase activity was significantly greater from interleukin (IL)-1β-induced cells transfected with constructs containing G at position -2510. Conclusions: The G carrier of the -2510A/G promoter polymorphism was found to be associated with primary knee OA, and could be a susceptibility factor in the development of primary knee OA in the Korean population.
